Differential Expression of Three Catalase Genes in Hot Pepper (Capsicum annuum L.)

  • Three different catalase cDNA clones (CaCat1, CaCat2, and CaCat3) were isolated from hot pepper (Capsicum annuum L.), and their expression patterns were analyzed at the levels of mRNA and enzyme activity. Northern hybridization showed that the three catalase genes were differentially expressed in various organs, and that expression of CaCat1 and CaCat2 was regulated differently by the circadian rhythm. In situ hybridization revealed different spatial distributions of CaCat1 and CaCat2 transcripts in leaf and stem. In response to wounding and paraquat treatment, CaCat1 mRNA increased at 4-12 h in both paraquat-treated and systemic leaves. In contrast, wounding had no significant effect on expression of the catalase genes. The increase of catalase activity in the paraquat-treated and systemic leaves paralleled that of CaCat1 mRNA, but did not match that of CaCat1 mRNA in paraquat-treated stems. Our results suggest that CaCat1 may play a role in responses to environmental stresses.

  • New prednisolone derivatives including methyl $20{\xi}-acetoxy-prednisolonate$ have been synthesized and evaluated for local anti-inflammatory agents with reduced systemic side effect. Methyl $20{\beta}-dihydroprednisolonate$ and methyl $20{\xi}-acetoxyprednisolonate$ showed comparable anti-inflammtory activity in vivo with significantly reduced systemic side effects.

  • Objectives : We studied the effects of Radix Asteris herbal acupuncture solution (RAHAS) on the type I hypersensitivity. Methods : In vivo, we measured compound 48/80 induced active systemic anaphylactic shock, anti-DNP IgE induced passive cutaneous anaphylaxis (PCA) and acetic acid induced microvascular permeability using ICR mice. In vitro, we showed effects on cytotoxicity and ${\beta}$-hexosaminidase release from RBL-2H3 cells. Results : In vivo, RAHAS pretreatments at $BL_{13}$ and optional points inhibited active systemic anaphylactic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. PCA was only inhibited by RAHAS pretreatments at $BL_{13}$. In vitro, RAHAS treatments inhibited ${\beta}$-hexosaminidase release. Conclusion : These results suggest that RAHAS may be beneficial in the prevention of type I hypersensitive inflammatory response.

승마 추출물의 항알레르기 효과 (Antiallergic Effect of Cimicifuga heracleifolia Extract)

  • Effects of the aqueous extract of Cimicifuga heracleifolia (CHAE) on the allergic reactions were investigated. CHAE inhibited systemic anaphylaxis induced by compound 48/ 80 in mice dose-dependently. Especially, CHAE inhibited compound 48/80-induced systemic anaphylaxis 100% with a dose of 0.5mg/g body weight. CHAE significantly inhibited serum histamine levels induced by compound 48/80. CHAE inhibited histamine release from the rat peritonea] mast cells activated by compound 48/80 or anti-DNP IgE. Our studies provide evidence that CHAE will be beneficial in the treatment of anaphylias.

황금약침(黃芩藥鍼)이 Type 1 Hypersensitivity에 미치는 영향 (Effect of scutellariae radix pharmacopuncture on the type 1 hypersensitivity)

  • Objectives : We studied the effects of Scutellariae Radix pharmacopuncture solution (SRHAS) on the type 1 hypersensitivity. Methods : In vivo, we measured compound 48/80-induced active systemic anaphylactic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rats. In vitro, we showed effects on cytotoxicity and ${\beta}-hexosaminidase$ release from RBL-2H3 cells. Results : In vivo, SRHAS pretreatments (100% or 50%) at BL13 inhibited active systemic anaphylactic shock induced by compound 48/80. PCA was only inhibited by pretreatments of SRHAS at optional points. In vitro, $0.1{\sim}2%$ SRHAS treatments did not affect cell viability while ${\beta}$-hexosaminidase release was significantly inhibited. Conclusions : These results suggest that SRHAS may be beneficial in the inhibition of type I hypersensitive inflammatory response.

경구투영된 김치 유산균 파쇄액이 쥐의 면역활성에 미치는 효과 (Immunostimulation Effects of Mice Fed with Cell Lysate of Lactobacillus plantarum Isolated from Kimchi)

  • Immunositmulation effects of the mice fed with the cell lysate of Lactobacillus plantarum isolated from Kimchi were studied. The mice group fed with cell lysate was different from the control group on the degree of immune responses, e.g. 1) proliferation of splenocytes and Peyer's patch cells, 2) production of nitric oxide (NO) by peritoneal macrophages, 3) production of intestinal secretory lgA (slgA), 4) variation of TNF-${\alpha}$ and IL-2 concentration in blood, 5) production of specific lgG against sheep red blood cells. A general enhancement in enteric and systemic immune responses was observed with a simple oral administration of immunostimulators. With the oral feeding of L. plantarum, not only the total amount of gut secretion antibody, but also the binding capacity of antibodies to the enteric microorganisms including L. plantarum was increased. These experimental results clearly showed that the oral feeding of immunostimulators gave multifunctional effects on the mucosal and systemic immune systems of mice.

전신성 건선 환자의 한방 치료 임상 보고 1례 (A Clinical Case of Systemic Psoriasis with Korean Medical Treatment)

  • Objectives : The purpose of this study is to report the Sik-yang-seng(食養生) therapy with Korean Medicine and acupuncture on a chronic psoriasis patient who is occurred to latrogenic cushing's syndrome. Methods : The patient who has been diagnosed psoriasis had symptoms with latrogenic Cushing's syndrome. The patient was treated with Sik-yang-seng(食養生) therapy including Korean Medicine and acupuncture. Evaluation was done by PASI score, itching score, statistics and pictures. Results : After treatment, the PASI score of patients with Sik-yang-seng(食養生) therapy including Korean Medicine and acupuncture was 1.8 which means 95.5% improvement. Conclusions : Korean medical treatment performed with Sik-yang-seng(食養生) therapy can help recovery of Biwi(脾胃)function therefore can increase effect of the treatment of psoriasis and latrogenic Cushing's syndrome.

Mouse 모델 알레르기 반응에서 삼백초(三白草)가 미치는 영향 (Inhibitory Effect of Saururus chinensis (Lour.) Baill Extracts on Allergy in Mouse Models)

  • We investigated the effect of Saururus chinensis (Lour.) Baill (SCB) on allergy in mice. We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the plasma histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability and anti-DNP IgE-mediated passive cutaneous anaphylaxis. In addition, SCB reduced IL-10 mRNA expression of the lung on ovalbumin-induced allergy. These results indicate that SCB inhibits allergy.

  • Purpose: The purpose of this study was to find the factors influencing adolescents' adult media engagement. The study is based on Agnew's General Strain Theory (GST) and a micro-systemic perspective. Methods: The study examined 479 middle school freshmen who participated in the fourth wave of the Korean Children & Youth Panel Survey (KCYPS) conducted by National Youth Policy Institute. Data were analyzed through descriptive statistics, factor analysis, and reliability analysis using SPSS 22.0 Ver. Also, hypothesis testing was performed via covariance-based structure equation modeling (CB-SEM) using Amos 21.0 Ver. Results: Strain factors had positive effects on adolescent depression, and depression had a positive effect on adult media engagement. Also, cell-phone dependency had a positive mediation effect between depression and adult media engagement. Conclusion: The study's result identified the factors affecting adult media engagement. Therefore, we suggest intervention programs considering adolescents' strains, depression and cell-phone dependency to improve their health.

Lidocaine을 사용한 상박신경총 차단시 Clonidine을 첨가하면 마취와 제통시간이 연장된다 (Clonidine Added to Lidocaine Prolongs the Duration of Anesthesia and Analgesia during Brachial Plexus Block)

  • Background: Clonidine, a selective ${\alpha}_2$ adrenergic agonist, increases the duration of anesthesia and analgesia when it is used with local anesthetics. This study was undertaken to evaluate whether clonidine, which was mixed with lidocaine for the brachial plexus block (BPB), has a local (peripheral) or a systemic (central) anesthetic effect. Methods: Seventy patients scheduled for upper extremity surgery were randomly allocated to two groups. In group IV (n = 35) an axillary perivascular BPB was performed with 40 ml of 1% lidocaine and 1:200,000 epinephrine, and just after BPB clonidine $2{\mu}g/kg$ was administered intravenously. In group BPB (n = 35) the same BPB was performed with 40 ml of 1% lidocaine, 1:200,000 epinephrine and clonidine $2{\mu}g/kg$. The following variables were recorded: onset time, duration of anesthesia and analgesia, and adverse effects. Results: Onset time was comparable in both groups. The duration of anesthesia and analgesia significantly increased to 306 min and 354 min in group BPB, when compared to 119 min and 151 min in group IV, respectively. No side effects such as hypotension, bradycardia, and sedation were reported. Conclusions: The duration of anesthesia and analgesia is prolonged by adding clonidine to lidocaine during brachial plexus block, which suggests that its effect is local rather than systemic.
