Effects of Selenium, Vitamin E, and Their Combination on Growth, Hematological Changes, and Biological Blood Parameters in Orchidectomized Rat Model (Vitamin E와 Selenium이 정소적출 포유동물모델의 성장, 혈액 및 생화학적인 변화에 미치는 영향)

  • The present study was devised to determine the effects of vitamin E and selenium (Selevit) on body weight, organ weight, hematological values and biochemical parameters in the orchidectomized (Orch) rats. Intact group (n=15) received no treatment and operation. Orch+Selevit received operation and Selevit. The body weights of each group increased, but that of the Orch+Selevit group were significantly lower than those of all the other groups. There were significantly different decreased (p<0.001) of body weights between Orch+Selevit group and all the other groups. Also, organ weights such as heart, liver, spleen, kidney, lung and skeletal muscle were measured. The heart and liver weights in the Orch+Selevit group were significantly different decreased (p<0.001) in comparison with those in the Intact and Sham groups. The kidney weights in the Orch+Selevit group were significantly different decreased (p<0.01, p<0.001) in comparison with those in all the other groups. The number of white blood cell (WBC) was significantly higher (p<0.05) in the Orch+Selevit group than in all the other groups. The hematological values of 12 parameters were not significantly different in any of the groups. The concentrations of serum total protein, albumin and alkaline phosphatase only increased significantly (p<0.05, p<0.01) in the Orch+Selevit group as compared to that in the Orch group. We conclude that Selevit was significantly decreased the body weight in the Orch rats. Our findings suggest that Selevit may influence the process of lipid packaging and absorption in the Orch rats.

Print Mottle : Causes and Solutions from Paper Coating Industry Perspective

  • The principal reasons for applying a pigment coating to paper are to improve appearance and printability. The pigment coating provides a surface that is more uniform and more receptive to printing ink than are the uncoated fibers and, in turn, both facilitates the printing process and enhances the graphic reproduction. The improvement in print quality is readily apparent, especially in image areas or when multiple colors are involved. Although pigment coating of paper is to improve the printability, coated paper is not completely free from printing defects. Actually there are a number printing defects that are observed only with the coated papers. Among the printing defects that are commonly observed for coated papers, print mottle during multi-color offset printing is one of the most concerned defects, and it appears not only on solid tone area but also half dot print area. There are four main causes of print mottle ranging from printing inks, dampening solution, paper, and printing press or its operation. These indicates that almost every factors associated with lithographic printing can cause print mottle. Among these variation of paper quality influences most significantly on print mottle problems in multicolor offset printing, and this indicates that paper is most often to be blamed for its product deficiency as far as print mottle problems are concerned. Furthermore, most of the print mottle problems associated with paper is observed when coated papers are printed. Uncoated papers rarely show mottling problems. This indicates that print mottle is the most serious quality problems of coated paper products. Overcoming the print mottle is becoming more difficult because the operating speeds of coating and printing machines are increasing, coating weights are decreasing, and the demands on high-quality printing are increasing. Print mottle in offset printing is caused by (a) nonuniform back trap of ink caused by a nonuniform rate of ink drying, referred as "back trap mottle, and (b) nonuniform absorption of the dampening solution. Furthermore, both forms of print mottle have some relationship to the structure of the coated layer. The surest way of eliminating ink mottling is to eliminate unevenness in the base paper. Coating solutions, often easier to put into practice, should, however, be considered. In this paper the principal factors influencing print mottle of coated papers will be discussed. Especially the importance of base paper roughness, binder migration, even consolidation of coating layers, control of the drying rate, types of binders, etc. will be described.

Production and Characterization of a Transgenic Mouse Model of Human Liver Cancer (형질 전환 기법을 이용한 인체 간암의 마우스 모델 제작 및 특성 규명)

  • Transgenic mice were generated by microinjecting a plasmid DNA containing the SV40 (simian virus 40) large T antigen (Tag) gene fused with mouse albumin promoter/enhancer sequences into fertilized one-cell mouse embryos. Among eleven founder transgenic animals, four developed hepatocellular carcinoma, two showed kidney cancer and one developed skin and brain tumors. Three stable transgenic lines, #1-2, #1-6 and #1-11 were established. Members of the lines #1-6 and #1-11 reproducibly developed liver tumors by 8 to 10 weeks of age but did not exhibit any phenotypic changes in other tissues. Histological changes loading to liver tumor formation occurred with predictable kinetics and could be classified into three distinct stages; (a) newborn to 3 weeks of age, characterized by hyperplastic hepatocytes with reduced amounts of cytoplasm without any nuclear alterations, (b) between 4 to 8 weeks of age, characterized by diffuse liver cell dysplasia without observable tumor nodules, and (c) 9 weeks of age and thereafter, characterized by hepatocellular carcinomas in the background of extensive liver dysplasia. Metastasis to the lung from a liver carcinoma was observed in #1-11 founder animal. This transgenic mouse system displays similarities with human liver cancers in a number of aspects and provides a useful model for the study of molecular events involved in hepatocarcinogenesis.

A New Paradigm to Mitigate Osteosarcoma by Regulation of MicroRNAs and Suppression of the NF-${\kappa}B$ Signaling Cascade

  • Osteosarcoma (OS) is one of the most common malignant primary bone tumors and NF-${\kappa}B$ appears to play a causative role, but the mechanisms are poorly understood. OS is one of the pleomorphic, highly metastasized and invasive neoplasm which is capable to generate osteoid, osteoclast and osteoblast matrix. Its high incidence has been reported in adolescent and children. Cell signal cascade is the pivotal functional mechanism acquired during the differentiation, proliferation, growth and survival of the cells in neoplasm including OS. The major limitation to the success of chemotherapy in OS is the development of multidrug resistance (MDR). Answers to all such queries might come from the knock-in experiments in which the combined approach of miRNAs with NF-${\kappa}B$ pathway is put into use. Abnormal miRNAs can modulate several epigenetical switching as a hallmark of number of diseases via different cell signaling. Studies on miRNAs have opened up the new avenues for both the diagnosis and treatment of cancers including OS. Collectively, through the present study an attempt has been made to establish a new systematic approach for the investigation of microRNAs, bio-physiological factors and their target pairs with NF-${\kappa}B$ to ameliorate oncogenesis with the "bridge between miRNAs and NF-${\kappa}B$". The application of NF-${\kappa}B$ inhibitors in combination with miRNAs is expected to result in a more efficient killing of the cancer stem cells and a slower or less likely recurrence of cancer.

  • The estrogens and estrogenic endocrine disrupting chemicals(EDCs) function through a steroid nuclear receptor-mediated process and subsequently regulate the transcription of mRNA for a number of target proteins. The estrogen receptor-related receptors(ERRs), which are structurally similar to estrogen receptors, are members of orphan nuclear receptor in the nuclear receptor superfamily and their functions are known to be involved in the formation of extra-embryonic ectoderm. To investigate effects of EDCs on early embryogenesis and ERR gene expression in marine invertebrates, we examined morphological changes and the mRNA expression of $ERR{\beta}$ like 1 in sea urchin Strongylocentrotus nudus exposed to estradiol-$17{\beta}(E_2)$ or nonylphenol(NP). The $E_2$ and NP-exposed embryos showed a delayed development compared to control embryos. Furthermore, they showed abnormal embryonic developments at late stages, i.e., blastular, gastrula and plutei stages. The mRNA level of $ERR{\beta}$ like 1 at the gastrula stage was significantly lower in $E_2$ and NP-exposed embryos than those of control group. These results suggest that NP and $E_2$ are potent chemicals causing abnormal embryonic development of S. nudus through at least in part down-regulated $ERR{\beta}$ like 1.

Expression of Cytochrome P450 Aromatase Genes during Sex Differentiation in Korean Rockfish, Sebastes schlegeli (조피볼락, Sebastes schlegeli의 성분화 기간 중 Cytochrome P450 Aromatase 유전자의 발현)

  • Sex determination and sex differentiation are influenced by genotype in many gonochoristic fish. Cytochrome P450 aromatase (CYP19) is the terminal enzyme in steridogenic pathway that converts androgens into estrogens. In this study, partial fragments of aromatase genes (ovarian aromatase, P450aromA and brain aromatase, P450aromB) were cloned and sequenced in Korean rockfish (Sebastes schlegeli), and gene specific primers were designed based on their sequences. Using these primers, aromatase gene expression during sex differentiation was investigated by RT-PCR. Expression of these aromatase genes were detected both in the head and body parts at 35 dab (days after birth). The number of fish that expressed the aromatase genes decreased at 52 dab, implying down-regulation of these genes. However, these genes were expressed at 59 dab in almost all fish studied here. The expression patterns of both genes are similar throughout the investigated period except for 45 dab where the expression of P450aromB was detected in more fish than that of P450aromA both in the head and body parts. Timing of sex differentiation in this species has been shown to be at around $50{\sim}65$ dab by histological analysis. However, the results from this study suggest that sex differentiation of rockfish may take place $1{\sim}2$ weeks earlier than the period proposed previously. The results also suggest that the mechanism of sex differentiation in viviparous fish may be similar to that in oviparous fish in terms of the importance of aromatase action during the critical period.

Reproductive Ecology of an Endangered Species Gobiobotia macrocephala (Pisces: Cyprinidae), in Seom River, Korea (멸종위기 어류 꾸구리 Gobiobotia macrocephala (Pisces: Cyprinidae)의 번식생태)

  • The reproductive ecology of an endangered species, Gobiobotia macrocephala (Pisces: Cyprinidae), was investigated in the Seom River, a tributary of the Han River drainage system, from March 2010 to February 2011. During the non-spawning season, no difference in the external morphology was observed between males and females; however, during the spawning season, clear differences merged for the depth and width of the body, preventral length and preanal length in the standard length. The sex ratio of females to males was 1 : 1.01. The spawning season was estimated to last from late April to middle of June, at water temperatures of $15{\sim}25^{\circ}C$. The number of mature eggs in the ovary averaged about $2,134{\pm}930$ (mean${\pm}$SD), and the mean diameter was approximately $0.88{\pm}0.04$ mm. The spawning area was composed of pebble and cobble bottoms (3~10 cm), with current velocities of 13~24 cm $s^{-1}$ and water depths of 12~18 cm. Spawning behavior between males and females was observed in a water tank 14~15 h after injecting females with Ovaprim to promote reproduction. Fish spawned near the water surface with monogamous pairing.

Efficient Establishment of Presumptive Embryonic Stem Cells from Bovine Blastocysts by Exposure to Calcium Ionophore (Calcium Ionophore를 이용한 소 배반포로 부터의 배아주 유사세포의 효과적인 분리)

  • It is crucial to remove trophectoderm (TE) cells of blastocysts for an efficient isolation of pluripotent embryonic stem (ES)-like cells from bovine blastocysts. We evaluated the effectiveness of chemosurgery using calcium ionophore A23l87 (CIPA) by investigating the viability and pluripotency of ES-like cell lines isolated from in vitro-produced bovine blastocysts after CIPA treatment. The blastocysts treated with 50 $\mu$M CIPA for 25 min colonized most efficiently (51% of blastocysts) and developed to ES-like cell lines through 10 passages (4.8% of blastocysts) among CIPA-treated groups with different concentration and duration. In comparison with CIPA-untreated blastocysts, the colonization rate and overall viability of the CIPA-treated blastocysts were five times higher, suggesting that CIPA treatment condition defined in this study was highly efficient for establishing ES-like cell lines without apparent toxicity of CIPA. We evaluated in vitro pluripotency of the established three ES-like cell lines by examining alkaline phosphatase (AP) activity, capability of embryoid body formation, and chromosomal euploidity of the cells. Our cells showed a heterogeneous AP activity similarly to other reports. The cells were able to form simple embryoid bodies during suspension culture and majority of them showed a normal chromosome number of 60, the euploid chromosomal complement of bovine Therefore, our data suggest that CIPA treatment can be safely used for an efficient isolation of ES-like cell lines from bovine blastocysts.

Differentiation of Dopaminergic Neurons from Mesenchymal-Like Stem Cells Derived from Human Umbilical Cord Vein

  • One of the most extensively studied populations of multipotent adult stem cells are mesenchymal stem cells (MSCs). MSCs derived from the human umbilical cord vein (HUC-MSCs) are morphologically and immunophenotypically similar to MSCs isolated from bone marrow. HUC-MSCs are multipotent stem cells, differ from hematopoietic stem cells and can be differentiated into neural cells. Since neural tissue has limited intrinsic capacity of repair after injury, the identification of alternate sources of neural stem cells has broad clinical potential. We isolated mesenchymal-like stem cells from the human umbilical cord vein, and studied transdifferentiation-promoting conditions in neural cells. Dopaminergic neuronal differentiation of HUC-MSCs was also studied. Neural differentiation was induced by adding bFGF, EGF, dimethyl sulfoxide (DMSO) and butylated hydroxyanisole (BHA) in N2 medium and N2 supplement. The immunoreactive cells for $\beta$-tubulin III, a neuron-specific marker, GFAP, an astrocyte marker, or Gal-C, an oligodendrocyte marker, were found. HUC-MSCs treated with bFGF, SHH and FGF8 were differentiated into dopaminergic neurons that were immunopositive for tyrosine hydroxylase (TH) antibody. HUC-MSCs treated with DMSO and BHA rapidly showed the morphology of multipolar neurons. Both immunocytochemistry and RT-PCR analysis indicated that the expression of a number of neural markers including NeuroD1, $\beta$-tubulin III, GFAP and nestin was markedly elevated during this acute differentiation. While the stem cell markers such as SCF, C-kit, and Stat-3 were not expressed after neural differentiation, we confirmed the differentiation of dopaminergic neurons by TH/$\beta$-tubulin III positive cells. In conclusion, HUC-MSCs can be differentiated into dopaminergic neurons and these findings suggest that HUC-MSCs are alternative cell source of therapeutic treatment for neurodegenerative diseases.

Distribution and Habitat Characteristics of Tadpole Shrimp (Crustacea: Notostraca; Triops longicaudatus (LeConte)) in Korea (한국산 긴꼬리투구새우 (Crustacea: Notostraca; Triops longicaudatus (LeConte))의 분포 및 서식지 특성)

  • Tadpole shrimps are aquatic crustaceans adapted to temporary water pool. Since they have been recorded as the living fossil, there are a number of reports on their past and present distribution as well as their morphology and reproduction in foreign countries. However, there are few studies of biology and ecology of tadpole shrimps despite registration as an endangered species by Ministry of Environment, in Korea. This study was carried out to examine the distribution and habitat characteristics of tadpole shrimps (Triops longicaudatus) collected from May 2006 to July 2008 in Korea. Tadpole shrimps were found from the paddy fields and ephemeral pools at 21 countries in South Korea. They appeared between May and July and survived for 30 days, and temperatrue range was $10.7^{\circ}C$ to $33.5^{\circ}C$. The average turbidity of experimental paddy fields was 176.8 (${\pm}239.3$) NTU, and controlled field was 28.9 (${\pm}26.7$) NTU. The life pattern of tadpole shrimps is adapted to a temporary water pool and unstable environmental conditions, and also there was great effect of agricultural activity on the development and growth of tadpole shrimps.
