Investigation of the Transforming Theories of Diseases and the Need of Teaching Them in the Pathology Class (동의병리학에서의 병리전변이론(病理傳變理論) 고찰 및 교육의 필요성)

  • In order to investigate the meanings and bases of transforming theories of diseases(病傳論), several relevant theories were collected from medical books since . The outside-to-inside changing process(pyoree jeon) was the fundamental pattern in febrile diseases regardless of slang han and wen bing after Han dynasty. But in case of numb disease the three exogenous pathogenic factors of wind, cold and dampness got into each viscera through five tissues like skin, vessesl, flesh, tendons, bones respectively. It was called corresponding changing process(sanghap jeon) here. The pathogenic changing process(byungsa jeon) had complicated details to explain pathologic processes, but could give useful informations on evaluating relative strength of pathogens and tendency ahead. The changing process of diseases of internal injuries were explained in case of emotional distress which is outbroken abruptly didn't follow regular order of viscera-emotion relations. So it was named random changing process(bulcha jeon) after 's usage. And marasmus and asthenic disease followed top-to-down or down-to-top changing process(sangha jeon) based on . There are many types of changing process of diseases between viscera and viscera or viscera and bowels like generation changing process, restriction changing process, changing process between couple, changing process of mutual transmission based on attributions of each element by the theory of 5 phases(五行論). And changing process to a neighbor doesn't have any special relations within two organs but has anatomical contiguity and physiological continuity between them. The transforming theories of diseases bring forth useful understanding on comings and goings of pathogenic factors and tendency and prognosis of disease, so they are needed to be taught in the course of pathology class.

Establishment of a diagnostic method for porcine proliferative enteropathy using polymerase chain reaction (중합효소연쇄반응을 이용한 돼지 증식성 장염 진단기법 확립)

  • Porcine Proliferative Enteropathy(PPE) is an infectious enteric disease and a major cause of economic loss in swine industry due to weight loss, poor growth and sudden death in growing and finishing pigs at 6 to 20 weeks of age. PPE has been diagnosed by clinical signs, syndrom and lesions in the intestine in Korea. However, the diagnostic method had several problems in the detection of infected or carrier pigs. Therefore, in this study, we established the polymerase chain reaction(PCR) which was a fast, specific and sensitive method for identification of Lawsonia intracellularis (L intracellularis). We designed and synthesized primer on the 16S rDNA and p78 gene encoding L intracellularis. Specificity of the method was confirmed by comparison of the PCR results using other enteric bacteria and the study has shown that PCR method was sensitive to detect 1ng of genomic DNA as a template. Identity of the PCR products was confirmed by comparison of pattern of restriction endonuclease analysis with restriction enzyme Hae III and Pst I. Also, the PCR method was applicable to the naturally affected pigs with PPE. Based on the results from this study, the PCR method could be used as a fast and specific diagnostic tool for PPE.

Application of 16S rDNA PCR-RFLP Analysis for the Rapid Identification of Weissella Species (Weissella 속 유산균의 빠른 동정을 위한 16S rDNA PCR-RFLP 분석법의 적용)

  • A polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP) analysis was applied to detect and identify ten Weissella spp. frequently found in kimchi. The previously reported genus-specific primers designed from 16S rDNA sequences of Weissella spp. were adopted but PCR was performed at the increased annealing temperature by $4^{\circ}C$. The sizes of amplified PCR products and restricted fragments produced by AluI, MseI, and BceAI endonucleases were well correspond with the expected sizes. W. kandleri, W. koreensis, W. confusa, W. minor, W. viridescens, W. cibaria, and W. soli were distinguished by AluI and MseI and W. hellenica and W. paramesenteroides were identified by BceAI. W. thailandensis was distinguished when restriction pattern of other species was compared but identified by the single use of MspI.

Bacterial Community Monitoring of Commercial Kimchi Produced in Korea and China with Evidence of Bacilli Spore Formation during Fermentation (한국산 및 중국산 김치의 Bacteria 군집 분석 및 발효과정 중 Bacilli 포자 형성 규명)

  • Terminal Restriction Fragment Length Polymorphism (T-RFLP) analysis was adopted to explore rapid differentiation in the diversity and dynamics of bacteria in kimchi made in Korea and China for future application in kimchi origin discrimination. T-RFLP analysis supported the reproducible and rapid detection of major lactic acid bacteria known to be involved in kimchi fermentation. The taxonomic resolution level of this T-RFLP analysis was between the species and genus level, but was not specific enough for the detection of a bacterium found only in one origin, either Korea or China. The bacterial community structure successions in kimchi samples from Korea and China analyzed by T-RFLP analysis occurred with a similar pattern. Bacillus spp. which were not detected in the early microbial studies of kimchi were constantly detected until the late fermentation stage of kimchi in our T-RFLP analysis and their existence was proved by culture-based identification. Additionally, sporulation of Bacillus spp. during kimchi fermentation was discovered.

A Simple Method for Extraction of High Molecular Weight DNA fromPorphyra Tenera (Rhodophyta) Using Diatomaceous Earth

  • The innate soluble polysaccharides and phenolic compounds of marine macroalgae are serious contaminants which interfere with experimental procedures such as restriction enzyme digestion, polymerase chain reaction (PCR) and other enzymatic reactions using extracted DNA samples. The viscous polysaccharides are co-precipitated with DNA samples by isopropanol or ethanol precipitation in conventional experiment. To overcome the problem, a method for the isolation of high molecular weight DNA from Porphyra tenera is developed with the application of diatomaceous earth column. The isolated DNAs by this method were about 50-100 kb in size and could be digested well with restriction enzymes. The nuclease activity seemed to be minimal, and high reproducibility in the arbitrary primed PCR for RAPD analyses was a distinctive feature. These results suggest that this method is very efficient in isolating nucleic acid from macroalgae including Porphyra.

  • Branching growth pattern of Mugnolza denuduta is likely to be originated from two growth strategies: environment overcoming strategy and life maintenance strategy, which coexist in a tree. Growth rate of branches was strongly correlated with relative light intensity (P<0.001) and physical contact (P<0.01), however there is no significant correlation between growth rate and direction of branch. When relative light intensity is less than 1%, the growth was restricted by physical contact with the surrounding branches. In contrast, the growth was rarely restricted by physical contact when relative light intensitiy was 10% or more. The branching rate was significantly affected by the presence or absence of physical contact (P<0.05), but it was not significantly affected by relative light intensity nor by the direction of branch. In the beginning stage of the growth, the ratio of the material allocation from main branch to subbranch was large and varied with the influence of surrounding environment. These various growth rates, which implicate a variety of material allocation ratios (0.16~0.98), affect branch growth pattern through the optimum growth strategies. The growth and arrangement of branches of Magnolia denudata display the solar collectors to maximize the total amount of energy absorbed and to overcome the restriction of the environment.

Effect of exercise and energy restriction on blood pattern in obese children (운동과 에너지 제한이 비만아동의 혈액 성상에 미치는 영향)

  • The purpose of this study was to examine the effect of exercise and diet control program on blood pattern in obese children. The subjects of this study were five obese boys aged from 8 to 12 participating in the control period(C) for 3 days, exercise period(E) for one week, and exercise+energy restriction period(EER) for another one week, consecutively. The intensity of the exercise was 60~70% of HRmax and the energy was restricted at 493kcal/day. Daily mean total energy intake was 2,152${\pm}$138kcal. 1,861${\pm}$138kca1, and 1,368${\pm}$87kcal for the period C, E and EER, respectively. Body weight of after the program was significantly decreased from 48.94$\pm$5.11kg to 45.94${\pm}$4.74kg(P<0.01). And skinfold thickness. %fat. lean body mass, body mass index were significantly decreased(p<0.01). Blood sugar concentration was not significantly affected by weight loss, but alkaline phosphatase activity was significantly decreased. Concentrations of total lipid, LDL-cholesterol, triglyceride, phospholipid were not significantly decreased. But concentrations of HDL-cholesterol, %HDL-cholesterol, free fatty acid were significantly increased and concentration of VLDL-cholesterol, atherogenic index were significantly decreased. The results of this study showed that the obese children had a tendency to decrease coronary heart disease risk in the respect of plasma HDL-cholesterol and VLDL-cholesterol concentration by the exercise plus energy restriction program. Thus if we apply the lower intensity or duration of exercise for them this program might be more effective on the obese children.

Effects of Dietary Treatment, Gender, and Implantation on Calpain/Calpastatin Activity and Meat Tenderness in Skeletal Muscle of Korean Native Cattle

  • The objectives of this study were to examine calpain activity and meat tenderness by three different feeding patterns in Korean native cattle (KNC). Total forty-five animals were assigned each fifteen in long term restriction feeding (LTFR), long-term restriction feeding and hormone treatment (LTFR-tH), and short term non-restriction feeding (STFNR), respectively. Concentrate was restricted based on body weight in exp 1 and 2. However, it was fed ad libitum in exp. 3. Hormonal implantation was made with $M-PO^{TM}$ for bulls and with $F-TO^{TM}$ for heifers at 18, 20, 22 months of age in exp. 2. Animals were purchased (3-5 month old) from local cattle market and managed in two local farms and university research unit at three different years. Animals were slaughtered at 24 months for long-term trial and at 18 month for short term-trial. Loin and tender loin muscle was used for calpain activity and meat quality. Calpain proteolytic system was not changed by treatment. However, calpastatin activity was low in short-term trial. The calpain and calpastatin activity is reciprocal relationship, therefore, the high calpain activity may effect on quality grade. The shear force value was decreased as the processing of aging after postmortem. On the other hand, the cooking loss was significantly higher in short-term than in long-term trial, and then gradually decreased by the aging. Hormone implants to increase meat yield influenced to calpastatin activity more powerfully than calpain activity to meat tenderness. In meat color-a*, there was not significant difference in loin. Meat color-b* was decreased as postmortem aging time increased in tenderloin. Western blots were done to learn whether these proteins are degraded during postmortem storage and whether this degradation temporally parallels the decrease of shear force value. Vinculin was detected at 0 day and 1 day and degraded after 3 day. In conclusion, Calpain activity was affected slightly on meat tenderness. But meat tenderness was influenced by calpastatin, more effectively.

Effects of Feeding Patterns and Sexes on Growth Rate, Carcass Trait and Grade in Korean Native Cattle

  • The objectives of this study were to examine growth performance and meat quality by three different feeding patterns in Korean native cattle (KNC). In each of 3 years, fifteen KNC were randomly assigned in a (3 feeding management)${\times}$(3 sex) factorial design experiment; thus, in total, there were 5 animals in each of the 9 treatments. The three feeding management treatments were longterm (24 month) restriction feeding (LTFR), long-term restriction feeding-hormone implant (LTFR-tH), and short-term (18 month) nonrestriction feeding (STFNR). Three sexes were bull, steer, and heifer. Concentrate diet was fed restriction-feeding method based on body weight in LTFR and LTFR-tH. However, the diet was fed ad libitum in STFNR. Hormonal implantation was made three times with M-$PO^{TM}$ for bulls and with F-$TO^{TM}$ for heifers at 18, 20, 22 month of age in LTFR-tH. Animal were purchased from the local cattle market and managed in two local farms and at the university research unit. Animals were slaughtered at 24 months for long-term trial and at 18 month for short-term trial. The growth rate was the highest in bulls and the lowest in heifers. However, the differences were diminished in F-$TO^{TM}$ implanted heifers. The average daily gain was high in STFNR due to ad libitum feeding. The carcass grade was similar among the treatments on percentage bases. Hormonal implants improved significantly the meat quality grade in all sexes. Castration increased body fat content and improved meat quality grade by intramuscular fat deposition. In conclusion, long-term feeding and hormone treatment increased meat quality grade more than short-term feeding. However, ADG was higher in the short-term trial although feedefficiency was lower.

Seasonal Differences of Bacterial Communities Associated with the Marine Sponge, Hymeniacidon sinapium (주황해변해면(Hymeniacidon sinapium) 공생세균 군집의 계절적 차이)

  • Seasonal differences of the cultivable bacterial communities associated with the marine sponge, Hymeniacidon sinapium, between spring and summer were analyzed through the Amplified Ribosomal DNA Restriction Analysis (ARDRA). For the cultivation of the bacterial isolates, modified Zobell and MA media were used. The 16S rDNA of individual strains were amplified and fragmented by using two restriction enzymes, HaeIII and MspI. As a result, 23 ARDRA types from the spring sponge and 28 types from the summer sponge were obtained. The partial sequencing result of 1 to 3 selected strains from each types showed over 94% similarities with the known species from the public database. The bacterial communities from the sponge, captured on spring, contained 4 phyla: Actinobacteria, Alphaproteobacteria, Gammaproteobacteria, and Firmicutes. There were 5 phyla observed from the bacterial communities associated with the sponge, captured on summer: Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Firmicutes, and Bacteroidetes. Gammaproteobacteria was predominant group in both spring and summer, accounted for 33.8% of total in spring and 67.4% in summer, showed increase pattern on summer. Because Firmicutes and Actinobacteria participated in 30.2% and 8.3% of the spring sponge while they represented only 6.9% and 0% of the summer sponge, both bacterial groups showed decrease drift on summer. Betaproteobacteria (4.7%) and Bacteroidetes (4.7%) were only observed on the sponge captured on summer. On the sponge, Hymeniacidon sinapium, more diverse bacterial communities were shown on summer than on spring, and even from the same sponge, there were seasonal differences.
