Interspecific Hybrids from Wild $\times$ Cultivated Triticum Crosses - A Study on the Cytological Behaviour and Molecular Relations -

  • Genetic diversity of cultivated wheat is narrowing down and is increasingly becoming non-complacent in tackling new pathogenic races and adverse environmental situations. Wild relatives of wheat are rich repositories of beneficial genes that are capable of defying adverse situations. However, these wild species are not readily crossable with cultivated ones. The present study attempted to cross three wild wheat species as females with three cultivated species of varying ploidy to understand the intricate behaviour of hybrids in relation to cytology, morphology, and molecular recombination. Post-fertilization barriers caused hybrid recovery in wild species in contrast to cultivated species. Triticum monococcum did not produce hybrids in any of the crosses. Various degrees of chromosome anomalies and hybrid sterility were seen with hybrids of T. timopheevi and T. sphaerococcum. Cytoplasmic factors were suspected to add more to the abnormality. G genome from T. timopheevi could enhance more pairing between Band D of cultivated species. Precocity of certain chromosomes in laggard formation was evident, pointing towards evolutionary self balance of the genomes which prevented homeologous pairing. They are eliminated in hybrids. Molecular diversity clearly corroborated with genetic proximity of the species, which distinguished themselves by maintaining the genome homeology.

Comparative Study of Two Congestion Management Methods for the Restructured Power Systems

  • Congestion management is one of the most challenging tasks of a system operator to ensure the operation of transmission system within operating limits. In this paper, cluster/zone method and relative electrical distance (RED) method for congestion management are compared based on the considered parameters. In the cluster/zone method, rescheduling of generation is based on user impact on congestion through the use of transmission congestion distribution factors. In the RED method, the desired proportions of generations for the desired overload relieving are obtained. Even after generation rescheduling, if congestion exists, load curtailment option is also introduced. Rescheduling cost, system cost, losses, and voltage stability parameter are also calculated and compared for the above two methods of congestion management. The results are illustrated on sample 6-bus, IEEE 30-bus, and Indian utility 69-bus systems.

Library Professionals' Perception on the ICT Applications in Engineering College Libraries: A study on Tamil Nadu, India

  • This study discusses the library professionals' perception of the ICT Applications in engineering college libraries in Tamil Nadu. The relevant data was collected from the library professionals in the self financing engineering colleges situated in Kanchipuram and Thiruvallur district of Tamil Nadu. 625 questionnaires were distributed, 504 replied with a response rate of 80.64%. It was found that the respondents with experience 'Below 5 years'gave 'Lack of infrastructure' as the first priority. 'Lack of interest on the part of users' and 'No support from administration in training library professionals' were the second and third preferences indicated by the respondents. The least preferences were given for 'Fear of ICT application'. Similarly, respondents with experience '6-10 years' indicated 'No support from administration in training library professionals' as the first priority. The least preference was given for 'Inadequate training in ICT applications' by the above respondents. It can be inferred that the professionals accepted and need the training in ICT applications.

Generation of single stranded DNA with selective affinity to bovine spermatozoa

  • Objective: This study was conducted to generate single stranded DNA oligonucleotides with selective affinity to bovine spermatozoa, assess its binding potential and explore its potential utility in trapping spermatozoa from suspensions. Methods: A combinatorial library of 94 mer long oligonucleotide was used for systematic evolution of ligands by exponential enrichment (SELEX) with bovine spermatozoa. The amplicons from sixth and seventh rounds of SELEX were sequenced, and the reads were clustered employing cluster database at high identity with tolerance (CD-HIT) and FASTAptamer. The enriched nucleotides were predicted for secondary structures by Mfold, motifs by Multiple Em for Motif Elicitation and 5' labelled with biotin/6-FAM to determine the binding potential and binding pattern. Results: We generated 14.1 and 17.7 million reads from sixth and seventh rounds of SELEX respectively to bovine spermatozoa. The CD-HIT clustered 78,098 and 21,196 reads in the top ten clusters and FASTAptamer identified 2,195 and 4,405 unique sequences in the top three clusters from the sixth and seventh rounds, respectively. The identified oligonucleotides formed secondary structures with delta G values between -1.17 to -26.18 kcal/mol indicating varied stability. Confocal imaging with the oligonucleotides from the seventh round revealed different patterns of binding to bovine spermatozoa (fluorescence of the whole head, spot of fluorescence in head and mid- piece and tail). Use of a 5'-biotin tagged oligonucleotide from the sixth round at 100 pmol with 4×106 spermatozoa could trap almost 80% from the suspension. Conclusion: The binding patterns and ability of the identified oligonucleotides confirms successful optimization of the SELEX process and generation of aptamers to bovine spermatozoa. These oligonucleotides provide a quick approach for selective capture of spermatozoa from complex samples. Future SELEX rounds with X- or Y- enriched sperm suspension will be used to generate oligonucleotides that bind to spermatozoa of a specific sex type.

Molecular Profiling of Rhizosphere Bacterial Communities Associated with Prosopis juliflora and Parthenium hysterophorus

  • Prosopis juliflora and Parthenium hysterophorus are the two arid, exotic weeds of India that are characterized by distinct, profuse growth even in nutritionally poor soils and environmentally stressed conditions. Owing to the exceptional growth nature of these two plants, they are believed to harbor some novel bacterial communities with wide adaptability in their rhizosphere. Hence, in the present study, the bacterial communities associated with the rhizosphere of Prosopis and Parthenium were characterized by clonal 16S rRNA gene sequence analysis. The culturable microbial counts in the rhizosphere of these two plants were higher than bulk soils, possibly influenced by the root exudates of these two plants. The phylogenetic analysis of V1_V2 domains of the 16S rRNA gene indicated a wider range of bacterial communities present in the rhizosphere of these two plants than in bulk soils and the predominant genera included Acidobacteria, Gammaproteobacteria, and Bacteriodetes in the rhizosphere of Prosopis, and Acidobacteria, Betaproteobacteria, and Nitrospirae in the Parthenium rhizosphere. The diversity of bacterial communities was more pronounced in the Parthenium rhizosphere than in the Prosopis rhizosphere. This culture-independent bacterial analysis offered extensive possibilities of unraveling novel microbes in the rhizospheres of Prosopis and Parthenium with genes for diverse functions, which could be exploited for nutrient transformation and stress tolerance in cultivated crops.

The Perception of Women Consumers on Buying Behaviour with Reference to Vellore District of Tamil Nadu, India

  • This paper portrays the perception of buying behaviour, factors of product preferences and product switch-over among women consumers in Vellore district of Tamil Nadu. To conduct this study, 300 respondents from 20 village blocks of Vellore district were selected using stratified random sampling as the technique and descriptive sampling as the design. The study came out with interesting results and conclusions. It was found that more than half of the respondents were purchasing the product without a bill. It was also found that the respondents who repaired their product in the authorized service centre during warranty period did not continue the same after the warranty period and preferred nearby repair shops. It was further found that more than half of the respondents were checking expiry date and quality standards while purchasing a product. The study's scope was limited to women consumers at Vellore district. It could be used for further research in other aspects such as gender comparison in Vellore district and comparison with other districts of Tamil Nadu and other states of India. It was suggested that there is still a necessity to upgrade the standard of the people in many aspects like education, occupation and level of income, which shall help them to compete with the neighboring cities on living standards.

  • 지구상에서 가장 중요한 자원 중 하나인 토양은 지형조건에 따라 서로 다른 다양한 형태를 가지고 있기 때문에 최적화되고 지속 가능한 토양 자원의 활용을 위해서는 정확하고 포괄적인 정보가 필요하게 된다. 그러나 연구대상지역인 인도 Tamil Nadu지역의 경우 지형적인 영향으로 토양에 대한 정보가 많이 누락되어 있었다. 따라서 본 연구에서는Tamil Nadu 지역 Eastern Ghat의 Kolli Hill에 대한 지형 측량과 원격탐측을 통한 토양조사와 지도제작이 이루어졌으며 토양 샘플의 물리화학적 특성은 미국 농무부 (USDA) 기준에 따라 분석이 이루어졌다. 연구 결과로 토양을 5개의 대분류와 10개의 부분류로 구분할 수 있었으며 토양의 분포 특성을 보면 Entisol, Inseptisol 그리고 Alfisol의 세 계층 중 Entisol의 경우 전체 지역에 대하여 75%의 분포를 보였으며 5개의 대분류에 대해 Ustorthent 가 73% 로서 대부분 지역에 나타나고 있다. 또한 Lithic Ustorthents(40%), Typic Ustorthents(26%)의 분포를 나타내었다. 앞으로도 대상지역에 대한 토양자원에 대한 지속적인 연구가 요구되며 이를 통하여 토양에 대한 많은 정보를 활용할 수 있을 것이다.

Perception on Plagiarism among Faculty Members and Research Scholars in Higher Educational Institutions of Dindigul District: A Study

  • These papers surveyed the respondents' Awareness and perception of Plagiarism among the Faculty members and research scholars of higher educational institutions in Dindigul District, Tamil Nadu. The 500 questionnaires were distributed among the participants, 456 questionnaires were filled and returned for usable by the respondents and remaining not replied. The responses rate is 91.2%. Further among 222 Male re- spondents, 11.18% of them are having Aware, 10.75% of them in Learning, 10.53% are with Fair and 76(16.23%) of them are Expert. Similarly out of 234 Female respondents, 16.45% of them has No Idea, 7.02% of them have Aware, 10.09% of them in Learning, 8.99% are with Fair and 8.77% of them are Expert.

A Structural Compendium on Perceptual Displays of Rural India towards the Role and Impact of ICT

  • The core purpose of the paper is to explore perceptual displays of rural India towards the role and impact of ICT in transforming the lives through education and telecommunications. After meticulous review of pertinent literature on ICT, and its policy framework, the researcher administers both quantitative and qualitative field data collected from the villages of Andhra Pradesh, Karnataka, Kerala, and Tamil Nadu with a sample of 250 from each state. More than 90 percent of respondents from the four southern states in India clearly thought that the role and impact of ICT is very momentous. Predominantly, cent percent respondents from Kerala evinced that ICT is the major reason for achieving the tag of "100% Literacy State". And also in the remaining three states viz., Andhra Pradesh, Karnataka, and Tamil Nadu, the literacy rate has crossed mind-boggling figures only because of ICT's reinforcement. Even in the arena of telecommunications, these four states average telecom density is more than the country's average. The paper concentrated only on how ICT function empirically to build human capacity in rural south-India through the sectors of only education and telecom. This study is first of its kind and precious to all stakeholders of the ICT sector like educational institutional, telecom firms, customers, employees, and governments, etc.

Thymic Hassall's corpuscles in Nandanam chicken - light and electronmicroscopic perspective (Gallus domesticus)

  • The present study was aimed to study the light and electron microscopic studies of thymic Hassall's corpuscles was done in various age groups of Nandanam Chicken ranging from day-old to forty weeks. Hassall's corpuscles are special, unique structures present in thymic medulla and also in the cortex of all the age groups of Nandanam chicken (from hatch to forty weeks) in the present study. Size of the Hassall's corpuscles in the medulla is larger than the ones present in the cortical region of thymus. The Hassall's corpuscles are made up of structureless eosinophilic mass surrounded by concentrically arranged reticuloepithelial cells. Under electron microscope, the Hassall's corpuscles were composed of reticuloepithelial cells interconnected by many desmosomes. The epithelial cells had abundance of cytoplasmic fibrils and desmosomes with few mitochondria and ribosomes. The nucleus was oval or round which was slightly indented. The centre of the Hassall's corpuscles was appeared either solid or cystic. The cystic corpuscles had cell debris within the cyst lumen.
