Cottonseed Extract Improves the Function of Osteoblastic MC3T3-E1 Cells

  • We have investigated the effects of cottonseed extract on the proliferation, differentiation and lipopolysaccharide (LPS)-induced production of local factors in murine clonal osteoblastic MC3T3-E1 cells. Ethanol extract of cotton seed ($4{\sim}63{\mu}g/mL$) significantly increased the proliferatin of MC3T3-E1 cells (p<0.05). Moreover, cottonseed extract ($10{\sim}50{\mu}g/mL$) caused a significant elevation of alkaline phosphatase (ALP) activity and collagen content in the cells. Lipopolysaccharide (LPS) is a potent stimulator of bone resorption in inflammatory diseases. We examined the effect of cottonseed extract on the LPS-induced production of tumor necrosis factor a (TNF-$\alpha$) and nitric oxide (NO) in MC3T3-E1 cells. Treatment with cottonseed extract ($10{\sim}50{\mu}g/mL$) decreased the $5{\mu}g/mL$ LPS-induced production of TNF-$\alpha$ and NO in osteoblasts, suggesting that the antiresorptive action of cottonseed extract may be mediated by decrease in these local factors. This study suggests that cottenseed may contribute to antiresorptive action against osteoblastic cells, resulting in a beneficial effect in promoting the function of osteoblastic cells.

Phosphorus removal by lime-natural mineral dissolved solutions

  • In previous studies, solely ferric (Fe3+) and calcium (Ca2+) ions were commonly used for removal of PO4-P (considered as T-P in this study) in wastewater via chemical precipitation. Herein, the removal of total phosphorus (T-P) in wastewater was performed using various mineral and lime dissolved solutions. The dissolution kinetics of different minerals (feldspar, olivine, elvan, illite, sericite, and zeolite) and lime was compared and used their solutions for T-P removal of real wastewater. The highest T-P removal (almost 90%) was obtained by the lime dissolved solution and followed by zeolite, illite, feldspar, and others. We observed a significant co-relationship (R of 0.96) between the amount of initial Ca2+ and T-P removal. This was induced by formation of hydroxyapatite-like mineral via Ca-P precipitation reaction at high pH solution. Furthermore, additional removal of suspended solid (SS) and chemical oxygen demand (COD) was achieved by only lime dissolved solution. Finally, the lime-feldspar dissolved solutions were prepared at different ratios (10-50%), which showed a successive T-P removal up to two times by samples of 40 and 50%.

Bioequivalence of Losazol Tablet to Pletaal Tablet (Cilostazol 50 mg) (프레탈 정(시로스타졸 50 mg)에 대한 로사졸 정의 생물학적 동등성)

  • Bioequivalence of two cilostazol tablets, the $Pletaal^{TM}$ (Korea Otsuka Pharmaceutical Co., Ltd.) and the $Losazol^{TM}$ (Kyoung Dong Pharmaceutical Co., Ltd.), was evaluated according to the guideline of KFDA. Fourteen normal male volunteers (age $20{\sim}28$ years old) were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After two tablets containing 50 mg of cilostazol were orally administered, blood was taken at predetermined time intervals and the concentration of cilostazol in plasma was determined with an HPLC method using UV detector. The pharmacokinetic parameters $(C_{max},\;T_{max}\;and\;AUC_t)$ were calculated and ANOVA was utilized for the statistical analysis of parameters. The results showed that the differences in $C_{max},\;T_{max}\;and\;AUC_t$ between two tablets were 3.14%, 10.0% and 7.35%, respectively. The powers $(1-{\beta})$ for $C_{max},\;T_{max}\;and\;AUC_t$ were 89.67%, 80.97% and 83.87%, respectively. Detectable differences $({\Delta})$ and confidence intervals were all less than 20% except $T-{max}$, but confidence interval of $T_{max}$ was also less than 20% at the significance $level({\alpha})$ of 0.1. All of these parameters met the criteria of KFDA for bioequivalence, indicating that $Losazol^{TM}$ tablet is bioequivalent to $Pletaal^{TM} tablet$.

  • Today, the phase of modem war is very different from past war. That is, the winning of war depends on the ability to obtain information and high technology. The purposes of this research are to propose an effective R&D governance model in national defense sector and to present R&D strategy for obtaining core national defense technology. As a part of collaborative innovation, the strategy to exchange R&D results actively between the defense sector and the private sector will be discussed. The main contribution of this research is dearly defining the concept of R&D governance in national defense sector and applying it to an actual case. The national defense R&D governance model proposed in this paper is based on the characteristics of national defense R&D which are different from other industries. The analysis of business success factors for national defense R&D through the T-50 case study is presented in detail. The T-50 case study reveals the importance of strategic intent, core technology knowledge base, organizational structure, and project management.

Identification of anti-HIV and anti-Reverse Transcriptase activity from Tetracera scandens

  • We report here that an ethanol extract of Tetracera scandens, a Vietnamese medicinal plant, has anti-HIV activity and possesses strong inhibitory activity against HIV-1 reverse transcriptase (RTase). Using a MT-4 cell-based assay, we found that the T. scandens extract inhibited effectively HIV virus replication with an $IC_{50}$ value in the range of 2.0-2.5 ${\mu}g$/ml while the cellular toxicity value (CC50) was more than 40-50 ${\mu}g$/ml concentration, thus yielding a minimum specificity index of 20-fold. Moreover, the anti-HIV efficacy of the T. scandens extract was determined to be due, in part, to its potent inhibitory activity against HIV-1 RTase activity in vitro. The inhibitory activity against the RTase was further confirmed by probing viral cDNA production, an intermediate of viral reverse transcription, in virus-infected cells using quantitative DNA-PCR analysis. Thus, these results suggest that T. scandens can be a useful source for the isolation and development of new anti-HIV-1 inhibitor(s).

Inhibitory Effects of Leachianone A from Sophora fIavescens Ait. against Cadmium Induced Cytotoxicity (고삼으로부터 분리된 Leachianone A의 카드뮴에 대한 독성억제 효과)

  • In this study, cytotoxicity of cadmium on NIH 3T fibroblasts was utilized in order to discover antitoxic compound in methanol extract of Sophora fIavascens Ait. There were treatment groups; control (medium only), $MTT_{50}$ group and five experimental groups. MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H- tetrazoliumbromide} assay was performed to evaluate the cytotoxicity of cell organelles and $IC_{50}$ was also measured. Accordingly we have examined the detoxification effects of methanol extract of S. flavescens Ait. and leachianone A (LA) on cadmium-treated NIH 3T3 fibroblasts ($IC_{50}=\;12.5{\mu}M$) to observe morphological changes by the light microscopy. Both S. flavescens Ait. methanol extract and LA showed inhibitory effects on cadmium-induced cytotoxicity. Furthermore, LA showed dose-dependency in detoxication. From these results, it is conceivable to suggest that LA from S. flavescens Ait. methanol extract is a potential antitoxic agent.

Investigation of Co- and Pr-doped yttria-stabilized cubic zirconia (YSZ) single crystal grown by skull melting method (스컬용융법에 의해 성장시킨 Co와 Pr이 첨가된 이트리아안정화큐빅지르코니아(YSZ) 단결정의 연구)

  • Co-(0.7 wt%) and Pr-(2.0, 3.5 or 5.0 wt%) doped cubic zirconia ($ZrO_2:Y_2O_3=50:50wt%$) single crystals grown by a skull melting method were heat-treated in $N_2$ at $1150^{\circ}C$ for 5 hrs. The brown colored as-grown single crystals were changed into either dark brownish green, greenish blue and light green color after the heat treatment. Before and after the heat treatment, the YSZ (yttria-stabilized zirconia) single crystals were cut for wafer form (${\phi}7.5mm{\times}t3mm$). The optical and structural properties were examined by UV-VIS spectrophotometer and X-ray diffraction. Absorption by $Co^{2+}$(${\fallingdotseq}589nm$: ${\Gamma}_8[^4A_2(^4F)]{\rightarrow}{\Gamma}_8+{\Gamma}_7[^4T_1(^4F)]$, ${\fallingdotseq}610nm$: ${\Gamma}_8[^4A_2(^4F)]{\rightarrow}{\Gamma}_8[^4T_1(^4F)]$], ${\fallingdotseq}661nm$: ${\Gamma}_8[^4A_2(^4F)]{\rightarrow}{\Gamma}_6[^4T_1(^4F)]$]) and $Pr^{3+}$(${\fallingdotseq}450nm$: ${^3}H{_4}-{^3}P{_2}$, ${\fallingdotseq}473nm$: ${^3}H{_4}{\rightarrow}{^3}P{_1}$, ${\fallingdotseq}484nm$: ${^3}H{_4}{\rightarrow}{^3}P{_0}$), change of ionization energy and lattice parameter were confirmed.

Rat Lens Aldose Reductase inhibitory of Taraxacum mongolicum and two Cirsium species (민들레와 두종의 엉겅퀴의 Rat lens aldose reductase 억제활성)

  • To evaluated active principles for diabetic compolications from the Taraxacum mongolicum (T. mongolicum), Cirsium japonicum (C japonicum), and Cirsium setidens (C. setidens) and its constituents, as well as silymarin tested for their effect on rat lens aldose reductase. As a result, the MeOH extract of T. mongolicum ($IC_{50}\;8.71\;{\mu}g/mL$) have strong lens aldose reductase inhibition abilities. Also, Aldose reductase inhibitory showed an isolated luteolin ($IC_{50}\;20\;{\mu}M$) from the T. mongolicum and sliymarin ($IC_{50}\;13\;{\mu}M$).
