  • Rehmannia glutinosa(Gaertn.) DC. is a herbaceous perennial plant and belonging to the Scrophulariaceae and used as roots for medicinal part and purpose. R. glutinosa is and usually used for fresh rehmannia root or prepared rehmannia root. However, it is very difficult to propagate using the seeds because of lack germination so it is propagated using the vegetative method as the rootstock. Currently, propagation and harvesting using the rootstock of R. glutinosa has difficulties about production of the high quality and quantity in R. glutinosa because of root rot disease. To optimize in vitro cultures and to improve the rootstock and seedling of R. glutinosa after morphological and genetical determination, 5 plant culture media (MS, DJ, LS, QL, and WPM) were used in this study then WPM was selected for better growth, for multiplication condition(WPM + IAA 1.0 mg/L + IBA 0.5 mg/L), and for root enlargement condition(WPM + NAA 0.1 mg/L) of R. glutinosa. Based on these results, in vitro seedlings of R. glutinosa were transferred to soil for acclimation with environment adaptation and shown the positive effects about root enlargement and root formation. Therefore, it can be used for high quality of R. glutinosa production and production of the rootstock based on propagation using in vitro seedlings of R. glutinosa.

  • Alzheimer's disease, a representative neurodegenerative disorder, is characterized by the presence of senile plaques and neurofibrillary tangles accompanied by neuronal damages. b-Amyloid peptide is considered to be responsible for the formation of senile plagues that accumulate in the brains of patients with Alzheimer's disease. There has been compelling evidence supporting that b-amyloid-induced cytotoxicity is mediated through generation of reactive oxygen species. In this study, we have investigated the possible protective effect of Rehmannia glutihosaagainst b-amyloid-induced oxidative ceil death in cultured human neuroblastoma SH-SY5Y cells. SH-SY5Y cells treated with b-amyloid underwent apoptotic death as determined by morphological features and positive in situterminal end-labeling (TUNEL staining). Rehmannia glutinosawater extract, wine, and vinegar pretreatments attenuated b-amyloid-induced cytotoxicity and apoptosis. Rehmannia glutinosa vinegar exhibited maximum protective effect by increasing the expression of anti-apoptotic protein, Bcl-2. in addition to oxidative stress, b-amyloid-treatment caused nitrosative stress via marked increase in the levels of nitric oxide, which was effectively blocked by Rehmannia glutinosa. To further explore the possible molecular mechanisms underlying the protective effect of Rehmannia glutinosa, we assessed the mRNA expression of cellular antioxidant enzymes. Treatment of Rehmannia glutinosa vinegar led to up-regulation of heme oxygemase-1 and catalase. These results suggest that Rehmannia glutinosa could modulate oxidative neuronal cell death caused by b-amyloid and may have preventive or therapeutic potential in the management of Alzheimer's disease. Particularly, Rehmannia glutinosa vinegar can augment cellular antioxidant capacity, there by exhibiting higher neuroprotective potential.

  • Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae and is used in traditional oriental medicine. This study was carried out current status of breeding of R. glutinosa, its problems, and the matters to be improved were made. Based on the research data published so far, this study summarized the characteristics of breeds cultivated in Korea and compared and analyzed the status of cultivation area, production, import, and price changes for 15 years from 2008 to 2021. The cultivated area of R. glutinosa is gradually decreasing, and the amount of import is increasing and decreasing repeatedly. Amount of import has declined significantly. There was a lot of disinfection and disposal in the middle of import, and dried Jihwang and Sukjihwang were imported without distinction. R. glutinosa must be cultivated and supplied as a variety desired by the industry, cultivation techniques need to be taught to producers. In order to clearly identify the amount of import, import code of dried Jihwang and Sukjihwang must be classify. In addition, it is necessary to develop and supply harvesting agricultural machines for mechanization. For the stable income of producers, supply control through contract cultivation will ensure stable farm income.

  • While rehmannia (Rehmannia glutinosa Libosch) was identified as a host of at least five viruses, including Rehmannia mosaic virus (ReMV), Youcai mosaic virus (YoMV), Broad bean wilt virus 2 (BBWV2), Plantago asiatica mosaic virus (PlAMV), and Rehmannia virus 1 (ReV1), viral incidence surveys have not been performed yet in rehmannia fields in Korea. In this study, we performed field surveys during 2017-2018 to investigate the incidence of 5 major viruses in rehmannia. A total of 145 symptomatic samples were collected from the rehmannia fields in major cultivation areas of Korea. Molecular diagnosis assays showed that all the collected leaf samples were infected with more than two viruses. Particularly, two species of Tobamovirus, ReMV and YoMV, were detected in all the samples. In addition, our analysis showed that the root stocks of 4 rehmannia cultivars were infected with at least two viruses. Since rehmannia is propagated by vegetative propagation, it is highly important to produce virus-free root stocks of rehmannia to control virus diseases in rehmannia.

  • Chitinase, which is one of the pathogenesis-related proteins, was examined from Rehmannia glutinosa. Rehmannia chitinases were classified into basic and acidic groups by chitin affinity column chromatography. According to the chitinase pattern by native PAGE, basic chitinases of low Rf values were extracted more under an acidic condition (pH 2.9) than a basic condition (pH 8.8), while acidic chitinases having high Rf values were mainly detected in basic extracts. There were in total seven chitinase isoforms of three basic and four acidic isoforms in Rehmannia. The range of molecular weight of Rehmannia chitinase was from 28 to 32 kD. It showed the possibility of tissue specific expression of acidic chitinases and basic chitinases were mainly detected in root.

  • This experiment was carried out to investigate the cost reduction of rhizome through the stable storage of Jiwhang #1(Rehmannia glutinosa Libosch). The growth characteristics of Jiwhang #1 are different from those of local Rehmannia glutinosa L. varieties in Korea. Various storage methods was introduced and analyzed for 120 days. Among storage methods, changes in mean daily temperature in storage location were the least from 7.2 to 13.8$^{\circ}C$ in underground storage, while those were wide in styrofoam box filled with soil, and changes in mean daily relative humidity were the highest 98.2% in underground storage. As the storage period increased, the rhizome brix and the rate of rhizome loss and spoiled rhizome increased. The brix and loss rate of rhizome were higher in net sack filled with hull and styrofoam box filled with hull, but the rate of spoiled rhizome was the greatest in underground storage among storage methods. As the storage period increased, the hardness of rhizome epidermis increased, while that of rhizome endodermis decreased.
