Carbon and Nitrogen Stocks of Trees and Soils in a 'Niitaka' Pear Orchard ('신고'배 재배지 내 수체 및 토양의 탄소 및 질소 저장량)

  • To report country-specific carbon and nitrogen stocks data in a pear orchard by Tier 3 approach of 2006 IPCC guidelines for national greenhouse gas inventories, an experimental pear orchard field of the Pear Research Station, National Institute of Horticultural & Herbal Science, Rural Development Administration, Naju, Korea ($35^{\circ}01^{\prime}27.70N$, $126^{\circ}44^{\prime}53.50^{\prime\prime}E$, 6 m altitude), where 15-year-old 'Niitaka' pear (Pyrus pyrifolia Nakai cv. Niitaka) trees were planted at a $5.0m{\times}3.0m$ spacing on a Tatura trellis system, was chosen to assess the total amount of carbon and nitrogen stocks stored in the trees and orchard soil profiles. At the sampling time (August 2012), three trees were uprooted, and separated into six fractions: trunk, main branches, lateral branches (including shoots), leaves, fruits, and roots. Soil samples were collected from 0 to 0.6 m depth at 0.1 m intervals at 0.5 m from the trunk. Dry mass per tree was 4.7 kg for trunk, 13.3 kg for main branches, 13.9 kg for lateral branches, 3.7 kg for leaves, 6.7 kg for fruits, and 14.1 kg for roots. Amounts of C and N per tree were respectively 2.3 and 0.02 kg for trunk, 6.4 and 0.07 kg for main branches, 6.4 and 0.09 kg for lateral branches, 6.5 and 0.07 kg for roots, 1.7 and 0.07 kg for leaves, and 3.2 and 0.03 kg for fruits. Carbon and nitrogen stocks stored between the soil surface and a depth of 60 cm were 138.29 and $13.31Mg{\cdot}ha^{-1}$, respectively, while those contained in pear trees were 17.66 and $0.23Mg{\cdot}ha^{-1}$ based on a tree density of 667 $trees{\cdot}ha^{-1}$. Overall, carbon and nitrogen stocks per hectare stored in a pear orchard were 155.95 and 13.54 Mg, respectively.

Expressional Analysis of Superoxide Dismutase in Olive Flounder (Paralichthys olivaceus) against Viral Hemorrhagic Septicemia Virus Infection (Viral hemorrhagic septicemia virus (VHSV) 감염에 대한 넙치 superoxide dismutase(Of-SOD)의 발현분석)

  • Superoxide dismutase is a family of important antioxidant metalloenzymes and catalyzes the dismutation of toxic superoxide anions into dioxygen and hydrogen peroxide. A recent study identified the partial superoxide dismutase (SOD) gene in olive flounder (Paralichthys olivaceus). The same study reported that it strongly induced benzo[a]pyrene and that it was an indicator of aquatic oxidative stress responses. However, its transcriptional response against viral infection has not been investigated. In the present study, the spatial and temporal expression profiles were analyzed to investigate the function of Of-SOD in the antiviral response. The Of-SOD transcripts were ubiquitously detected at various levels in diverse tissues in a real-time PCR. The expression of Of-SOD was significantly higher in the muscles, liver, and brain but extremely low in the stomach and spleen. Following a VHSV challenge, the expression of Of-SOD increased within 3 h in the kidneys and decreased to the original level 2 days postchallenge. In muscle, liver, and brain, Of-SOD mRNA was similarly up-regulated at 3-6 h postchallenge and then decreased to the basal level. Although the expression pattern and induction time differed slightly depending on the tissue, the transcript of Of-SOD consistently increased in the acute infection response, but the expression was low in the chronic response. The expression of Of-SOD was induced after the VHSV infection, and Of-SOD was probably involved in the immune response against the viral challenge. These results suggest that SOD may play important roles in the immune defense system of P. olivaceus and perhaps contribute to the protective effects against oxidative stress in olive flounder.

The Physico-chemical Changes and Sensory Characteristics of Kimchi Added with the Mashed Red Pepper (마쇄고추를 첨가한 김치의 이화학적 성분 변화 및 관능적 특성)

  • We investigated the physico-chemical and sensory characteristics of Kimchi made with red pepper that was washed and mashed. The pH of juice from Kimchi made with red pepper powder was the highest on the day of Kimchi preparation. In the case of Kimchi made with mashed red pepper, the pH of juice and liquid was lower than that of other samples. A similar decrease in pH of juice and liquid was observed up to the second week of fermentation, but the significant difference between both samples wasn't found. The total acidity of Kimchi with mashed red pepper was significantly increased during early fermentation, but was similar during the second week, compared with that of Kimchi with red pepper powder. From the third week of storage, both juice and liquid from Kimchi made with red pepper powder was relatively increased. L and a value of liquid was highest in the case of Kimchi made with mashed red pepper, but b value was lowest during fermentation. In the case of organic acids, acetic acid and lactic acid contents were increased in Kimchi made with mashed red pepper while fermentation progressed. In addition, citric acid content was constant up to the second week in Kimchi made with mashed red pepper but from the third week wasn't detected in both Kimchi made with mashed red pepper and Kimchi made with red pepper powder. In the case of QDA(Quantitative Descriptive Analysis) profiles, the values of Kimchi made with mashed red pepper were significantly higher than those of Kimchi made with red pepper powder in respect to redness, pungency and fresh flavor immediately after the preparation of Kimchi and during the second week of fermentation, but during the fifth week the values were higher in respect to redness and fresh flavor of Kimchi. Appearance and overall acceptability was remarkably increased in Kimchi made with mashed red pepper, compared with that of Kimchi made with red pepper powder immediately after pickling, during the second and the fifth week of fermentation. Therefore, these results indicate that mashed red pepper increased more citric acid content, L and a value of Kimchi in comparison with red pepper powder, resulting in the good effects on overall acceptability due to the significant increase of redness and fresh flavor.

Effects of Dietary ${\omega}$-3 and ${\omega}$-6 Polyunsaturated Fatty Acids on Fatty Acid Composition of Immune Organs in Young Chicks (${\omega}$-3 및 ${\omega}$-6계 지방산 첨가 사료의 급여가 어린 병아리에서 면역기관 내 지방산 조성에 미치는 영향)

  • Effects of various combinations of corn oil (CO) and perilla oil (PO) as respective dietary sources of ${\omega}$-6 and ${\omega}$-3 polyunsaturated fatty acids on fatty acid profiles of immune organs were studied in young chicks. Seventy-five 1-day-old male (ISA Brown) chicks were assigned to five treatments with three replications. Semi-purified-type diets containing glucose and soybean meal as major ingredients were added with 8% CO, 6% CO+2% PO, 4% CO+4% PO, 2% CO+6% PO and 8% PO and fed for 7 weeks. There were no significant differences in body weight gain, feed intake and relative weights of liver and immune organs (g/100g weight) among dietary groups. Dietary fatty acid patterns were generally reflected in the fatty acid compositions of all immune organs such as spleen, thymus and bursa of Fabricius. The levels of a-linolenic acid(LNA), eicosapentaenoic acid (EPA) and docosahexaenoic acid in various immune organs increased with increasing levels of perilla oil in the diets, whilet the levels of linoleic acid (LA) and arachidonic acid (AA) decreased. Thymus appeared to have capacity to retain remarkably higher (P<0.05) levels of LA and LNA up to 37 and 22%, respectively, compared to the other organs. Thymic tissue contained ${\omega}$-3 fatty acid and ${\omega}$-6 fatty acid 10~36 times and 3~5 times higher than the other organs, respectively. Spleen tissue was specifically higher (P<0.05) in the levels of AA and EPA and the ratios of AA/LA and EPA/LNA, compared to the other organs, suggesting that the tissue might have high desaturase activity to convert LA or LNA to AA or EPA, respectively. BSA antibody production tended to increase by 18 ~ 32% with higher levels of perilla oil in diet, although the increase was not statistically significant. In conclusion, fatty acid compositions of immune organs very depending on the lipid composition of the diets and each organ appears to respond differently for its fatty acid profile to dietary lipids. Considering AA and EPA are precursors of many important eicosanoids, further studies are required to clarify the responses of the immune organs to the dietary fatty acids.

Growth Performance and Carcass Characteristics of Two Different Broiler Strains by Different Levels of Metabolizable Energy (사료 내 대사 에너지 수준 차이가 계통이 다른 육계의 성장과 도체 특성에 미치는 영향)

  • The objective of this study was to evaluate the effects of different levels of dietary ME on growth performance and carcass characteristics in two different strains of broiler chicken. A total of one thousand, 1-day-old A strain and R strain male chicks were randomly assigned into 8 treatments in a $2{\times}4$ factorial arrangement. They were fed iso-nitrogenic (CP 21%) crumbled diets formulated to contain metabolizable energy (ME) 2,950 to 3,250 kcal/kg in increment of 100 kcal/kg in the starter phase (1 to 21d) and iso-nitrogenic (CP 19%) pelleted diets containing same ME levels as in the finishing phase (22 to 38d). The body weight (BW) gain of chicks fed the lower ME diets (2,950 or 3,050 kcal/kg) were higher than those of the higher ME groups. The dietary energy level showed significant effects on feed intake and feed conversion rate (FCR) from 1 to 38 days of age (p<0.05). With the increment of dietary energy, feed intake tended to be reduced, whereas FCR was improved in the two strains of broiler chickens. The lowest FCR was observed at 3,250 kcal/kg diet groups in both of the two strains from 1 to 38 days of age. Feed intake and BW gain during 38 days were significantly affected by the strain factor. Increasing dietary energy up to 3,250 kcal/kg had no effect on the relative weights of breast meat and abdominal fat. The dietary energy and strains showed significant effects on the dressing percentage. There were no significant differences in various blood profiles except for GPT activity.

Stereotactic Target Point Verification in Actual Treatment Position of Radiosurgery (방사선수술시 두개내 표적의 정위적좌표의 치료위치에서의 확인)

  • Purpose : Authors tried to enhance the safety and accuracy of radiosurgery by verifying stereotacitc target point in actual treatment position prior to irradiation. Materials and Methods : Before the actual treatment, several sections of anthropomorphic head phantom were used to create a condition of unknown coordinates of the target point. A film was sandwitched between the phantom sections and punctured by sharp needle tip. The tip of the needle represented the target point. The head phantom was fixed to the stereotactic ring and CT scan was done with CT localizer attached to the ring. After the CT scanning, the stereotactic coordinates of the target point were determined. The head phantom was secured to accelerator's treatment couch and the movement of laser isocenter to the stereotactic coordinates determined by CT scanning was performed using target positioner. Accelerator's anteroposterior and lateral portal films were taken using angiographic localizers. The stereotactic coordinates determined by analysis of portal films were compared with the stereotactic coordinates previously determined by CT scanning. Following the correction of discrepancy the head phantom was irradiated using a stereotactic technique of several arcs. After the irradiation, the film which was sandwitched between the phantom sections was developed and the degree of coincidence between the center of the radiation distribution with the target point represented by the hole in the film was measured. In the treatment of the actual patients, the way of determining the stereotactic coordinates with CT localizers and angiograuhic localizers was the same as the phantom study. After the correction of the discrepancy between two sets of coordinates, we proceeded to the irradiation of the actual patient. Results : In the phantom study, the agreement between the center of the radiation distribution and the localized target point was very good. By measuring optical density profiles of the sandwitched film along axes that intersected the target point, authors could confirm the discrepancy was 0.3 mm. In the treatment of an actual patient, the discrepancy between the stereotactic coordinates with CT localizers and angiographic localizers was 0.6 mm. Conclusion : By verifying stereotactic target point in actual treatment position prior to irradiation, the accuracy and safety of streotactic radiosurgery procedure were established.

Development of Heat Exchanger for Fermentation Heat Utilization from Waste Woody Biomass (목질계 폐바이오메스의 발효열이용 열교환기의 개발)

  • It is urgently required to develop the production of fermentation-heat energy from the waste agricultural and forest biomass and its effective heat exchanging system for the supply of warm water to rural households and greenhouses. In this study 3 helical-type and 1 plate-type heat exchangers using 3 different waste biomasses [e.g. hardwood (HW) sawdust (100%), softwood (SW) sawdust : HW sawdust (50 : 50) and HW sawdust : grass (90 : 10)] were applied in order to find out the best heat recovery system. The heat exchanger was basically considered to improve the overall heat recovery efficiency, to minimize heat loss and to simplify manufacturing, assembling and breaking up the fermenting beds. The helical-type heat exchanger (HX-H3) installed in fermenting bed of HW sawdust : grass (90 : 10) showed relatively higher temperature profiles, in particular mid- and upper-parts than lower and surface parts during 45-day fermentation process. The maximum temperature was ranged from $40^{\circ}C$ to $65^{\circ}C$ with average $60^{\circ}C$. The water temperature of tank outlet was ranged to $33{\sim}48^{\circ}C$ during whole measuring periods. By the way plate-type one (HX-P) installed in same biomass compositional fermenting bed showed $64.5{\sim}76.5^{\circ}C$ at center part, and $43{\sim}56^{\circ}C$ and $42{\sim}58^{\circ}C$, water tank and tank outlet temperatures, respectively, during 100 day measurement. It could be concluded that the plate-type heat exchanger (HX-P) provides not only the effective heating for the rural households and greenhouses, but also having the best heat recovery performance, easy manufacturing, assembling and breaking up the systems.

Development of Decision Tree Software and Protein Profiling using Surface Enhanced laser Desorption/lonization - Time of Flight - Mass Spectrometry (SELDI-TOF-MS) in Papillary Thyroid Cancer (의사결정트리 프로그램 개발 및 갑상선유두암에서 질량분석법을 이용한 단백질 패턴 분석)

  • Purpose: The aim of this study was to develop a bioinformatics software and to test it in serum samples of papillary thyroid cancer using mass spectrometry (SELDI-TOF-MS). Materials and Methods: Development of 'Protein analysis' software performing decision tree analysis was done by customizing C4.5. Sixty-one serum samples from 27 papillary thyroid cancer, 17 autoimmune thyroiditis, 17 controls were applied to 2 types of protein chips, CM10 (weak cation exchange) and IMAC3 (metal binding - Cu). Mass spectrometry was performed to reveal the protein expression profiles. Decision trees were generated using 'Protein analysis' software, and automatically detected biomarker candidates. Validation analysis was performed for CM10 chip by random sampling. Results: Decision tree software, which can perform training and validation from profiling data, was developed. For CM10 and IMAC3 chips, 23 of 113 and 8 of 41 protein peaks were significantly different among 3 groups (p<0.05), respectively. Decision tree correctly classified 3 groups with an error rate of 3.3% for CM10 and 2.0% for IMAC3, and 4 and 7 biomarker candidates were detected respectively. In 2 group comparisons, all cancer samples were correctly discriminated from non-cancer samples (error rate = 0%) for CM10 by single node and for IMAC3 by multiple nodes. Validation results from 5 test sets revealed SELDI-TOF-MS and decision tree correctly differentiated cancers from non-cancers (54/55, 98%), while predictability was moderate in 3 group classification (36/55, 65%). Conclusion: Our in-house software was able to successfully build decision trees and detect biomarker candidates, therefore it could be useful for biomarker discovery and clinical follow up of papillary thyroid cancer.

Effects of Different Roughage to Concentrate Ratios on the Changes of Productivity and Metabolic Profiles in Milk of Dairy Cows (조사료와 농후사료의 급여 비율이 착유유의 우유생산성과 대사산물에 미치는 영향)

  • This study was conducted to evaluate roughage to concentrate ratio on the changes of productivity and metabolic profiling in milk. Six lactating Holstein cows were divided into two groups, T1 group was fed low-concentrate diet (Italian ryegrass to concentrate ratio = 8:2) and T2 group was fed high-concentrate diet (Italian ryegrass to concentrate ratio = 2:8). Milk samples were collected and its components and metabolites were analyzed by 1H-NMR (Nuclear magnetic resonance). The result of milk components such as milk fat, milk protein, solids-not-fat, lactose and somatic cell count were not significantly different between two groups. In carbohydrate metabolites, trehalose and xylose were significantly higher (P<0.05) in T1 group, however lactose was not significantly different between two groups. In amino acid metabolites, glycine was the highest concentration however, there was no significant difference observed between two groups. Urea and methionine were significantly higher (P<0.05) in the T2 group. In lipid metabolites, carnitine, choline and O-acetylcarnitine there were no significant difference observed between the two groups. In benzoic acid metabolites, tartrate was significantly higher (P<0.05) in T2 group. In organic acid metabolites, acetate was significantly higher (P<0.05) in T1 group and fumarate was significantly higher (P<0.05) in T2 group. In the other metabolites, 3-methylxanthine was only significantly higher (P<0.05) in T2 group and riboflavin was only significantly higher (P<0.05) in T1 group. As a result, milk components were not significantly different between two groups. However, metabolites concentration in the milk was significantly different depends on roughage to concentrate ratio.

Formation and Evolution of the Paleo-Seomjin River Incised-Valley System, Southern Coast of Korea: 1. Sequence Stratigraphy of Late Quaternary Sediments in Yosu Strait (한반도 남해안 고섬진강 절개곡 시스템의 형성과 진화: 1. 여수해협의 후기 제 4기층에 대한 순차층서)

  • Detailed interpretation of some high-resolution seismic profiles in Yosu Strait reveals that Late Quaternary deposits consist of three allostratigraphic units (UH, LH, PL) formed by fluvial and tidal controls. The top mud unit, UH, thins onshore, and overlies the backstepping modem Seomjin delta deposits, which is interpreted as a transgressive systems tract (757) related to Holocene relative sea-level rise. The unit LH below the unit UH is composed of delta, valley- and basin-fill facies. The delta facies (Unit $LH_1$) occurs only in Gwangyang Bay and shows two prograding sets retrogradationaly stacked, thus it is also interpreted as a transgressive systems tract(757). On the contrary, the valley- and basin-fill facies (Unit $LH_2$), interpreted as 757, occur between the units UH and PL (Pleistocene deposits) in Yosu Strait. The bounding surface between UH and $LH_2$ can be interpreted as a tidal ravinement surface on the basis of trends thinning toward inner bay and becoming young landward. Furthermore its geomorphological pattern is similar to that of recent tidal channels. This allostratigraphy in'ffsu Strait suggests that two 757 deposits (UH and $LH_2$), divided by tidal ravinement surface, have been formed in Yosu Strait, whereas in Gwangyang Bay backstepping delta deposits ($LH_1$) without tidal ravinement surface have been formed during Holocene sea-level rise. These characteristics indicate that different stacking patterns could be formed in these two areas according to different increasing rate of accommodation space caused by different geomorphology, sediment supply and tidal-current patterns even in the same period of Holocene sea-level rise.
