Effect of Electron Beam Irradiation on Selected Vegetable Seeds and Plant-Pathogenic Microorganisms (전자선 조사의 채소 종자 및 식물병원성 미생물에 대한 영향)

  • Electron beam (EB) irradiation was tested to determine the dose required to eradicate plant pathogens, such as Botrytis cinerea and Agrobacterium rhizogenes, from the infected seeds without affecting the germination rate of the irradiated vegetable seeds, including crown daisy, cucumber, hot pepper, green onion, leaf lettuce, and radish seeds. EB irradiation of 1.5 kGy and 2 kGy was sufficient to kill 100% of hairy root disease bacteria and gray mold conidia, respectively. EB irradiation showed no effect or minimal effect on the germination rate of the crown daisy, cucumber, green onion, and radish seeds. However, the germination rate of the hot pepper and leaf lettuce seeds was significantly reduced by using 2 kGy of EB irradiation. Difference in susceptibility to EB irradiation appears not to be related to the weight of each seed, but to the intrinsic characteristic of each plant. Conclusively, EB irradiation might be a useful way to decontaminate crown daisy, cucumber, green onion, and radish seeds.

  • In order to determine the optimum planting density of Job's Tear, We investigatedagronomic characters, yield components and yields by using Ewon and Heuksuk cultivar. The characters, plant height, stem diameter, number of stems per $m^2$ and number of leaves per $m^2$, etc., were excellent in $60{\times}15cm$ planting space for the Ewon and Heuksuk local cultivar. Both the weight of 100 grains and that of 1 liter also higher for the Ewon local cultivar, and yield of stem and seed were the most excellent in $60{\times}15cm$ planting space for the Ewon and Heuksuk local cultivar. For the Ewon and Heuksuk, the analysis of variance for agronomic characters following the difference of planting density were acknowledged to be worth of attention.

Variation of Tocopherol Composition and Morphology in Soybean [Glycine max (L.) Merr.] Germplasms

  • In order to get information on tocopherol content and composition in fifty-six soybean germplasms were evaluated by HPLC. From the principle component analysis, the first three components accounted for 71.6% of the total variance of tocopherol content in the germplasms. Principal component 1 showed significant correlations with all the morphological markers except 100-seed weight. Soybean germplasms were divided into three groups by the first two principal components. The highest content of ${\alpha}$-tocopherol was $38.6{\mu}g$ per g in Tanner, while that of ${\gamma}$-tocopherol was $195.6{\mu}g$ in PI 91073. In case of ${\delta}$-tocopherol, IT 105622 showed the highest value as $29.8{\mu}g$. The contents of tocopherol were gradually increased from the late August to late September, which was 10 days before maturity, in Alchankong and PI 96322. The tocopherol content was higher in seeds from the plants sown early than those sown late. Total tocopherol contents of PI 96188, Geomjeongkong 2, and Suwon 183 grown in Gimje were higher than those grown in Iksan, but the difference was not observed in PI 96322. These results suggest that the contents of tocopherol in soybean were affected not only by the genotypes but also by environment.

  • In this study, A dust inhibitor for $PM_{10}$ was prepared by using molecular structure, molecular weight, viscosity, hydrophilicity, swelling and wetting / moisturizing properties of natural polysaccharide which is widely used as food and cosmetic materials. Various dust control effects were confirmed and superior results were obtained than simple water spraying and synthetic PVA as a control. In addition, the comparison of water evaporation, pot test of cabbage seed germination, effects on fish and aquatic plants, and effects on herb plant growth were studied and the safety of soil and water quality was studied. The availability of the dust inhibitor was confirmed.

Plant Regeneration via Organogenesis from Leaf Explant Culture of Taraxacum coreanum Nakai (흰민들레 잎조직으로 부터 기관형성을 통한 식물체 재생)

  • Plant Regeneration via organogenesis from leaf disk of Korean dandelion was investigated. Leaf disk cultured on MS medium with various combinations of BA (0-4 mg/L) and 2,4-D (0-1 mg/L). Shoot regeneration from leaf explant was observed after 3 weeks of culture. The highest shoot regeneration frequency from leaf disk was obtained with 2 mg/L BA. To analyze the effect of leaf age along shoot formation, we measured number of shoots per explant, shooting rate, fresh and dry weight of leaf explant. The highest number of shoots (11.5) per explant were obtained leaf from 7 weeks old plantlets after seed germination. The regenerated shoots were transferred in 1/2 MS medium with 0.5 mg/L NAA for root formation. Regeneratied plantlets thought organogenesis were growing to whole plants in the pots with acclimation.

Humic Acid and Synthesized Humic Mimic Promote the Growth of Italian Ryegrass

  • Humic acid (HA) is a complex organic matter found in the environments, especially in grassland soils with a high density. The bioactivity of HA to promote plant growth depends largely on its extraction sources. The quality-control of HA and the quality improvements via an artificial synthesis are thus challenging. We recently reported that a polymeric product from fungal laccase-mediated oxidation of catechol and vanillic acid (CAVA) displays a HA-like activity to enhance seed germination and salt stress tolerance in a model plant, Arabidopsis. Here, we examined whether HA or CAVA enhances the growth of Italian ryegrass seedling. Height and fresh weight of the plant with foliar application of HA or CAVA were bigger than those with only water. Interestingly, enhanced root developments were also observed in spite of the foliar treatments of HA or CAVA. Finally, we proved that HA or CAVA promotes the regrowth of Italian ryegrass after cutting. Collectively, CAVA acts as a HA mimic in Italian ryegrass cultivation, and both as a biostimulant enhanced the early growth and regrowth after cutting of Italian ryegrass, which could improve the productivity of forage crops.

A Study of Optimum Conditions in Preparing Gruel with Black Bean Germ Sprout Source (검정콩의 발아물을 이용한 죽 제조의 최적화 조건에 관한 연구)

  • Three different gruels; $100\%$ rice, $70\%$ rice and $30\%$ sprouted black bean with sprout, and $100\%$ rice and $70\%$ sprouted black bean, were prepared. Organoleptic, chemical and rheological properties were compared according to the heating time and the quantity of water. Regarding to the the rheological properties, the gruel with $30\%$ rice and $70\%$ sprouted black bean with sprout revealed the lowest level of solidity and the highest level of viscosity among these three kinds of gruels. The chemical properties; pH, total sugar content and amylose content were similar each other. In terms of organoleptic properties, the best gruel was made with $30\%$ rice and $70\%$ sprouted black bean with sprout. In addition, the best cooking condition was found that 9 times of water to the weight of rice and sprouted black bean with sprout, was used follows by 40 minutes' heating.

  • Wild ginseng production is increasing due to forest recovery for last 30 years. Total number of Symmani (traditional mountain ginseng digger) was 558 in 2001. Provincial distribution of Symmani in 2001 was highest in Kangwon $(32\%),$ next in Choongbook $(21\%)$ and least in Jeonnam $(0.7\%)$ and Kyoungnam $(0.9\%).$ Age distribution of Symmani was $33\%\;for\;fourties,\;32\%$ for fifties and $20\%$ for sixties. There were 8 persons in eighties. Symmanies are still keeping traditional ritual for mountain god serving clothes of colored ribbons and foods. Increased production induced open market system from underground dealing of mountain ginseng. Korea Mountain Ginseng Association established mountain ginseng assessment committee with professional Symmanies in 2001. From September to November in 2001, 987 roots were requested for quality assessment to the committee and 476 roots $(48\%)$ were passed and graded and others were rejected. Highest frequency of rejection was foreign origin. Pass rate was highest $(74\%)$ in Choongnam suggesting best place for quality. Number of collected roots in each province was positively correlated (p=0.05) with number of Symmanies. There are 3 quality groups of mountain ginseng, Heaven (pure natural), Earth (from seeding of wild ginseng) and Man (from seeding or seedling of wild ginseng with slight environmental modification). The relationship between price and age was polynomial in high quality root, Heaven, Earth and seed long head of Man group, and linear in low quality group, seedling long head of Man. The best one in 2001 was 26 g, 124 years old and sold with 109 million won. Quality criteria are age, shape, weight, color and healthy outlook. Fine roots are criteria for health status of roots and taproot is criteria for efficacy and called as medicine barrel. The implication is that ginsenosides have rarely been experienced for efficacy. The quality criteria of cultivated ginseng were originated from those of mountain ginseng. It is unique for mountain ginseng that only fresh one can be on market. Since quality criteria of mountain ginseng must be based on the efficacy experience it is well expected that present criteria might almost be established at the age of Shinnong Materia Medica.

Storage Life Comparison of late Maturing Medium Sheridan, Tano Red and Muscat Bailey A Seed Grapes (만생종 중립계 포도 품종 Sheridan, Tano Red 및 Muscat Bailey A의 저장성 비교)

  • In an attempt to test the storage life of late maturing-medium grapes, Sheridan, Tano Red and Muscat Bailey A cultivated widely in Korea, were selected as test samples. These grapes were treated with SO2 gas and packed with polyethylene film, then stored at 0$^{\circ}C$ under 90% RH. Total weight decreased with the storage days and the loss extent of Sheridan was the least. Abnormal grapes increased along with the storage days and occurred rapidly after 90% days. After 135 days, these were produced as the following order. Sheridan < Muscat Bailey A < Tano Red. The external appearances were deteriorated with the storage days. After 135 days, these indices of Sheridan, Muscat Bailey A and Tano Red wen 6.3, 5.0 and 1.7, respectively, showing the most satisfactory result from Sheridn. Water content decreased along with the storage days. The soluble solids content and the proportionality of sugar to acids increased with the storage days, however the titratable acidity decreased. Considering these results, the storage lifes were excellent by the order Sheridan, Muscat Bailey A and Tano Red.

Chracteristics of Mulberry Fruits on Daeseongppong, Daebungppong, Daeokppong and Shingwangppong(Morus Spp.) (대성뽕, 대붕뽕, 대옥뽕 및 신광뽕 오디의 과실적 특성)

  • Daeseongppong, Daebungppong, Daeokppong were bred, and Shingwangppong was selected among the recommended mulberry varieties of the grown silkworm, in order to meet the increasing the interest about the mulberry fruit, The major characteristics of the mulberry fruits were also studies. The chromosome number of Daeseongppong, Daebungppong and Daeokppong are 2n=56 recognized autotetraploid, and that of Shingwangppong is 2n=42 as a triploid. Seed forming rate of Daeseongppong, Daebungppong, and Daeokppong are more than 58%, but that of the Shingwangppong is very low with 6.7%. Concerning the single fruit weight, Daeseongppong is 4.05g, Daeokppong 3.38g, Daebungppong 2.99g which belonged to big fruit group. However Shingwangppong is medium as 2.28g. The sugar contents of Shingwangppong, Daeokppong, Daebungppong, Daeseongppong are 14.8%,13.7%,13.1% and 12.7% respectively. The sugar/acidity rate of them are above 21. The fruit maturity of Shingwangppong is early, those of Daebungppong, Daeokppong are medium and that of Daeseongppong is late. Yielding Potentiality of Daebungppong is high, those of Daeseongppong, Shingwangppong are comparatively high, and that of Daeokppong is moderate. In the case of cold hardiness, those of Shingwangppong, Daeokppong, Daebungppong are comparatively good, that of Daeseongppong is weak slightly.
