Antiulcerogenic effects of Gymnosporia rothiana

  • Gymnosporia rothiana (walp) Lawson (celastraceae), commonly known as Maytenus rothiana, is used in Indian folk medicine as an antiulcerogenic agent. However, there have been no scientific reports regarding its antiulcer activity. Therefore, this study was intended to evaluate the antiulcer property of petroleum ether, chloroform, and methanolic extract of leaves of Gymnosporia rothiana at different dose levels in ethanol induced and indomethacin induced gastric ulcer models. It was observed that oral administration of all the extract of Gymnosporia rothiana produces significant reduction in ulcer lesion index as well as increase in volume and pH of gastric content in both experimental models, being petroleum ether extract the most effective at dose of 250 mg/kg; it significantly reduced gastric lesion index (70.06%), in comparison to omeprazole (71.20%) and methanolic extract at a dose of 500 mg/kg (67.22%). Increased gastric mucosal defense mechanism by petroleum ether extract is probably due to its high levels of terpenoids like $\beta$ amyrin, lupeol acetate. The present results clearly shows antiulcer effect of Gymnosporia rothiana against various irritants has been mainly due to cytoprotective effect mediated through prostaglandin and partly due to free radical scavenging activity.

  • The present study deals with the amelioration by Lagerstroemia speciosa Linn. leaf extract against hepatotoxicity induced by carbon tetrachloride ($CCl_4$), which was evaluated in terms of serum marker enzymes like serum glutamate pyruvate transaminase, serum glutamate oxaloacetate transaminase, alkaline phosphatase, serum total bilirubin, total protein levels along with concomitant hepatic and antioxidants like superoxide dismutase, catalase, glutathione, glutathione peroxidase and lipid peroxidation enzymes were monitored. These biochemical parameters altered by the single dose level of $CCl_4$ (0.75 ml/kg body weight, i.p). Pre treatment with L. speciosa prior to the administration of $CCl_4$, at the doses of 50 and 250 mg/kg. body weight/day, p.o. for 7 days, significantly restored all the serum and liver tissue parameters near to the normal levels, respectively. Silymarin was used as a reference standard, prior to the administration of $CCl_4$ to rats. These findings indicate the protective potential of L. speciosa against hepato toxicity which possibly involve mechanism related to its ability of selective inhibitors of (reactive oxygen species like antioxidants brought about significant inhibition of TBARS suggesting possible involvement of $O_2{\cdot}-$, $HO_2{\cdot}$, and ${\cdot}OH$. In conclusion, the amelioration may be attributed to the synergistic effects of its constituents rather than to any single factor as the leaves are rich in tannins, sterols, flavonoids, saponins etc.

Antioxidant Effect of Bee Venom Herbal Acupuncture (봉독약침액의 항산화 효능)

  • Bee venom has been clinically used to control the pain of inflammation disease etc. in general. Although the effect of bee venom herbal acupuncture has been reported, its mechanism has not been fully elucidated yet. Free radical metabolism seems to occupy a remarkably common position in the mechanisms of inflammation and ageing related disease. Oxidative damage to DNA, lipids, proteins and other molecules may contribute to the development inflammation disease. NO or DPPH is one of the free radicals and a mediator in inflammation diseases. The objective of this study is to investigate the scavenging effect of bee venom herbal acupuncture against NO and DPPH. The followings are the summary of the results: (1) There is no significant scavenging effect of bee venom herbal acupuncture on NO in BVS and BVP group. (2) There is a significant scavenging effect of bee venom herbal acupuncture on DPPH in BVS-1 and BVP-1 group. These results suggest that bee venom herbal acupuncture can be used for inflammation diseases such as rheumatoid arthritis. This study would provide important basic data on the possibility of the clinical treatment of bee venom herbal acupuncture. Further studies are required to investigate the antioxidative effects of it.

Structure of Edge Flame in a Methane-Oxygen Mixing Layer (메탄/순산소 혼합층에서 edge flame의 구조)

  • Structure of edge flame established in a mixing layer, formed between two uniformly flowing pure $CH_4$ and pure $O_2$ streams, is numerically investigated by employing a detailed methane-oxidation mechanism. The numerical results exhibited the most outstanding distinction of using pure oxygen in the fuel-rich premixed-flame front, through which the carbon-containing compound is found to leak mainly in the form of CO instead of HC compounds, contrary to the rich $CH_4-air$ premixed flames in which $CH_4$ as well as $C_2H_m$ leakage can occur. Moreover, while passing through the rich premixed flame, a major route for CO production, in addition to the direct $CH_4$ decomposition, is found to be $C_2H_m$ compound formation followed by their decomposition into CO. Beyond the rich premixed flame front, CO is further oxidized into $CO_2$ in a broad diffusion-flame-like reaction zone located around moderately fuel-rich side of the stoichiometric mixture by the OH radical from the fuel-lean premixed-flame front. Since the secondary CO production through $C_2H_m$ decomposition has a relatively strong reaction intensity, an additional heat-release branch appears and the resulting heat-release profile can no longer be seen as a tribrachial structure.

  • The phase-transfer polymerization of methyl methacrylate with tricaprylmethylammonium chloride-$Na_2S_2O_4-CCl_4$ initiator system was investigated in an aqueous-organic two-phase system. The observed rates of polymerization were compared with those obtained from the polymerization mechanism proposed with a cyclic phase-transfer initiation step. The rate of polymerization was found to be proportional to the concentration of $Q^+$ and square root of ${S_2O_4}^{-2}$ in the aqueous solution and the feed quantity of $CCl_4$ and MMA.

  • Abies nephrolepis(Pinaceae) extracts were tested for determined immune system regulating activity based on antiinflammatory activity, antioxidant activity and anti-proliferative effect on HeLa cell line. The A. nephrolepis extracts increased dose-dependently anti-proliferation of HeLa cell line. The DM fraction of the extracts having anti-proliferatative effects of HeLa cell line was fractionalized four subfractions($D1{\sim}D4$). Inflammation-induced NO production was inhibited by D2 and D4 in LPS-activated RAW264.7 macrophages. And also, this fractions showed antioxidant activity examined by DPPH radical scavenging effects. These results suggest that the potential use of DM fraction of A. nephrolepis in chemoprevention and regulation overproduction of NO on pathogenic conditions. The mechanism of the inflammatory effects, however, must be evaluated through various parameters in the induction cascade of NO production.

  • Quantum information processing using superconducting qubit based on Josephson junction has become one of the most promising candidates for possible realization of a quantum computer. In the heart of the qubit circuits, the superconducting microwave resonator plays a key role in quantum operations and measurements, which enables single-photon level microwave quantum optics. During last decade, the coherence time, or the lifetime of the quantum state, of the superconducting qubit has been dramatically improved. Among several technological innovations, the improvement of superconducting microwave resonator's quality has been the main driving force in getting the qubit performance almost ready for elementary quantum computing architecture. In this paper, I will briefly review very recent progresses of the superconducting microwave resonators especially aimed for quantum device applications during the last decade. The progresses have been driven by ingenious circuit design, material improvement, and new measurement techniques. Even a rather radical idea of three-dimensional large resonators have been successfully implemented in a qubit circuit. All those efforts contributed to our understanding of the qubit decoherence mechanism and as a result to the improvement of qubit performance.

  • A novel, polysaccharide-based, superabsorbent hydrogel was synthesized through crosslinking graft copolymerization of methacrylic acid (MAA) onto kappa-carrageenan ($_{k}C$), using ammonium persulfate (APS) as a free radical initiator in the presence of methylenebisacrylamide (MBA) as a crosslinker. A proposed mechanism for $_{k}C$­g-polymethacrylic acid ($_{k}C$-g-PMAA) formation was suggested and the hydrogel structure was confirmed using FTIR spectroscopy. The effect of grafting variables, including MBA, MAA, and APS concentration, was systematically optimized to achieve a hydrogel with the maximum possible swelling capacity. The swelling kinetics in distilled water and various salt solutions were preliminarily investigated. Absorbency in aqueous salt solutions of lithium chloride, sodium chloride, potassium chloride, calcium chloride, and aluminum chloride indicated that the swelling capacity decreased with increased ionic strength of the swelling medium. This behavior can be attributed to the charge screening effect for monovalent cations, as well as ionic crosslinking for multivalent cations. The swelling of super absorbing hydrogels was measured in solutions with pH ranging from 1 to 13. In addition, the pH reversibility and on-off switching behavior, at pH levels of 3.0 and 8.0, give the synthesized hydrogels great potential as an excellent candidate for the controlled delivery of bioactive agents.

A New Chemiluminescence Method for Determination of Cytosine Arabinoside in Pharmaceutical Preparations

  • A novel chemiluminescence (CL) system was established for the determination of cytosine arabinoside (Ara-C) in pharmaceutical preparations. It was showed that a clear CL signal was observed when Eosin Y mixed with Fenton reagent. The CL intensity was decreased significantly when Ara-C was added to the reaction system and partially scavenged the hydroxyl radicals in the solution. The extent of decrease in the CL intensity had a good stoichiometrical relationship with the Ara-C concentration. Based on this, we developed a new method for the determination of Ara-C using a flow injection analysis (FIA) technique with CL detection. Under the optimal conditions, the linear range of Ara-C concentration was $6.0{\times}10^{-9}\sim1.0{\times}10^{-7}mol/L$ (R = 0.9982) with a detection limit of $7.6{\times}10^{-10}mol/L$ (S/N=3), the RSD was 5.6% for $6.0{\times}10^{-8}mol/L$ Ara-C (n = 11). The method was successfully applied to the determination of Ara-C in injection samples. The possible chemiluminescence reaction mechanism was discussed.

$N_2$-Laser Flash Photolysis study of Photosensitizing Properties of Benoxaprofen ($N_2$-레이저 플래쉬 광분해 방법에 의한 Benoxaprofen의 광증감 작용에 관한 연구)

  • The nitrogen-laser flash photolysis apparatus has been set up to detect directly the transient produced from the excited Benoxaprofen (BXP). The function of the instrument has been tested by using anthracene as a standard sample. The laser photoexcitation of BXP in ethanol has been carried out under a constant stream of $N_2$ gas and a transient absorption spectrurn has been recorded over the 400-600 nm. The effects of oxygen and ${\beta}-carotene$ on the transient spectrum reveal that thee maximum transient absorption at 415 nm is attributed to the triplet state of BXP and the triplet energy of BXP is above 22 Kcal/mole. In addition to the triplet state, another transient species has been observed to be produced, which is probably a radical form of BXP. The photosensitization mechanism of BXP is discussed based on these results.
