Evaluation of Soil Carbon Storages in the Organic Farming Paddy Fields (유기 논토양의 토양탄소 저장효과 평가)

  • This study was conducted to investigate the differences in carbon storage capacity of soil between the conventional and the organic agricultural cultivation followed by the assessment of their economic values. An analysis of 107 samples in the organic and the conventional rice cultivation soils in six regions across South Korea showed that the five regions, Buyeo-II, Gimhae, Sancheong-I, II and Suncheon, had higher organic soil carbon contents than those of values observed on the conventional soils with the exception of the Buyeo-I areas. Based on the results from soil carbon contents, the carbon storage were estimated to be 36.1 megagram carbon (MgC) per ha in the organic paddy soils, while its conventional paddy soils were 29.4 MgC per ha. It showed that the organic paddy soils were 23 % greater than that of its conventional paddy soils. It was estimated that the carbon trading price for economic assessment was ₩758,100 per ha in the organic paddy soil and ₩617,400 per ha in the conventional paddy soil.

Proximate Composition, Cholesterol and $\alpha$-Tocopherol Content in 72 Species of Korean Fish (72종 어류의 일반성분조성과 콜레스테롤 및 $\alpha$-토코페롤함량)

  • Proximate composition, cholesterol (CHOL) and $\alpha-tocopherol$ ($\alpha-Toc$) content of seventy-two species of fish muscle (sea water fish, 59 species; fresh water fish, 13 species), which caught off Tongyeong coast of the Southern Sea ( Nam-Hae) and caught Kyeongho river, Sancheong, Korea, were determined. Lipid content was ranked from $0.53\%$ to $20.4\%$ and was high in the following order; mid-surface dwelling and migratory fish ($6.09{\pm}4.25\%$) > fresh water fish ($4.40{\pm}5.90\%$) > coastal and reep dwelling fish ($3.41{\pm}2.68\%$) > demersal fish ($2.12{\pm}2.41\%$). There was a negative correlation between the lipid and moisture content in all fish groups (p< 0.001). Protein was approximately $17\~18\%$ in all fish group, and no correlation was between the protein and moisture content in all but the demersal fish, which there was a negative correlation between both contents. Ash content was almost constant in all fish, accounting for $1\~2\%$, Cholesterol (CHOL) content was shown a great difference according to the fish species from 6.7 mg/100 g muscle (grass fish) to 249 mg/100 g (eel) and there was a positive correlation between the lipid and CHOL content. $\alpha-Toc$ content was less than 3 mg/100 g muscle in all fish, except that of eel (4.1 mg/100 g) and was relatively rich in the fish species contained a large amount of lipid.

Enhancing Production of Terpenoids in Metabolically Engineered Transgenic Spearmint (Mentha spicata L.) by Salt and Fungal Elicitors

  • Forest tree species usually takes for long periods to be harvested and cultivated but spearmints are a good model system for woody plant because of reducing and shortening cultivation time. Spearmints are good model plants (Mentha species) for research about terpenoids production and industrial essential oil manufacture. Isopentenyl pyrophosphate isomerase (Iso) and limonene synthase (Limo) are the key enzymes of terpenoid biosynthesis pathway. Transgenic and wild spearmints (Mentha spicata, MS) were cultured in vitro and assessed for the essential oil contents. The content of essential oil of transgenic spearmint also was enhanced slightly depending on the target terpenoid genes. In an attempt to increase productivity of terpenoids further, salt and fungal elicitation strategy was adopted on transgenic Mentha spicata. The salt (800 mM NaCl) as abiotic and two fungi (Botrytis cinerea and Glomerella cingulata) as biotic were used for elicitors. In the absence of salt stress four terpenoids were detected from the spearmint extracts, all of them being monoterpenes. On the other hand, the transgenic (MSIso) extracts contained eleven terpenoids (10 monoterpenes and 1 phenylpropene) while transgenic (MSLimo) extracts contained seven monoterpenes. After 3 days of fungal infection, the resistance indices further increased to 4.38, 3.89 and 2.04 for wild type, MSIso and MSLimo, respectively. The salt and fungal elicitators proved beneficial towards modifying both the terpenoids profile and improvement in the composition of essential oil. These results have important applications for the large-scale production of essential oils and forest biotechnology with respect to spearmint.

Ingredients Analysis and Biological Activity of Fermented Angelica gigas Nakai by Mold (곰팡이 발효 참당귀의 유효성분 분석 및 생리활성 작용)

  • Three mold strains, Aspergillus oryzae (AOFAG), Aspergillus kawachii (AKFAG), and Monascus purpureus (MPFAG) were used for fermentation of Angelica gigas Nakai powder. The contents of polyphenolic compounds, flavonoids, minerals, decursin and decursinol angelate and the activities of DPPH (${\alpha},{\alpha}'$-diphenyl-$\beta$-picrylhydrazyl) free radical scavenging, reducing power, and tyrosinase were measured. The highest contents of phenolic compound and flavonoid were NFAG at 2.78% and MPFAG at 1.18%, respectively. Major minerals were K, Mg, Fe, Na and Ca. Decursin and decursinol angelate were the major ingredients of Angelica gigas according to HPLC analysis. Decursin area was higher in all fermented Angelica gigas than in NFAG. The activities of free radical scavenging and tyrosinase were stronger in all fermented Angelica gigas than NFAG. However, the Fe/Cu reducing powers were stronger in NFAG than all fermented Angelica gigas. Overall, these results may provide the basic data needed to understand the biological activities and chemical characteristics of Angelica gigas fermented by mold for the development of functional foods.

Distribution of Agamermis unka (Nematoda: Mermithidae), a Mermithid Parasite of Brown Planthopper (Nilapawata lugens) in Korean Rice Paddies (우리나라 벼논에서 벼멸구선충(Agamermis unka)의 분포)

  • Agamermis unka, a mermithid parasite of brown planthopper (BPH), Nilaparvata lugens, is the most important natural enemy of BPH and white-backed planthopper (WBPH), Sogatella furcifera in Korea. Distribution of A. unka was investigated in Korean rice fields from 1992 to 1998 and in 2001. Overwintering population of A. unka in Gosung, Namhae, and Tongyoung from 1992 to 1998 was different depending on locality and year. In the survey of A. tanka distribution in the spring of 2001, A. unka was found only at Jangheung and Haenam in Jeonnam province and Namhae, Jinju, Sancheong, Sacheon, Gosung, Tongyoung, Uiryeong, Changwon, Gimhae, and the experimental field of Gyeongsangnamdo Agricultural Research and Extension Services at Jinju in Gyeongnam province out of 30 observed regions in 5 provinces. The number of A. unka was 1,045/㎥ at Namhae, 947/㎥ at the experimental field of Gyeongsangnamdo Agricultural Research and Extension Services, and 395/㎥ at Gosung. Density of A. unka at the rice paddies of Gyeongnam province after harvest in 2001 was higher at the rice fields of Namhae, Gosung, and the experimental field of Gyeongsangnamdo Agricultural Research and Extension Services. Although density of A. unka was higher in the pesticide-untreated plots than fungicide-treated or insecticide-treated plots of forecasting paddies, there were no significant differences. After rice harvest A. unka was found from the forecasting paddies of Gosung, Jinju, Namhae and Sacheon out of 19 observed localities in Gyeongnam province.

Pathogenicity of Korean Entomopathogenic Nematodes (Steinermematidae and Heterorhabditidae) against Local Agricultural and Forest Insect Pests (한국산 곤충병원성 선충 (Steinernematidae와 Heterorhabditidae)의 지역농림해충에 대한 병원성)

  • Pathogenicity of Korean entomopathogenic nematodes against local insect pests was different depending on strains or target-insect pests. Mortalities of diamondback moth, Plutella xylostella larvae were 51.8%, 77.8%, 96.3% or 100% in Hanrim Steinernema sp. and 50.0%, 74.1%, 96.3% or 98.1% in Hamyang Heterorhabditis sp. on filter paper when larvae were exposed to 3, 6, 12, or 24 nematodes per larva. Mortalities of them on kale leaves at the same concentration were 44.4%, 63.0%, 76.1%, or 94.5% in Hanrim Steinernema sp. and 79.7%, 81.6%, 94.4%, or 100% in Hamyang Heterorhabditis sp., respectively. In field test, control value of Hanrim Steinernema sp. was 72.0% and that of Hamyang Heterorhabditis sp. was 84.1% in 14 days when 300,000 nematodes were sprayed to each plot ($13.27\m^2$). Although mortalities of rice stem borer, Chilo suppressalis larvae showed no difference at high concentration, Hamyang Heterorhabditis sp. (47.3~100%) was more effective than Hanrim Steinernema sp. (34.3~83.3%) at low concentration, 50~200 nematodes/ml. When chestnut curculio, Curculio sikkimensis larvae were treated with Sancheong Steinernema sp. and Hamyang Heterorhabditis sp., respectively. When pellucid zygaenid, Pryeria sinica larvae were exposed to nematodes, Pocheon Steinernema sp. was effective ranging from 96.7% to 100% but mortalities of them were 63.3~76.7% in Dongrae Steinernema sp..

Investment Beneficial Analysis of Culture of Mountain Medical Plant Resources - Open field Culture of Schizandra - (산지약용식물자원의 재배 투자 수익성 분석 - 오미자 노지재배 -)

  • This study is to give informations about management Situations and investment returns to someone who will newly cultivate schizandra or expand area of culture. This study used IRR, B/C Ratio and NPV for beneficial analysis. We surveyed fifty one among schizandra cultivation farmhouses in chief producing districts Gangwon Inje, Gyeongbuk Munkyung, Kyungnam Sancheong and Jeonnam Whasoon in Korea from May till September in 2009. So this study surveyed about inputted labors and materials, land price and etc. by working process to calculate producing and operating cost. Fruits of schizandra are sold from 5,000 won to 6,000 won per kg at locals. And IRR(Internal Rate of Returns), B/C Ratio and incomes ratio is calculated by three senarios 5,000 won, 5,500 won and 6,000 won per kg at locals. Discount rate is 3.00%. As the result of this study IRRs are -16.00%, -6.91%, 0.40% one after another and B/C Ratios are 0.81, 0.89, 0.97 one after another. And schizandra cultivation isn't profitable. When price of schizandra fruits is 6,200 won per kg, IRR is 3.00% and B/C Ratio is 1.00. Therefore, if price of schizandra fruits is over 6,200 won per kg, we judge that it is profitable. And income ratio is 23.9% on 6,200 won per kg.

Occurrence and Damage of Japanese Gall-forming Thrips, Ponticulothrips diospyrosi Haga et Okajima (Thysanoptera: Thripidae) in Persimmon Orchards (감 과원에서 감관총채벌레(Ponticulothrips diospyrosi Haga et Okajima)의 발생과 피해)

  • Occurrence of Japanese gall-forming thrips, Ponticulothrips diospyrosi was surveyed from 143 orchards at 11 cities or counties in 6 provinces, Korea from 2008 to 2009. Occurrence of P. diospyrosi was not observed from Jeju (4 persimmon orchards) in Jeju-do and Wanju (12 persimmon orchards) in Jeollabuk-do. However, P. diospyrosi occurred newly in the orchards in Sanchung, Gyeongsangnam-do and in Gangreung, Gangwon-do. Rate of damaged orchards by P. diospyrosi in Gumi, Mungyeong, Sangju and Euseong in Gyeongsangbuk-do (73.3%) was higher than that of in Gyeongsangnam-do or Youngdong areas in Chungcheongbuk-do. A maximum rate of damaged trees by P. diospyrosi was 23.5% in Gumi, while the damage was lower in Sancheong and Jinju representing 0.2 and 1.6%, respectively. Rate of damaged leaves by P. diospyrosi was the highest in Sangju (0.12%). P. diospyrosi occurred between $N35^{\circ}07'50"$ and $35^{\circ}51'24"$ and between $E127^{\circ}38'36"$ to $128^{\circ}53'02"$. Persimmon orchards close to mountain were more damaged by P. diospyrosi. Edge parts of persimmon orchards were more damaged by P. diospyrosi than inner parts, especially in orchards close to mountain. Also, the middle and lower parts of tree canopy were more damaged by P. diospyrosi than the upper parts of canopy.

The Components of the Sap from Gorosoe(Acer mono Max.) and Sugar Maple(Pseudo-sieboldianum Kom.) (고로쇠나무 및 당단풍나무 수액의 성분조성)

  • Even though the saps have been consumed as beverage in Korea for a very long period of time, a little research has been conducted on the chemical composition. We determined mineral, free sugar, composition amino acids and nucleotides in the sap of Gorosoe(Acer mono Max.) and Sugar Maple(Pseudo-sieboldianum Kom.) collected from Sancheong, Kurye and Hamyang. The contents of solid, crude protein and ash were 1.1~3.3%, 0.03~0.12% and 0.02~0.05% in the sap of Gorosoe and Sugar Maple, respectively. The detectable nucleotides were CMP, UMP, IMP, GMP, AMP and hypoxanthine. Free sugar that we determined in the sap of Gorosoe and Sugar Maple were sucrose, fructose and glucose, but maltose was not detected. The chief component of saccharides in the sap was sugar and ranged from 7.1 to 36.6g/L. Especially, the sucrose contents in the sap of Gorosoe and Sugar Maple from Hamyang were higher 3.7 and 2.2 times than those from the other samples. The prominent minerals in the sap were calcium and potassium. The calcium concentration was ranged from 99.0~153.3mg/L in the sap of Gorosoe and was ranged from 21.2~32.4mg/L in that of sugar maple. The sap of Gorosoe and Sugar Maple were composed of 18 and 15 kinds of amino acid respectively, and the total contents of amino acid were in the range of 0.7~29.3mg%. The major amino acids in the sap of Gorosoe were taurine, serine, glutamic acid and aspartic acid in decreasing order. Taurine contents in the sap of Gorosoe and Sugar Maple was found to be the dominant amino acid.

Nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice (갈근 및 녹차추출물의 아질산염 소거작용 및 간독성 보호효과)

  • This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.
