Anti-obesity effect of resveratrol-amplified grape skin extracts on 3T3-L1 adipocytes differentiation

  • Resveratrol (3,4,5-trihydroxy-trans-stilbene), a phytoalexin found in grape skin, grape products, and peanuts as well as red wine, has been reported to have various biological and pharmacological properties. The purpose of this study was to investigate the anti-obesity effect of resveratrol-amplified grape skin extracts on adipocytes. The anti-obesity effects of grape skin extracts were investigated by measuring proliferation and differentiation in 3T3-L1 cells. The effect of grape skin ethanol extracts on cell proliferation was detected by the MTS assay. The morphological changes and degree of adipogenesis of preadipocyte 3T3-L1 cells were measured by Oil Red-O staining assay. Treatment with extracts of resveratrol-amplified grape skin decreased lipid accumulation and glycerol-3-phosphate dehydrogenase activity without affecting 3T3-L1 cell viability. Grape skin extract treatment resulted in significantly attenuated expression of key adipogenic transcription factors, including peroxisome proliferator-activated receptor, CCAAT/enhancer-binding proteins, and their target genes (FAS, aP2, SCD-1, and LPL). These results indicate that resveratrol-amplified grape skin extracts may be useful for preventing obesity by regulating lipid metabolism.

Research Trends in the Development of Cosmetic Ingredients for Skin Barrier Improvement

  • In 2022, the domestic production performance of functional cosmetics in South Korea reached 4.6 trillion won, accounting for 33.85% of the total cosmetics production. The number of functional cosmetics reviewed increased by about 7.5% from the previous year, totaling 974 items. Especially with the increasing importance of the skin barrier function due to skin sensitivity caused by various environmental pollutants, domestic cosmetic companies are showing interest in the development of new ingredients and products related to this area. This study aims to analyze academic research trends related to in vitro experiments for the development of cosmetics improving the skin barrier, to provide practical information for the cosmetic industry. The findings are as follows: Academic research mainly focused on the efficacy of natural ingredients in improving the skin barrier, but there is a significant lack of quantitative accumulation of research. For the development of skin barrier-improving cosmetic ingredients, efficacy evaluation indicators were set, including hyaluronic acid production, expression of filaggrin gene, loricrin, formation of cornified envelope (CE), and expression of ceramide synthesis enzyme genes. Moreover, effective cosmetic ingredients for improving the skin barrier included lemongrass and perilla leaf extracts, flavonoids, Lactococcus lactis subsp. lactis, Exosomelike Nanovesicles derived from apple callus, Eleutherococcus sessiliflorus, Acanthopanax sessiliflorus, Eleutherococcus gracilistylus, Acer okamotoanum extracts, Aloe vera adventitious root extract, ethanol extract of Aruncus dioicus, and organic solvent fraction of Dracocephalum argunense.

  • This study was performed to measure the effect of carotenoid polarity on absorption and Pigmentation in blood, muscle, and skin of laying hens. Carotenoids used in this study and Polarity were ${\beta}$-8-Apo-carotenoic acid ethyl ester(ACAEE) > astaxanthin > canthaxanthin > ${\beta}$-carotene. The chickens used in this study were 61∼78 weeks old ISA brown laying hens. Experiment #1 was designed to measure the effect of carotenoid level on the accumulation of carotenoids in carcass of laying hens after feeding for 6 weeks. D-carotene was accumulated in skin only at a detectable level when it was fed at 300 mg/kg feed. The skin was pigmented as yellow when it was measured by colorimeter. The concentration of ${\beta}$-carotene in blood was proportional to that in the feed. Pigmentation of muscle by 9-carotene was not effective. Canthaxanthin significantly increased redness of the skin(p<0.05). However, canthaxanthin did not pigment muscle. The level of canthaxanthin in the blood and skin increased as the concentration in feed increased. ACAEE at 200 and 300 mg/kg feed significantly increased yellowness of the skin(p<0.05). At all levels of ACAEE used($\geq$50 mg/kg feed) the b values of colorimeter increased. With increases in the contents of ACAEE, the concentration of ACAEE in the blood and skin increased. Compared to ${\beta}$-carotene, ACAEE and canthaxanthin were absorbed 9- and 3-fold more into the blood, respectively. The concentration of ACAEE and canthaxanthin in the skin was 1/10 of those in the blood. The lower were the concentrations of carotenoids in the feed, the higher were the absorption rates(from feed to blood and from blood to skin) The results indicated that the higher was the polarity of carotenoids, the more effective were the absorption and pigmentation. In experiment #2, the effect of carotenoid levels of feed on the accumulation of carotenoids in each body part of laying hens was determined. The colorimeter values for redness and yellowness significantly increased when canthaxanthin was fed at $\geq$50 mg/kg feed(p<0.05). Breast and thigh were not affected by feeding of canthaxanthin at the levels used. The L values of muscle but not the a and b values were significantly affected by feeding at $\geq$200 mg/kg feed for wings and breasts, respectively. The yellowness of skin and muscle significantly increased when ACAEE was fed at $\geq$ 100 and $\geq$ 200 mg/kg feed, respectively(p<0.05).

Effect of Toluene Application to Skin on the Liver Injury in Rats

  • To investigate an effect of the topical toluene application to .at skin on the liver injury, toluene (35 mg/$cm^2$) was sequentially applied for 3 or 5 days to rat skin and then the animals were sacrificed. 5 day toluene-treated rats showed the slight increase of live. weight per body weight(%) compared with control. Serum levels of xanthine oxidase and alanine aminotransferase activity were significantly increased both in 3 days and 5 days toluene-treated animals compared with control. In the histopathological findings, cytoplasmic degeneration of hepatocytes around the central vein was noted in the liver of rats applied with toluene to the skin. These results indicate toluene application to rat skin feds to somewhat slight liver injury. On the other hand, the hepatic benzylalcohol or aldehyde dehydrogenase activities were significantly decreased by toluene application to rat skin. In conclusion, the liver min was induced by toluene application to rat skin, and it can be hypothesized that accumulation of benzaldehyde in liver cell may be responsible for liver injury.

Anti-Inflammatory Effects of Water Chestnut Extract on Cytokine Responses via Nuclear Factor-κB-signaling Pathway

  • Water chestnut (Trapa japonica Flerov.) is an annual aquatic plant. In the present study, we showed that the treatment of water chestnut extracted with boiling water resulted in a significant increase 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity and decrease the intracellular $H_2O_2$-induced accumulation of reactive oxygen species. In addition, water chestnut extract (WCE) inhibited lipopolysaccharide (LPS)-induced nitric oxide production and suppressed mRNA and protein expression of the inducible nitric oxide synthase gene. The cytokine array results showed that WCE inhibited inflammatory cytokine secretion. Also, WCE reduced tumor necrosis factor-${\alpha}$- and interleukin-6-induced nuclear factor-${\kappa}B$ activity. Furthermore, during sodium lauryl sulfate (SLS)-induced irritation of human skin, WCE reduced SLS-induced skin erythema and improved barrier regeneration. These results indicate that WCE may be a promising topical anti-inflammatory agent.

A rare case report of neonatal calcinosis cutis induced by distant and delayed extravasation of intravenous calcium gluconate

  • A 3,480 g male neonate showed tachypnea symptom with a serum ionized calcium level of 0.66 mmol/L by routine clinical analysis. He was injected calcium gluconate intravenously through femoral vein catheter to treat the hypocalcemia. On second day after the injection, he started to show erythema in the flank area. The lesion became firm and changed into whitish crust consist of small crystals. Abdominal X-ray and ultrasonography showed the accumulation of calcium deposit in the subcutaneous layer of the lesion. Surgical debridement was performed to remove the crust with calcium deposit and acellular fish skin graft rich in omega-3 (Kerecis) was applied to the defect site for secondary intention of the defect wound. After 2 months, the skin and soft tissue defect were fully covered with healthy normal skin without depression or contracture. This report is a first case of iatrogenic calcinosis cutis without extravasation symptom.

  • Melanin is synthesized by tyrosinase to protect the skin from ultraviolet light. However, overproduction and accumulation of melanin can result in hyperpigmentation and skin melanoma. Tyrosinase inhibitors are commonly used in the treatment of hyperpigmentation. Natural tyrosinase inhibitors are often favoured over synthetic ones due to the potential side effects of the latter, which can include skin irritation, allergies, and other adverse reactions. Nuciferine, an alkaloid derived from Nelumbo nucifera, exhibits potent antioxidant and anti-proliferative properties. This study focused on the in silico screening of nuciferine for anti-tyrosinase activity, using kojic acid, ascorbic acid, and resorcinol as standards. The tyrosinase protein target was selected through homology modeling. The residues of the substrate binding pocket and active site pockets were identified for the purposes of grid box optimization and docking. Therefore, nuciferine is a potent natural tyrosinase inhibitor and shows promising potential for application in the treatment of hyperpigmentation and skin melanoma.

Functional Analysis of a Grapevine UDP-Glucose Flavonoid Glucosyl Transferase (UFGT) Gene in Transgenic Tobacco Plants (담배 형질전환체를 이용한 포도 UDP-glucose flavonoid glucosyl transferase (UFGT) 유전자의 기능 분석)

  • Anthocyanin, a phenolic compound, is a pigment that shows blue or red color in the fruit, petal and other tissues. It is an important factor in grape berry skin pigment and accumulates only in the skin. This skin-specific accumulation of anthocyanin has been reported to be regulated by the ufgt gene which encodes UDP-glucose: flavonoid 3-O-glucosyltransferase that participates in the biosynthesis of anthocyanin. The ufgt gene is expressed only in berry skin, while the other genes involved in the biosynthetic pathway are expressed in both skin and flesh tissues. In order to determine whether anthocyanin accumulation is primarily regulated by compartment of UFGT, a ufgt cDNA clone was isolated from grape berry, its open reading frame was ligated in pBI121 vector in either a sense or an antisense orientation under the control of the CaMV35S promoter and the recombinant constructs were incorporated into tobacco plants. Several transgenic lines were selected and characterized to determine the level of expression of the grapevine ufgt transcript and endogenous homologs of tobacco. Compared to the wild-type, the amount of anthocyanins in sense transgenic plants increased by 44%, while the amount of anthocyanins in antisense transgenic plants decreased by 88%. In addition, the color of flowers became intense in the sense transgenic plants. These results suggest that over-expression or repression of the ufgt gene affected the accumulation of anthocyanin in flowers of tobacco.

Surgical Treatment of Congenital Chest Wall Defects (선천성 흉벽질환의 교정)

  • Fifty-nine cases of congenital chest wall defects experienced in the department of thoracic surgery of Seoul National University Hospital were analyzed and the relevant literatures were reviewed. They are 52 cases of funnel chest, 3 cases of pigeon breast, one case of superior sternal fissure, one case of costochondral incurvation, one case of Cantrell`s pentalogy, and one case of Poland`s syndrome. Funnel chest affected males more frequently than females by 44 to 8. All of the funnel deformities were corrected by Ravitch operation or its modification except one which was the first case of this series and was corrected by a sterno-turnover. Two cases required a mechanical ventilation for 3 days and 5 days respectively. Four minor complications which were two cases of skin wound infection and 2 cases of fluid accumulation were noted. Skin would infection was repaired by a secondary closure and fluid accumulation was treated by aspiration only. The result are all excellent without recurrence or reoperation. In 3 cases of pigeon breast, they were treated by subperichondrial resection of all of the involved costal cartilages and shortening their course with reefing sutures in the perichondrium with excellent result. The superior sternal fissure which was combined by a ventricular septal defect was treated by a simple wire closure with a good result. The costochondral incurvation was corrected by subperichondrial resection of deformed cartilages and a rib graft removed from the contralateral normal side. The Poland syndrome and the Cantrell`s pentalogy was already presented previously.

Histological Change and Collagen Formation on Laser Wounded Rat using 808 nm Diode Laser and $CO_2$ Laser

  • Lasers are necessity in our life related to the fields of medicine and cosmetic surgery. With 808 nm diode laser and $CO_2$ laser, we made some wounds on a dorsum of rat by laser irradiation. All of irradiations shows thermal effects on the whole region of skin tissues. They make wound damage depending on laser power and irradiation time. Because a collagen is plays an important role in tissue repair, we studied collagen accumulation in wound tissue. For wound healing, collagen accumulation was found in the near region of damage in epidermis and dermis layer of the rat skin. In case of the quantitative analysis of collagen in wound tissue, the amount of collagen in wound tissue by $CO_2$ laser irradiation is higher than that of 808 nm diode laser irradiation. And re-epithelialization was significantly faster in wound by $CO_2$ laser irradiation compared with that of 808 nm diode laser irradiation.
