  • This study investigated the structural characteristics of Taxus cuspidata communities on Mt. Hambaek. The vegetation consisted of 9 species in the T$_1$(tree) layer, 35 species in the T$_2$(subtall tree) layer, 28 species in the S(shrub) layer, and 69 species in the H(herb) layer. The dominant species of the T$_1$ layer was Taxus cuspidata and that of the T$_2$ layer was Acer tschonoskii var, rubripes. The dominant species of the S layer was Tripterygium regelii, and the dominant species of the H layer was Pseudostellaria palibiniana. The diameter distribution of the dominant five species in the T$_1$ and T$_2$ layers indicates that these Taxus cuspidata forests may be gradually dominated by Quercus mangolica and by Tripterygium regelii. The composition of biological type was Ph-D$_1$-R$_{5}$-e.

  • In this study, the characteristics and mechanism of microwave-assisted drying were investigated to improve the efficiency of the storage and extraction of biomass through the removal of moisture from plant cell Taxus chinensis. The efficiency of microwave-assisted drying increased with increasing microwave power. When the experimental data were fitted to typical drying kinetic models, the page and modified Page models were the most appropriate. The microwave-assisted drying was determined to be a spontaneous endothermic process, and randomness increased during the drying process. The effective diffusion coefficient (3.445 × 10-9~7.163 × 10-7 ㎡/s) and mass transfer coefficient (3.1529 × 10-5~1.2895 × 10-2 m/s) increased with increasing microwave power. The small Biot number (0.3890~0.7198) indicated that the mass transfer process was externally controlled.

  • The suspension cells of Taxus cuspidata were transformed with Agrobacterium tumefaciens harboring binary vector pCAMBIE1302 encoding mgfp. Transient transfection efficiency was compared by using the fluoremetric measurement. The transient transfection efficiency was improved by transformation with DMSO and/or sonication treatment. Optimum conditions for DMSO and sonication treatment were 3% and 30sec, respectively. selection and maintenance of transformed cells were continued for 3 months. An insertion of the mgfp gene in transformed cells was detected by PCR and an expression of GFP confirmed by the western blot analysis.

  • Allelopathy is an ecological phenomenon that refers to the beneficial or harmful effects of one plant on another plant, both crop and weed species, by the release of organic chemicals (allelochemicals) from plant parts by leaching, root exudation, volatilization, residue decomposition in soil and other processes in both natural and agricultural systems. Allelopathy can affect many aspects of plant ecology including occurrence, growth, plant succession, the structure of plant communities, survival, dominance, diversity, and plant productivity. In this review, we describe the concept of allelopathy, some mechanisms of operation within plants and then focus on a select number of gymnospermous tree genera: Ephedra, Pinus, Taxus, Cedrus, Juniperus, Picea, Cunninghamia and Araucaria. Pinus, Taxus (yew) and Cedrus (cedar) trees have a strong negative allelopathic effect on the germination, growth, or development of other plant species in the forest community.

  • Influence of fructose addition to the cultivation medium on the production of taxanes and the growth of callus culture of Taxus baccata was studied. The cultures showed an ability to adjust to the substitution of some of the sucrose in the media by fructose and the fresh biomass accumulation was higher on the media containing different concentrations of fructose during the second cultivation period.

  • We have isolated endophytic fungi from the Indian yew tree, Taxus baccata, and then screened for taxol production. Out of the 40 fungal cultures screened, one fungus Gliocladium sp. was found to produce taxol and 10-DABIII (10-deacetyl baccatin III). These compounds were purified by TLC and HPLC and characterized using UV-spectroscopy, ESI-MS, MS/MS, and proton NMR. One liter of Gliocladium sp. culture yielded $10\;{\mu}g$ of taxol and $65\;{\mu}g$ of 10-DABIII. The purified taxol from the fungus showed cytotoxicity towards cancer lines HL-60 (leukemia), A431 (epidermal carcinoma), and MCF-7 (breast cancer).

  • Undifferentiated callus were induced from the young leaves of Taxus cuspidata by treatment with various combinations of plant growth hormones. The effects of light and culturing temperature on production of baccatin III, the precursor of taxol, were studied. The contents of baccatin III in the cultured callus were analysed by HPLC. The illumination of fluorescence and administration of isoleucine, one of the possible substrates in biosynthesis of terpenoids, to the culturing media increased the production of baccetin III.

  • Endophytic fungi from Ginkgo biloba and Taxus cuspidata in Korea were screened for production of taxol. Eighteen and twelve fungal isolates from G. biloba and T. cuspidata, respectively, were shown to produce immunologically detectable amount of taxol. The highest production of taxol at 260 ng/l was achieved by stationary culture of an Alternaria isolate from G. biloba. The strain also produced unidentified antifungal agent(s) against Pythium ultimum. However, the activity gradually decreased when the strain was stored at $4^{\circ}C$ for 6 months.
