Effects of Altitude and Planting Time on Tuber Bulking of Potato (감자 고랭지 재배 시 표고 및 파종시기에 따른 괴경의 비대반응)

  • Field experiments were carried out to improve the cultural practice of potato by analysing its tuberization patterns. Tuberization patterns affected by different altitudes was analyzed at two potato cultivation regions, Jinbu (600 m) and Daegwallyung (800 m) using two potato cultivars, 'Superior' and 'Atlantic'. To analyse tuberization patterns affected by different planting time, seed potatoes were planted at every 10 days from April 19 to May 19 in Daegwallyung. Total dry weight was greater in plant grown at the altitude of 800 m than 600 m during the entire growth period and the highest increase was observed at the early growth period, July 6, comparing to other growth period. The total dry weight was the greatest at 110~112 days after planting (DAP) at the altitude of 800 m and 108~111 DAP at 600 m. There was no significant differences between altitudes and between cultivars. Tuber dry weight per plant at the altitude of 600 m was lower than 800 m on July 6 (58 DAP), but it increased rapidly from July 21 (73 DAP). At both altitudes, the increase of tuber dry weight per plant from July 6 to August 8 was higher than the other growth period. The time of growth period at which tuber dry weight per plant was the highest was similar at both altitudes that was 118~125 DAP at the altitude 800 m and 118~124 DAP at the altitude 600 m. Dry weight per tuber at the altitude 800m was higher than 600 m due to the number of tubers per plant. A higher increase of crop growth rate (CGR) was shown at the altitude 600 m on July 6 (58 DAP), comparing to at 800 m. The highest tuber dry weight per plant of each cultivar was shown when the planting time was April 29 for 'Superior' and was April 19 for 'Atlantic'. Both the tuber dry weight of plant and the total dry weight were lower at a later planting time. Dry weight per tuber increased quickly during the period between June 30 to August 8. Tuberization period was shortened as the planting time was delayed.

Consideration on coexistence strategy of GM with non-GM, environmentally friend crops in South Korea (GM과 non-GM, 친환경작물의 공존을 위한 제도 보완의 필요성)

  • The current legislation in South Korea clearly states that the tolerance threshold on the adventitious presence of GMO in environment-friendly agricultural products is 3.0% and no GM seed should be detected in their planting seed batches. To date, in Korea, there is no approved GM crop for commercial cultivation in field. However, several GM crops including rice, Chinese cabbage, potato and wild turf grass are currently under risk assessment for their environmental release. Also Korean government (Rural Development Administration, RDA) announced that 11 institutes including universities have been currently certified to carry out a risk assessment of GM crops. Meanwhile, the cultivated area and certified quantities of environment-friendly crops (organic, pesticide-free and low-pesticide) are sharply increasing every year according to the report of National Agricultural Products Quality Management Service (NAQS). In detail, in 2007, the certified quantities of environment-friendly agricultural products were elevated up to 100-fold for organic, 171-fold for pesticide-free and 2,324-fold for low-pesticide crops when compared with those in 1999. The total certified quantity of environment-friendly cereal crops in 2007 was equivalent to 6.4% of total production of cereal crops. Moreover, 24% of total production of root and tuber crops such as potato and sweet potato were certified for environment-friendly agricultural products. In these circumstances, I strongly suggest that current legislations on GM crop's safety management should be revised to include strategies for the coexistence of GM with non-GM crops, especially environment-friendly crops before GM crop is approved to be cultivated for commercialization. Since all types of crops are grown in an open environment, the adventitious presence of GM crops among non-GM crops is inevitable if appropriate measures for coexistence are not established for species by species such as isolation distance, workable management measures to minimize admixture.

Growth Characteristics and Yield of Sweet Potato Cultivars between Virus-free and Farmer's Slips in Late Season Cultivation (고구마 바이러스 무병묘와 농가묘의 만기재배에서 품종 간 생육 및 수량특성)

  • This work was conducted to obtain some information about stable production of high quality seed-tubers in the late season cultivation of virus-free sweet potato [Ipomoea batatas (L.) Lam.]. Growth characteristics and storage root yield between virus-free and farmer's slips in 9 cultivars were investigated using black-film vinyl mulching cultivation with $75{\times}25cm$ planting density on July 10. At 30 days after planting, vine length, vine diameter, number of node, and number of branch in virus-free slips were significantly increased than those in farmer's slips. The vine growth was significantly different among cultivars, and vine elongation was excellent in 'Kogeonmi', 'Shincheonmi', 'Shinhwangmi', 'Shinyulmi', and 'Yeonhwangmi' compared to the other cultivars. At 110 days after planting, vine length, vine diameter, number of node, number of branch, and fresh weight were significantly different among cultivars, but no significant differences between virus-free and farmer's slips were seen except number of node. Total yield in virus-free slips was increased by 12-49% among cultivars than that in farmer's slips. The mean yields between virus-free and farmer's slips were 1,625 kg/10a and 1,230 kg/10a, respectively, and it was significantly different between virus-free and farmer's slips. Percentage of marketable storage root in virus-free slips was 65.6%, and it was significantly higher than 57.8% in farmer's slips. Marketable yields ($40g{\leq}$) between virus-free and farmer's slips were 1,067 kg/10a and 710 kg/10a, respectively. Marketable yield in 'Shincheonmi', 'Shinyulmi' and 'Shinzami' was more than 1,300 kg/10a, and these cultivars showed to be highly adaptable for the late-season cultivation among 9 tested cultivars.

Field test on PGPRM was on going. Varictus methods formality improvement through cultivation were developed. Management research of ginseng production was rare Extension was active throuch official and private organizations and through workshop for the extension specialists, and direct lectures to grower's. Extension services made the researcher to understand the existing problems at grower's fields. Research environment for ginseng production was in prime time only for three years when Korea Ginseng Research Institute was established then gradually aggravated.

Comparison of Sudden Death Syndrome in Responses to Fusarium solani f. sp. glycines between Korea and U.S. Soybean Lines

  • In order to identify the responses of Korean soybean cultivars to sudden death syndrome (SDS), forty-two Korean cultivars and three check cultivars (Hartwig and PI 520733 are resistant; Hartz 6686 is susceptible) were tested with sorghum seed inoculum infested with Fusarium solani f. sp. glycines isolate 171 in the greenhouse. This isolate has blue pigment cultural shape on potato dextrose agar (PDA) medium. All Korean cultivars inoculated with F. solani isolate 171 showed the typical SDS symptoms and disease severity on soybean leaves in each cultivar varied at 4 weeks after inoculation. Nine cultivars were included in the most SDS susceptible group and six cultivars were included in the most susceptible group based on Duncan's multiple range tests (P$\leq$0.05). In results of the LSD analysis for SDS the resistant group, a total of twenty-five Korean cultivars were included in the same SDS resistant group as PI 520733 or Hartwig and fourteen Korean cultivars were included in the same SDS susceptible group as Hartz 6686. In the second experiment, ten Korean cultivars, ten U.S. cultivars, and one introduced line were compared in the same way as the first experiment Disease severity ranking of check cultivars, Hartwig, PI 520733, and Hartz 6686, were the same as in the first experiment. Within Korean cultivars, seven cultivars showed the consistent severity proportions of leaf symptoms. Disease rankings of these cultivars in this experiment were the same as those in the first experiment. Three US cultivars: Hartwig, Hartz 5454, and Forrest, three Korean cultivars: Keunolkong, Myeongjunamulkong, and Jinpumkong 2, and one introduced line, PI 520733, were included in the highest SDS resistant group. Shinphaldalkong 2, Milyang 87, and Samnamkong consistently showed the highest SDS susceptibility in both experiments. Average disease severity in the first and the second experiment were 49.56% and 45.39%, respectively.

Forecasting Late Blight of Potatoes at the Alpine Area in Korea (한국의 고랭지대에 있어서의 감자역병 발생예찰에 관하여)

  • Late blight incited by Phytophthora infestans (Mont.) de Bary, is an important problem for seed potato prodcution in Korea. At the alpine Daekwanryeong area, unprotected potatoes are often defoliated within 14 days after late blight is first observed in the field. Since regular spraying can control late blight, the forecasting service is needed for timely initiation of the spraying program. Climatological data and notes on late blight incidence were recorded during 1970-1977 at the Alpine Experiment Station. The moving graph method using 7-day average mean temperature and 7-day total rainfall did not give highly accurate forecasts. Adding data on relative humidity and 7-day average minimum temperature increased the usefulness of the moving graph. Yields of late blight susceptible varietieties in sprayed plots were related to late blight occurrence and to the rainfall distribution pattern.

  • The disaccharide trehalose ($\alpha$-D-glucopyranosyl-$\alpha$-D-glucopyranoside) is found in variety of organ-isms that are able to withstand almost complete desiccation. In order to identify the function of trehalose in plants, we isolated Arabidopsis trehalase (AtTRE) gene that encodes the enzyme able to hydrolyze trehalose to glucose, and trehalose-6-phosphate synthase isolog, TPS3 gene by RT-PCR. The AtTRE had the substrate specificity to hydrolyze only trehalose, and a broad pH range of enzyme activity. The AtTRE promoter/GUS reporter gene was expressed in cotyledons, mature leaf tissues including guard cells, and developing siliques. The GUS expression driven by AtTPS3 promoter was significant in root tissues, and the level of GUS activity was much higher than that of the pBll 21 control seedlings. The knockout of AtTPS3 gene in Arabidopsis resulted in the retarded root development, whereas the overexpression of AtTPS3 increased the root elongation in the presence of sucrose in MS medium. Possible functions of AtTRE and AtTPS3 in plant will be discussed. In addition, ectopic expression of yeast TPS1 driven by the inducible promoters in tobacco and potato conferred the plants on the drought and freezing tolerances.

  • Non-heading Chinese cabbage (Brassica rapa L. ssp. chinensis) is a popular vegetable in Asian countries. The diamondback moth (DBM), Plutella xylostella (L.), an insect with worldwide distribution, is a main pest of Brassicaceae crops and causes enormous crop losses. Transfer of the anti-insect gene into the plant genome by transgenic technology and subsequent breeding of insect-resistant varieties will be an effective approach to reducing the damage caused by this pest. We have produced transgenic non-heading Chinese cabbage plants expressing the potato proteinase inhibitor II gene (pinII) and tested the pest resistance of these transgenic plants. Non-heading Chinese cabbages grown for 45 days on which buds had formed were used as experimental materials for Agrobacterium-mediated vacuum infiltration transformation. Forty-one resistant plants were selected from 1166 g of seed harvested from the infiltrated plants based on the resistance of the young seedlings to the herbicide Basta. The transgenic traits were further confirmed by the Chlorophenol red test, PCR, and genomic Southern blotting. The results showed that the bar and pinII genes were co-integrated into the resistant plant genome. A bioassay of insect resistance in the second generation of individual lines of the transgenic plants showed that DBM larvae fed on transgenic leaves were severely stunted and had a higher mortality than those fed on the wild-type leaves.

  • Short-preservation of basidiomycetes is generally being conducted in slant tubes containing solid medium based on agar. In this study, we investigated the vitality of 28 species and 76 strains preserved on potato dextrose agar (PDA) at $4^{\circ}C$ for 2~7 years. The survival rates of the fungi were 82%, 86%, 94%, 96%, 94%, and 94% for seven, six, five, four, three, and two years old preservation, respectively. The volume of medium in Lentinula edodes showed decrease after 2 years preserved. The pH of preserved medium was 5.42 in 2007 (two years old), but it became nearly neutral as increasing preservation term.

Studies on the Induction of Sprouting of Dormant Seed Potato in Fall Crop Production (추작감자의 최아법에 관한 연구)

  • To find out the most effective method of treatments for the induction of sprouting of dormant seed potato pieces for the fall crop production of Irish cobbler, this experiment was carried out with sprout inducing bed and field performance trial. In GA treatments, about 10 days were required to sprout and resulted uniform and thin 3-4 sprouts per tuber piece, but sprouts were slender and rooting was not observed. In Ethrel treatments, 20-25 days were required, and sprouts were inferior than that of GA treatment in uniformity of sprouting, and percentage of rotten pieces and of healthy sprouted tuber pieces, but number of sprouts per tuber pieces was low, being 1-2, and sprouts were short, thick, and healthy, and showed good rooting. In GA and Ethrel mixture treatments, 1-2 more days were required to sprout than GA treatments, but sprouts were relatively healthy, and other sprouting pattern were like that of GA treatments. In Ethylene chlorohydrin and 6-Benzyl-adenine treatments, sprouting was like that of Ethrel treatments, but much more days were required than Ethrel treatments and tendency of severe rotting was observed. Optimum treating methods of promising chemicals found to be 1-2 and 2-5 ppm GA solution, 500 and 1000-2000 ppm Ethrel solution, and 1-2+250-500 and 5+250-500 ppm GA and Ethrel mixture solution for 60 min. treatment of tuber piece and whole-tuber, respectively. Induction of sprouting in dry and hot time resulted severer rotting of tuber pieces during the induction of sprouting and with the advancement of dormancy, being delayed in date of treatment, tendency of promotion of sprouting and rooting was observed. When sprouted tuber piece was transplanted at the same date, yields were in order of Ethrel, GA and Ethrel mixture, and GA treatment, indicating the correlation between yield and healthiness of sprout and rooting status of sprouted tuber piece. In all treatments, earlier transplanting resulted higher yields.
