The objectives of the trial were to investigate the effects of supplementing vitamin E (VE) on nutrient digestion, nitrogen (N) retention and plasma parameters of beef cattle in feedlot. Four growing Simmental bulls, fed with a total mixed ration composed of corn silage and concentrate mixture as basal ration, were used as the experimental animals. Four levels of VE product, i.e. 0, 150, 300, 600 mg/head/d (equivalent to 0, 75, 150, 300 IU VE/head/d), were supplemented to the basal ration (VE content 38 IU/kg dry matter) in a $4{\times}4$ Latin square design as experimental treatments I, II, III and IV, respectively. Each experimental period lasted 15 days, of which the first 12 days were for pretreatment and the last 3 days for sampling. The results showed that supplementing VE did not affect the nutrient digestibility (p>0.05) whereas decreased the urinary N excretion (p<0.01), increased the N retention (p<0.05) and tended to increase the microbial N supply estimated based on the total urinary purine derivatives (p = 0.057). Supplementing VE increased the plasma concentrations of VE, glucose and triglycerol (TG) (p<0.05) and tended to increase the plasma concentration of total protein (p = 0.096) whereas did not affect the plasma antioxidant indices and other parameters (p>0.05). It was concluded that supplementing VE up to 300 IU/head/d did not affect the nutrient digestibility whereas supplementing VE at 150 or 300 IU/head/d increased the N retention and the plasma concentrations of VE and TG (p<0.05) of beef cattle.
The crystal and molecular structure of theophylline hydrochloride has been determined from X-ray data by Patterson techniques. The structure has been refined by block-diagonal least-squares and Fourier synthesis on three dimensional data. The unit cell is orthorhombic, space group $P_{na21}$, with a = 14.01, b = 11.49, c = 6.77${\AA}$, and contains four molecules. The final R value based on 743 observed reflexions is 12.2%. The intramolecular distances are similar to those in other compounds containing a purine or pyrimidine group. The molecules are nearly planar and are stacked in layers parallel to the (001)plane. The chlorine atom is coordinated to N(1) atom at a distance of 3.06${\AA}$. The structure is stabilized mainly by van der Waals interactions; however, a short N${\cdot}{\cdot}{\cdot}$Cl contact of length $3.06\AA$, which is slightly less than the expected van der Waals separation, suggest that weak charge transfer interaction may be present. The relationship between this structure and the known structures of theophylline monohydrate and caffeine monohydrate are discussed.
This study aimed to investigate dietary concentrate:forage ratios (C:F) and undegraded dietary protein (UDP) on nitrogen balance and urinary excretion of purine derivatives (PD) in lambs. Four Dorper${\times}$thin-tailed Han crossbred castrated lambs with $62.3{\pm}1.9$ kg body weight at 10 months of age were randomly assigned to four dietary treatments in a $2{\times}2$ factorial arrangement of two levels of C:F (40:60 and 60:40) and two levels of UDP (35% and 50% of CP), according to a complete $4{\times}4$ Latin-square design. Each experimental period lasted for 19 d. After a 7-d adaptation period, lambs were moved into individual metabolism crates for 12 d including 7 d of adaption and 5 d of metabolism trial. During the metabolism trial, total urine was collected for 24 h and spot urine samples were also collected at different times. Urinary PD was measured using a colorimetric method and creatinine was measured using an automated analyzer. Intake of dry matter (DM) (p<0.01) and organic matter (OM) (p<0.01) increased as the level of UDP decreased. Fecal N was not affected by dietary treatment (p>0.05) while urinary N increased as the level of UDP decreased (p<0.05), but decreased as dietary C:F increased (p<0.05). Nitrogen retention increased as dietary C:F increased (p<0.05). As dietary C:F increased, urinary excretion of PD increased (p<0.05), but was not affected by dietary UDP (p>0.05) or interaction between dietary treatments (p>0.05). Daily excretion of creatinine was not affected by dietary treatments (p<0.05), with an average value of $0.334{\times}0.005$ mmol/kg $BW^{0.75}$. A linear correlation was found between total PD excretion and PDC index ($R^2$ = 0.93). Concentrations of creatinine and PDC index in spot urine were unaffected by sampling time (p>0.05) and a good correlation was found between the PDC index (average value of three times) of spot urine and daily excretion of PD ($R^2$ = 0.88). These results suggest that for animals fed ad libitum, the PDC index in spot urine is effective to predict daily excretion of PD. In order to improve the accuracy of the spot sampling technique, an appropriate lag phase between the time of feeding and sampling should be determined so that the sampling time can coincide with the peak concentration of PD in the urine.
Four rumen-fistulated crossbred beef cattle (Brahman native) were randomly assigned according to a $4{\times}4$ Latin square design experiment to be fed plant herb supplements in their concentrate mixture. The treatments were: without herb supplementation (Control), lemongrass meal supplementation at 100 g/d (L), lemongrass meal supplementation at 100 g/d plus peppermint powder at 10 g/d (LP), and lemongrass meal supplementation at 100 g/d plus peppermint powder at 10 g/d with garlic powder 40 g/d (LPG), respectively. Based on the present study, the DMI and apparent digestibility of DM, OM, aNDF and ADF were not affected by dietary herb supplementation while CP digestibility tended to be decreased by herb supplement. Moreover, $NH_3$-N and BUN were decreased in all herb supplemented treatments and there was a tendency to an increase in ruminal pH in all herb supplemented groups. While there was no change in TVFA and C4 among lemongrass treatments, C2 was decreased in all herb supplemented treatments while C3 was increased. Methane production by calculation was the lowest in the LP and LPG groups. Population sizes of bacteria and protozoa were decreased in all herb supplemented groups, but not fungal zoospores. In all supplemented groups, total viable and proteolytic bacteria were decreased, while amylolytic and cellulolytic bacteria were similar. More importantly, in all herb supplemented groups, there were higher N balances, while there was no difference among treatments on purine derivative (PD) excretion or microbial N. Based on the results above, it could be concluded that there was no negative effect on ruminal fermentation characteristics and nutrient utilization by plant herb supplement, but protozoal population and $CH_4$ production were reduced. Thus, lemongrass alone or in combination with peppermint and garlic powder could be used as feed additives to improve rumen fermentation efficiency.
The effects of four types of tropical protein-rich trees on nutrient digestibility, nitrogen (N) balance, urinary purine derivative (PD) excretion and blood metabolites in four Thai Brahman cattle (290${\pm}$2.5 kg) were studied. The animals were fed twice daily, with each feeding consisting of 1 kg (fresh weight) rice straw and one of the four dietary supplements: i) 1.98 kg oven-dried rain tree pods (RTP) and 20 g premix (RTPP), ii) 980 g RTP and 1 kg sun-dried leucaena leaves and 20 g premix (LLRT), iii) 980 g RTP and 1 kg sun-dried cassia leaves and 20 g premix (CLRT) and iv) 980 g RTP and 1 kg sun-dried mulberry leaves and 20 g premix (MLRT). The apparent dry matter (DM) and organic matter (OM) digestibilities were higher (p<0.05) in cattle fed the CLRT supplement than in those fed the other supplements, whilst the apparent digestibility of neutral detergent fibre (NDF) was higher (p<0.05) in cattle fed the CLRT and MLRT supplements than in those fed the other supplements. The N-balance of cattle fed LLRT and CLRT supplements was higher (p<0.05) than in cattle fed RTPP and MLRT supplements, whilst the apparent digestibility of N was highest (p<0.05) in cattle fed RTPP supplement, compared to the other supplements. Allantoin and PD excretion in the urine, and the ratios of allantoin/DOMI and PD/DOMI were higher (p<0.05) in cattle fed RTPP and MLRT than for those fed LLRT and CLRT supplements. Plasma ${\beta}$-hydroxy butyrate (${\beta}$-HBA) and insulin concentrations were higher (p<0.05) in cattle fed RTPP supplement than in those fed the other supplements. The study demonstrated the value of using local multipurpose trees (MPTs) to improve Brahman cattle feeding systems in the tropics.

Five male dairy goats (Saanen), 4.6 month old with a body weight of 21.4 ($SD{\pm}1.6$) kg, were used to examine 5 dietary urea treatments in a $5{\times}5$ Latin Square experimental design. The five levels of urea were 10, 20, 30, 40 and 50 g urea/kg DM of steam-treated oil palm fronds (OPF) and dry matter intake tended (p>0.05) to increase with increasing urea supplementation up to 30 g/kg OPF (77.7 g/kg $BW^{0.75}$), but decreased (p<0.05) with 40 and 50 g urea/kg OPF (67.4 and 63.7 g/kg BW0.75, respectively) supplementation. Similarly, dry matter, organic matter, crude protein, neutral detergent fiber and hemicellulose digestibilities increased (p<0.05) with the addition of urea to 30 g/kg OPF but thereafter decreased (p<0.05) with 40 and 50 g/kg OPF. Ruminal pH, ruminal $NH_3$-N concentration and plasma urea concentration increased linearly (p<0.01) and quadratically (p<0.01) as a consequence of addition of urea to the diet. Excretion of total purine derivatives (PD) by goats fed 30 g of urea/kg OPF was highest (p<0.05) followed by goats fed 20, 40, 10 and 50 g of urea/kg OPF. Microbial N (g N/day) and efficiency of microbial N supply expressed as g N/kg organic matter apparently digested in the rumen were higher (p<0.05) in goats fed 30 g of urea/kg OPF (5.5 g N/day and 22.0 g N/kg DOMR, respectively) than in goats on 10 and 50 g of urea/kg OPF treatments. However, the former did not differ from goats fed 20 g of urea/kg OPF (3.9 g N/day and 16.6 g N/kg DMOR, respectively). Ruminal VFA concentration, protein/energy ratio, N absorption and N retention increased (p<0.05) with the addition of urea to the diet up to 30 g/kg OPF but decreased (p<0.05) with 40 and 50 g/kg OPF. This implies that the optimal level of urea supplementation in an OPF based diet was about 30 g urea/kg OPF.

Until now, it has not been well known for Far-infrared(FIR) how to affect to the human body. We introduced and presumed the mechanism of FIR based on molecular biology in this study, as below. The human body is composed of proteins which get easily changed by a thermal factor (about 42 $^{\circ}$C over). FIR with low temperature can deeply penetrate on the human body composed things without troublesome, since FIR has effectively operated on the human body at low temperature (35-40 $^{\circ}$C). When FIR penetrated on the human body, it would inhibit the abnormal genes and cells expression, and then information of DNA and RNA would be reexpressed for arranging DNA and RNA abnormal state. As FIR's receptors in the body, it colud be presumed that N-glycosyl linkage of purine and deoxyribose, RNA splicing process, and heat shock protein.
In this study, we established a novel somatic embryogenesis and plant regeneration system through cell suspension culture of white dandelion (Taraxacum coreanum NAKAI.). Embryogenic calli could be initiated from leaf and root explants of sterile seedlings on solid Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) after 3-week cultures. To proliferate embryogenic calli rapidly, cell suspension culture was performed with transferred to liquid MS medium with various combinations of plant growth regulators (PGRs) including 2,4-D, ${\alpha}$-naphthalene acetic acid (NAA), indole-3-acetic acid (IAA), $N^6$-benzylamino purine (BAP), thidiazuron (TDZ), and kinetin. During suspension cultures, embryogenic calli not only greatly proliferated, but shoot organogenesis also simultaneously occurred from the surface of somatic embryos. Among them, TDZ at lower concentration, 0.1 mg/L produced the highest efficiency of somatic embryo formation and shoot organogenesis. Rooting of embryogenic calli with adventitious shoots was done on solid MS medium containing 0.1 mg/L NAA and 0.3% activated carbon. Nearly 80% of embryogenic calli with shoot organogenesis could be rooted normal. Well-rooted plantlets were transferred into pots under a greenhouse condition, and plants derived from this system appeared phenotypically normal.
Depurination, the release of purine bases from nucleosides by hydrolysis of the N-glycosidic bond, gives rise to alterations of the cell genome. Although, cells have evolved mechanisms to repair these lesions, unrepaired apurinic sites have been shown to have two biological consequences: lethality and base substitution errors. 2-Bromopropane (2-BP) is used as an intermediate in the synthesis of pharmaceuticals, dyes, and other organics. In addition, 2-BP has been used as a cleaning solvent in electronics industry. But, 2-BP was found to cause reproductive and hematopoietic disorders in local workers exposed to it. We observed massive depurination after incubation of 2'-deoxyadenosine (dA) and 2'-deoxyguanosine (dG) with the excess amount 2-BP at the physiological condition (pH 7.4, $37^{\circ}C$), which were analyzed by HPLC and LC-MS/MS. In addition, time and dose response relationship of depurination in dA and dG induced by 2-BP at the physiological condition were investigated.
Four Holstein$\times$Indigenous cows with ruminal canulas were used in a 4$\times$4 Latin square design with 28 d periods to determine the effect of replacing nonforage fiber source (NFFS) from cassava residues for non-fiber carbohydrates (NFC) on ruminal fermentation characteristics and milk production. Dietary treatments contained 17% forage neutral detergent fiber (FNDF) from corn silage and 0, 3, 6 and 9% nonforage NDF from cassava residues and 11% nonforage NDF from other NFFS, so that levels of nonforage NDF were 11, 14, 17 and 20% dry matter (DM). Intakes of DM and net energy for lactation, average daily gain and milk fat percentage were not different (p>0.05). Ruminal pH, ammonia concentrations, acetate to propionate ratios, 24 h in sacco fiber digestibility significantly increased with increasing contents of nonforage NDF from cassava residues. Concentrations of VFA, urinary excretion of purine derivatives, milk protein percentage, production of milk and 4% FCM significantly decreased. These results suggest that NFC in diets is one of the limiting factors affecting productivity of dairy cows in the tropics and thus NFFS is better used as partial replacements for FNDF.
