Crossbreeding Combination Test for the Production of New Synthetic Korean Native Commercial Chickens (신품종 토종닭 생산을 위한 교배조합 검정시험)

  • In this study, we performed a cross-combination test using offspring from two paternal and five maternal parent stock lines to produce a new synthetic breed of Korean native commercial chickens. The survival rate, body weight, feed intake, and feed efficiency were measured for 10 crosses, with 837 Hanhyup-3 chickens used as a control. Most chickens had brown-based feathers, and there was little difference in color phenotypes among combinations. The average survival rate of the combinations was 94.9%, which was significantly different from that of Hanhyup-3 at 75.3% (P<0.01). There was significant differences in body weight among the combinations at all weeks of age (P<0.01). At 10 weeks of age, the FHHY combination was the heaviest with 2,300 g, while the weight of Hanhyup-3 was 1,922 g. In addition, the weight difference between the male and female chickens was 458 g. As a result of regression analysis, the duration needed to reach 1.9 kg was about 61 days for the FHFY, FHHY, and HFFY combinations, and 70 days for Hanhyup-3, exhibiting a 9-day difference. The average feed conversion ratio of the combinations until 10 weeks was 2.42, which was significantly different from that of Hanhyup-3 at 2.97 (P<0.01). In conclusion, FHHY, FHFY, and HFFY were determined to be the best new breed combinations based on production capacity. When these combinations are used as new commercial breeds, they are expected to increase the average weight by 18% and improve feed efficiency by 19%, as compared to the existing Hanhyup-3 breed.

Empirical Selection of Informative Microsatellite Markers within Co-ancestry Pig Populations Is Required for Improving the Individual Assignment Efficiency

  • The Lanyu is a miniature pig breed indigenous to Lanyu Island, Taiwan. It is distantly related to Asian and European pig breeds. It has been inbred to generate two breeds and crossed with Landrace and Duroc to produce two hybrids for laboratory use. Selecting sets of informative genetic markers to track the genetic qualities of laboratory animals and stud stock is an important function of genetic databases. For more than two decades, Lanyu derived breeds of common ancestry and crossbreeds have been used to examine the effectiveness of genetic marker selection and optimal approaches for individual assignment. In this paper, these pigs and the following breeds: Berkshire, Duroc, Landrace and Yorkshire, Meishan and Taoyuan, TLRI Black Pig No. 1, and Kaohsiung Animal Propagation Station Black pig are studied to build a genetic reference database. Nineteen microsatellite markers (loci) provide information on genetic variation and differentiation among studied breeds. High differentiation index ($F_{ST}$) and Cavalli-Sforza chord distances give genetic differentiation among breeds, including Lanyu's inbred populations. Inbreeding values ($F_{IS}$) show that Lanyu and its derived inbred breeds have significant loss of heterozygosity. Individual assignment testing of 352 animals was done with different numbers of microsatellite markers in this study. The testing assigned 99% of the animals successfully into their correct reference populations based on 9 to 14 markers ranking D-scores, allelic number, expected heterozygosity ($H_E$) or $F_{ST}$, respectively. All miss-assigned individuals came from close lineage Lanyu breeds. To improve individual assignment among close lineage breeds, microsatellite markers selected from Lanyu populations with high polymorphic, heterozygosity, $F_{ST}$ and D-scores were used. Only 6 to 8 markers ranking $H_E$, $F_{ST}$ or allelic number were required to obtain 99% assignment accuracy. This result suggests empirical examination of assignment-error rates is required if discernible levels of co-ancestry exist. In the reference group, optimum assignment accuracy was achievable achieved through a combination of different markers by ranking the heterozygosity, $F_{ST}$ and allelic number of close lineage populations.

Production of Virus-Free Stocks from Citrus Plant by the Shoot-Tip Grafting and Heat Treatment (열처리와 Shoot-Tip Grafting에 의한 감귤 바이러스 무독묘 생산)

  • Virus-free stocks was produced by the combination of the heat treatment of virus infected plant and shoot-tip grafting (STS). To produce virus-free stocks, the plants infected with citrus viruses were used for virus-free stock production using the modified method of STG in thermotherapy at $40^{\circ}C$ for 16 hours in the light, and at $30^{\circ}C$ for 8 hours of darkness for 4 weeks. Trifoliate orange (P. trifoliata) were used as rootstock seedling for STG. Percentages of virus-free stocks against citrus tristeza virus (CTV), satsuma dwarf virus (SDV) and citrus tatter leaf virus (CTLV) were $75.7\%,\;100.0,\%\;82.6\%$ respectively. Shoot tip size for successful STG were as small as possible. Less than $0.3\;\cal{mm}$ of shoot tips gave the hight efficiency of virus free plants but survival rates were low. And, survival rate after shoot-tip culture was analyzed and the rates were dependant on the cultivars; Yuzu cultivar showed the hight survival rate ($74.6\%$) and early satsuma mandarin (Iwasagi) was $13.3\%$ as the lowest cultivar. But citrus trees were not succeed to grown, turned brown, and died.

Influence of Density and Feeding Frequency on Early Life History and Cannibalism of River Puffer, Takifugu obscurus (황복, Takifugu obscurus의 초기 생활사와 공식 행위에 있어 밀도와 먹이 공급 횟수의 영향)

  • To investigate the influence of density and feeding frequency (food amount) on growth, cannibal dynamics and survival of puffer Takifugu obscurus (TL $25.0{\pm}0.5$ mm, BW $0.53{\pm}0.03$ g) during early life history, we conducted one experiment composed with 6 density groups set up as 1.43 inds./${\ell}$, 2.14 inds./${\ell}$, 2.86 inds./${\ell}$, 3.57 inds./${\ell}$, 4.29 inds./${\ell}$ and 7.14 inds./${\ell}$. The experiment was triplicately performed in 1 ton FRP tank for 30 days. Daily food intake (DFI), feed efficiency (FE), growth, cannibal frequency, and survival rates were examined. Although not find any significant relation of density to DFI and FE, we found that growth decreased significantly with the increasing of density under 3.57 inds./${\ell}$, but significantly increased with the increasing of density over 4.29 inds./${\ell}$. Cannibal frequency and daily mortality rate was significantly increased with increasing density and decreasing feeding frequency, inducing the most poor survival rate in the highest density group and low feeding frequency. Therefore, we concluded that the density and the feeding frequencies affect sibling cannibal dynamics and survival of puffer stock.

Micropropagation of Juvenile and Mature Tree of Corylopsis coreana by Axillary Bud Culture (액아배양에 의한 유묘 및 성숙 히어리나무의 기내번식)

  • We have developed an in vitro micropropagation system via shoot formation from axillary buds using nodal segments of Corylopsis coreana. Explants from both juvenile tree (one-year-old greenhouse stock seedlings) and mature tree (ten-years-old tree in nursery) were compared with regard to propagation efficiency. Combined treatment of both BA and zeatin were effective on shoot proliferation since the best result was obtained on MS medium supplemented with 0.5∼3.0 mg/L zeatin and 0.2 mg/L BA. Generally, juvenile explants were better in both shoot proliferation and growth than mature explants. However, as the duration of in vitro culture was proceed to 6 months, explants from mature tree also produced three shoots per explant. Distinctive differences in rooting and adaptability to soil of shoots obtained from mother trees. Whereas shoots originated from juvenile explants rooted as high as 97%, those from adult explants showed 62% rooting. Similar result was also observed in soil acclimatization. The plantlets derived from juvenile plants survived 67%, while only 48% of those from adult trees survived. The results showed a possibility of the micropropagation of Corylopsis coreana through shoot formation from axillary buds. In addition, the advance of the research still remain to enhance the frequency of acclimatization of plantlets from mature trees for practical application.

Advancement and Application of Somatic Cell Nuclear Transfer Technique in Dog

  • The cloning of canids was succeeded in 2005, several years after the birth of Dolly the sheep and also after the cloning of numerous other laboratory and farm animal species. The delay of successful somatic cell nuclear transfer (SCNT)was due to the unique reproductive characteristics of the female dogin comparison to other domestic mammals, such as ovulation of immature canine oocyte and a requirement of 25 days for the completion of meiosis within the oviduct (Holst & Phemister, 1971). When the technology for the recovery of in vivo matured oocyte was established, the application of cloning also became possible and cloned dog offspring were obtained. This report summarizes the progress of technical procedures that are required for cloning canids and the application of this technique. The first cloned dog, Snuppy, was achieved using an in vivo-matured oocyte which was enucleated and transferred with an adult skin cell of male Afghan hound. After establishment of a criterion of well-matured oocyte for the improvement of SCNT efficiency, we obtained three cloned female Afghan hound and a toy poodle cloned from 14 year-old aged Poodle using SCNT through this factor. To date, cloned dogs appeared to be normal and those that have reached puberty have been confirmed to be fertile. Through application of canine SCNT technique, first, we demonstrated that SNCT is useful for conserving the breed of endangered animal from extinction through cloning of endangered gray wolves using inter-species SCNT and keeping the pure pedigree through the cloning of Sapsaree, a Korean natural monument. Secondly, we showed possibility of human disease model cloned dog and transgenic cloned dog production through cloning of red fluorescent protein expressing dog. Finally, SCNT can be used for the propagation of valuable genotypes for making elite seed stock and pet dog. In summary, dog cloning is a reproducible technique that offers the opportunity to preserve valuable genetics and a potential step towards the production of gene targeted transgenic cloned dogs for the study of human diseases.

  • To improve the efficiency and safety of railway systems, the train control system has considerably evolved from the ground-equipment-based control system (e.g. track circuit, interlocking system, etc.) into the on-board-equipment-based control system. In addition, this train control system enables the rolling stock to intelligently control the trackside facilities by introducing information and communication technologies (ICT). Accordingly, since the ICT-based train control system simplifies the railway system (i.e. the heavy ground-equipment can be removed), an efficient and cost-effective railway system can be realized. In this paper, we perform a feasibility test of the ICT-based train control system using a simulation. To this end, we implement a test-bed consisting of prototype machines of on-board/ground equipment and introduce an integrated operation scenario for the train control. The simulation results satisfy all the requirements of train operation according to the scenario and show the effectiveness of the proposed train control system.

Comparison of the Practical Use Condition of e-finance Portal Site between Korea and U.S.A. (한.미간 e-finance 금융포털사이트의 활용실태 비교)

  • For increasing the competitiveness and efficiency of Korea's finance industry under the new e-finance paradigm, this paper compared the practical use of finance portal site' on service parts and stage between Korea and U.S.A.. The services which can be served from site are banking, mortgage and credit loan, stock, card, retirement tax, PFM(Personal Finance Management), EBPP(Electronic Bill Presentment and Payment) and Account Aggregation and so on. The stage of site can be divided as the information provide stage which only gives information about service parts, on-line transaction stage which real-time transaction is possibile and PFM services provide stage according to development process. As a result, the beginning of finance portal service in Korea was lated about 10years and more than it of U.S.A. So the development stage of domestic portal site is still staying in the first step and the providing services and contents or business model development parts are also in the same stage than U.S.A. Resides, Korea's sites mainly focus on their first service parts even though they recently aim internet finance portal, and provide not real time transaction but finance information. On the other hand, the U.S.A. site support substantially not only various on-line transactions but also distinctive personal services like PFM(Personal Finance Management), EBPP(Electronic Bill Presentment and Payment), Account Aggregation and Trans-account, brokerage, education center, mortgage loan, mutual fund, option, pension fund and IPOs and so on. Thus, the site of Korea need to establish real type of internet finance portal which provides one-stop services on every type of finance to customers in the real time and also require the strategic integration among finance institutions. The next turn, they need to build information system and education center to give best satisfaction to customers and acquire customer information and marker environment changes and need to provide distinctive services to quality customers throughout database from this. Also the site should provide various type of banking services which refereed above like PEM, EBPP and education center etc, and the government of Korea should support the building of IT infrastructure to Physical, legal, systematic, sociocultural, technical and human resource sections. This paper provided the future movement direction of the domestic finance portal through comparison and analysis on the practical use of it between Korea and U.S.A. and also wanted to contribute for developing and reading of Korea finance portal in the new era of the finance paradigm.

The Effects of Ownership Structure on Analysts' Earnings Forecasts (기업지배구조가 재무분석가의 이익 예측오차와 정확성에 미치는 영향)

  • This paper analyzes empirically how analysts' forecasts affected by ownership structure. This study examine a sample of 1,037~1,629 the analysts' forecasts of firms registered in Korean Stock Exchange in the period from 2000 to 2006. The empirical results are summarized as follows. First, from the analysis, companies which have higher major shareholder's holdings tend to increase earnings forecast errors and earnings forecast accuracy. Meanwhile, companies which have higher institution shareholder's holdings tend to decrease earnings forecast errors and earnings forecast accuracy. This result is in line with the view of previous works that companies with higher major shareholder's holdings look towards more of analysts' optimistic forecasts in order to maintain friendly relations with major shareholders. Because of analysts' private information use from major shareholders, earnings forecast accuracy is higher in high major shareholder's holdings firm than in high institution shareholder's holdings it. Second, this analysis is whether the minimal required selection condition of outside directors, audit committee adoption and audit quality affect the relation between ownership structure and analysts' forecasts. This result is that variables related corporate governance do not affect statically the relation between ownership structure and analysts' forecasts. The meanings of this paper is to suggest the positive relations between ownership structure and analysts' forecasts. After this, if analysts will notice forecasts of more many firms, capital market will be more efficient and this field works are plentiful. Also it will need monitoring systems not to distort market efficiency by analysts' dishonest forecasts.

On-site Inventory Management Plan for Construction Materials Considering Activity Float Time and Size of a Stock Yard (공정별 여유시간과 야적장 규모를 고려한 건설자재의 현장 재고관리 방안 연구)

  • The inventory of many materials requires a large storage space, and the longer the storage period, the higher the potential maintenance cost. When materials are stored on a construction site, there are also concerns about safety due to the reduction of room for movement and working. On the other hand, construction sites that do not store materials have insufficient inventory, making it difficult to respond to demands such as sudden design changes. Ordering materials is then subject to delays and extra costs. Although securing an appropriate amount of inventory is important, in many cases, material management on a construction site depends on the experience of the site manager, so a reasonable material inventory management plan that reflects the construction conditions of a site is required. This study proposes an economical material management method by reflecting variables such as the status of the preceding and following activities, site size, material delivery cost, timing of an order, and quantity of orders. To this end, we set the appropriate inventory amount while adjusting related activities in the activity network, using float time for each activity, the size of the yard, and the order quantity as the main variables, and applied a genetic algorithm to this process to suggest the optimal order timing and order quantity. The material delivery cost derived from the results is set as a fitness index and the efficiency of inventory management was verified through a case application.
