S. platensis로부터 phycobiliprotein의 분리에 관한 연구 (Studies on the Isolation of Phycobiliprotein from S. platensis)

  • S. platensis로부터 phycobiliprotein을 $30-60{\%}$ 황산암모늄 분별침전법과 Sephadex G-100 gel filtration 및 DEAE-Sephacel anion exchange chromatography의 순서로 4.23의 정제도로 분리하였다. 분리된 phycobiliprotein은 최대흡수 파장이 620 nm인 c-phycocyanin이었다. 분리된 phycobiliprotein은 SDS-PAGE상에서 $({\alpha}$$({\beta}$의 두개의 소단위체로 구성되어 있었으며. $({\alpha}$$({\beta}$ 소단위체의 분자량은 각각 14.5 kDa과 16 kDa 부근이었다. 또한 gel filtration을 사용한 변성 전 분자량은 약 100 kDa이었다. 이러한 결과는 본 연구에서 분리한 phycobiliprotein이 $({\alpha}{\beta})_{3}-trimer$로 이루어졌음을 보여준다.

  • 본 논문에서는 Digital PFC Controller에 적합한 11비트 100KS/s의 Algorithmic ADC를 설계하였다. 설계한 Algorithmic ADC는 PFC controller에 적합한 11비트 해상도를 만족하면서 반복적인 순환구조의 동작으로 인해 전체 크기를 줄일 뿐 아니라 소비 전류를 최소화 할 수 있다. 본 논문의 Algorithmic ADC는 0.18um 1Poly-3Metal의 CMOS 공정으로 제작 되었으며 100KS/s의 동작 속도에 SNDR 66.7dB, ENOB 10.78비트의 성능을 가진다. 또한 소비전류는 5V 전원 전압에서 780uA이며 설계된 ADC의 칩 면적은 $0.27mm^2$이다.

자하거약침(紫河車藥鍼)의 Bell's palsy에 대한 치험보고(治驗報告) (The Effect of Hominis Placenta Herbal Acupuncture on Bell's palsy)

  • This report was done to observe the effect of Hominis placenta herbal acupuncture on Bell's palsy. The study group comprised 16 patients who arrived at Woo-suk university oriental hospital from January, 1999 till January, 2000 for Bell's palsy. All patients were divided into two group. One was herbal acupunture group, and the other was control group. Acupunture group was done herbal acupuncture therapy on the facial acupuncture points. Followings are achievement and a term of each group. In herbal acupuncture group, 100% motor recovery was 7 case, 75% was 1 case, and 25% motor recovery term was $7.38{\pm}5.21$ days, 50% was $11.00{\pm}6.16$ days, 75% was $15.13{\pm}9.55$ days, 100% was $23.14{\pm}7.97$ days. In control group, 100% motor recovery was 4 case, 75% was 2 case, 25% below was 2 case and 25% motor recovery term was $11.17{\pm}4.96$days, 50% was $18.17{\pm}6.82$ days, 75% was $29.50{\pm}6.95$ days, 100% was $44.00{\pm}11.49$ days. The above results indicate that Hominis placenta herbal acupuncture is a useful effect on Bell's palsy. thus, continuous herbal acupunture study will be needed for more clinical application on Bell' palsy.

한국인 모유의 수유기간별 비중, 충고형분 및 단백질 함량의 변화 (Changes in Specific Gravity, Total Solid and Protein Contents of Human Milk During the Course of Lactation in Korean Women)

  • 수유기간의 경과에 따른 모유중의 비중, 총고형분, 단백질 함량 변화를 살펴 보고자 수유부 27명을 대상으로 분만후 15일부터 150일까지 추적하여 채취한 총112개 모유시료로 부터 얻은 결과는 다음과 같다. 즉 모유 비중의 경시적인 변화는 15일경에 1.0298(S.D. 0.0044, n=25), 30일경에 1.0304(S.D. 0.0046, n=24), 60일경에 1.0270(S.D. 0.0035, n=20) 90일경에 1.0293(S.D. 0.0055, n=18), 120일경에 1.0254(S.D. 0.0038, n=16), 150일경에 1.0268(S.D. 0.0039, n=9)로서 나타났고 수유기간이 경과함에 따라 유의하게 감소하는 경향을 보였다. 총고형분은 모유 100$m\ell$중에 15일 경에 12.48g(S.D. 1.23, n=25), 30일경에 12.31g(S.D. 1.31, n=24) 60일경에 12.47g (S.D. 1.45, n=20), 90일경에 11.81g (S.D. 1.85, n=18), 120일경에 11.44g(S.D. 0.92, n=16) 150일경에 11.88 (S.D. 0.89, n=9)으로 나타났고 수유기간의 경과에 따른 유의한 차이는 보이지 않았다. 모유 100$m\ell$중 단백질함량은 1.07~1.46g의 범위였고, 수유기간별 변화는 15일경에 1.46g (S.D. 0.20, n=25), 30일 경에 1.32g (S.D. 0.19, n=24) 60일경에 1.15g (S.D. 0.10, n=20), 90일경에 1.09g (S.D. 0.17, n=18), 120일경에 1.11g (S.D. 0.17, n=16), 150일 경에 1.07g (S.D. 0.18, n=9)으로 수유기간이 경과 함에 따라 유의하게 감소하는 경향을 보였다. 또한 수유기간 15, 30, 50, 90, 120, 150일에 수집한 모유시료 전체의 총평균 단백질함량은 1.20g (S.D. 0.14, n=27, 총시료수 112)로 나타났다.

Bioscrubber와 Biofilter 병합 시스템에 의한 H2S 제거특성 ([ H2S ] Removal Characteristics by Combined System of Bioscrubber and Biofilter)

  • This study compared the performance of a bioscrubber, a biofilter, and a combined system of bioscrubber and biofilter employed being operated at the laboratory-scale. for the removal of hydrogen sulfide. The bioscrubber maintained 100% removal of hydrogen sulfide up to inlet load of $56\;g-S/m^3{\cdot}hr$, while the removal efficiency was decreased with the increase of inlet load. The biofilter showed 100% removal efficiency up to inlet load of $126\;g-S/m^3{\cdot}h$ and the maximum elimination capacity of $126\;g\;S/m^3{\cdot}h$ for the inlet load of $224\;g-S/m^3{\cdot}h$. On the other hand, the combined system of bioscrubber and biofilter showed 100% removal for an inlet hydrogen sulfide load of up to $85\;g-S/m^3{\cdot}h$ and the maximum elimination capacity of $153\;g-S/m^3{\cdot}h$ for inlet loads of $224\;g-S/m^3{\cdot}h$.

인체 S100A2 단백질에 특이적인 단일클론 항체 (Characterization of the Monoclonal Antibody Specific to Human S100A2 Protein)

  • Background: The S100A2 gene, also known as S100L or CaN19, encodes a protein comprised of 99-amino acids, is a member of the calcium-binding proteins of EF-hand family. According to a recent study, this gene was over-expressed in several early and malignant carcinomas compared to normal tissues. To elucidate the role of S100A2 protein in the process during carcinogenesis, production of monoclonal antibody specific to the protein is essential. Methods: First, cDNA sequence coding for ORF region of human S100A2 gene was amplified and cloned into an expression vector to produce GST fusion protein. Recombinant S100A2 protein and subsequently, monoclonal antibody to the protein were produced. The specificity of anti-S100A2 monoclonal antibody was confirmed by immunoblot analysis of cross reactivity to other recombinant proteins of S100A family (GST-S100A1, GST-S100A4 and GST-S100A6). To confirm the relation of S100A2 to cervical carcinogenesis, S100A2 protein in early cervical carcinoma tissue was immunostained using the monoclonal antibody. Results: GST-S100A2 recombinant protein was purified by affinity chromatography and then fusion protein was cleaved and S100A2 protein was isolated. The monoclonal antibody (KK0723; Korean patent pending #2001-30294) to the protein was produced and the antibody did not react with other members of EF-hand family proteins such as S100A1, S100A4 and S100A6. Conclusion: These data suggest that anti-S100A2 monoclonal antibody produced in this study can be very useful for the early detection of cervical carcinoma and elucidation of mechanism during the early cervical carcinogenesis.

Application of rDNA-PCR Amplification and DGGE Fingerprinting for Detection of Microbial Diversity in a Malaysian Crude Oil

  • Two culture-independent methods, namely ribosomal DNA libraries and denaturing gradient gel electrophoresis (DGGE), were adopted to examine the microbial community of a Malaysian light crude oil. In this study, both 16S and 18S rDNAs were PCR-amplified from bulk DNA of crude oil samples, cloned, and sequenced. Analyses of restriction fragment length polymorphism (RFLP) and phylogenetics clustered the 16S and 18S rDNA sequences into seven and six groups, respectively. The ribosomal DNA sequences obtained showed sequence similarity between 90 to 100% to those available in the GenBank database. The closest relatives documented for the 16S rDNAs include member species of Thermoincola and Rhodopseudomonas, whereas the closest fungal relatives include Acremonium, Ceriporiopsis, Xeromyces, Lecythophora, and Candida. Others were affiliated to uncultured bacteria and uncultured ascomycete. The 16S rDNA library demonstrated predomination by a single uncultured bacterial type by >80% relative abundance. The predomination was confirmed by DGGE analysis.

  • A total of 220 adult male mice (18-20g) of the S.M. strain were divided into ten experimental and control groups. The total-body X-ray irradiation doses used were 50 r, 100r, 200r, 400r, 600r, 800r, 1,000r, 1,200r, 1,400r, and1,600r. The respiratory arrest (mortality) caused by each irradiation doses were observed for 30 days. Relationships between irradiation doses and survival time and percentage of response were examined. 맥주오염미생물의 동정과 specific PCR primer의한 신속한 검출 방법 (Characterization of beer-spoilage microorganism and its rapid detection by specific PCR primer)

  • Several contaminated bacteria such as Lactobacillus brevis and Pediococcus damnosus in beer production cause beer spoilage by producing off flavours and turbidity. Detection of these organisms is complicated by the strict anaerobic conditions and lengthy incubation times required for their cultivation, consequently there is a need for more rapid detection methods. Recently, two contaminated strains were isolated from vessel of beer production and identified as Lactobacillus species by API kit identificaton as well as 16S-23S ITS sequencing analyses. Two isolated strains were named as Lactobacillus sp. HLA1 and Lactobacillus HLB2, respectively. A polymerase chain reaction (PCR) method was developed for the rapid and specific detection of Lactobacillus sp.. Two sets of primer pairs (HLA1-F/HLA1-R and HLB2-F/HLB2-R) were designed for the amplification of a 1576 base pair (bp) fragment of the HLA1 16S-23S rRNA gene and 1888 bp fragement of the HLB2 16S-23S rRNA. Amplified PCR products were highly specific to detect corresponding bacteria when other contaminated strains were used as PCR templates. However, detection of both strains were limited when $100{\mu}{\ell}$ of cultured samples were mixed with $100m{\ell}$ of beer sample in arbitrary manner. The sensitivity of the assay still needs to be improved for direct detection of the small amounts of bacteria present in beer.

일부 학교 건축물의 석면함유 건축자재(ACM) 특성과 위해등급에 관한 연구 (A study of asbestos containing material characteristics and grade of risk assessment in schools, Korea)

  • 본 연구의 목적인 우리나라 일부 학교의 석면함유 건축자재을 조사하고 EPA AHERA rule과 ASTM rule을 적용하여 위해성평가를 실시하였다. 총 100개 학교를 2010년 1월부터 12월까지 조사하였다. 건축년도에 따른 ACM 검출율은 1980년대 이전 건축물은 100%, 1990년대 이전은 94.1%, 2000년대 이전은 100%, 2000대 이후는 62.5%로 나타났다. 학교별 ACM 검출율은 유치원과 고등학교는 100%, 초등학교는 97.1%, 중학교는 92.9%, 특수학교는 80%로 나타났다. 천장 텍스는 2~8 %의 백석면과 갈석면, 밤라이트 보드는 6~11 %의 백석면이 검출되었다. 또한 개스킷과 슬레이트 지붕은 각각 16~17 %, 10~13 %의 백석면이 검출되었다. EPA AHERA rule을 이용하여 ACM의 위해성 평가를 실시한 결과, 모든 건축자재는 "Pool" 등급이 나왔으며, ASTM rule 평가에서는 모든 ACM이 "Q&M program"이 필요한 것으로 나타났다.
