Potential and Significance of Ammonium Production from Helicobacter pylori

  • Glutamine and urea, abundant in body fluids or plasma, yield net ammonium ions upon hydrolysis by ${\gamma}-glutamyl$ transpeptidase (${\gamma}-GTP$) and urease, respectively, and these two enzymes are largely produced from Helicobacter pylori. To investigate bacterial potential of ammonium production, we first quantified those in whole-cell systems and found that the relative ratio of their amounts varied greatly, especially with pH values and the cell's aging. During the H. pylori cultivation, the ratio appeared to be inversely proportional to each other, showing a progressive increase of the ${\gamma}-GTP$ with decreasing of the urease. Under the urease-defective conditions due to low pH or coccoids, the bacterial cells still possessed a considerable amount of ${\gamma}-GTP$, which was found exclusively in the external compartment, therefore, the cell's ammonium production was found to be solely dependent upon glutamine, and the external ammonium concentration was constant without any contribution of urea concentration. Such ammonium constancy would definitely have an adverse effect on the host, because of its absolute requirement for vacuolar degeneration by H. pylori VacA, maximized at approximately 10 mM $NH_4Cl$. It was also found that, by using the metal-saturated membrane vesicles, ammonium ions were likely to be involved in the pH-dependent cation-flux across the H. pylori membrane, where the role of ${\gamma}-GTP$ in ammonium homeostasis around cells was suggested, especially under the hostile conditions against H. pylori.

Structure of Station Class Lightning Arresters and Electrical Characteristics of ZnO Varistor Blocks (발변전용 피뢰기의 구조 및 ZnO 바리스터 소자의 전기적 특성)

  • This paper presents structural characteristics of station class lightning arresters and electrical characteristics of manufactured ZnO varistor blocks which are usable in those arresters. Three types of station class lightning arresters were investigated and those are a ceramic arrester, a FRP tube type polymer arrester, and a FRP rod type polymer arrester. Each arrester has merits and demerits with structural characteristics. In general, polymer arresters were made of silicon rubber for housing materials, FRP tube or rod for mechanical strength, ZnO blocks for electrical characteristics, and metal parts for electrical contact and the silicon rubber, the housing materials, was directly injected to the arrester module which was assembly composed of electrodes, ZnO blocks and FRP tube or rod, and to prevent the nonlinear electric fields distribution on upper parts of arresters, the grade ring was adopted to the upper electrodes. The reference voltage, nonlinear coefficient, residual voltage, and voltage ratio of manufactured ZnO varistors are 4.90kV, 50, 9.54kV, 1.94, respectively. Compared to designed electrical characteristics, the reference voltage was low for 600v and the voltage ratio was slightly high. However, the characteristics of discharge withstand was so excellent that the mechanical destruction does not occur at the impulse current of $8/20{\mu}s$ 10kA for 100 times.

Determining Kinetic Parameters and Stabilization Efficiency of Heavy Metals with Various Chemical Amendment (중금속 안정화제의 반응 매개변수 결정 및 중금속 안정화 효율성 평가)

  • In this study, total of 5 different chemical amendments were evaluated for determining kinetic parameters and stabilization efficiency of heavy metals in aqueous phase. Standard solution of Cd and Pb ($100mg\;L^{-1}$) was mixed with various ratio of amendments (1, 3, 5, 10%) and heavy metal stabilization efficiency was monitored for 24hrs. All examined amendments showed over 90% of removal efficiency for both Cd and Pb except zerovalent iron (ZVI) for Cd (43-63%). Based on result of heavy metal stabilization efficiency, it was ordered as $CaCO_3$ > Dolomite > Zeolite > Steel slag > ZVI for both Cd and Pb in aqueous phase. For kinetic study, first order kinetic model was adapted to calculate kinetic parameters. In terms of reaction rate constants (k), zeolite showed the fastest reaction rate (k value from 0.4882 for 1% to 2.0105 for 10%) for Cd and ZVI (k value from 0.2304 for 1% to 0.5575 for 10%) for Pb. Considering reaction rate constant and half life for heavy metal stabilization, it was ordered as Zeolite > $CaCO_3$ > Dolomite > Steel slag > ZVI for Cd and $CaCO_3$ > Dolomite > Steel slag > Zeolite > ZVI for Pb. Overall result in this study can be interpreted that lime containing materials are more beneficial to remove heavy metals with high efficiency and less time consuming than absorbent materials.

Emission Characteristics of Elemental Constituents in Fine Particulate Matter Using a Semi-continuous Measurement System (준 실시간 측정시스템을 이용한 미세입자 원소성분 배출특성 조사)

  • Fine particulate matter < $1.8{\mu}m$ was collected as a slurry using the Semicontinuous Elements in Aerosol Sampler with time resolution of 30-min between May 23 and 27, 2002 at the Sydney Supersite, Florida, USA. Concentrations of 11 elements, i.e., Al, As, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Se, and Zn, in the collected slurry samples were determined off-line by simultaneous multi-element graphite furnace atomic absorption spectrometry. Temporal profiles of $SO_2$ and elemental concentrations combined with meteorological parameters such as wind direction and wind speed indicate that some transient events in their concentrations are highly correlated with the periods when the plume from an animal feed supplement processing facility influenced the Sydney sampling site. The peaking concentrations of the elemental species during the transient events varied clearly as the plume intensity varied, but the relative concentrations for As, Cr, Pb, and Zn with respect to Cd showed almost consistent values. During the transient events, metal concentrations increased by factors of >10~100 due to the influence of consistent plumes from an individual stationary source. Also the multi-variate air dispersion receptor model, which was previously developed by Park et al. (2005), was applied to ambient $SO_2$ and 8 elements (Al, As, Cd, Cr, Cu, Fe, Pb, and Zn) measurements between 20:00 May 23 and 09:30 May 24 when winds blew from between 70 and $85^{\circ}$, in which animal feed processing plant is situated, to determine emission and ambient source contributions rates of $SO_2$ and elements from one animal feed processing plant. Agreement between observed and predicted $SO_2$ concentrations was excellent (R of 0.99; and their ratio, $1.09{\pm}0.35$) when one emission source was used in the model. Average ratios of observed and predicted concentrations for As, Cd, Cr, Pb, and Zn varied from $0.83{\pm}0.26$ for Pb to $1.12{\pm}0.53$ for Cd.

The Variation of Packing Density According to Powder Size Distribution and Epoxy Resin in Soft Magnetic Composite (연자성 복합체에서 파우더 크기 분포와 Epoxy Resin에 따른 Packing Density 변화)

  • There is growing interest in power inductors in which metal soft magnetic powder and epoxy resin are combined. In this field, the process technology for increasing the packing density of magnetic particles in an injection molding process is very important. However, little research has been reported in this regard. In order to improve the packing density, we investigated and compared the sedimentation heights of pastes for three types of soft magnetic alloy powders as a function of the mixing ratios and the type of resin used. Experimental results showed that the packing density was the highest (71.74%) when the mixing ratio was 80 : 16 : 4 (Sendust : Fe-S : CIP) according to the particle size using an SE-4125 resin. In addition, the packing density was found to be inversely related to the layer separation distance. As a result, it was confirmed that the dispersion of solid particles in the paste was important for curing; however, the duration of the curing process can greatly affect the packing density of the final composite.

Nano Dispersion of Aggregated Y2O3:Eu Red Phosphor and Photoluminescent Properties of Its Nanosol (응집된 Y2O3:Eu Red 형광체의 나노분산 및 나노졸의 형광특성)

  • Nanosized and aggregated $Y_2O_3:Eu$ Red phosphors were prepared by template method from metal salt impregnated into crystalline cellulose. The particle size and photoluminescent property of $Y_2O_3:Eu$ red phosphors were controlled by variation of the calcination temperature and time. Dispersed nanosol was also obtained from the aggregated $Y_2O_3:Eu$ Red phosphor under bead mill wet process. The dispersion property of the $Y_2O_3:Eu$ nanosol was optimized by controlling the bead size, bead content ratio and milling time. The median particle size ($D_{50}$) of $Y_2O_3:Eu$ nanosol was found to be around 100 nm, and to be below 90 nm after centrifuging. In spite of the low photoluminescent properties of $Y_2O_3:Eu$ nanosol, it was observed that the photoluminescent property recovered after re-calcination. The dispersion and photoluminescent properties of $Y_2O_3:Eu$ nanosol were investigated using a particle size analyzer, FE-SEM, and a fluorescence spectrometer.

Dispersion and Shape Control on Nanoparticles of Gd2O3:Eu3+ Red Phosphor Prepared by Template Method (주형법으로 제조된 Gd2O3:Eu3+ 적색 형광체의 나노입자 분산 및 형상제어)

  • $Gd_2O_3:Eu^{3+}$ red phosphors were prepared by template method from crystalline cellulose impregnated by metal salt. The crystallite size and photoluminescence(PL) property of $Gd_2O_3:Eu^{3+}$ red phosphors were controlled by varying the calcination temperature and $Eu^{3+}$ mol ratio. The nano dispersion of $Gd_2O_3:Eu^{3+}$ was also conducted with a bead mill wet process. Dependent on the time of bead milling, $Gd_2O_3:Eu^{3+}$ nanosol of around 100 nm (median particle size : $D_{50}$) was produced. As the bead milling process proceeded, the luminescent efficiency decreased due to the low crystallinity of the $Gd_2O_3:Eu^{3+}$ nanoparticles. In spite of the low PL property of $Gd_2O_3:Eu^{3+}$ nanosol, it was observed that the photoluminescent property was recovered after re-calcination. In addition, in the dispersed nanosol treated at $85^{\circ}C$, a self assembly phenomenon between particles appeared, and the particles changed from spherical to rod-shaped. These results indicate that particle growth occurs due to mutual assembly of $Gd(OH)_3$ particles, which is the hydration of $Gd_2O_3$ particles, in aqueous solvent at $85^{\circ}C$.

Photoluminescence Characteristics of $Y_3Al_5O_{12}:Ce^{3+},Eu^{3+}$ Phosphors by $Eu^{3+}$ ions ($Eu^{3+}$ 농도에 따른 $Y_3Al_5O_{12}:Ce^{3+},Eu^{3+}$ 형광체의 광학적 특성)

  • For this study, Yttrium aluminum garnet (YAG) particles co-doped with $Ce^{3+}$ and $Eu^{3+}$ were prepared via the combustion process using the 1:1 ratio of metal ions to reagents. The characteristics of the synthesized nano powder were investigated by means of X-ray diffraction (XRD), Scanning Electron Microscope (SEM), and photoluminescence (PL). The various YAG peaks, with the (420) main peak, appeared at all Eu concentrationin XRD patterns. The YAG phase crystallized with results that are in good agreement with the JCPDS diffraction file 33-0040. The SEM image showed that the resulting YAG:Ce,Eu powders had uniform sizes and good homogeneity. The grain size was about 50nm. The photoluminescence spectra of the YAG:Ce,Eu nanoparticles were investigated to determine the energy level of electron transition related to luminescence processes. It was composed a broad band of $Ce^{3+}$ activator into the weak line peak of $Eu^{3+}$ in YAG host. The PL intensity of $Ce^{3+}$ has the wavelengths of 480-650 nm and The PL intensity of $Eu^{3+}$ has main peak at 590nm.

Discrimination of the Heated Coconut Oil using the Electronic Nose (전자코를 사용한 가열처리 야자유의 판별)

  • Effect of heat (160, 190, and $220^{\circ}C$ for 24 hr) on coconut oil was examined by principal component analysis using electronic nose consisting of six metal oxide sensors. Increase in heating temperature decreased ratio of resistance and first principal component score (from +0.952 to -0.325), indicating rancidity of coconut oil increased at high heating temperature. Result of electronic nose based on GC with surface acoustic wave sensor showed significant changes in volatile profiles of coconut oil. High resolution olfactory imaging $(VaporPrint^{TM})$ was particularly useful for evaluating oil quality. Peak numbers and areas increased with increasing heating time and temperature (160, $220^{\circ}C$). Electronic nose analysis can provide simple, fast, and straightforward results and is best suited for quality control and process monitoring in flavor field of food industry.

Development of Strain-gauge-type Rotational Tool Dynamometer and Verification of 3-axis Static Load (스트레인게이지 타입 회전형 공구동력계 개발과 3축 정적 하중 검증)

  • In this task, the tool dynamometer design and manufacture, and the Ansys S/W structural analysis program for tool attachment that satisfies the cutting force measurement requirements of the tool dynamometer system are used to determine the cutting force generated by metal cutting using 3-axis static structural analysis and the LabVIEW system. The cutting power in a cutting process using a milling tool for processing metals provides useful information for understanding the processing, optimization, tool status monitoring, and tool design. Thus, various methods of measuring cutting power have been proposed. The device consists of a strain-gauge-based sensor fitted to a new design force sensing element, which is then placed in a force reduction. The force-sensing element is designed as a symmetrical cross beam with four arms of a rectangular parallel line. Furthermore, data duplication is eliminated by the appropriate setting the strain gauge attachment position and the construction of a suitable Wheatstone full-bridge circuit. This device is intended for use with rotating spindles such as milling tools. Verification and machining tests were performed to determine the static and dynamic characteristics of the tool dynamometer. The verification tests were performed by analyzing the difference between strain data measured by weight and that derived by theoretical calculations. Processing test was performed by attaching a tool dynamometer to the MCT to analyze data generated by the measuring equipment during machining. To maintain high productivity and precision, the system monitors and suppresses process disturbances such as chatter vibration, imbalances, overload, collision, forced vibration due to tool failure, and excessive tool wear; additionally, a tool dynamometer with a high signal-to-noise ratio is provided.
