Factors Affecting Efficiency of Shoot Induction in Citrus junos Sieb.

  • To enhance the shoot induction efficiency from nodal stem of yooza, the culture conditions such as basal medium, carbohydrate source, solidifying agent and the optimum concentration of plant growth regulators for shoot induction were investigated. The nodal explants were cultured better on MS medium than on MT, SH, B5 or W media in considering of shoot induction rate and mean shoot length. Solidifying agent in medium was better with 0.8% agar than with 0.3% agar, 1.2% agarose or 0.2% gelrite. Carbohydrate source in shoot induction medium was efficient with 30 g/L sucrose. The optimum concentrations of plant growth regulators were determined that 0.1 mg/L NAA as auxin was effective on the shoot induction, and 1.0 mg/L BAP as cytokinin induced multiple shoots efficiently. Shoot induction was the most effective on MS medium supplemented with 4 mg/L $GA_3$ in yooza.

인삼근 신아의 암하생육에 미치는 영향 (Effect of Temperature on Growth of new Shoot in Panax ginseng under Dark)

  • New shoot growth of Panax ginseng root was investigated comparing with burley and soybean from l0$^{\circ}C$ to 30$^{\circ}C$ under dark. Shoot growth ceased by 12days at 30$^{\circ}C$ and optimum temperature appeared to be 15$^{\circ}C$/20$^{\circ}C$ (15hrs/9 hrs) , and 15$^{\circ}C$/15$^{\circ}C$ for ginseng. Shoot growth seems to be Poor below l0$^{\circ}C$. Temperature for maximum growth 20$^{\circ}C$/20$^{\circ}C$ for barley and 20$^{\circ}C$ /25$^{\circ}C$ for soybean. Barley did not germinate above 25$^{\circ}C$/25$^{\circ}C$, but grow better than soybean below 15$^{\circ}C$/25$^{\circ}C$. Fresh weight of 2 weeks suggesting cessation of water uptake at higher temporal use. Ginseng showed greater root ply s shoot of ginseng was linearly increased at 15$^{\circ}C$ but did not increased at 25$^{\circ}C$ after occurence of die-back of new shoot or root rot above 25$^{\circ}C$.

Medium compositions reveal potential organogenesis in the diploid and tetrploid Codonopsis lanceolata

  • Medium composition plays a key role on influencing organogenesis in plant tissue culture. This study was carried out to examine the effects of medium composition on organogenesis in diploid and tetraploid Codonopsis lanceolata and obtain in-vitro mass propagation of superior species of C. lanceolata. Diploid C. lanceolata was found to be declined regarding MS medium composition for each concentration. However, shoot and adventitious root formation were suppressed with higher mineral salt concentration, and active growth of shoot and adventitious root was exhibited as 4.9 cm and 3.2 cm respectively in 1/2 MS medium. While in tetraploid C. lanceolata, it showed 2.9 cm and 3.2 cm respectively in 1/4 MS medium. In the case of sucrose concentration, no consistent decrease was observed for growth of shoot and the adventitious root of diploid both at high and low concentration. The growth of shoot (at 3% concentration) and adventitious root (at 7% concentration) was 2.3 cm and 2.0 cm respectively. Although there was no difference in shoot formation of tetraploid C. lanceolata in all concentrations with the range of 1.7~1.8, there was a slight decrease in shoot growth at high concentration. Results revealed that the adventitious root formation was suppressed at high concentration. The concentration of agar exhibited no significant difference in shoot formation of diploid C. lanceolata at all concentrations. The maximum result of adventitious growth (4.1 cm) was observed at 0.8% concentration. Slight inhibition of shoot formation and root formation of tetraploid C. lanceolata was observed at higher concentration. Shoot formation of diploid C. lanceolata also exhibited inhibition at higher concentration. Shoot formation of diploid C. lanceolata was increased at lower pH and shoot growth was the highest (2.3 cm) at pH 3.8. Adventitious root formation was higher at lower pH. However, both the adventitious root formation and growth exhibited comparatively higher result at pH 5.8. Taken together, the levels of pH had an effect on shoot and root formation in diploid and tetraploid of C. lanceolata

Perennial Ryegrass 품종의 계절적 생육특성 II. 여름철 생육의 품종간 차이 (Seasonal Growth Chrateristics of Perennial ryegrass varieties II. Varietal differences in summer growth)

  • This experiment was carried out to study the biological yields(BY) of varieties by the developmental growth stages and shoot weight(SHW), leaf area(LA), root dry weight(RW), number of tillers(NT), shoot and root ratio(S/R), weight of a tiller(WT) as factors of yield components with 5 varieties of perennial ryegrass. The results are summarized as follows; 1. The biological yields in summer were about 115 of spring yield. 2. The biological yields of Manhattan were the highest until1 third developmental growth stage among perennial ryegrasses, and those for Maprima and P-2 varieties were the highest in the following stages. In terms of shoot dry weight, the similarly tendency appeared in the fifth developmental growth stage. 3. The shoot dry weight increased highly with the root dry weight. 4. The Tempo variety of biological yield was generally low but the weight of a tiller was the highest. 5. The highest increasing stage of biological yields was between fifth and sixth developmental growth stage, but the lowest increasing stage of biological yields was between first and second developmental growth stage, respectively. The dominant factors of biological yields were influenced to shoot and root dry weight.

비슬산 진달래군락지의 생육활성화 방안 (Management Methods on the Growth Activation of Rhododendron mucronulatum Habitat in Mt. Biseul)

  • This study was carried out to find growth increasing methods of Rhododendron mucronulatum in Mt. Biseul. 4 treatments(heavy pruning, light pruning, non-pruning+fertilization, non-pruning(control)) were used in the study area with different pruning methods. Growth characteristics of new shoot, number of creation of flower buds and leaf buds in new shoot were analysed. Significant differences were observed among the treatments and stem diameter degree. Diameter, length of new shoots and the number of flower buds, leaf buds per new shoots increased in the pruning treatments. But heavy pruning showed more effective growth increasing way than light pruning. Non-pruning and fertilization treatments were not effective to increase growth of Rhododendron mucronulatum. And pruning at the end of May was the most effective way for growth of new shoot.

한라산 구상나무림 복원을 위한 묘목식재시험 (Experimental Planting of the Seedlings for the Restoration of Abies koreana forest in Hallasan)

  • 구상나무의 묘목을 이식하여 활착률을 높이는 방법을 찾고자, 한라산국립공원 어리목대피소(해발 900m)에서 양묘된 2~6년생 묘목을 식재시기별(하기와 춘기), 지표처리별(코이어 매트. 코이어 네트 및 무처리)로 시험식재하고 활착률과 신초생장량을 조사.비교하였다. 하기식재의 경우 묘목의 연령이 높을수록 활착율이 높았으며, 지표처리에 따른 차이는 없었다. 춘기식재의 경우에는 묘목의 연령이 낮을수록 활착률이 높았으며, 지표처리 및 차이는 없었다. 하기식재된 구상나무 묘목의 신초생장에 있어서는 지표처리의 효과가 통계적으로 인정되었다. 코이어 매트를 처리한 시험구가 평균 2.11mm로 가장 많이 성장하였고, 대조구는 평균 1.47mm 성장하는 데 그쳤다. 묘령은 신초생장에 영향이 없었으며. 상대적으로 3년생 묘목을 코이어 매트 처리한 경우가 2.26mm로 가장 잘 자랐다. 춘기식재된 구상나무 묘목의 신초생장에 있어서는 지표처리나 묘령의 통계적 유의성은 인정되지 않았다.

Growth Properties of Central and Peripheral Ramets in a Zoysia sinica's Clone

  • A natural, tidal-flat clone of Zoysia sinica was studied to compare ramet growth properties in central area with in peripheral area. In new stolon and rhizome, internode length, weight, shoot height and weight, and spike production were monitored on July 25, 2004. The weight/height rate of shoot between stolon and rhizome, the shoot/stolon (or rhizome) rate in weight between central and peripheral area were not different. However, other properties differed in stolon from rhizome or between central and peripheral area significantly differed: 1. The rhizome in central area had a larger node number, shorter internode length, higher shoot height, larger shoot biomass, and higher rate of non-shoot nodes than that in peripheral area. 2. The stolon in central area had a smaller node number, shorter internode length, and smaller biomass than that in peripheral area. 3. In the same area, the rhizome had a larger node number (except for central area), shorter internode, higher shoot height, larger shoot biomass, higher rate of non-shoot node, and higher rate of node having over two shoots than the stolon. No relationship could be found between shoot size and spike production in shoot on vertical rhizome (lower node of old shoot).

작약(Paeonia lactiflora Pall.) 동아의 경정배양을 통한 기내증식 (In vivo propagation of Paeonia lactiflora Pall. Through Shoot-Tip Culture of Winter Buds)

  • 작약동아의 정아 및 액아의 기내배양에 의한 유묘의 증식에 필요한 배양조건을 구명코자 실험한 결과는 다음과 같다. 정아배양의 경우, 생장조절재의 조성에 무관하게 두지방종에서 100% 신초가 신장하였으나 생장은 의성지방종 은 NAA 0.1 mg/L 단용배지, 영천지방종은 NAA 0.01 mg/L 단용배지에서 가장 양호하였다. 액아배양의 경우, NAA 0.01 mg/L와 zeatin 5.0 mg/L 혼용배지에서 의성지방종은 100%, 영천지방종은 50%의 가장 높은 신초신장율을 보였고 신초의 생장도 양호한 경향이었다. 정아 및 액아배양으로부터 유도된 신초는 vermiculite를 지지물로 한 NAA 0.1 mg/L 첨가배지에서 30.0%의 발근율을 보였으나 뿌리의 생장은 양호하였다.

정단 및 마디조직 배양을 통한 지황의 기내 증식 (In Vitro Propagation by Shoot-tip and Node-bud Culture of Rehmannia glutinosa)

  • 정단 및 마디조직을 이용한 지황의 기내증식방안을 마련하기 위하여 일련의 시험을 실시하였다. 재생된 신초의 정단을 재배양하였을 경우 BA 5.0 mg/L에서 7.8개의 신초가 형성되었으며 투명화된 묘의 발생은 Gelrite 보다는 Bacto-agar 첨가구에서 감소하였다. 광도와 한천의 농도가 증가할수록 신초형성수 및 생장은 억제되었고 생체중 : 건물중의 비율도 감소하였다. 광도가 2000 lx일 때는 Bacto-agar 0.6-1.2%, 3,000 lx에서는 한천 0.4-0.6%에서 건전한 신초를 10개 이상 생산할 수 있었다. 활성탄소 0.1-0.3%첨가는 신초의 생장촉진과 뿌리형성에 촉진적으로 작용하였으며 투명화 발생률도 감소시켰으나 신초형성수는 현저히 감소시켰다. MS배지내 첨가되는 다량원소의 농도를 0.8-1.0배 하였을때 신초형성수 및 생장이 촉진되었다. 기관형성에 미치는 오옥신의 종류 및 농도별 효과를 조사한 결과 NAA나 IBA 보다는 IAA 0.3mg/L 와 BA 5.0mg/L를 혼합처리 하였을 때 증식률이 16배에 달하였다.

Temperature Effects on Shoot Growth and Flowering of Kumquat Trees

  • This study investigated the effects of temperature on the shoot growth and flowering of potted kumquat [Fortunella margarita ( Lour.) Swingle] trees grown in subtropical conditions of I-Lan County in Taiwan. Temperature treatments included T 25-32, T 17-25, T 22, and T 18. The T 25-32 treatment trees were to the day/night temperatures of $25/18^{\circ}C$ for 2 weeks, followed by 28 weeks at $32/25^{\circ}C$. T 17-25 was exposed for 4 weeks to $17/10^{\circ}C$ followed by 26 weeks at $25/18^{\circ}C$. T 22 and T 18 were exposed at $22/18^{\circ}C$ and $18/13^{\circ}C$, respectively, for the entire duration of the experiment. Control trees were placed in a plastic greenhouse under conditions similar to the natural environment. The kumquat trees exposed to high-temperature environment of $32/25^{\circ}C$ showed more frequent and speedy sprouting of new buds, but induced the earlier termination of shoot elongation growth, resulting in decreased vegetative growth. The temperature treatments lower than $22^{\circ}C$ suppressed the new shoot production but increased the shoot growth period, resulting in increased shoot length and diameter. Temperatures higher than $25/18^{\circ}C$ readily induced flowering, with flowering being advanced under the higher temperature conditions such as $32/25^{\circ}C$. However, flowering was substantially inhibited under temperature conditions lower than $22/18^{\circ}C$, indicating the negative role of relatively lower temperatures on flowering of kumquat trees.
