GABAA Receptor- and Non-NMDA Glutamate Receptor-Mediated Actions of Korean Red Ginseng Extract on the Gonadotropin Releasing Hormone Neurons

  • Korean red ginseng (KRG) has been used worldwide as a traditional medicine for the treatment of various reproductive diseases. Gonadotropin releasing hormone (GnRH) neurons are the fundamental regulators of pulsatile release of gonadotropin required for fertility. In this study, an extract of KRG (KRGE) was applied to GnRH neurons to identify the receptors activated by KRGE. The brain slice patch clamp technique in whole cell and perforated patch was used to clarify the effect of KRGE on the membrane currents and membrane potentials of GnRH neurons. Application of KRGE (3 ${\mu}g$/${\mu}L$) under whole cell patch induced remarkable inward currents (56.17${\pm}$7.45 pA, n=25) and depolarization (12.91${\pm}$3.80 mV, n=4) in GnRH neurons under high $Cl^-$ pipette solution condition. These inward currents were not only reproducible, but also concentration dependent. In addition, inward currents and depolarization induced by KRGE persisted in the presence of the voltage gated $Na^+$ channel blocker tetrodotoxin (TTX), suggesting that the responses by KRGE were postsynaptic events. Application of KRGE under the gramicidin perforated patch induced depolarization in the presence of TTX suggesting its physiological significance on GnRH response. Further, the KRGE-induced inward currents were partially blocked by 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX; non-NMDA glutamate receptor antagonist, 10 ${\mu}M$) or picrotoxin (PIC; $GABA_A$ receptor antagonist, 50 ${\mu}M$), and almost blocked by PIC and CNQX mixture. Taken together, these results suggest that KRGE contains ingredients with possible GABA and non-NMDA glutamate receptor mimetic activity, and may play an important role in the endocrine function of reproductive physiology, via activation of $GABA_A$ and non-NMDA glutamate receptors in GnRH neurons.

Comparative Transcriptome Analysis Reveals Differential Response of Phytohormone Biosynthesis Genes in Glumous Flowers of Cold-Tolerant and Cold-Sensitive Rice Varieties Upon Cold Stress at Booting Stage

  • Low temperature stress is one of the major negative factors affecting vegetative and reproductive growth of rice. To better understand responses of rice plants to low temperature we analyzed transcriptome expression patterns in glumous flower of cold-tolerant japonica rice variety, Stejaree45, and cold-susceptible variety, HR19621-AC6 at booting stage under cold water irrigation. A total of 2,411 probes were differentially expressed by low temperature in glumous flowers of the two varieties. Some important genes involved in hormone biosynthesis showed variety-specific regulation. Expression of GA20ox3 and GA2ox, among the genes involved in GA biosynthesis, was regulated differentially in the two varieties. Among the genes involved in IAA biosynthesis, YUCCA1 and TAA1:1 showed variety-specific regulation. Among the genes involved in cytokinin biosynthsis and signaling, expression of LOG, HK1 and HK3 was significantly down-regulated only in the cold-susceptible variety. Among the genes involved in ABA biosynthesis, NSY and AAO3 were down-regulated only in the cold-tolerant variety. In general, genes involved in GA, IAA and cytokinin biosynthesis responded to cold temperature in such a way that capacity of those bioactive hormones is maintained at relatively higher levels under cold temperature in the cold-tolerant variety, which can help minimize cold stress imposed to developing reproductive organs in the cold-tolerant variety.

Protection provided by a commercial modified-live porcine reproductive and respiratory syndrome virus (PRRSV) 1 vaccine (PRRSV1-MLV) against a Japanese PRRSV2 field strain

  • Background: Porcine reproductive and respiratory syndrome virus (PRRSV) vaccines do not provide full cross-protection, mainly due to the virus genetic variability. Despite this, vaccines based on modified-live PRRSV (PRRSV-MLV) reduce the disease impact. Objectives: To assess the efficacy of two commercial vaccines-one based on PRRSV1 (PRRSV1-MLV) and another on PRRSV2 (PRRSV2-MLV)-against a Japanese PRRSV2 field strain. Methods: Two groups of three-week-old piglets were vaccinated (G1: PRRSV1-MLV; G2: PRRSV2-MLV) and two were kept as non-vaccinated (INF and CTRL). One month later, G1, G2, and INF were challenged with a PRRSV2 field strain. Results: After the challenge, clinical signs were only observed in INF. Moreover, the highest rectal temperatures and values for the area under the curve (AUC) were observed in INF. Regarding viral detection, both AUC and the proportion of positive samples in blood were higher in INF. In G1, viremic animals never reached 100%. At necropsy (21 d after the challenge), differences for titers among groups were only found in tonsils (G1 < G2 and INF). One animal (belonging to G1) was negative in all tissues. Regarding humoral responses, G1 and G2 seroconverted after vaccination, as detected in the corresponding enzyme-linked immunosorbent assay. Specific neutralizing antibodies (NA) against PRRSV1-MLV were already detected at 14 d after vaccination in G1, showing a significant booster after the challenge, while PRRSV2-MLV NA were detected in G2 at the end of the experiment. Conclusions: Despite genetic differences, PRRSV1-MLV has been demonstrated to confer partial protection against a Japanese PRRSV2 strain, at least as good as PRRSV2-MLV.

Effects of dietary energy and lysine levels on physiological responses, reproductive performance, blood profiles, and milk composition in primiparous sows

  • The adequate intake of energy and lysine for primiparous sows are necessary for maternal growth of sows and growth of their progeny. This study was conducted to evaluate the effects of dietary energy and lysine levels on primiparous sows and their progeny. A total of 48 gilts (Yorkshire × Landrace), with an initial body weight (BW) of 168.1 ± 9.71 kg and at day 35 of gestation, were allotted to eight treatment groups with a 2 × 4 factorial arrangement. The first factor was metabolizable energy levels in diet (3,265 or 3,365 kcal of ME/kg), and the second factor was lysine levels in diet (gestation 0.55%, 0.65%, 0.75%, 0.85%, lactation 0.70%, 0.85%, 1.00%, 1.15%). The BW gain (p = 0.07) and backfat thickness (p = 0.09) in the gestation period showed a tendency to be increased in sows fed the high-energy diets. In the lactation period, sows fed the high-energy diets tended to be greater BW (p = 0.09) and less BW loss (p = 0.05) than those of sows fed the low-energy diets. Sows fed high-energy diets had a tendency of greater piglet weight at day 21 of lactation and greater piglet weight gain (p = 0.08 and p = 0.08, respectively). Although the blood urea nitrogen (BUN) was increased linearly as dietary lysine level increased at day 110 of gestation (Linear, p = 0.03), the BUN was decreased linearly as dietary lysine level increase at day 21 of lactation (Linear, p < 0.01). In the composition of colostrum, sows fed high-energy diets had greater casein, protein, total solid, solid not fat, and free fatty acid concentrations than those of sows fed low-energy diets (p < 0.05). Supplementation of total lysine 0.75% for gestation and 1.00% for lactation with 3,365 kcal of ME/kg energy level could be applied to the primiparous sows' diet to improve performance of sows and growth of their progeny.

$CO_2$농도와 온도 상승이 멸종위기식물 단양쑥부쟁이의 종생태적 반응에 미치는 영향 (Effect of Elevated $CO_2$ Concentration and Temperature on the Ecological Responses of Aster altaicus var. uchiyamae, Endangered Hydrophyte)

  • 단양쑥부쟁이(Aster altaicus var. uchiyamae)는 한국특산식물이자 환경부 지정 멸종위기식물 II급으로 하천변에 서식하는 식물이다. 본 연구는 지구온난화가 단양쑥부쟁이의 생태적 반응에 미치는 영향을 알아보기 위해 $CO_2$농도와 온도가 상승된 처리구와 대조구에서 종자를 파종하고, 유식물을 키워 발아율, 식물계절학, 영양생장과 생식생장을 관찰하였다. 그 결과, 단양쑥부쟁이의 발아율은 대조구보다 처리구에서 더 낮았다. 처리구에서 살아있는 잎의 지속 시기는 대조구보다 더 길었다. 개화와 꽃의 지속 시기, 종자 성숙 시기는 처리구에서 더 느렸다. 지상부와 지하부 무게는 처리구에서 더 가벼웠다. 개체 당 꽃대 수와 꽃대 당 종자개수는 처리구에서 더 적었다. 개체 당 꽃대무게, 꽃대 당 종자무게 그리고 종자 한 개 무게는 처리구에서 더 가벼웠다. 이상으로 볼때, $CO_2$농도와 온도상승은 단양쑥부쟁이의 생태에 부정적인 영향을 준다.

여성건강을 위한 개념적 모형 (Conceptual Model for Women s Health)

  • There has recently been an increased interest in women's health from, various disciplines, with different perspectives presented according to each profession's academic background. This has led to many instances of incorrectly defining, or misinterpretation, of the issues even among professionals. Nurse scholars as well as practitioners who work in women's health care need to have a clear conceptual understanding of women's health in order to build a body of knowledge, delineate curricular activities, and set directions for professional nursing interventions. In addition, a conceptual model that may be directly utilized in practice is needed to maintain and promote women's health issues. The purpose of this study was to apply a Hybrid model, analyzing conceptual definitions and discussions related to women's health gathered from review of the literature. Further to compare analyticals the concepts and properties observed from field work, so as to present a final definition of women's health and, build a conceptual framework for a united comprehensive perspective on the concept as well as on nursing practice. Data collection and analysis consisted of a theoretical stage, field work stage, and final analysis. A heterogeneous group of professionals and lay persons, 39 in all, participated in the field work. Study findings Include several subconcepts under the concept of women's health : a woman's whole life, holistic health, quality of life, awareness of being a woman, individual nursing, self care ability, reproductive health, and family health. Thus, a comprehensive definition was built, 1. e., "Women's health care be defined as improvement in the quality of life of women through attainment of holistic health throughout the life span. With reproductive health at the core, the concept is directly related to family and national health, and includes taking care of one's own health based on awareness of being a woman and utilizing self care activities. Women's health care issues are unique and allow various responses, therefore women's health professionals need to apply individual approaches to reach solutions in attaining holistic health and improving quality of life." The constructual factors of women's health were found to be reproductive functions, diseases more common in woman, self actualization, mental health, women's health policies, sexuality, midlife changes, and marital relations, with each factor having more than three properties. Positive factors affecting women's health were found to be a normal childbearing process, a healthy lifestyle, active health management, health information, support, and resources, and interpersonal relationships. Negative factors were found to be overwhelming role stress, cultural oppression, gender inequality, distorted sexual identity, economic difficulties, misuse and/or abuse of substances, and stress. The model of women's health may be visualized as a balance scale set upon a woman's life, supporting 4 concentric circles. The innermost circle and second circle incorporate conceptual definitions of women's health, and the outer two circles represent the constructional factors and properties of women's health. Each circle has its own color that symbolizes the conceptual meaning. Positive and negative factors are represented as weights at either end of the scale, and are affected by nursing intervention, i. e., health and wellness increase when positive factors are stronger, whereas disease and illness increase when negative factors are stronger. This model is only a preliminary effort and requires much discussion and testing to be further developed. Continuous research is also required.

기상환경과 병해충 발생 및 그 대책 (Meteorological Condition and Pest Management)

  • The effects of climatic factors on organisms lire variable and complex, and it, however, can be interpreted in terms of those on the distribution and those on the population densities. The distribution of an organism may largely be determined by the temperatures, except some temporal organisms which are depended on the air mass movements. Population density of an organism is determined by various climatic factors, such as previous winter temperature, temperature of growing season and rainfall. The start of growing season of the rice plants has been shifted to earlier since last decade in Korea. This may mean that the overall climatic condition during the growing season might be considerably different from those in past years, and such a difference in climatic conditions might have close relation with the recent status of the diseases and insect pests through direct effects on the physiology and population dynamics of the organisms, as well as through on the biotic associations of the pest organisms. The white back planthopper and brown planthopper have become the key insect pests in Korea in recent years. They are migratory and have high reproductive pontentials and more generations than average residential insects. The synchronization of the migrants and physiological condition of the rice plants seems to be the important factors in relation to the recent outbreaks of these insects; the high reproductive rate can be obtained with the growth stage of rice being 30-50 days after transplanting. The modication of the microclimate associated with high plant density and some other introduced new cultural techniques also have some relation with the outbreak. The key diseases of the rice are the blast disease, sheath blight and the bacterial leaf blight. For the rice blast, the seedling blast and leaf blast during the early growing season and the neck blast, have become more serious, the former may be related to hotbed nursery and the later may be related to the high humidity in early August, and synchronization of the heading time which has been shifted to early part from middle or late part of August. In general, for the rice diseases, the development of the new races have been the most serious which are largely resulted from the introduction of the new varieties, but it also seems to be related with the prolonged periods of the favorable condition associated with the shifted growing seasons. In general, the diseases and insect pest problems have become much more variable and complex, and control measures should be based on the thorough knowledge of the ecology of the pest organisms, that is, effects of various environmental factors on the disease cycle; spore release, spore deposition, infection, colonization and sporulation of the disease organisms, and those on the development, reproductive potentials, dispersal, age specific responses of the insects. The well organized real-time pest management systems, such as alfalfa weevil management system developed at the Purdue University in U.S., is the prime importance for the implementation of the pest management principles.

토양의 수분과 유기물이 멸종위기식물 큰바늘꽃(Epilobium hirsutum L.)의 번식계절 및 생리 반응에 미치는 영향 (Effect of Moisture and Nutrient of Soil on Reproductive Phenology and Physiological Response of Epilobium hirsutum L., an Endangered Plant)

  • 본 연구에서는 멸종위기식물인 큰바늘꽃(Epilobium hirsutum L.)의 효과적인 보전 및 복원을 위한 기초자료를 얻기 위해 토양의 수분함량과 유기물함량이 번식계절과 생리 반응에 어떠한 영향을 주는지에 대하여 알아보았다. 큰바늘꽃은 다년생식물이지만 모든 구배에서 한 해에 생식생장을 하였다. 꽃봉오리, 꽃 그리고 열매주머니는 수분구배와 영양소구배에서 각각 높은 수분 조건과 높은 유기물 조건에서 가장 이른 시기에 성숙하였다. 그리고 꽃 수와 열매주머니 수는 높은 수분 조건과 높은 유기물 조건에서 더 빨리 증가하였다. 엽록소 함량은 수분구배에서 높은 중간 수분 조건과 높은 수분 조건에서 가장 많았고, 영양소구배에서는 차이가 없었다. 최소엽록소형광 값은 수분구배와 영양소구배 모두 차이가 없었고, 최대엽록소형광 값은 높은 수분 조건과 높은 유기물 조건에서 가장 높았다. 광계 II의 광화학적 효율 값은 모든 수분구배에서 0.75로 차이가 없었고, 영양소구배에서의 경우 높은 유기물 조건에서 0.78로 가장 높았다. 큰바늘꽃은 수분이 증가할수록 엽록소 함량이 많아지고, 유기물이 증가할수록 Fv/Fm 값이 높아졌다. 이상의 연구결과는 토양의 충분한 수분과 유기물 함량은 큰바늘꽃의 번식계절을 앞당겨 주고 생식생장을 촉진한다는 것을 보여준다. 추후 멸종위기종인 큰바늘꽃의 개체군 유지와 서식지를 관리하는데 중요한 정보가 될 것으로 판단된다.

보리 생식생장기의 수분부족이 수량 및 몇 가지 생리적 반응에 미치는 영향 (Grain Yield and Physiological Responses of Water Stress at Reproductive Stage in Barley)

  • 보리는 생식생장기에 수분 스트레스에 민감하게 반응하여 수량에 크게 영향을 준다. 보리 5품종 (겉보리 : 밀양 1002, 올보리, 두루보리, 쌀보리: 백동, 맥주보리: 향맥)을 공시하여 생식생장기에 해당되는 출수전 20일, 출수전 10일 및 출수기에 각각 10일간씩 단수처리를 하여 한발을 유발하였으며 처리 후에는 관수를 하여 정상적으로 재배하였다. 보리 품종들을 10월에 포장에 파종하여 익년 2월에 대형 폿트에 이식하여 재배하였으며, 건물중, 유효경비을, 간장, 수량 및 수량 구성요소를 조사하였고, 유리 proline함량과 상대팽압도를 측정하였다. 1. 한발에 의한 피해는 출수전 10일>출수기>출수전 20일 처리의 순으로 나타났으며, 출수전 10일 처리구의 경우 대조구에 비하여 간장은 85∼98%, 주당수수는 52∼83%, 일수립수는 71∼86%, 천립중은 80∼84%, 그리고 10주당 수량은 60∼94%를 나타내었다. 2. 출수 10일전 단수처理에서 수량 및 수량구성 요소들의 감소율이 가장 컸고, 수량구성요소 중에 주당수수와 일수립수가 비교적 한해에 민감하게 반응하였다. 3. 올보리>밀양 1002ㆍ두루보리>향맥>백동의 순으로 내한성이 약하였다. 4. 한발처리를 끝낸 후에 다시 관수하여 지엽의 유리 proline함량의 감소율과 상대팽압도의 회복율을 품종별로 보면 피맥 3품종이 높고 백동과 향맥이 매우 낮았다.

The Vomeronasal Organ and Adjacent Glands Express Components of Signaling Cascades Found in Sensory Neurons in the Main Olfactory System

  • The vomeronasal organ (VNO) is a sensory organ that influences social and/or reproductive behavior and, in many cases, the survival of an organism. The VNO is believed to mediate responses to pheromones; however, many mechanisms of signal transduction in the VNO remain elusive. Here, we examined the expression of proteins involved in signal transduction that are found in the main olfactory system in the VNO. The localization of many signaling molecules in the VNO is quite different from those in the main olfactory system, suggesting differences in signal transduction mechanisms between these two chemosensory organs. Various signaling molecules are expressed in distinct areas of VNO sensory epithelium. Interestingly, we found the expressions of groups of these signaling molecules in glandular tissues adjacent to VNO, supporting the physiological significance of these glandular tissues. Our finding of high expression of signaling proteins in glandular tissues suggests that neurohumoral factors influence glandular tissues to modulate signaling cascades that in turn alter the responses of the VNO to hormonal status.
