Isolation and characterization of EST-SSR markers for Astilboides tabularis (Saxifragaceae), endangered species in Korea

  • Genetic assessments of rare and endangered species are among the first steps necessary to establish the proper management of natural populations. Transcriptome-derived single-sequence repeat markers were developed for the Korean endangered species Astilboides tabularis (Saxifragaceae) to assess its genetic diversity. A total of 96 candidate microsatellite loci were isolated based on transcriptome data using Illumina pair end sequencing. Of these, 26 were polymorphic, with one to five alleles per locus in 60 individuals from three populations of A. tabularis. The observed and expected heterozygosity per locus ranged from 0.000 to 0.950 and from 0.000 to 0.741, respectively. These polymorphic transcriptome-derived simple sequence repeat markers would be invaluable for future studies of population genetics and for ecological conservation of the endangered species A. tabularis.

Ecological Characteristics and Distribution of Native Phytolacca insularis in Ulleung Island (울릉도의 섬자리공 분포와 자생지의 생태적 특성)

  • This study was conducted to investigate the native Phytolacca insularis in Ulleung Island for their distribution and ecological characteristics. P. insularis, called as "Sum-Ja-Ri-Gong", is very rare plant which is only restricted in Ulleung Island. It is a plant out of 217 endangered plant species designated by the Korea Forest Service (1996). The native sites were discovered several plants in the small communities at the area from 32m to 116m above the sea level in the seashore of Ulleung Island. The average vegetation height of herb layer in the native sites was 0.77m and average coverage percentage of vegetation layer was 75.56%. Average 11.89 species were emerged on a plot. P. insularis has a tendency to emerged with Galium spurium var. echinospermon and Artemisia lavandulaefolia community.

Resource Plants of Mt. Midong in Chungcheongbuk-do, Korea (충청복도 미동산의 자원식물상)

  • This study was carried out to investigate the resource plants of Mt. Midong. The resource plant of Mt. Midong was surveyed from March, 2002 to October, 2003. In the result of survey, the vascular plant species in Mt. Midong were summarized as 400 taxa; 85 families, 266 genera, 349 species, 48 varieties and 3 forms. And in the results of survey on resource plants among 400 taxa, we confirmed 162 taxa of ornamental plants(40.5%), 250 taxa of edible plants(62.5 %), 257 taxa of medicinal plants(64.3%) and 167 taxa of the others(41.8 %). The six taxa were recorded in the surveyed sites as the Korean endemic plants; Hytomecon hylomeconoides, Filipendula palmata var. glabra, Ajuga spectabilis, Weigela subsessilis, Cirsium setidens and Saussurea seoulensis. The rare and endangered plants designated by Korea Forestry Service were two taxa; Hylomecon hylomeconoides and Polygonatum stenophyllum.

The Flora of Vascular Plants and Vegetation Status of Geojedo (거제도의 관속식물상과 주요 식생 현황)

  • The flora of vascular plants in Geojedo revealed to be 550 taxa based on only the voucher specimens to be collected. It comprise 108 families, 341 genera, 478 species, 69 varieties and 3 forms respectively. Among them, 13 taxa of Korean endemics and 8 taxa of rare and endangered plants were investigated in addition to 75 taxa of specially designated plants announced by the Ministry of Environment. Furthermore, 36 taxa of induced and naturalized plants were also observed in this area. One of the characteristics of forest vegetation is the broadly occupation of Pinus thunbergii on whole area of Geojedo. Although it's well condition and conservation status, the destruction area of vegetation was rapidly increasing by the industrial development, the deforestation and the tramp as well as by the basic disturbances of human beings. Therefore, more active and positive conservation strategy such as expansion of conservation area should be prepared rapidly by the results of long-term monitoring studies.

Distribution of vascular plant in Mt. Cheonsung, Yangsan (양산 천성산지역 관속식물의 분포)

  • 588 taxa of vascular plants including 121 families, 399 genera, 510 species, 67 varieties, 2 subspecies, 7 forma, and 2 hybrid species were investigated in Mt. Cheonsung. Among them, 116 species were newly added in this research. The area for Gyeongbu High-speed Railway passes Mujechi 3, 4, 5, and 6 and Daeseongsa around the wetlands were investigated 6 species of rare and endangered plants, 9 species of endemic plants and 37 species of special flora plants in Mt. Cheonsung. Appear due to the construction of the Gyeongbu High-speed Railway vegetation changes, especially about the impact of wetland plants for long-term monitoring is expected to need. Moreover, plant preservation plan about water source change in upper wetland should be established.

Genomic Analysis of the Xanthoria elegans and Polyketide Synthase Gene Mining Based on the Whole Genome

  • Xanthoria elegans is a lichen symbiosis, that inhabits extreme environments and can absorb UV-B. We reported the de novo sequencing and assembly of X. elegans genome. The whole genome was approximately 44.63 Mb, with a GC content of 40.69%. Genome assembly generated 207 scaffolds with an N50 length of 563,100 bp, N90 length of 122,672 bp. The genome comprised 9,581 genes, some encoded enzymes involved in the secondary metabolism such as terpene, polyketides. To further understand the UV-B absorbing and adaptability to extreme environments mechanisms of X. elegans, we searched the secondary metabolites genes and gene-cluster from the genome using genome-mining and bioinformatics analysis. The results revealed that 7 NR-PKSs, 12 HR-PKSs and 2 hybrid PKS-PKSs from X. elegans were isolated, they belong to Type I PKS (T1PKS) according to the domain architecture; phylogenetic analysis and BGCs comparison linked the putative products to two NR-PKSs and three HR-PKSs, the putative products of two NR-PKSs were emodin xanthrone (most likely parietin) and mycophelonic acid, the putative products of three HR-PKSs were soppilines, (+)-asperlin and macrolactone brefeldin A, respectively. 5 PKSs from X. elegans build a correlation between the SMs carbon skeleton and PKS genes based on the domain architecture, phylogenetic and BGC comparison. Although the function of 16 PKSs remains unclear, the findings emphasize that the genes from X. elegans represent an unexploited source of novel polyketide and utilization of lichen gene resources.

The Specific Plant Species and Conservation of the Bukhansan National Park (북한산국립공원의 특정식물과 그 보전 대책)

  • The Specific Plant Species such as N degree (2 species) and V degree (5 species) which is categorized by the Ministry of Environment are appeared as 7 species. II degree species among Endangered species of wild flora is Abeliophyllum distichum, and the Rare and Endangered Species which is categorized by the Korea Forest Service are enlisted as the five taxa. The Bukhansan is one of the phytogeographic ally important areas among national parks of Korea because this region is located in the northern limit of Southern Plant Types, such as Carpinus laxiflora, Cornus kousa, Styrax japonica and Zanthoxylum piperitum. The flora of the area was enlisted as 638 taxa, 103 families, 362 genera, 541 species, 87 varieties, and 10 forms. Naturalized Species was identified as 58 taxa, 16 families, 52 genera, 54 species and 4 varieties. The Ratio of Naturalized and Urbanization Index was 9.09% and 20.21%, respectively.

The Flora of Coastal Dune Area in Shinduri, Korea (신두리 해안 사구 지역의 식물상)

  • This study was carried out to investigate and analyze the vascular flora and injury status in Shinduri coastal dune area $(N36^{\circ}\;49',\;E126^{\circ}\;11')$, where designated as Natural Monuments and the result are as follows. The vascular plants identified and surveyed were 125 taxa including 108 species, 14 varieties, 2 forms and 1 hybrid of 92 genera of 40 families and there are 20 taxa including 17 species, 2 varieties and 1 hybrid of 15 genera of 10 families for woody plants, while 105 toxa including 91 species, 12 varieties and 2 forms of 78 genera of 34 families for the herbaceous plants at the . study sites. Seventeen taxa including 17 species of 15 genera of 9 families were investigated for naturalized plants and ratio of naturalization was 13.6 percents at the study sites. Glehnia littorolis Fr. Schm. and Lilium callosum S. et Z. appeared as rare and endangered plants designated in Korea Forest Service. Nine taxa in degree 1, 2 taxa in degree 2, 4 taxa in degree 3, 2 taxa in degree 4 and 2 taxa in degree 5 were represented for especialized species.

Vascular Plants Distributed in Bukcheon Stream, Urban Stream in Gyeongju-si (경주시의 도시하천인 북천에 분포하는 관속식물상)

  • The purpose of this study is to present the basic data for maintenance and management of river ecosystem by surveying and analysing the kinds and their characteristics of vascular plants in Bukcheon stream, Gyeongju-si, South Korea. The numbers of vascular plants were summarized as 428 taxa including 92 families, 265 genera, 3 subspecies, 24 varieties, 6 forms, 5 hybrids and 5 cultivars. Among the identified taxa, there were 90 planted species. The endangered wild species was Cicuta virosa, and the rare plants were 5 taxa including Prunus × yedoensis, Juniperus chinensis L. var. sargentii and so on. The Korean endemic plants were 5 taxa Populus × tomentiglandulosa, Salix koriyanagi, Lespedeza maritima, Forsythia koreana and Aster pseudoglehni. The specific plants by floristic region were 26 taxa including 2 taxa of grade V, 3 taxa of grade IV, 2 taxa of grade III, 8 taxa of grade II and 11 taxa of grade I. The naturalized plants were 81 taxa including Euphorbia maculata, Erigeron strigosus, Vulpia myuros and so on. The invasive alien plants were 7 taxa including Humulus scandens, Rumex acetosella, Ambrosia artemisiifolia, Ambrosia trifida, Lactuca seriola, Symphyotrichum pilosum and Paspalum distichum.

The Flora and Rare Plants in Kayasan National Park (가야산국립공원의 관속식물상 및 희귀식물)

  • The flora of the Kayasan National Park was surveyed from March to September, 1997. This area has very rich and diverse vascular plants; 613 taxa with 102 families, 323 genera, 522 species, 1 subspecies, 83 varieties, 5 forms and 2 hybrids. The rare and endangered plants species such as Lilium cernum Kom., Thymus quiquecostatus Celak., Cypripedium macranthum Sw., Tricyrtis dilatata Nakai, Abies koreana Wils., etc. were recorded. The plant species of 100 taxa such as Typha orientalis Presl, Corydalis maculata B. Oh et Y. Kim, Ajuga spectabilis Nakai, etc. were recorded for the first time in the area. The introduced plant species were found along the traffic road sides. The plant species of 14 taxa such as Lilium cernum Kom., Thymus quiquecostatus Celak., Abies koreana Wils., etc. which categorized as rare and endangered plants by the Office of Forestry were also recorded in the area(This species were mainly distributed in the habitats where easily threatened by human's activities).
