In vitro Folding of Recombinant Hepatitis B Virus X-Protein Produced in Escherichia coli: Formation of Folding Intermediates

  • The folding of recombinant hepatitis B virus X-protein (rHBx) solubilized from Escherichia coli inclusion bodies was investigated. By sequential dialysis of urea, rHBx was folded into its native structure, which was demonstrated by the efficacy of its transcriptional activation of the adenovirus major late promoter (MLP), fluorescence spectroscopy, and circular dichroism (CD) analysis. The decrease in CD values at 220 nm and a corresponding blue shift of the intrinsic fluorescence emission confirmed the ability of rHBx to refold in lower concentrations of urea, yielding the active protein. Equilibrium and kinetic studies of the refolding of rHBx were carried out by tryptophan fluorescence measurements. From the biphasic nature of the fluorescence curves, the existence of stable intermediate states in the renaturation process was inferred. Reverse phase-high performance liquid chromatography (RP-HPLC) analysis further demonstrated the existence of these intermediates and their apparent compactness.

  • For the purpose of developing capacitive humidity sensor having interdigit electrodes, interdigit electrode was modeled and simulated to obtain capacitance and sensitivity as a function of geometric parameters like the structural gap and thichness. For the development of ASIC, switched capacitor signal conditioning circuits for capacitive humidity sensor were designed and simulated by cadence using 0.25um CMOS process parameters. The signal conditioning circuits are composed of amplifier for voltage gain control, and clock generator for sensor driving and switch control The characteristics of the fabricated sensors are; 1) sensitivity is 9fF/%R.H., 2) temperature coefficient of offset(TCO) is 0.4%R.H./$^{\circ}C$, 3) nonlinearity is 1.2%FS, 4) hysteresis is 1.5%FS in humidity range of 3%R.H. ${\sim}$ 98%R.H.. The response time is 50 seconds in adsorption and 70 seconds in desorption. Fabricated process used in this capacitive humidity sensor having interdigit electrode are just as similar as conventional IC process technology. Therefore this can be easily mass produced with low cost, simple circuit and utilized in many applications for both industrial and environmental measurement and control system, such as monitoring system of environment, automobile, displayer, IC process room, and laboratory etc..

  • An ultra wide band ultra-high frequency (UHF) signal processing module was designed for on-line UHF partial discharge (PD) monitoring systems for gas-insulated switchgears (GIS). Major advantage of the unit is an improved PD detection sensitivity through minimizing the effect of surrounding interference signals. The detection sensitivity of the unit was <-60 dBm that is sufficient to detect UHF PD signals as low as 1 pC. Precise detection of PD occurred in the GIS, due to internal defects, is possible by using the signal processing unit.

  • Pulse oximetry is a non-invasive method for monitoring how much oxygenated hemoglobin is present in the blood. The principle of pulse oximetry is based on the red infrared light adsorption characteristics of oxygenated and deoxygenated hemoglobin. Even through the convenience of a pulse oximeter, its weak signal-to-noise ratio against motion artifacts and low perfusion makes it difficult to be accepted by execs devices. Several researchers have suggested the use of an adaptive noise cancellation (ANC) algorithm. They have demonstrated that ANC is feasible for reducing the effects of motion artifacts. Masimo Corporation developed a discrete saturation transformation (DST) algorithm that uses a reference signal and ANC. In commercial devices, it is very hard to escape it because Masimo's patents are very powerful and a better method is yet to be developed. This study proposes a new method that can measure noise saturation as well as accurate oxygen saturation from signals with high motion artifacts without using ANC and DST. The proposed algorithm can extract a normal signal without noise from a signal with motion artifacts. The reference signal from a pulse oximeter simulator was used for the evaluation of our proposed algorithm and achieved good results.

A study on manganese health hazards among experienced welders (용접경력자의 망간에 의한 건강 장해에 관한 연구)

  • This study was conducted to evaluate the health hazards and to develop early diagnostic methods of the manganism in experienced welders and to know the meaning of signal intensities on the brain Magnetic Resonance images. It was carried out from December 1996 to february 1997 with 277 male welders, the duration of welding was at least 5 years or more. The study was consisted of a questionnaire, physical examination and measurements of blood & urine manganese concentrations. Brain Magnetic Resonance imaging was done on 19 study subjects by random sampling. As the duration of welding increases, the positive rates of clinical symptoms, neurological examinations and blood manganese concentrations were also increased. However, physical examinations and urine manganese concentrations were not statistically significant with the duration of welding. Authors couldn't observe any Parkinsonism-like diseases. There were statistically significant correlations between duration of welding and blood manganese concentration(r=0.16, p<0.01). There were not statistically significant correlations between duration of welding and urine manganese concentrations (r=0.06). There were statistically significant correlations between blood & urine manganese concentration(r=0.34, p<0.01). By viewing brain Magnetic Resonance images, 13 welders(68.4 %) among 19 welders were found to have signal intensities. The positive rates of clinical symptoms, physical examinations, neurological examinations and blood & urine manganese concentrations were not statistically different between those with signal intensities and those without signal intensities. We would like to suggest that some non-specific clinical symptoms and neurological signs are correlated with the duration of welding but any Parkinsonism-like diseases had not been observed with these welders. Next we suggest that the high signal intensities on TlWI of brain Magnetic Resonance images are not the sign of manganese intoxication but the sign of manganese deposition.

  • We have examined and analyzed the status of policy, R&D investments, patents and market share of physiological signal measurement technologies for major countries including Korea, the United States, European Union and Japan. Korea is generally inferior to the others in terms of priority of industrial policy, R&D investment, number of patents, technological level and world market share. However, Korea could recover competitiveness, with intensive government supports for this technology.

  • In the spectral analysis, Fourier coeffcients are very important to give informations for the original signal f on a finite domain, because they recover f. Also Fourier analysis has extension to wavelet analysis for the whole space R. Various kinds of reconstruction theorems are main subject to analyze signal function f in the field of wavelet analysis. In this paper, we will present a new reconstruction theorem of functions in $L^1(R)$ using a nonharmonic Fourier series. When we construct this series, we have used dyadic subdivision schemes.

  • We have developed a system which can analyze the status of signal sent from/received to a signalling service equipment within the fully electronic switching system such as TDX-10. The system has such functions that acquire PCM signal of the preferred channel from the subhighway of the thirth-two channel by which a Universal Signal Tr ansceiver Unit(USTU) is connected with Time Switch Unit (TSU), and then classify the type of signal such as R2MFC/DTMF/CCT/VOICE, and determine the digit. Up to now, we have analyzed the frequency spec trum using the FFT. This paper describes the developement of PCM acquirer which can analyze the signal characteristics by acquiring the PCM signal in SHW(subhighway), and proposes the CZT(Chirp Z-Transform) algorithm. An algorithm which analyzes the acquired signal and determines the signal frequency and the corresponding power spectrum using the CZT is also discussed here.

Phosphate Deficiency Stress Response Mediated by Pho Regulon in Bacillus subtilis (Bacillus subtilis의 Pho Regulon을 통한 인산 결핍 스트레스 반응)

  • Bacillus subtilis PhoP-PhoR two-component system (TCS) senses phosphate deficiency conditions, and then controls expression of the Pho regulon to prolong survival. The sensor histidine kinase, PhoR, is autophosphorylated and transfers the phosphate to the response regulator, PhoP. Phosphorylated PhoP (PhoP~P) binds to repeated 6-bp consensus PhoP binding sequences of Pho regulon promoters and activates or represses gene expression. Pho signal transduction systems are part of interconnected signal transduction network involving at least three TCSs (PhoP-PhoR, ResD-ResE TCS, SpoOA phosphorelay), a global carbon metabolism regulator (CcpA), and transition state regulators (AbrB, ScoC). In addition, PhoP-PhoR TCS is cross related with YycF-YycG TCS by cross-regulation. While indescribable progress has been made in understanding phosphate deficiency stress response through refined expression of the Pho regulon in the recent past years, many important questions still remain. Solving these questions may provide important information for application study using B. subtilis.

Evaluating the ANSS and ATS Values of the Multivariate EWMA Control Charts with Markov Chain Method

  • Average number of samples to signal (ANSS) and average time to signal (ATS) are the most widely used criterion for comparing the efficiencies of the quality control charts. In this study the method of evaluating ANSS and ATS values of the multivariate exponentially weighted moving average (EWMA) control charts with Markov chain approach was presented when the production process is in control state or out of control state. Through numerical results, it is found that when the number of transient state r is less than 50, the calculated ANSS and ATS values are unstable; and ATS(r) tends to be stabilized when r is greater than 100; in addition, when the properties of multivariate EWMA control chart is evaluated using Markov chain method, the number of transient state r requires bigger values when the smoothing constatnt ${\lambda}$ becomes smaller.
