The agronomic traits, such as days to flowering and maturity, plant height, 100-seed weight and seed filling period, are quantitatively inherited and important characters in soybean(Glycine max L. Merr.). A total of 126 $F_5$ recombinant inbred lines(RILs) developed from the cross of PI 171451$\times$Hwaeomputkong were used to identify quantitative trait loci(QTLs) for days to flowering(FD), days to maturity(MD), plant height(PH), 100-seed weight(SW), number of branches(NB) and seed filling period(FP). A total of 136 simple sequence repeat(SSR) markers segregated in a RIL population were distributed over 20 linkage groups(LGs), covering 1073.9 cM of the soybean genome with the average distance between adjacent markers of 7.9 cM. Five independent QTLs were identified for FD, three for MD, two for PH, three for SW, one for NB and one for FP. Of these, three QTLs were related to more than two traits of FD, MD, PH, NB and FP and mapped near the same positions on LGs H and O. Thus, these traits could be correlated with biologically controlled major QTLs in this soybean RIL population.
Halenia coreana is an endangered, endemic species that is distributed in only a few locations in Korea, such as Mts. Hwaaksan and Daeamsan. It has been recently segregated from H. corniculata, broadly distributed in cold temperate regions that include northern Japan, the Russian Far East, northeastern China, Mongolia, and eastern Europe, where population sizes are usually large. To examine the genetic diversity of H. coreana and evaluate the level of genetic differentiation of the species compared with that of H. corniculata, we surveyed 183 candidate simple sequence repeats (SSR) motif markers for H. coreana and H. corniculata from sequence data of amplified fragments of a specific length in the genome. A total of 17 genomic-SSR markers were selected to examine the levels of genetic diversity and differentiation using 17 samples of H. coreana and 60 samples of three populations of H. corniculata. The results here suggest that the genetic diversity of H. coreana is very low with a high frequency of inbreeding within its population. We found that H. coreana is genetically differentiated from H. corniculata, supporting the recognition of the geographically isolated H. coreana as a distinct species.
본 연구는 총 50개의 SSR 마커를 이용하여, 찰옥수수 및 종실용 옥수수 핵심집단(찰옥수수 40계통, 종실용 옥수수 40계통)의 자식계통들의 유전적 다양성, 집단구조 및 계통유연관계를 분석하였다. 1. 그 결과 65bp에서 225bp 크기의 범위로 총 242개의 대립단편들을 증폭시켰다. SSR primer들에서 증폭된 대립단편의 수는 최소 2개에서부터 최대 9개까지의 범위로 나타났고, 평균 4.84개가 증폭되었다. 그리고 GD값은 0.420에서 0.854의 범위로 나타났고, 평균 0.654의 값을 나타내었다. 2. 80개의 옥수수 자식계통들의 집단구조를 분석한 결과, 13개의 찰옥수수 자식계통은 group I에 포함되었고, Group II는 7개의 찰옥수수 자식계통과 38개의 종실용 옥수수 자식계통들이 포함되었다. 나머지 22개의 자식계통들은 admixed group에 포함되었으며, 20개 찰옥수수 자식계통과 2개의 종실용 옥수수 자식계통으로 구성되어있다. 3. UPGMA법에 의한 계통유연관계 분석 결과, 80개 옥수수 자식계통들은 유전적 유사성 31.7% 수준에서 크게 3개의 그룹으로 나누어졌다. Group I은 40개의 찰옥수수 자식계통과 11개의 종실용 옥수수 자식계통을 포함하고 있었고, Group II는 27개의 종실용 옥수수 자식계통을, 그리고 Group III은 단지 2개의 종실용 옥수수 자식계통을 포함하고 있었다. 따라서 본 연구의 결과는 앞으로 강원도 농업기술원 옥수수시험장에서 육성한 찰옥수수 및 종실용 옥수수 자식계통들에 대한 유전자원 관리 및 선발 그리고 교배조합 구성 및 예측 등에 유용한 정보를 제공할 것으로 기대된다.
소나무의 2년생 인공교배(人工交配) 전형매차대목(全兄妹次代木) 114개체를 대상으로 화분친(花粉親)이 아닌 개체의 화분에 의해 생성된 차대목(次代木)을 식별하기 위하여 부계유전(父系遺傳)되는 반수체(半數體) 표지자인 cpSSR 표지자 분석을 실시하였다. 3개의 cpSSR primer를 이용한 PCR 분석을 통하여 화분친(花粉親)과 3개 모수(母樹)의 haplotype 조합을 결정하고, 이에 의해 각 개체의 DNA 지문이 확인되었다. 동일한 cpSSR primer를 사용하여 전형매차대(全兄妹次代) 114개 개체목의 haplotype을 확인하고 이를 화분친(花粉親) 및 3개 모수(母樹)에서 확인된 haplotype 조합과 비교한 결과, 이들 중 14개체에서 인공교배(人工交配) 화분친(花粉親)과 다른 cpDNA haplotype이 확인되어 이들이 교배에 사용된 화분친(花粉親)이 아닌 타개체로부터 유입된 화분에 의해서 생성된 개체로 동정되었다. 특히, 강원30으로부터 생산된 차대(次代) 중 한 개체목은 불완전한 제웅(除雄)이나 인공교배(人工交配)시 모수(母樹)에서 생산된 화분의 유입으로 인해서 야기된 자가교배(自家交配)에 의해서 생성되었을 가능성이 매우 높은 것으로 나타났다. 본 연구에서 분석된 cpSSR 지문분석은 향후 자연림내 친계차대목(親系次代木) 감별과 삽목, 접목 및 조직배양에 의한 무성번식묘(無性繁殖苗)의 동정, 순수(純粹) 전형매차대(全兄妹次代)의 확인 등 식물법의학적(植物法醫學的) 분석법(分析法)에 유용하게 활용될 수 있을 것으로 기대된다.
국내 5개 지역에서 채집한 비자나무(Torreya nucifera Siev. et Zucc.) 95개체를 대상으로 I-SSR 표지자를 분석하였다. 총 62개의 I-SSR 증폭산물(增幅産物)이 관찰되었으며, 그 중 7개의 증폭산물(增幅産物)은 분석된 95개 개체에서 단형성(單形性)이었다. 관찰된 전체 I-SSR 증폭산물(增幅産物)을 통합(統合)하여 분석한 결과 개체목에 대한 DNA지방판별(指放判別)이 가능하였다. 대부분의 유전다양성(遺傳多樣性)이 임분(林分)내의 개체목 간에 존재하는 것으로 나타났고(90.65%), 전체 5개 임분(林分)에서 유사한 수준의 유전다양성(遺傳多樣性)을 보였다. 집단간의 유전적(遺傳的) 분화(分化)정도는 심하지 않았다(${\phi}_{ST}=9.35%$). UPGMA법에 의한 유집분석(類集分析) 결과 각 집단의 유전적(遺傳的) 유연관계(類緣關係)는 임분(林分)의 지리적(地理的) 분포양상(分布樣相)과 일치(一致)하지 않았으며, 각 교점(交點)의 형성(形成)에 있어서 통계적 유의성이 없었고 따라서 전체 집단들이 유전적(遺傳的)으로 크게 분화(分化)되지 않았음을 알 수 있었다.

The production of spring wheat, the major crop in Mongolia, is accounting for 98% of the cultivated area. Collection, conservation and utilization of wheat germplasm resources play an important role in wheat breeding and production in Mongolia. Understanding genetic variability in the existing genebank accessions is important for collection and conservation of wheat germplasms. To determine the genetic diversity and population structure among a representative collection of Mongolian local wheat cultivars and lines, 200 wheat accessions were analyzed with 15 SSR markers distributed throughout the wheat genome. A total of 85 alleles were detected, with 3 to 5 alleles per locus and a mean genetic diversity value of 5.66. The average genetic diversity index was 0.68, with values ranging from 0.37 to 0.80. The 200 Mongolian wheat accessions were divided into two subgroups based on STRUCTURE, un-rooted NJ cluster and principal coordinate analyses. The results from this study will provide important information for future wheat germplasm conservation and improvement programs with Mongolian genebank.

콩에서 경장과 연관된 DNA 표지인자를 개발하여 품종육성에 활용함으로서 육종효율 증진에 기여하고자 수행하였다. 본 시험은 육성된 큰올콩과 신팔달콩의 RIL 계통 및 SSR marker를 이용하여 유전자지도를 작성하고, 이를 바탕으로 경장과 관련된 양적형질 유전자좌(QTt)를 탐색하였다. 시험재료로 이용된 큰올콩과 신팔달콩은 경장이 각각 30.57 cm와 49.75 cm로 매우 큰 차이를 보였다. 경장과 연관된 QTL은 개별마커들과의 분산분석 결과, 연관군 F, J, N 및 O에서 전체변이의 37.83%를 설명할 수 있는 4개의 QTL을 탐색하였다. 특히, 연관군 J와 O에서 각각 14.25%와 10.68%를 설명할 수 있는 주요 QTL을 확인하였다. 따라서 경장 관련 QTL중 연관군 J와 O에서 확인된 주요 QTL은 품종 육성과정에서 경장 관련 선발 마커로서 활용가치가 높은 것으로 판단된다.
Agaricus bisporus, commonly known as the button mushroom, is the most widely cultivated species of edible fungi. Low frequency of recombination ratio and homokaryotic or monokaryotic spore on meiotic basidia form obstacles for breeding programs. Since the first hybrid varieties for white button mushrooms were released in Europe, new varieties released afterwards were either identical of very similar to these first hybrids on morphologies. Therefore, different DNA markers have been used to define unique varieties of A. bisporus strains. Aim of this study is to assess the genetic diversity of different A. bisporus strains in Korea. Twelve UFP (Universal fungal primer, JK BioTech. Ltd), 12 simple sequence repeat (ISSR) and 30 SSR primers were used to assess genetic diversity of monokaryotic and dikaryotic Agaricus bisporus strains including other 19 Agaricus spp. Of them, four UFP, four SSR primers, $(GA)_8T$, $(AG)_8YC$, $(GA)_8C$ and $(CTC)_6$ and seven SSR markers produced PCR polymorphic bands between the Agaricus species or within A. bisporus strains. PCR polymorphic bands were inputted for UPGMA cluster analysis. Forty five strains of A. bisporus are genetically clustered into 6 groups, showing coefficient similarity from 0.75 to 0.9 among them. In addition, genetic variations of monokaryotic and dikaryotic Agaricus bisporus strains were partially detected by PCR technologies of this study. The varieties, Saea, saedo, Saejeong and Saeyeon that have recently been developed in Korea were involved in the same group with closely genetic relationship of coefficient similarity over 0.96, whereas, other strains were genetically related to A. bisporus strains that were introduced from USA, Eroupe and Chinese.
Understanding the genetic variation among landrace collections is important for crop improvement and utilization of valuable genetic resources. The present study was carried out to analyse the genetic diversity and associated population structure of 621 foxtail millet accessions of Korean landraces using 22 EST-SSR markers. A total of 121 alleles were detected from all accessions with an average of 5.5 alleles per microsatellite locus. The average values of gene diversity, polymorphism information content, and expected heterozygosity were 0.518, 0.594, and 0.034, respectively. Following the unweighted neighbor-joining method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 3 clusters, and population structure analysis also separated into 3 populations. Principal coordinate analysis (PCoA) explained a variation of 13.88% and 10.99% by first and second coordinates, respectively. However, in PCoA analysis, clear population-level clusters could not be found. This pattern of distribution might be the result of gene flow via germplasm exchanges in nearby regions. The results indicate that these Korean landraces of foxtail millet exhibit a moderate level of diversity. This study demonstrated that molecular marker strategies could contribute to a better understanding of the genetic structure in foxtail millet germplasm, and provides potentially useful information for developing conservation and breeding strategies.
Little millet (Panicum sumatrense) is well known for its salt and drought stress tolerance and high nutritional value, but very limited knowledge of genetic variation and genomic information is available. In this study, a total of 779 primer pairs were designed from the 22,961 EST sequences of switchgrass (Pancium virgatum), of which 48 EST-SSR markers were developed based on the trials of transferability of these primers in little millet. The EST-SSR amplicons showed reproducible single band polymorphism and produced a total of 160 alleles with an average of 3.3 alleles per locus in 37 accessions of little millet. The average values of expected and observed heterozygosities were 0.266 and 0.123, respectively. The polymorphic information content (PIC) values were observed in range of 0.026 to 0.549 with an average of 0.240. The genetic relatedness among the little millet accessions was evaluated by neighbor-joining dendrogram, which grouped all accessions into two distinct groups. The validation thus demonstrated the utility of the switchgrass EST-SSR markers in assessing genomic relationships in little millet. The findings from this study could be useful for designing strategies for the identification of diverse germplasm for conservation and future molecular breeding programs for little millet.
