Studies on the anti-inflammatory action of steamed Rehmannia glutinosa in central nervous system (중추신경계(中樞神經系)에서 숙지황(熟地黃)의 항염증작용(抗炎症作用)에 관(關)한 연구(硏究))

  • We investigated whether an aqueous extract of Rehmannia glutinosa steamed root (RGAE) inhibits secretion of inflammatory cytolanes from primary cultures of mouse astrocytes. RGAE dose-dependently inhibited the $TNF-{\alpha}$ secretion by astrocytes stimulated with substance P (SP) and lipopolysaccharide (LPS). Interleukin-1 (IL-1) has been shown to elevate $TNF-{\alpha}$ secretion from LPS-stimulated astrocytes while having no effect on astrocytes in the absence of LPS. We therefore also investigated whether IL-1 mediated inhibition of $TNF-{\alpha}$ secretion from primary astrocytes by RGAE. Treatment of RGAE to astrocytes stimulated with both LPS and SP decreased IL-1 secretion to the level observed with LPS alone. Moreover, incubation of astrocytes with IL-1 antibody abolished the synergistic cooperative effect of LPS and SP. These results suggest that RGAE has an antiinflammatory activity on the central nervous system curing some pathological disease states.

  • Psoriasis is a chronic skin disease characterized by angiogenesis. It has been reported that growth factor as vascular endothelial growth factor(VEGF) and insulin like growth factor(IGF) II are overexpressed in psoriatic epidermis. To investigate the inhibitory effects of IGF-II induced VEGF and HIF-1${\alpha}$ expression by RR extracts, we performed MTS assay, western blots using HaCaT cells. RR extracts significantly reduced IGF-II induced HIF 1${\alpha}$ protein level via MAPK pathway in HaCaT cells. Also, RR extracts inhibited IGF-II induced VEGF mRNA and protein expression levels in the HaCaT keratinocytes. These results suggest that inhibition of HIF-1${\alpha}$ and VEGF expressions by RR extracts contributes to the anti angiogenic effects.

  • Ascorbate peroxidase (APX) plays a crucial role in the detoxification of hydrogen peroxide. APX activity is maintained significantly higher in the paraquat­treated leaves of the paraquat-tolerant Rehmannia glutinos. This study was conducted to understand structural and regulatory characteristics of APX gene in R. glutinosa. A putative APX cDNA clone (RgAPX1) was isolated from a leaf cDNA library using a partially sequenced expressed sequence tag clone. RgAPX1 is consisted of 1148 bp nucleotides and contains an open reading frame encoding a 250 amino acid-long polypeptide. Deduced RgAPX1 amino acid sequence shares higher sequence similarity to cytosolic APXs. RgAPX1. expression was higher in the leaf than in the flower and root. Southern blot result indicates the presence of one or two RgAPX1-related genes in R. glutinosa genome. RgAPX1 transcription was affected differentially by various stresses and phytohormone. The results indicate that RgAPXl is constitutively expressed in most tissues and its expression is modulated for the immediate and efficient detoxification of $H_2O_2$ under normal and stress conditions.

Anti-inflammatory Activities of Ethylacetate Extract of Rehmannia glutinosa in LPS-induced RAW 264.7 Cells

  • This study is to investigate the anti-inflammatory effects of the ethylacetate extract of Rehmannia glutinosa (RGEAE). The anti-inflammatory activities using nitric oxide (NO), cytokine, and chemokine production in lipopolysaccharide (LPS)-induced RAW 264.7 cells were checked. Results indicated that RGEAE suppressed the NO, interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1) production in a dose-dependent manner. Inhibition of NO formation was due to a decrease in inducible NOS (iNOS) expression. It was also found that the anti-inflammatory activities of RGEAE resulted from its inhibitory role on the nuclear factor $(NF)-{\kappa}B$ activation and reactive oxygen species (ROS) production. Therefore, it is suggested that RGEAE has potential as a therapeutic material to attenuate the inflammatory disease such as rheumatoid arthritis.

Effects of Black Polyethylene Film Mulching and Planting Density on Growth and Yield of Rehmannia glutinosa Libosch. (흑색비닐 피복 및 재식밀도가 지황의 생육 및 수양에 미치는 영향)

  • These studies were carried out to clarify the effects of black P.E. mulching and planting density on the growth and rhizome yield of Jihwang #1, a new high yielding Rehmannia glutinosa variety. Emergence ratio was higher 3.1% in mulching than in non-mulching, but was not affected by planting density. Plant height and fresh leaf weight per plant were increased by mulching and at lower planting density. However, the fresh leaf weight per unit area was increased at higher planting density. Dry rhizome weight was increased 28% in mulching than that of non-mulching. Dry rhizome weight per plant was increased, but that of per unit area was decreased at lower planting density.

Antioxidant Activities and Quality Characteristics of Sauce Added with Rehmannia Radix Preparata and Rubus coreanus Wine (숙지황 농축액을 첨가한 복분자 소스의 품질 특성 및 항산화성)

  • This study was performed to evaluate the antioxidant activities as well as quality and sensory characteristics of Rubus coreanus wine sauce prepared with different amounts of Rehmannia Radix Preparata concentrate (RPC, 0, 0.5, 1.0, 1.5 %). Moisture content, salinity and acidity of control were 83.84%, 0.57% and 0.10%, respectively. Sugar content of control was 15.6 $^{\circ}Brix$. Acidities, and reducing sugar contents of sauces increased according to the added amount of RPC, whereas pH and color values of lightness, redness and yellowness in the Hunter color system of sauces decreased. Viscosity of sauces added with RPC significantly increased, compared to that of control. Total phenol contents of sauces increased according to RPC amount. Antioxidant activities such as DPPH and hydroxyl radical scavenging activities improved according to the amount of RPC, due to the increased amount of total phenol contents. Sensory results showed that the overall-preference scores were highest in sauces containing 0.1% RPC. Based on these results, it was suggested that 1.0% RPC addition to sauce is the most suitable to improve quality.

Genotoxicological Safety Evaluation of the Solvent Extracts for Medicinal Herbs that are of Highly Domestic Spendings (국내 소비 주요 생약재의 유기용매 추출물에 대한 유전독성평가)

  • This study is carried out to evaluate the genotoxicity of herbs (Angelica decursiva, Polypori umbellate, Astragalus membranaceua, Paeonia iactiflora, Glycyrrhiza uralensis, Cnidium officinale, Rehmannia glutinosa, Cyperus rotundus, Dioscorea batatas and Platycodi Radix) by using the Ames test. The Salmonella typhimurium reversion assay is being performed by using the Sal. typhimurium TA98, TA100, TA102, TA1535 and TA1537 as tester strains. Among 70% of ethanol extracts from 10 herbs, the number of revertant colonies is being increased in Astragalus membranaceua, Cnidium officinale and Dioscorea batatas in a dose-dependent manner, as compared with negative controls of the metabolic activation. In case of dichloromethane and ethyl acetate fraction from 70% of ethanol extracts, the number of revertant colonies is increased in Angelica decursiva, Astragalus membranaceua, Cnidium officinale, Rehmannia glutinosa and Dioscorea batatas in a dose-dependent manner, as compared with negative controls of metabolic activation. Such results indicate that Angelica decursiva, Astragalus membranaceua, Cnidium officinale, Rehmannia glutinosa and Dioscorea batatas all show genotoxic effects when being extracted with the solvent extractions such as 70% of ethanol, dichloromethane and ethyl acetate, and thus, they might be genotoxically- non-safe.

  • Rehmannia glutinosa is one of the most important medicinal crops in Korea. However, various plant pathogens, including Fusatium spp., cause great damage on R. glutinosa and result in enormous economic losses. This study was conducted to breed Fusarium-resistant plants by using Agrobacterium tumefaciences and AFP (anti-fungal protein) gene. The plant material used was a native accession of R. glutinosa. The PCR analysis was conducted to verify transgenicity. Based on the PCR analysis, nptII band was observed in transgenic plant genome. Southern blot and AFP protein analyses also showed the expression of this gene in transgenic plants. Expression of AFP in transgenic plants offers the possibility of developing resistance to fungal infection.

Changes in Antioxidant Activity of Rehmannia radix Libosch with Heat Treatment

  • This study evaluated the effects of heat treatment on antioxidant activity of Rehmannia radix Libosch (RRL). RRL was heated at various temperatures ($110-150^{\circ}C$) for various times (1-5 hr), and the total polyphenol, flavonoid content, and antioxidant activity were investigated. With increased heating temperature and exposure time, total content of polyphenol, flavonoid, as well as antioxidant activity increased. The highest total polyphenol and flavonoid contents were 21.65 and 3.56 mg/g, respectively, these values were occurred after heating for 3 hr at $150^{\circ}C$ (RRL was 5.09 and 0.83 mg/g, respectively). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest value of 83.46% after heating for 3hr at $150^{\circ}C$. The 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation radical scavenging activity was highest value of 20.43mg ascorbic acid (AA) eq/g after heating for 2 hr at $150^{\circ}C$. There were highly significant differences in the total polyphenol, flavonoid content, and antioxidant activity among heating temperatures and times (p<0.001), with heating temperature having the greater effect.

Charactrization of Biological Activities of Rehmannia glutinosa Extracts (숙지황(Rehmannia glutinosa) 추출물의 생리활성)

  • The content phenolic compounds in extracts from Rehmannia glutinosa was the highest in 40% ethanol extracts as $5.1{\pm}0.2mg/g$. DPPH scavenging activity of R. glutinosa extracts was high in water extracts and 40% ethanol extracts as 85~93%, ABTS radical cation decolorization of water extracts and 40% ethanol extracts was about the same as 55~62%, antioxidant protection factor (PF) was confirmed in water extracts and 40% ethanol extracts as 1.6~1.9 PF, and TBARs of water extracts and 40% ethanol extracts were concluded to have the similar antioxidant effects. The hypertension inhibitory activity of water extracts and 40% ethanol extracts from R. glutinosa indicated the activities as 87.2% and 81.1%, anti-gout activity was determined very low in R. glutinosa extracts and antimicrobial activity against skin microorgasm was confirmed, and tyrosinase inhibitory activity was determined as 70.2% in 40% ethanol extracts, it was expected the whitening effects in 40% ethanol extracts. The elastase inhibitory activity which are related to the wrinkle cause was observed in water extracts and 40% ethanol extracts as 76.2% and 57.2%. The hyaluronidase inhibitory activity to R. glutinosa extracts was observed weakly in only 40% ethanol extracts of $200{\mu}g/ml$ phenolic content as 5.1%.
