FtsZ, a tubulin homologue, is an essential protein of the Z-ring assembly in bacterial cell division. It consists of two domains, the N-terminal and C-terminal core domains, and has a conserved C-terminal tail region. Lateral interactions between FtsZ protofilaments and several Z-ring associated proteins (Zaps) are necessary for modulating Z-ring formation. ZapD, one of the positive regulators of Z-ring assembly, directly binds to the C-terminal tail of FtsZ and promotes stable Z-ring formation during cytokinesis. To gain structural and functional insights into how ZapD interacts with the C-terminal tail of FtsZ, we solved two crystal structures of ZapD proteins from Salmonella typhimurium (StZapD) and Escherichia coli (EcZapD) at a 2.6 and $3.1{\AA}$ resolution, respectively. Several conserved residues are clustered on the concave sides of the StZapD and EcZapD dimers, the suggested FtsZ binding site. Modeled structures of EcZapD-EcFtsZ and subsequent binding studies using bio-layer interferometry also identified the EcFtsZ binding site on EcZapD. The structural insights and the results of bio-layer interferometry assays suggest that the two FtsZ binding sites of ZapD dimer might be responsible for the binding of ZapD dimer to two protofilaments to hold them together.

The effects of postnatal exposure to octylphenol(OP) on the expressions of the steroidogenic enzymes and testosterone production were evaluated. Postnatal male mice (15-day-old) were injected with 2 or 20mg $kg^{-l}$ body weight (BW) of OP for 5 days and sacrificed on postnatal day 21. Testosterone concentration was measured by radioimmunoassay and the expressions of the testicular genes were determined by RT-PCR analyses. Significant reductions in the mean body and testis weight were observed in the OP treated animals. No marked alteration in the histological structure of the testis were observed, however, slight reduction in the seminiferous tubule diameter and the number of Leydig cells and several pyknotic cells could be identified in the 20 mg $kg^{-l}$ BW of the OP treated animals. Serum testosterone concentration was dramatically reduced and the mRNA expressions of the steroidogenic acute regulatory protein (StAR), cholesterol side-chain cleavage enzyme (P450scc) and $17\beta$-hydroxylase/Cl7-20 lyase $(P450_{17\alpha})$ were decreased. No significant changes of the gene expressions of the steroidogenic factor-l (SF-I) and estrogen and androgen receptor after the OP treatment showed that the decreased expressions of the steroidogenic enzymes in the present study did not correlate with these genes. Altogether, the present study demonstrates that postnatal treatment of OP inhibits steroidogenesis by decreasing the transcriptional expressions of the StAR and steroidogenic enzymes. The alteration in steroidogenesis may adversely affect the normal development of the testis and sper- matogenesis.
Chemical composition and nutritive values of napier grass (Pennisetum purpureum Schum.) silages subjected to two cutting intervals were studies; 1st harvest in July (A), and 2nd (B) and 1st (C) harvests in November. Each forage was ensiled with 4% molasses in plastic bags and stored for 5 or 9 months. A feeding experiment with castrated goats was conducted from April to June of the following year. Dry matter (DM) and crude protein (CP) content of the harvests varied from 9.5 to 22.8% and 6.6 to 13.6% of DM, respectively. The dry matter content of the silages fed to the goats were 13.0 to 24.4%, because some effluent was removed from each silage before the feeding trial. The pH values of the silages were between 4.03 and 4.29. Goats were given sufficient silage to meet maintenance nitrogen requirements from napier grass silage. Silage C was not completely consumed, and the silage had low digestibilities of DM, CP, hemicellulose and cellulose. Nitrogen balance was slightly positive for goats consuming silage B and was negative for goats consuming silages A and C. Nitrogen utilization was discussed in terms of ruminal $NH_3-N$ and volatile fatty acid concentration in the rumen fluids. It is concluded that goats could not maintain N-equilibrium not only when a younger forage was consumed at a level of N requirement by a restricted feeding, but also when an older forage could not be consumed enough for N requirement because of feed intake limitation.

A protease inhibitor from fish eggs was fractionated using chromatographic methods. The fractionation efficiency was evaluated in terms of specific inhibitory activity (SIA, U/mg), purity (fold), total inhibitory activity (TIA, U), and recovery (%). The protease inhibitor (PI) from egg extracts of skipjack tuna (ST Katsuwonus pelamis), yellowfin tuna (YT Thunnus albacares) and Alaska pollock (AP Theragra chalcogramma) was fractionated using Sephadex G-50 gel filtration and DEAE-Sepharose CL-6B anion exchange chromatography based on protein size exclusion and net charge, respectively. Fractions exhibiting strong inhibitory activity were contained in the 30-50 kDa fraction on gel filtration and in the range of 0.4-0.7 M NaCl gradient fraction on anion exchange chromatography. The respective TIA and percent recovery of the fraction obtained with gel filtration toward trypsin and $N{\alpha}$-benzoyl-L-arginine-p-nitroanilide (BAPNA) were 2,758.7 U and 29.6% for ST, 1,005.5 U and 25.6% for YT, and 1,267.5 U and 26.0% for AP. Gel filtration chromatography was more effective at fractionating PI than using ion exchange chromatography. These results suggest that fish eggs act as serine protease inhibitors and might be useful for protease inhibition in foodstuffs.
0.374(1/day) of specific growth rate and 0.435(mg/108 viable cells) of Anti-Microbial factor (AMF) productivity were obseaved for the batch cultivation of human promyelocytic cells in 10% serum containing medium. The crude protein was purified 10 folds by a serial purification steps of ion exchange chromatography, Bio-Rex 70 and gel filtration chromatography, Sephadex G-70 and 100. The ranges of MIC(Minimal Inhibitory Concentration) of commercially available antibiotics, penicillin G, streptomycin and ampicillin was estimated as 40 to ($70\mu\textrm{g}$/ml) on Gram (-) E. coli and Gram (+) Streptococcus aureus. The values of the MBC (Minimal Bactericidal Concentration) of Purified AMF was ($0.5\mu\textrm{g}$/ml) and 0.4($\mu\textrm{g}$/ml), respectively. The molecular weight of the AMF was estimated as 15,000 dalton by SDS-PAGE.
To investigate effects of Paeonia Radix Rubra aqua-acupuncture on Adjuvant Athritis in rats, the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum were measured and histological test of the muscular tissue were done in the arthritis part. 1. After arthritis of Sprague dawley(SD) rats was induced by injecting Freund's complete adjuvant for 2 weeks, normal saline was injected for the Control group and Paeonia Radix Rubra acua-acupuncture was injected for the Exp. group during 30 days. Selected point was $D{\acute{u}}bf(ST_{35})$ in both groups. And then the inframmator volume of plantar were checked. The volume of the plantar was $0.84{\pm}0.14mm$ in Control group and $0.57{\pm}0.09mm$ in Exp. group, the swelling of plantar was restricted significanily in Exp, group(P<0.05). 2. The number of WBC was $10.34{\pm}0.14(10^3/ml)$ in normal group and $37.47{\pm}5.46(10^3/ml)$ in Control group. It was $25.21{\pm}3.45(10^3/ml)$ in Exp. group. This fact showed that Exp group. with Paeonia Radix Rubra acua-acupuncture was more effective than Control group in the treatment of arthritis(P<0.05). 3. The content of total protein in the blood serum were $6.14{\pm}0.43g/d{\ell}$ in normal group, $7.95{\pm}0.94g/d{\ell}$ in Control group, and $6.82{\pm}1.23g/d{\ell}$ in Exp. group. There was no significance in total protein between Exp. group and Control group from the statistical analysis. 4. The contents of albumin in the blood serum were $2.94{\pm}0.13g/d{\ell}$ in normal group, $2.01{\pm}0.48g/d{\ell}$ in Control group and $2.75{\pm}0.14g/d{\ell}$ in Exp, group. This fact showed that Exp. group had significant increase in the serum albumin from the statistical analysis, in comparison with Control group(P<0.05). 5. The contents of globulin in the blood serum were $3.19{\pm}0.48g/d{\ell}$ in normal group, $4.70{\pm}1.26g/d{\ell}$ in Control group and $3.58{\pm}0.73g/d{\ell}$ in Exp. group. There was no significance in the serum globulin between Exp. group and Control group from the statistical analysis. 6. In histological examinadon, because inflammatory reaction was active, remarkably irregular tissue and large amount of inflammatory cells were found in Control. group. But Exp. group showed that tissues were in some degree recovered, the number of inflamed cells was decreased and inflammation was restrained. From these results, it is shown Paeonia Radix Rubra aqua-acupuncture restrained inflammatory reaction and muscular tissue necrosis in SD rat's plantar was restrained by Freund's complete adjuvant.
Objectives : To investigate effects of Achyranthis Radix aqua-acupuncture on adjuvant arthritis in rats. Methods : After eliciting arthritis in Sprague dawley(SD) rats by injection of Freund's complete adjuvant for 2 weeks, normal saline was injected for the Exp. I group and Achyranthis Radix aqua-acupuncture was injected for the Exp. II group during 30days. The selected point was Dokbi (ST35) in both groups. Then the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum, and histological test of the muscular tissue were measured in the arthritis stricken areas. Results : I) The volume of the paws were checked. The volume of the paws was $0.84{\pm}0.l4mm$ in the Exp. I group and $0.47{\pm}0.14mm$ in the Exp. IIgroup. The swelling of the paws was restricted significantly in the Exp. II group(P<0.05). 2) The number of WBC was $10.34{\pm}0.14(l03/ml)$ in the normal group and $37.47{\pm}5.46(103/ml)$ in the Exp. I group. It was $21.24{\pm}2.58(103/ml)$ in the Exp. II group. This fact showed that treatment with Achyranthis Radix aqua-acupuncture adminstered to the Exp. II group was more effective than the Exp. I group in the treatment of arthritis(P<0.05). 3) The content of the total protein in the blood serum was $6.14{\pm}43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.41{\pm}0.68g/dl$ in the Exp. II group. There was no significance in total protein between the Exp. II group and the Exp. I group from the statistical analysis. 4) The content of albumin in the blood serum was $2.94{\pm}0.l3g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in the Exp. I group and $3.15{\pm}0.27g/dl$ in the Exp. II group, which revealed a significant increase in the serum albumin from the statistical analysis of the Exp. II group compared with the Exp. I group. 5) The content of the globulin in the blood serum was $3.19{\pm}$0.48g/dl in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group and $3.26{\pm}0.57g/dl$ in the Exp. II group. There was no significance in the serum globulin between the Exp. II group and Exp. I group from the statistical analysis. 6) In histological findings, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed a small amount of inflammatory cells, the refrained inflammatory state and even recovering state. Conclusions : These results show that Achyranthis Radix aqua-acupuncture reduces the inflammatory reaction and muscular tissue necrosis in SD rat's paws induced by Freund's complete adjuvant.

Purpose : Since standard solution is the one that knows its exact concentration, the curve of the dissolution has been determined according to the amount of the solution, compared to the amount of the unknown sample. Therefore, the antigen that makes up standard materials should be made in a pure form. The configuration of the standard substance solution in the kit we use is a freeze-dried material, or made and comes as a liquid. Lyophilized reference material is used after dissolving in usually D.W. (Distilled Water), and if the antigen to use is too sensitive, reagents should be freeze-dried. Furthermore, when freeze-dried reference has to be frozen again after being dissolved, it should be kept under $-20^{\circ}C$ until the expiration date according to the reports. Since it is not expressed in the experiment if it is safe or stable to reuse the solution which was dissolved a few times, thus, this time it is tested and evaluated that the changes of the standard solution by freezing and melting several times, and its results and the effectiveness of it were compared to the solution which was kept in a fridge. Materials and Methods : Among Vitro diagnostic kits on the market made by radioimmunoassay, parathyroid hormone (PTH), adrenocorticotropic hormone (ACTH), luteinizing hormone (LH) are made of freeze-dried standard solution and all composed of the same Lot.NO. These hormones melted in D.W. and were separated into three groups. In the first group, melting and freezing were repeated, and in the second group, The solution only for one time use was put into a test tube after melting and freeze it. The third group was kept in the refrigerator. This experiment has been conducted from January to February in 2012. January to 2012. PH test was employed because ph is prone to changing depending on the change of protein. Each group of the standard solution, cpm (counter per minute), and the patient relative concentration values were compared by date, and Through the correlation coefficient and Paired t-test, the significant level of each group was analyzed. Results : ACTH, PTH, LH pH values were too subtle denaturation rather than numerical changes in the protein. In addition, when the standard solution of ACTH, PTH, LH was refrigerated, after 3 days and 7 days, there was a significant difference observed between the solution being kept in a refrigerator and a freezer within a significance level. Conclusion : Standard solution should be kept in a freezer, and being kept in a fridge, it is recommended to use the solution as soon as possible.
We investigated effects of a topical treatment of methoprene(0.5-5.0$\mul$/egg), a juvenLle horm mane analogue, on embryo development in the gypsy moth, Lymantri$\alpha$ dispar. Methoprene lowered egg hatching rate, and also reduced the mean wet weights of hatched 1st instar larvae w with the most effect shown at the highest concentration. The differences in protein(p < 0.01) and carbohydrate(p < 0.05) contents between control and methoprene(5$\mul$/ egg) treatment g groups were observed during embryo development.
Purpose : Urinary tract infection (UTI) in children is the most common disease during the infantile period, therefore early diagnosis and treatment are important. Pyuria is a useful clinical parameter for the initial diagnosis of a UTI. In this study we aimed to compare the clinical, laboratory, and imaging findings in relation to the duration of pyuria in infants with UTIs. Methods : Three hundred seventy-four infants <12 months of age who were admitted between January 1995 and December 2005 for the first episode of a febrile UTI were retrospectively reviewed. Patients were divided into two groups according to the duration of pyuria as follows: group 1, pyuria resolved <3 days after initial treatment; and group 2, pyuria lasted at least 3 days after initial treatment. Results : There were no significant differences between the two groups in relation to gender, age, total duration of fever, and organisms in the urine. Group 2 had a significantly higher peripheral blood leukocyte count ($14,360.86{\pm}5,526.16cells/mm^3$ vs. $11,822.55{\pm}5,687.26cells/mm^3$, P <0.001), erythrocyte sedimentation rate ($32.81{\pm}19.34mm/hr$ vs. $23.74{\pm}20.43mm/hr$, P <0.001), and C-reactive protein ($6.84{\pm}5.68mg/dL$ vs. $3.78{\pm}3.99mg/dL$, P <0.001) than group 1. There was a significantly higher incidence of hydronephrosis and a higher grade of vesicoureteral reflux (VUR) in group 2 compared to group 1. Conclusion : In infants with UTI, pyuria of longer duration is related to severe UTI and higher grade VUR, therefore aggressive radiologic studies may be necessary.

제 19 조 (관할 법원)
