Effects of Cryo-extenders for Spermatozoa Sorted by Percoll on In Vitro Fertility of in Miniature Pigs (Percoll 분리된 미니돼지 정액의 체외 수정 능력에 있어서 동결보존액의 영향)

  • The objective of this study was to evaluated the efficiency on sperm cryosurvival and ability of in vitro fertilization using Triladyl and Lactose Egg-Yolk(LEY) as extenders for cryopreservation of separated sperm by 65% percoll in miniature pig. Sperm viability was measured with SYBR-14/PI double stained sperm by flow cytometry. Ability on embryo cleavage rate and blastocyst development were observed by in vitro fertilization after frozen-thawing of sperm separated by 65% percoll. The experimental groups were designed that separated sperm by 65% percoll with Triladyl (ST) or LEY(SL) and unseparated sperm with Triladyl(UT) or LEY(UL) for cryopreservation. As a results, the viability was significantly(p<0.05) higher in ST(55.1%), SL(63.1%), UL(58.8%) than UT(38.2%) group. Sperm viability in SL(63.1%) group was significantly(p<0.05) higher than other experimental groups. On the other hand, embryo cleavage rate was significantly(p<0.05) higher in ST(79.1%), SL(83.2) than UT(74.1) and UL(75.7%) groups at 96h after in vitro fertilization. Blastocyst development was also significantly(p<0.05) higher in ST(21.5%), SL(20.9%) than UT(17.0%) and UL(18.8%) groups. In conclusion, cryopreservation of miniature boar sperm separated by 65% percoll were beneficial to viability and capacity on in vitro fertilization.

  • Effect of substrate composition, stocking density and food organisms on survival and growth of the surf clam, Spisula sachalinensis was studied. Spat collection from the substratum was the highest, when its sand size was less than 250 $\mu m4; at such substratum, survival and growth of the surf clam were also the highest. The spat (1.4 mm) grew to 4.6 mm and 8.2 mm in shell length on day 30 and 50, respectively. The relationship between shell length (SL) and shell height (SH), shell breadth (SB), total weight (TW) and shell weight (SW) of the spat are formulated by the following equations; SH : 0.7351SL+0.2272 ( $r^2$ =0.9600)i SB = 0.4037SL+0.2112 ($r^2$ = 0.9452); TW : 0.2824SL^{2.7020} ($r^2$ = 0.9820)i SW = $0.4021SL^{2.0217} ($r^2$ : 0.6169). In the substratum containing fine sand, growth and survival of the clam roared at the density of 1~2 ind./$cm^2$ were optimal. The quality of feed decreases in the following order; C. calcitrans, mixture group, I. galbana, P. lutheri, T. suecica.

Study on Dietary habits and Body Composition in University Students According to Salty Taste Preference (대학생의 짠 맛 선호도에 따른 식습관 및 체조성에 관한 연구)

  • This study investigated the effect of salt preference on dietary habits and the body composition of university students. The subjects were divided into two groups: 85 students who dislike salt (salt-dislike (SD) group) and 104 students who like salt (salt-like (SL) group). We found that the SL group showed a higher preference for sweet, sour, spicy, and bitter flavors compared to the SD group. There was no significant difference in the frequency of eating breakfast according to the salt taste preference. The meal speed of the SL group was significantly higher than the SD group (p<0.01) and the body mass index of the SL group ($22.59kg/m^2$) was higher than SD group ($21.04kg/m^2$). The fat mass of the SL group (15.30 kg) was higher than the SD group (12.80 kg) (p<0.01). Salt preference and snack intake frequency had a significant and positive correlation with fat mass. The frequency of eating breakfast, and meal speed also showed a significant and positive correlation with subcutaneous fat. In conclusion, the SL group eats relatively more, speedily and frequently intakes snacks and carbonated drinks, likely resulting in higher body and subcutaneous fat. These results suggest salt preference is related to food choice, influences unreasonable eating habits, and possibly changes body composition. Taste preferences should therefore be considered for dietary consulting and nutritional education.

Lipase-Catalyzed Synthesis of Structured Lipids with Capric and Conjugated Linoleic Acid in a Stirred-Batch Type Reactor (대두유로부터 Lipase를 이용한 재구성 지질의 합성 및 특성)

  • Structured lipid (SL) was produced from soybean oil with molar ratio of 1:2:2 (soybean oil:capric acid:CLA) using Chirazyme L-2 lipase (4% by weight of total substrates). The reaction was conducted for 24 hr at 55$^{\circ}C$ in a 1 L stirred-batch type reactor. SL-soybean oil contained 4.9 mol% capric acid and 4.1 mol% CLA, respectively. Iodine value of SL-soybean oil was reduced than that of soybean oil due to the incorporated capric acids. Tocopherol content in SL-soybean oil was 18.2 mg/l00 g. SL-soybean oil appeared more yellowish color than soybean oil. Reverse-phase HPLC showed that SL-triacylglycerol species containing capric acid consisted of about 12.6 area%.

Molecular Cloning of cDNA Encoding a Putative Eugenol Synthase in Tomato (Solanum lycopersicum 'Micro-Tom') and Prediction of 3D Structure and Physiochemical Properties (토마토 'Micro-Tom' 과실의 eugenol synthase 유전자 클로닝, 단백질의 3차 구조 및 생리화학적 특성 예측)

  • Eugenol is a volatile compound synthesized by eugenol synthase in various plants and belongs to phenylpropene compounds. However, characteristics of eugenol synthase in tomato has not been known. Therefore, we cloned a full length cDNA of a putative eugenol synthase from tomato 'Micro-Tom' using rapid amplification of cDNA ends (RACE) technique and named a clone SlEGS. Open reading frame of SlEGS was 921bp long and its deduced amino acid sequence was 307bp. The BLAST analysis indicated that SlEGS shared high similarity with PhEGS1 (67.1%) and CbEGS2 (69.4%). Amino acid composition of SlEGS was determined by CLC genomics workbench tool and 3D structure of SlEGS was constructed by homology modeling using Swiss-PDB viewer and validated using PROCHECK and ProSA-web tool. In addition, the physiochemical properties of SlEGS was evaluated using ExPASy's ProtParam tool. Molecular weight was 33.93kDa and isoelectric point was 5.85 showing acidic nature. Other properties such as extinction coefficient, instability index, aliphatic index, and grand average hydropathy was also analyzed.

Dairy Potential of Pabna Cows and Crossbreds with Sahiwal and Friesian and Within- and Between-Breed Sire Effects

  • Dairy performances of Pabna cows and its crossbreds with Sahiwal (SL) and Friesian (F) in the Baghabarighat dairying area of Bangladesh were evaluated. Four SL and three F sires were used for the crossbreeding. The exotic sire effect of individuals on their daughters were also evaluated in this study. It showed that the genetic group had a significant (p<0.05) effect on birth weight, age at first heat, calving interval, lactation length and lactation yield. The lowest birth weight (17.9 kg), longest age at first heat (39.2 m) and calving interval (16.3 m) shortest lactation length (199 d) and lowest lactation yield (728 L) were found in Pabna cows. The highest birth weight (22.5 kg). shortest age at first heat (25.5 m) and calving interval (13.7 m), longest lactation length (253 d) and highest lactation yield (1936 L) were found in the F crossbreds. Within the same breed, the effect of individual sire was significant (p<0.05) on age at first heat, lactation length and lactation yield. Individual sire effects of SL bulls differed significantly (p<0.05) but F sires had a non-significant (p>0.05) effect on calving interval in their progeny. The fat % and SNF % in milk and number of services required for conception did not differ (p>0.05) among inter-and intra- breed sire groups.

Heterologous Production of Paromamine in Streptomyces lividans TK24 Using Kanamycin Biosynthetic Genes from Streptomyces kanamyceticus ATCC12853

  • The 2-deoxystreptamine and paromamine are two key intermediates in kanamycin biosynthesis. In the present study, pSK-2 and pSK-7 recombinant plasmids were constructed with two combinations of genes: kanABK, and kanABKF and kacA respectively from kanamycin producer Streptomyces kanamyceticus ATCC12853. These plasmids were heterologously expressed into Streptomyces lividans TK24 independently and generated two recombinant strains named S. lividans SK-2/SL and S. lividans SK-7/SL, respectively. ESI/ MS and ESI-LC/MS analysis of the metabolite from S. lividans SK-2/SL showed that the compound had a molecular mass of 163 $[M+H]^+$, which corresponds to that of 2-deoxystreptamine. ESI/MS and MS/MS analysis of metabolites from S. lividans SK-7/SL demonstrated the production of paromamine with a molecular mass of $324[M+H]^+$. In this study, we report the production of paromamine in a heterologous host for the first time. This study will evoke to explore complete biosynthetic pathways of kanamycin and related aminoglycoside antibiotics.

Comparison of Effects of Rice Straw and Sewage Sludge Cake on Aerobic Composting of Food Wastes (음식물쓰레기의 호기성 퇴비화에 있어서 볏짚과 하수슬러지케이크가 미치는 영향에 관한 비교 연구)

  • This study was performed to compare the effects of rice straw and towage sludge cake as bulking materials on temperature, pH, weight and volume reduction, porosity, C/N ratio, salinity, and conductivity in aerobic composting of food wastes. Volume ratios of food wastes to rice straw in reactor control, RS-1, RS-2, RS-3 and RS-4 were 4:0, 4:1, 4:2, 4:3 and 4:4, respectively. Weight ratios of food wastes to sewage sludge rake in reactor control, SL-1, SL-2, SL-3 and SL-4 were 4:0, 4:1, 4:2, 4:3 and 4:4, respectively. Reactors were operated for 24 days with 1 hour stirring by 1 rpm and 2 hours aeration per day. The values of pH of food waters, rice straw and sewage sludge cake were 4.39, 7.40 and 5.79, respectively. The lowering of the volume ratio of food wastes to rice straw resulted in the high reaction temperature and the fast weight and volume reduction rates. The lowering of the weight ratio of food wastes to sewage sludge cake resulted in the slow weight and volume reduction rates. C/N ratio in control was larger than that in rice straw containing reactors, and that in rice straw containing reactors was larger than that in sewage sludge cake containing reactors. Salinity and conductivity in reactors were condensed and increased by reaction days.

A Mutation of a Putative NDP-Sugar Epimerase Gene in Ralstonia pseudosolanacearum Attenuates Exopolysaccharide Production and Bacterial Virulence in Tomato Plant

  • Ralstonia solanacearum species complex (RSSC) is a soil borne plant pathogen causing bacterial wilt on various important crops, including Solanaceae plants. The bacterial pathogens within the RSSC produce exopolysaccharide (EPS), a highly complicated nitrogencontaining heteropolymeric polysaccharide, as a major virulence factor. However, the biosynthetic pathway of the EPS in the RSSC has not been fully characterized. To identify genes in EPS production beyond the EPS biosynthetic gene operon, we selected the EPS-defective mutants of R. pseudosolanacearum strain SL341 from Tn5-inserted mutant pool. Among several EPSdefective mutants, we identified a mutant, SL341P4, with a Tn5-insertion in a gene encoding a putative NDP-sugar epimerase, a putative membrane protein with sugar-modifying moiety, in a reverse orientation to EPS biosynthesis gene cluster. This protein showed similar to other NDP-sugar epimerases involved in EPS biosynthesis in many phytopathogens. Mutation of the NDP-sugar epimerase gene reduced EPS production and biofilm formation in R. pseudosolanacearum. Additionally, the SL341P4 mutant exhibited reduced disease severity and incidence of bacterial wilt in tomato plants compared to the wild-type SL341 without alteration of bacterial multiplication. These results indicate that the NDP-sugar epimerase gene is required for EPS production and bacterial virulence in R. pseudosolanacearum.
