Cadmium에 의한 흰쥐의 간장 및 신장의 Metallothionein 변화와 방어효과 (Metallothionein Induction and Its Protective Effect in Liver and Kidney of Rats Exposed to Cadmium Chloride)

  • Tolerance to several toxic effects of cadmium, including lethality has been shown following pretreatment with cadmium and zinc. This study was designed to determine if tolerance also develops to Cd-induced hepatotoxicityandrenaltoxicity. Three groups of rats (A, B, C), each consisting of 16 rats, were studied and each group was divided into four subgroups (1, 2, 3, 4), 4 rats for each subgroup. Rats were subcutaneously pretreated with saline (A), $CdCl_2$ (0.5 mg/kg, B), and $ZnCl_2$ (13.0 mg/kg, C) during time periods of $1{\sim}6$ weeks. At the end of the period, rats were challenged with $CdCl_2$ (3.0, 6.0 and 9.0 mg/kg, ip). After giving the challenge dose, cadmium and metallothionein (MT) concentrations were determined and also observed the histologic change in liver and kidney. The concentration of cadmium in liver and kidney increased dose-dependently to the challenge dosage. These da indicate the kidney is a major target organ of chronic cadmium poisoning, and suggest that cadmium induced hepatic injury, via release of Cd-MT, may play an important role in the nephrotoxicity observed in response to long-term exposure to cadmium. In addition, histologic examination of group $A_2,\;A_3\;and\;A_4$ revealed moderate to severe cadmium toxicity, evidenced by infiltration of inflammatory cells, cell swelling, pyknosis, enlarged sinusoids and necrosis in liver, and tubule cell necrosis and degeneration in kidney. However, MT concentrations in liver and kidney were increased by the pretreatment of $CdCl_2$ and $ZnCl_2$, and their morphological findings were not significantly changed, comparing with control group. Higher MT concentration in liver and kidney observed in the pretreated groups constitutes a plausible explanation of the protective effects of pretreatment against the cadmium toxicity after challenge dosing.

보리 Sourdough의 제빵성 연구 (Study on Bread-making Quality with Barley Sourdough in Composite Bread)

  • 흰찰쌀보리가루를 이용한 sourdough의 물리 화학적 특성, 발효 양상 및 물성 등의 반죽특성과 이의 제빵성을 조사하였다. L. plantarum과 L. Brevis 및 이의 혼합처리로써 보리가루를 발효시킨 결과 pH, 총당 및 환원당 등은 감소하였고 젖산균수, 산도는 증가하였으며, 젖산균에 따라 환원당을 제외하고 유의적인 차이가 없었다. 보리 sourdough의 젖산균수는 각 처리구 모두 발효 후 증가하였으나 효모는 감소하였고 젖산균에 따라 이화학적 특성에 차이가 있었다. 특히 L. plantarum을 처리한 보리 sourdough는 반죽의 형성시간, 안정도 및 탄성 등에서 물성개량제를 활용한 보리가루 첨가 대조구에 비해 양호하였고 발효진행속도는 느리나 가스발생량, 가스보유율도 가장 높았다. 또한 보리 sourdough 빵의 pH는 발효하지 않은 빵에 비해 낮았고 수분 및 수분 활성도는 증가하였으며, 필연적으로 L. plantarum를 처리한 보리 sourdough빵의 부피, 경도, 탄성, 관능적 특성 등의 제빵특성에 있어서도 물성개량제를 첨가한 보리가루 첨가 대조구에 비해 우수하였고 이는 빵 내부 조직의 protein-starch matrix의 미세구조에서도 확인되었다.

IIA형 Phospholipase $A_2$에 의한 TNF-${\alpha}$와 IL-6 생성 기전 (The Production Mechanism of TNF-${\alpha}$ and IL-6 by Group IIA Phospholipase $A_2$)

  • Background: Secretory phospholipase $A_2$ ($sPLA_2$) are a group of extracellular enzymes that release fatty acids at the sn-2 position of phospholipids. Group IIA $sPLA_2$ ($sPLA_2$-IIA) has been detected in the inflammatory fluids, and its plasma level increases in the inflammatory disease. This study examined the effect of $sPLA_2$-IIA on mouse macropahges in order to investigate the potential mechanism of $sPLA_2$-induced inflammation. Materials and Methods: Wild type $PLA_2$ and mutant H48Q $PLA_2$ were purified from HEK293 cells transfected with the corresponding plasmids, and the $PLA_2$ activities were measured using 1-palmitoyl-2-[1-$^{14}C$]linoleoyl-3-phosphatidylethanolamine as substrates. The TNF-${\alpha}$ and IL-6 released in the supernatants were determined by ELISA. In addition, the TNF-${\alpha}$ and IL-6 mRNA were analyzed by RT-PCR. Results: $sPLA_2$-IIA stimulated the production of TNF-${\alpha}$ and IL-6 in a dose- and time-dependent manner. In addition, the effect of $sPLA_2$-IIA on cytokine production from the macrophage was found to be associated with the accumulation of their specific mRNA. The mRNA levels of TNF-${\alpha}$ and IL-6 peaked at 2 and 6 hours in a time-dependent manner, respectively. Conclusion: In conclusion, the production of proinflammatory cytokine might be mediated by the binding of $sPLA_2$-IIA to the receptors.

Preclinical Study of DA-5018, a Non-narcotic Analgesic Agent

  • DA-5018 is a synthetic capsaicin derivative under development as a non-narcotic a analgesic ag$\varepsilon$nt. DA-50 18 showed a potent analgesic activity against acute and chronic pain m model(Tablel, 2.), but it had a narrow margin of safety. DA-5018 did not bind to opioid(${\kappa}, {\delta}, {\mu}$), NKl, CGRP receptors in vitro and its analgesic effect was not antagonized by naloxone, a and it did not develop analgesic tolerance. In addition DA-5018 had no inhibitory effects against c cyclooxygenase and 5-lipooxygenase activities. DA-5018 significantly increased the relcase of substance P from the slices of the rat spinal cord. These results suggest that DA-50 18 is not a narcotic nor aspirin-like analgesic and the release of substance P is one of analgesic mechanism of action of DA-5018. We found that DA-5018 was almost ten times more potent and was at l least IOO-times less irritable compared to capsaicin. Accordingly development of topical formula was adopted. Topical formula was desiged and screened by flux test of DA-5018 using hairless mouse skin and several formulas were selected. With these topical formulas we a assessed the analgesic efficacy and carried out the toxicity, skin irritation and pharmacokinetic studies. In streptozotocin-induced hyperalgesic rat and 50 % galactose-fed hyperalgesic rat as diabetic pain models, DA-5018 cream increased the pain thresh이ds up to 77.0% and 24.4% respectively, while Zostrix-HP(capsaicin cream) incr$\varepsilon$as cd by 65.9% and 21.0%. DA-5018 c cream showed a good analgesic effect as welI in FCA-induced arthritic rat. DA-5018 cream did not show any toxicological signs in acute and chronic toxicity test and had little skin irritation in car swclIing and scratching t$\varepsilon$st. Pharmacokinetics of DA-50 18 were studied after topical application of ${14}^C$-Iabelled or unlabelIed DA-5018 cream. Plasma and skin concentrations c except applied skin wcre below the dctection limit and after 7-day cummulative application, plasma concentrations were also below detection limit DA-50 18 may have an advantag$\varepsilon$ ov$\varepsilon$r c capsaicin and is now being developed as a topical agent for the treatment of pains. DA-50 18 cream was approved for Korean IND and is now under a Phase II clinical study for arthritic pain a after finising Phase I study. 녹차추출물의 잇몸 질환 원인균에 대한 항염증 효능 연구 (The Anti-inflammatory Effect of Green Tea Extract Against Prevotella intermedia)

  • 치아의 세균들은 잇몸에서 염증반응을 일으켜서 치은염과 치주염같은 잇몸 질환의 원인이 된다. 따라서 잇몸질환의 예방과 치료를 위해서는 치아 세균에 의한 염증반응을 조절하는 것이 가장 효과적인 방법이다. 현재 대부분의 구강 관리 제품들은 살균제를 이용하여 구강 세균을 제거하는 방법을 사용하고 있다. 하지만 최근의 연구들은 심지어 열처리로 사멸된 구강 세균도 염증반응을 일으킬 수 있다는 사실을 보고하고 있다. 따라서 보다 효과적인 잇몸 염증반응억제를 위해서는 살균제를 이용한 방법에 대한 개선이 필요하다. 또한 아직까지 구강 세균에 의한 잇몸 염증반응의 기작과 효과적인 천연 염증 억제 물질들은 보고되어 있지 않다. 본 연구에서는 대표적인 치은염, 치주염 유발세균인 Prevotella intermedia와 인간의 잇몸상피세포를 이용하여, 실제로 잇몸에서 일어나는 염증반응의 기작을 연구하고, 이를 통해 효과적인 천연 잇몸 염증 완화 물질을 도출하려고 하였다. 실험 결과, Prevotella intermedia는 잇몸상피세포를 자극하여 염증매개인자인 IL-8을 분비하게 함으로써 잇몸 염증반응을 개시하였다. 또한 Prevotella intermedia에 의한 잇몸 염증반응은 기전적으로 COX-2, AP-1, TNF-${\alpha}$와 연관되어 있었으며, 녹차추출물은 Prevotella intermedia에 의한 잇몸 염증반응을 효과적으로 억제할 수 있음을 확인하였다. 따라서 본 연구는 치아 세균에 의한 잇몸 염증반응의 기전 연구를 통해서 효과적인 천연 잇몸질환 개선 물질을 도출했다는 점에서 중요한 의미를 가진다.

Effect of the different cover crop incorporation on glomalin-related soil protein and soybean and maize growth

  • The glycoprotein known as glomalin-related soil protein (GRSP) is abundantly produced on the hyphae and spores of arbuscular mycorrhizal fungi (AMF) in soil and roots. GRSP play a decisive role in the soil aggregation, but GRSP was also sensitive to agricultural managements. Thus, our objectives were to assess the effect of different cover crop incorporation on the GRSP content in soil and growth of subsequent soybean and maize. Pot experiments with the incorporation of four cover crops were set up. The same amount (666g) of aboveground plant parts of wheat (AMF host), hairy vetch (AMF host), mustard (non-host) and rapeseed (non-host) was separately incorporated into soils. The aboveground plant parts and roots of soybean and maize were grown in each incorporated pots and sampled at 6 and 9 weeks after sowing. Our results showed that the different cover crops incorporation affected soil biological and chemical properties such as EC, $NO_3-N$ content, ${\beta}-glucosidase$ activity, alkaline phosphatase (ALP) activity and GRSP content. The soil EC and $NO_3-N$ content in the hairy vetch, mustard and rapeseed was higher compared to the wheat. The ${\beta}-glucosidase$ activity in the wheat and hairy vetch was significantly higher than that in the mustard and rapeseed, and the ALP activity in the wheat was significantly higher than that in the hairy vetch, mustard, and rapeseed. The GRSP content in the mustard and rapeseed was significantly lower than that of the hairy vetch and wheat. Moreover, The top dry weight and leaf area of soybean and maize in the hairy vetch at 6 weeks were significantly higher compared to the other treatments. Our results indicated that the incorporation of mustard and rapeseed may cause indirectly the decrease of GRSP content and soil enzyme activity in soil. One possible explanation for the decrease of GRSP in non-AMF host crop treatments may be the decrease of AMF density in the soil. AMF are not able to form a symbiotic relationship with Brassicaceae roots due to the release of anti-fungal compounds. This means the AMF may not be able to produce GRSP in the soil. However, the differences in the benefit of cover crop incorporation were shown only by a pot experiment. Comparative investigations of crop residue managements would be applied to both pot experiment and field study to clarify a better selection of cover crops in rotation to encourage GRSP production.

  • Objectives : Substantial evidence suggests that reinforcing effects of nicotine can be mediated by the mesolimbic dopaminergic system. It has been shown that repeated injections of nicotine produce an increase in locomotor activity and expression of the immediate-early gene, c-fos, in the dopaminergic target areas. Herbal medicine as a therapeutic intervention has been widely used for the treatment of mental dysfunction. Many studies have shown that Radix Scutellariae (RS) can affect the biochemical balance in the central nervous system. Tn order to investigate whether RS has an influence on nicotine-induced behavioral sensitization, we examined the effect of RS on nicotine-induced locomotor activity and c-fos expression in the striatum and nucleus accumbens utilizing the fos-tike immunohistochemistry (FLI). Methods : Male SD rats received RS (200mg/kg, i.p.) 30min before repeated daily injections of nicotine (0.4mg/kg, s.c.) for 7 days. This was followed by withdrawal for 3 days and one challenge for 1 day. Results : System challenge with nicotine produced a much larger increase in locomotor activity and accumbal FLI. Pretreatment with RS significantly inhibited nicotine-induced locomotor activity and FLI in the striatum and nucleus accumbens. Conclusions : These results demonstrate that reduction in locomotor activity by RS may be reflected by reduction of dopamine release and postsynaptic neuronal activity in the striatum and nucleus accumbens. Our results suggest that RS may have therapeutic effect on nicotine addiction.

골모세포유사세포의 cytoskeletal change가 세포 활성에 미치는 영향에 관한 실험적 연구 (An Experimental Study on the Effect of Cytoskeletal Changes or Osteoblastic Cell Activities)

  • Cytoskeleton은 세포핵과 세포외 기질을 연결하고 있어서 기질에 가해지는 물리적 힘에 의해 cytoskeletal change가 유도되고 이에 의해 세포의 개조활성이 영향을 받는다고 생각되어 왔다. 본 연구는 골모세포 활성에 대한cytoskeletal change의 역할을 규명하기 위한 것으로서, 신생 백서로부터 조골세포양 세포를 분리, 배양하고 네가지 농도의 cytochalasin B(CB) 또는 colchicine(COL)을 3시간 처리하였다. 다시 배양액을 교환하고 24시간 동안 배양하여 prostaglandin $E_2\;(PGE_2)$, interleukin-6(IL-6), tumor necrosis factor-$\alpha$(TNF-$\alpha$) 및 matrix metalloproteinase-1 (MMP-1) 생산을 측정하고 통계적으로 비교하였으며 cytoskeletal protein actin 변화를 관찰하기위하여 면역형광염색하고 형광현미경으로 관찰하여 다음과 같은 결과를 얻었다: 1. CB 처리군에서 $PGE_2$ 생산이 증가되는 경향을 보였고 COL 처리군에서는 약물농도에 비례하여 증가하였다. 2. IL-6 생산은 CB농도 1.0 ${\mu}g/ml$일때를 제외하고 증가되었다. 3. TNF-$\alpha$도 CB 농도가 1.0 ${\mu}g/ml$ 일때를 제외하고 증가하였다. 4. MMP-1 생산은 CB 처리군에서 감소하는 경향을 보이고 COL 처리군에서는 변화되지 않았다. 5. CB처리군에서는 cytoskeletal actin stress fibers가 사라지고 세포모양이 둥글어지는 경향을 보였다. 이상의 결과로 미루어 보아 cytoskeletal rearrangement는 골모세포유사세포의 활성, 특히 $PGE_2$, IL-6, 및 TNF-$\alpha$같은 paracrine/autocrine factor의 생산과 관련있는 것으로 보인다.

Self-Assembled Nanoparticles of Bile Acid-Modified Glycol Chitosans and Their Applications for Cancer Therapy

  • This review explores recent works involving the use of the self-assembled nanoparticles of bile acid-modified glycol chitosans (BGCs) as a new drug carrier for cancer therapy. BGC nanoparticles were produced by chemically grafting different bile acids through the use of l-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC). The precise control of the size, structure, and hydrophobicity of the various BGC nanoparticles could be achieved by grafting different amounts of bile acids. The BGC nanoparticles so produced formed nanoparticles ranging in size from 210 to 850 nm in phosphate-buffered saline (PBS, pH=7.4), which exhibited substantially lower critical aggregation concentrations (0.038-0.260 mg/mL) than those of other low-molecular-weight surfactants, indicating that they possess high thermodynamic stability. The SOC nanoparticles could encapsulate small molecular peptides and hydrophobic anticancer drugs with a high loading efficiency and release them in a sustained manner. This review also highlights the biodistribution of the BGC nanoparticles, in order to demonstrate their accumulation in the tumor tissue, by utilizing the enhanced permeability and retention (EPR) effect. The different approaches used to optimize the delivery of drugs to treat cancer are also described in the last section.

무기 입자 강화제와 브롬(Br) 난연제에 따른 폴리프로필렌 복합재료의 난연성 향상에 관한 연구 (Fire Resistance Study of PP Thermoplastic Composites with Particulate Reinforcements and Br Flame Retardants)

  • 본 연구에서는 범용 열가소성수지인 폴리프로필렌에 다양한 입자 강화제로 무기질 폐기 제올라이트, 탈크, 탄산칼슘 등을 첨가하여 소재를 복합재료화 하였다. 또한 입자 사이즈에 따른 열안정성과 입자 강화제에 따른 난연 특성을 확인하였다. 본 연구에서는 입도 분석 결과 폐기 제올라이트가 85.34 $mu extrm{m}$, 탄산칼슘이 33.93 $mu extrm{m}$, 탈크가 18.51 $mu extrm{m}$의 평균 입자 크기를 가지고 있는 것을 확인하였다. 난연성 측정으로 산소지수(LOI, ASTM D2863)와 콘 칼로리미터 (ASTM E1354 ISO 5660)를 사용하였으며, 열 안정성 측정으로는 TGA를 사용하였다. 입자 강화제와 난연제 DBDPO를 사용한 결과 최대 열 방출 속도(M-HRR)는 탈크>탄산칼슘>폐기 제올라이트 순으로 감소됨을 확인하였다. 콘 칼로리미터 실험 결과, 난연제 DBDPO만 혼합하였을 경우보다 입자 강화제를 첨가 혼합하였을 경우가 난연 효율이 대략 2배 정도 향상됨을 확인하였다. 또한 산소지수 결과도 콘 칼로리미터와 유사한 경향을 보임을 확인하였다. 광학현미경(OM)과 주사전자현미경(SEM)을 사용하여 입자 강화된 복합재료의 연소되는 과정의 단면을 연소 단계별로 관찰함으로써 연소 표면에서의 입자 강화제의 배열 양상 및 산소 공급의 특성 등을 연구하였다.
