Effects of Mite Antigen and Toxic Shock Syndrome Toxin-1 on the Biological Actvity of Human Fibroblast (Toxic Shock Syndrome Toxin-1 및 Mite 항원이 사람섬유아세포의 생물활성에 미치는 효과)

  • The production of interleukin-1(IL-1)and nitric oxide(NO) by cultured fibroblast cells of human nasal turbinate was revealed by biological assay respectively. The cells were incubated for various periods of time in the presence of staphyloccocal toxic shock syndrome toxin-1(TSST-1) and house dust mite(Dermatophagoides farinae, HDM), and the culture supernatants were harvested. There was a little difference in the activities of IL-1beta and the amount of NO produced by the cells when stimulated with 0.002-0.1$\mu$g/ml of TSSTO-1 and 0.02-1$\mu$g/ml of HDM. The shapes of the time course curves for the production of IL-1beta and NO by the cells were different. Groups stimulated with TSST-1 or HDM produced more IL-beta in 2 h than no exposure group(Control). A certain mixed group(TSST-1, 10ng+mite, 100 ng) continued to produce IL-1beta highly throughout the entire incubation period. The cells stimulated with TSST-1 or HDM produced more NO in 2 h and 6 h than that produced in the end of incubation(48 h). Also, the mixed groups were generally similar. There results suggest that induction of IL-1beta by a certain mixed condition(TSST-1+mite) in fibroblast cell in vivo may play a role in inflammation.

PCR Detection of Virulence Genes Encoding Coagulase and Other Toxins among Clinical Methicillin-Resistant Staphylococcus aureus Isolates (Methicillin 내성 S. aureus 임상분리균주의 Coagulase와 주요 독소 유전자의 PCR 검출)

  • To characterize the genotypic traits of clinical methicillin-resistant Staphyiococus aureus (MRSA) isolates (n=49), major virulence-associated genes were detected by using PCR-based methods. All the MRSA isolates possessed coagulase gene and showed four polymorphism types [500bp ($6{\%}$),580bp ($27{\%}$), 660bp ($65{\%}$) and 740bp ($2{\%}$)] due to variable numbers of tandem repeats present within the gene. The four or five different loci of hemolysin gene family were dominant in the MRSA isolates,25 of which($51{\%}$) possessed a combination of hla / hlb / hld/ hlg / hlg-2 genes as the most prevalent type. The prevalence of enterotoxin genes was varied among the MRSA isolates. sea and seb genes were detected from all the MRSA isolates. But sei, tsst-1, seg, sec, and seh genes were detected from 31 ($63{\%}$), 16 ($33{\%}$), 14 ($29{\%}$), 8 ($16{\%}$), and 5 ($10{\%}$) isolates, respectively. sed and sej genes were detected from 1 ($2{\%}$) isolate, respectively. see, eta, and etb genes were not detected at all. sea / seb genes were co-detected from 11 ($23{\%}$) isolates, sea / seb / sei genes from 9 ($19{\%}$) isolates, and sea / seb / seg / sei / tsst-1 genes from 5 ($10{\%}$) isolates. Other genes were co-detected with below $10{\%}$ frequencies.

  • Most people suffer from stress during day life because modernized society is very complex and changes fast. Because stress can affect to many kind of physiological phenomena it is even considered as a disease. Therefore, it should be detected earlier, then must be released. When a person is being stressed several bio-signals such as heart rate, etc. are changed. So, those can be detected using medical electronics techniques. In this paper, stress assessment system is studied using random forest algorithm based on heart rate, RR interval and Galvanic skin response. The random forest model was trained and tested using the data set obtained from the bio-signals. It is found that the stress assessment procedure developed in this paper is very useful.

Toxins and Antibiotic Resistance of Methicillin-Resistant Staphylococcus aureus Isolated from Clinical Specimens (임상검체로부터 분리된 methicillin 내성 Staphylococcus aureus의 독소 및 항생제 내성)

  • Seventy five methicillin- resistant Staphylococcus aureus (MRSA) strains and 24 methicillin- susceptible S. aureus (MSSA) were isolated from clinical specimens obtained from a hospital in Suncheon, Jeonnam province, Korea, from July to December, 2009. Antibiotic resistance was determined using the disc diffusion method. Genes encoding enterotoxin (SE), toxic shock syndrome toxin-1 (TSST-1), exfoliative toxin (ET) and Panton-Valentine leukocidin (PVL) were detected by multiplex PCR-mediated amplification using specific primers. Sixty (80%) MRSA isolates possessed either one or more toxin genes and the most common pattern that coexisted in MRSA was seb, sec, seg, sei and tst (22.7%) followed by coexistence of sec, seg, sei and tst genes (18.7%). Gene pvl encoding leukocidin was not found. Significant correlation between the production of sec, seg, sei and tst genes was found. MRSAs were resistant to erythromycin (89% of the isolates), gentamicin (70.7%), ciprofloxacin (69.3%), clindamycin (61.3%) and tetracycline (58.7%), while MSSAs were susceptible to the antibiotics with the exception of erythromycin. Toxin genes seb, sec and tst were related to the tetracycline resistance of MRSA.

  • To provide microbial information for the safety of agricultural production, the presence of enterotoxin genes and antibiotic susceptibility of 14 isolated Staphylococcus aureus (11.7%) strains were investigated using PCR-based methods and disk diffusion method, respectively. Among enterotoxin-encoding genes, sea was detected from two isolates (14.3%), sea and sed genes were co-detected from three isolates (21.4%), and sea, sed, and see genes in seven isolates (50.0%), whereas seb, sec, and tsst were not detected in any isolate. Nine (64.3%), eight (57.1%), six (42.9%), two (14.3%), and one (7.2%) isolates were resistant to penicillin, novobiocin, amphicillin, erythromycin and oxacillin, and doxycycline and kanamycin, respectively. Methicilline-resistant S. aureus was found in roller of B farm and in hydroponic solution of D farm.

Detection and Identification of $\beta$-lactamase, Enterotoxin and Other Exotoxins Genes of Staphylococcus aureus by PCR

  • Staphylococcus aureus is a major pathogen for cattle, causing various forms of subclinical and clinical mastitis and could be a causative agent of food poisoning, it produces various superantigenic exotoxins which have a great public health significance. A total of 72 S. aureus clinical isolates from dairy farms located in Kyunggi Province Korea were examined for the species identification by biochemical method, and for the detection of $\beta$-lactamase, enterotoxin and other exotoxins genes by PCR. The results of species identification by biochemical method agreed with those of PCR done with species specific primer STA-AU. $\beta$-lactamase is an enzyme closely associated with the resistance to antibiotic penicillin, which is an important means of treatment of mastitis, all the isolates were positive for the presence of genes encoding $\beta$-lactamase, which were reproduced in penicillin susceptibility disc assay. Six types of toxin genes, Staphylococcal enterotoxin (SE)A, SEB, SEC, SEE, toxic shock syndrome toxin (TSST-1) and exfoliative toxin A (ET A) were detected in 72 isolates by PCR associated genotypic method in this study, none of the isolates carried the genes for enterotoxin D (SED) and exfoliative toxin B (ETB). The occurrence rate of exotoxin genes rated as 12.5%, and the precision of the PCR identification results has been confirmed using the reference strains.

Associated-Genes and Virulence Factors of Staphylococcus aureus Isolated from Nasal Cavity of Neonates (신생아 비강에서 분리된 황색포도구균의 병원성 인자와 관련 유전자)

  • Purpose : Nosocomial infection with Staphylococcus aureus, especially methicillin resistant S. aureus, has become a serious concern in the neonatal intensive care unit. The aim of this study is to investigate the virulence factors, and the relationship between the antibiotic resistance and the associated genes of Staphylococcus aureus isolated from nasal cavity of neonates. Methods : Fifty one isolates of S. aureus were obtained from nasal swab taken in 28 neonates in the NICU and nursery of Pusan National University Hospital between February and May, 2001. They were tested in regard to antibiotic susceptibility, coagulase test and typing, plasmid DNA profile, as well as reactivity to enterotoxin A-E(sea, seb, sec, sed, see) genes and toxic shock syndrome toxin-1(tst) gene by polymerase chain reaction(PCR). Associated genes such as mecA, mecR1, mecI, and femA were also determined by PCR. The origin of MRSA strains was assessed using DNA fingerprinting by arbitrarily-primed polymerase chain reaction(AP-PCR). Results : Twenty three(45.1%) and six(11.8%) isolates were resistant to oxacillin and vancomycin respectively. Multidrug resistance to three or more of the antibiotics tested was observed in 51.0% of the isolates. Forty two isolates were coagulase positive and twenty two isolates had mecA gene. Sixteen isolates had both mecA and femA genes and had type I-III plasmids. 64.7% of isolates carried sec gene, and 80.4% carried tst gene. DNA fingerprinting by AP-PCR for 12 MRSA strains showed 10 distinct patterns, suggesting different origins. Conclusion : We confirmed that the prevalence of nasal carriage of S. aureus and the incidence of antimicrobial-resistant S. aureus, especially vancomycin resistance, is very high in neonates who were admitted in NICU and nursery. It is possible that these pathogens are responsible for serious nosocomial infections in neonates. The need for improved surveillance and continuous control of pathogens is emphasized.

All isolates possessed coagulase gene and showed five polymorphism types [500 bp (2.4%), 580 bp (17.4%), 660 bp (61.6%), 740 bp (17.4%), and 820 bp (1.2%)] due to variable numbers of tandem repeats present within the gene. Two or three different loci of hemolysin gene family were dominant in isolates, 47 of which (55%) possessed combination of hla/hld/hlg-2 genes as the most prevalent types. Among enterotoxin-encoding genes, sea was detected from 32 isolates (37%), sed from 1 isolate (1%), and sea and sed genes were co-detected from 4 isolates (5%), whereas seb, sec, and tsst-1 genes were not detected. All isolates were susceptible to ciprofloxacin, trimethoprim/sulfamethoxazole, oxacillin, and vancomycin, 85 isolates (99%) to penicillin G, 54 isolates (63%) to chloramphenicol, 51 isolates (59%) to erythromycin, and 7 isolates (8%) to clindamycin. Among resistant isolates, seven displayed multiantibiotic-resistance against two different antibiotics.
