Studies on the Comparative Performance of Victory-1, S-36 and Kanva-2 Mulberry Genotypes and their Impact on Silkworm Rearing under Telangana Conditions of Andhra Pradesh.

  • A new high yielding mulberry genotype, Victory-1 (V-1) was recommended to the field for irrigated areas by Central Sericultural Research and Training Institute (CSRTI), Mysore during 1996-1997. As K-2 and S-36 mulberry genotypes are already in the field, a study on the comparative performance of V-1, 5-36 and K-2 varieties, in terms of leaf yield, yield attributes, leaf moisture and moisture retaining capacity and their impact on silkworm rearing was carried out under Telangana conditions of Andhra Pradesh during 1999-2001. Among the three genotypes the leaf yield was found to be significantly (P < 0.01) higher in V-1 (64,130 kg/ha/yr) followed by S-36 (44,064 kg/ha/yr) and K-2 (23,990 kg/ha/yr). The maximum leaf moisture was also observed in V-1 (71.84%) compared to S-36 (66.86%) and K-2 (64%). The moisture retaining ability was more in V-1 (70.17%) followed by S-36 (66.20%) and K-2 (60.08%). The yield attributes were found to be significantly (P < 0.01) enhanced in V-1 over S-36 and K-2. Silkworm rearing results (bioassay) indicated a significant (P < 0.01) reduction in larval duration and improvement in larval weight, effective rate of rearing by number and weight, single cocoon and shell weights and shell ratio in the silkworm lots fed on V-1 variety followed by S-36 and K-2. Hence, in this study, V-1 was found to be superior among the three varieties studied.

Evaluation of the anti-inflammatory effects of silkworm (Bombyx mori L.) pupal extracts against lipopolysaccharide-induced inflammation in the murine macrophage cell line RAW264.7

  • Silkworm pupal extracts (SPE) were prepared in different solvents (water, 30%, 50%, 70%, and 100% ethanol) and their anti-inflammatory effects were evaluated in the RAW264.7 cell line. The SPE composition was initially evaluated by determining the protein content and performing Fourier transform infrared (FTIR) analysis. The protein content of the different SPE ranged from 6.75-130.93 mg/g of extract. FTIR analysis exhibited distinguishable absorption peaks among the extracts and indicated the presence of lipids, proteins, carbohydrates, and nucleic acid moieties. The levels of released nitric oxide (NO) and interleukin-6 (IL-6) expression in lipopolysaccharide (LPS)-induced RAW264.7 cells were only attenuated by 100% ethanolic SPE to 19.44% and 16.77%, respectively. The other solvent extracts were ineffective. Hence, further studies were conducted with 100% ethanolic SPE from three distinct stages of male and female silkworm pupae belonging to four silkworm varieties (Baegokjam; B, GoldenSilk; G, Juhwangjam; J, and YeonNokjam; Y). The best reduction in NO release and interleukin-1β (IL-1β) expression levels was achieved by the SPE of early female pupae belonging to the Baegokjam variety (32.72%) and those of early female pupae belonging to the Baegokjam and GoldenSilk (59.93%) varieties, respectively. The best reduction in IL-6 expression by 49.70% was achieved by SPE from female pupae of the mid-pupal stage belonging to the Baegokjam variety.

  • [ ${\alpha}-Glycosidase$ ] inhibitors slows the velocity of the uptake of monosaccharides in the small intestine by retarding the speed of degradation of disaccharides to monosaccharides, which made it possible to develop ${\alpha}-glycosidase$ inhibitors as the antihyperglycemic ('antihyperglycemic' means 'blood-glucose-level-lowering') reagent for the diabetic patients such as acarbose and miglitol. Twenty kinds of ${\alpha}-glycosidase$ inhibitors have been reported to exist in mulberry, Morns alba, and some of them are also found in the silkworm, Bombyx mari, as the result of its daily feeding of mulberry leaves as the sole diet. 1-Deoxynojirimycin (DNJ), one of the most potent ${\alpha}-glycosidase$ inhibitor, is the most abundant among polyhydroxylated alkaloids with ${\alpha}-glycosidase$ inhibiting activity in both M alba and B. mari, therefore considered the antihyperglycemic criterion of the mulberry- or silkworm-based neutroceutical products. DNJ is thought to be accumulated in the body of silkworm because the its concentration in the silkworm body is two to three times as much as that in the mulberry leaves. Eighteen silkworm F1 varieties have been recommended for industrial rearing in Korea by some standards such as pathological strength. DNJ concentration in 18 recommendedwere measured at 3rd day in the 5th instar after lyophilization to determine the varieties that accumulates DNJ in its body most. GeumOk-Jam was the highest in the DNJ concentration of 5.45 mg/gDW among the recommended F1 varieties.

Effect of silkworm varieties on Paecilomyces tenuipes culture (누에 품종에 따른 눈꽃동충하초 배양 특성)

  • There is no specific data on Entomogenous fungus (DongChungHaCho) in accordance with the silkworm varieties, despite of very high value industrial use as functional materials at domestic and abroad. In this study, we investigated culture condition and characteristics of Paecilomyces tenuipes in 3 silkworm cultivars and 2 specific silkworm breeds. Infection rate of P. tenuipes for silkworm varieties was the highest Kumokjam, followed by Baegokjam, Daeseongjam, Golden silk, Yeonnokjam in that order. The optimum culture conditions were as follows: dark condition, $22^{\circ}C{\pm}1$, and about $4cm^2$ in planting density per pupa. Also, we have developed annual production technologies of P. tenuipes. First, in order that the moisture content of the infection pupa became 3% or less in the natural condition. Second, the dried pupa must be vacuum-packed and was kept under $4^{\circ}C$. Finally, by restoring the moisture content of the dried pupa and culture in optimal condition, the technique for annual production of the P. tenuipes was achieved. Therefore, we expect that the use of Kumokjam and annual production technique will contribute significantly to farm income.

  • The major dominant fluoride-endurance (Dfe) gene was introduced into the commercial varieties by crossing and pedigree selection to breed silkworm races that could normally develop in the area that polluted by fluoride. After backcrossed for two generations, the Dfe gene was made homozygous, and individuals with good economic characters were selected to generate next generation. After 8 generations of selection, their characters became stable, and the silkworm variety which is resistant to fluoride, Huayuan${\times}$Dongsheng, for spring rearing were bred.

  • Historical background and present status of Korean functional sericulture was reviewed. Five-year expansion projects began in 1962, and produced 41,700 tons of cocoon in 1976 and exported $271 million. However, the number of silkworm rearing households reduced to 59,800 by 1987 from 488,000 in 1976 due to rapid development of the secondary and the tertiary industries with higher labor cost. That's why functional sericulture was developed in Korea. Freeze-dried silkworm powder was found that it lowered the level of blood glucose. Nuedongchunghacho, Paecilomyces tenuipes, was cultured on silkworm larva. Odi, the fruit of mulberry trees, was studied as a new food ingredient. Mulberry tree varieties for improved Odi production were also developed.

  • Normally, silkworms, Bombyx mori, generate offspring by sexual activity. As we known, the hybrids of the first generation of the silkworm have higher cocoon production than pure lines. During the sericulture production, many processes are related with sex control. For example, sex sorting in the egg grainages, rearing of only male silkworm to save the mulberry leaf consumption and increase silk output and quality. Therefore it is very interested in understanding the sex control of the silkworm in theory and practice. Chinese sericultural scientists have been being engaged in the researches in the fields of artificifial parthogenesis, dispermic androgenesis, sex-limited varieties, sex linkage balanced lethal strain and high temperature sensitive male stocks for several decades and gained substantial achievement. Some of the achievements have been used in the commercial production. In this review, the authors introduced that the methods for control of the silkworm sex, and regulate the silkworm sex ratio according to different producing aim in the world and especially in China.

In vitro screening of anti-skin aging and antioxidant properties of aqueous/solvent extracts from distinctive stages of silkworm (Bombyx mori L.) pupae

  • Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.

  • Since 1992, Sericulture & Entomology Research Institute(NSERI) and Kyung Hee University group screened an activity of lowering blood-glucose levels with silkworm related materials such as silkworm larvae powder, dried feces, pupae and silkworm moth in other to guide laymans to rational and educated utilization of silkworm-related materials for the treatment of diabetes mellitus. In experiments examining several silkworms in different stages and prepared in different conditions, a freeze dried silkworm powder of 5th instar 3rd day showed a higher glucose lowering activity about 20% than heat dried matured silkworm powder. Among the three dosage of 500mg, 830mg and 1,160mg in ate preliminary clinical trial, the 830mg exhibited a significant effect on postprandial blood glucose level and did and did not cause any hypoglycemic side effect. In the blood glucose lowering activity of mulberry and silkworm varieties, the Yongcheonppong and Samkwangjam showed the highest activity for lowering blood glucose levels. In experiments to see the difference in blood-glucose lowering activity between either male and female or larvae and pupae, activity of larvae was higher than that of pupae and the male was higher than female. The heating dry and artificial diet showed lower than mulberry diet and normal freeze dry of the 5th instar 3rd day. Among the sericultural products, larvae showed the highest activity. We find out the fact that effect of silkworm powder attributed to the inhibition of ${\alpha}$-glucohydrolase catalyzed reaction in the small intestine.

Studies on the Effects of Atmospheric Pollution in Sericulture -Injuries of Sulphur Dioxide and Cadmium on Parent Silkworms Rearing- (대기공해가 양잠에 미치는 영향에 관한 연구 -원잠종 사육에 있어서 아황산 가스 및 카드미움의 해를 중심으로-)

  • Those studies were examined rearing two varieties on Japanese descent and two varieties of Chinese descent by feeding polluted mulberry leaves, non-polluted ones and water-cleaned ones respectively to find effects of air-pollution on the economic characters of silkworms and analysis of contents of Sulphur and Cadmium in the mulberry leaves and silkworms as followings; 1) Japanese descent of polluted part was delayed about 2.5 days than non-polluted part, Chinese descent was delayed about 4 days or inequal and water-cleaned part was medium in the silkworm larval duration. 2) Results of maximum weight of 5th instal, cocoon layer weight and cocoon weight were decreased in due order non-polluted, water-cleaned and polluted in the factors of mulberry 3) Pupal ratio of Japanese descent was not shown statistical significance, but Chinese descent was revealed it obviously in the factors of mulberry leaves. 4) In the resistance of polluted mulberry leaves, Chinese descent was feeble obviously than Japanese and there were some difference even through among the same varieties. 5) The content of S and Cd of polluted area mulberry leaves was increased about 30% respectively than non-polluted area. 6) The fed part of non-polluted mulberry leaves was S 0.41% and Cd 0.013 ppm water-cleaned part was S 0.47% and Cd 0.024 ppm and polluted part was S 0.52% and Cd 0.042 ppm in the contents of S and Cd of silkworm larvae. 7) The contents of S and Cd didn't make visible injury in mulberry leaves but made it seriously in silkworm larvae. 8) The injury of dust on mulberry leaves was more serious than that of quality of mulberry leaves by air-pollution in the economic characters of silkworm. 9) As above results, Chinese descent should avoid contaminated area and it rearing by water-cleaned mulberry leaves can get noticable results in inevitable case on selection of parents silkworm rearingzone.
