A Study on the Efficient Use of Fused Phosphate for the Improvement of Upland Soil Productivity (토양(土壤)의 생산력증진을 위(爲)한 용성인비(鎔成燐肥)의 효과적인 시용법에(施用法) 관(關)하여)

  • In order to obtain some informations on the improvement of fertility of poor red yellow soils widely distributed in hilly lands of Korean peninsula, a pot culture study was made with a hill land soil in comparison with a fertile soil commonly used for vegetables. The results are as follows; 1. In addition to the normal application of phosphate, the mixing of a large dose of fused phosphate with whole soil increased the yeild of chinese cabbage remarkably, having the promotion particulary of the available phosphate level in the acidic low fertility soil. At the sometime the mixing seemed to short down the period of soil maturing to fertile for good crop. It is, however, considered that such effect of fused phosphate must also be evaluated economically through a long term experiment. 2. A relatively close relationship between the soil phosphate soluble in dilute acid and chinese cabbage growth was observed.

Isolation of Antifungal Compound and Biocontrol Potential of Lysobacter antibioticus HS124 against Fusarium Crown Rot of Wheat

  • Fusarium graminearum is the main cause of substantial economic loss in wheat production. The aim of this study is to investigate biocontrol potential of Lysobacter antibioticus HS124 against F. graminearum and to purify an antifungal compound. In preliminary study, n-butanol crude extract revealed destructive alterations in the hyphal morphology of F. graminearum and almost degraded with $1,000{\mu}g\;mL^{-1}$ concentration. For further study, the antifungal compound extracted from the n-butanol crude extract of L. antibioticus HS124 was identified as N-Butyl-tetrahydro-5-oxofuran-2-carboxamide ($C_9H_{16}NO_3$) using NMR ($^1H-NMR$, $^{13}C-NMR$, $^1H-^1H\;COSY$, HMBC, and HMQC), and HR-ESI-MS analysis. To our knowledge, N-Butyl-tetrahydro-5-oxofuran-2-carboxamide may be a novel compound with molecular weight of 186.1130. The minimum inhibitory concentration value of antifungal compound was $62.5{\mu}g\;mL^{-1}$ against F. graminearum. In an in vivo pot experiment, crown rot disease from F. graminearum was inhibited when wheat seeds were treated with both HS124 culture and F. graminearum. Growth of wheat seedling was enhanced by treatment of HS124 compared to control. Our results suggest that L. antibioticus HS124 characterized in this study could be successfully used to control F. graminearum and could be used as an alternative to chemical fungicides in modern agriculture.

  • The relations of the growth responses of plants, i.e. 4 species of crops, 12 species of roadside trees and 5 species of horticultural plants to cadmium (Cd) to which added soil were studied in pot culture. Growth in dry weight of corn, soybean, barley and wheat plant were decreased with increase of Cd concentration. Damage of corn plant caused by Cd treatment was more or less recovered when it was grown in soil with calcium, but that of other three crops was not recovered when it was with calcium. Although crop plants used here absorbed small amount of Cd through root, Cd content in the shoot was directly proportionate to the concentration of Cd treated to soil. Additions of calcium and sulfur to soil were effectively to change of soil pH, and only of calcium increased exchangeable calcium and cation exchange capacity of its soil. The chlorosis on leaves caused by Cd treatment was observed in 2 species such as Euonymus japonica and Rhododendron yedoense out of 5 species of the horticultural plants, especially at 50ppm of Cd concentration, Euonymus japonica occurred symptoms of chlorosis and difoliation, and at the higher concentration than that the symtoms were sever more and more. At 200 ppm of Cd feeble damage was observed in Pinus koraiensis and Ginkgo biloba but severe chlorosis observed in Robinia pseudoacacia and Sabina chinensis, Buxus koreana, Abies holophylla and Platanus orientalis. Nevertheless those plants had serious damage at 200 ppm of Cd, such symptom was weakened by adding calcium to soil with Cd. There were many Cd tolerant species out of the plants used in thies xperiment, such as Crassula falcata, Chrysanthemum morfolium, Hibiscus syriacus, Ligustrum ovalifolium, Liriodendron tulipeferia, Lespedeza crytobotrys.

In Vitro Propagation of Anthurium andreanum ′Atlanta′ Developed for Pot Culture (분화용 Anthurium andreanum ′Atlanta′의 기내번식)

  • In order to establish micropropagation system Anthurium andreanum 'Atlanta', dwarf type, shoots of A. andreanum were cultured on medium supplemented with cytokinin. Callus was formed from the base of shoots. high frequency callus induction was obtained on medium with 10.0mg/L BA or 10.0mg/L TDZ(thidiazuron) at more than 71.8%. The shoots were cultured on media with various combinations and concentrations of TDZ, BA and 2.4-D to enhance callus induction. Callus was induced at more than 72.6% and grew vigorously on media containing 10.0mg/L BA and 0.0∼0.5mg/L 2.4-D, or 1.0mg/L TDZ. Stimulation effects of cytokinin by 2.4-D did not occur in combined treatments of cytokinin and 2.4-D. Callus was cut into sections(7${\times}$10mm), and then cultured on media with BA alone or BA and 2.4-D to regenerate shoots and to stimulate the callus growth. Shoot regeneration and callus growth were effective on media with 10.0mg/L BA alone, or 10.0mg/L BA and 0.1mg/L 2.4-D. In combined treatments of BA and 2.4-D, stmulation effects of cytocinin by 2.4-D also did not occur. Callus growth was decreased, accordiong to increasing the concentration of 2.4-D. In cimbined treatments of TDZ and 2.4-D in shoot regeneration and callus proliferation, stimulated effects of cytokinin by 2.4-D did not occur entirely. Media with 0.5∼1.0mg/L TDZ ingibited the regeneration and rooting of shoots, and callus growth from callus sections. Addition of 2.4-D on medium with TDZ ingibited the regeneration and rooting of shoots, and callus growth. Rooted plantdts were acclimatized in greenhouse. The plantlets were survived more than 98% in soil of vermiculite alone or mixed perlite 1 and vermiculite 1.

Plantlet Regeneration by Tissue Cultures of Cyclamen persicum Mill. (시클라멘 (Cyclamen persicum Mill.)의 조직배양에 의한 식물체 재분화)

  • This study was conducted to examine the effects of explant sources and plant growth regulators on mass propagation of Cyclamen persicum. Tuber, cotyledon, and petiole tissues were cultured on MS medium supplemented with different concentrations and combinations of auxins and cytokinins. Shoots were not induced from calli on cotyledon and petiole explants cultured on MS medium containing various concentrations of 2,4-D or NAA. However, multiple shoots were formed directly from tuber explants cultured on the medium containing 0.5 and 1.0 mg/L 2,4-D or NAA. In MS medium with cytokinin alone, TDZ was more effective in shoot formation than BA or kinetin in all explants. The combinations of NAA and BA was found to be most effective in shoot formation from tuber, cotyledon and petiole explants. Especially, shoots were formed from all the tuber explants on the medium containing 0.5 mg/L of NAA and BA. Hormonal effects on root formation were examined by subculturing single shoots on MS medium containing NAA or IBA. The medium with 0.5 mg/L IBA was most effective in root induction and subsequent plantlet regeneration.

  • Paper sludges collected from three different paper mills were physico-chemically analyzed in order to use them as raw materials for making bed soils and seedling pots. The sludge from a fine paper mill contained lots of inorganic pigment particles used for coating, as those from a newsprint mill and a tissue mill had not. It was clearly through XRD analysis confirmed that all sludges included calcium carbonate. The paper sludge from the tissue mill contained the greatest amount of particles, which would contribute to water absorption and nutrient storage. The sludge from the fine paper mill had the highest density due to many inorganic elements. While the ash content and the total nitrogen content were the highest in the sludge from the fine paper mill, the C/N ratio was the lowest in the fine paper mill sludge. All sludges seemed to have insufficient contents of potassium. The sludges from the newsprint mill and the tissue mill showed more silicon contents than that from the fine paper mill. It was concluded that the sludge from the fine paper mill would be able to be the most efficient raw materials for making bed soils and seedling pots and the other two sludges would be more efficient for intensive culture for crops such as rice and grain with additional supplement of nitrogen and other nutrients.

Biological Control of Meloidogyne hapla Using an Antagonistic Bacterium

  • We examined the efficacy of a bacterium for biocontrol of the root-knot nematode (RKN) Meloidogyne hapla in carrot (Daucus carota subsp. sativus) and tomato (Solanum lycopersicum). Among 542 bacterial isolates from various soils and plants, the highest nematode mortality was observed for treatments with isolate C1-7, which was identified as Bacillus cereus based on cultural and morphological characteristics, the Biolog program, and 16S rRNA sequencing analyses. The population density and the nematicidal activity of B. cereus C1-7 remained high until the end of culture in brain heart infusion broth, suggesting that it may have sustainable biocontrol potential. In pot experiments, the biocontrol efficacy of B. cereus C1-7 was high, showing complete inhibition of root gall or egg mass formation by RKN in carrot and tomato plants, and subsequently reducing RKN damage and suppressing nematode population growth, respectively. Light microscopy of RKN-infected carrot root tissues treated with C1-7 showed reduced formation of gall cells and fully developed giant cells, while extensive gall cells and fully mature giant cells with prominent cell wall ingrowths formed in the untreated control plants infected with RKNs. These histopathological characteristics may be the result of residual or systemic biocontrol activity of the bacterium, which may coincide with the biocontrol efficacies of nematodes in pots. These results suggest that B. cereus C1-7 can be used as a biocontrol agent for M. hapla.

Biocontrol of Late Blight (Phytophthora capsici) Disease and Growth Promotion of Pepper by Burkholderia cepacia MPC-7

  • A chitinolytic bacterial strain having strong antifungal activity was isolated and identified as Burkholderia cepacia MPC-7 based on 16S rRNA gene analysis. MPC-7 solubilized insoluble phosphorous in hydroxyapatite agar media. It produced gluconic acid and 2-keto-gluconic acid related to the decrease in pH of broth culture. The antagonist produced benzoic acid (BA) and phenylacetic acid (PA). The authentic compounds, BA and PA, showed a broad spectrum of antimicrobial activity against yeast, several bacterial and fungal pathogens in vitro. To demonstrate the biocontrol efficiency of MPC-7 on late blight disease caused by Phyto-phthora capsici, pepper plants in pot trials were treated with modified medium only (M), M plus zoospore inoculation (MP), MPC-7 cultured broth (B) and B plus zoospore inoculation (BP). With the sudden increase in root mortality, plants in MP wilted as early as five days after pathogen inoculation. However, plant in BP did not show any symptom of wilting until five days. Root mortality in BP was markedly reduced for as much as 50%. Plants in B had higher dry weight, P concentration in root, and larger leaf area compared to those in M and MP. These results suggested that B. cepacia MPC-7 should be considered as a candidate for the biological fertilizer as well as antimicrobial agent for pepper plants.

Effect of soil moisture on growth of P. ginseng (토양수분(土壤水分)이 인삼생육(人蔘生育)에 미치는 영향(影響))

  • Effect of soil moisture on growth of 2 year old Pan ax ginseng was investigated by pot culture using clay loam and sandy loam soils. In clay loam soil emergence rate of new shoot and stem growth were greatly decreased below 40% of field capacity. In sandy loam soil the growth of stem and leaf were greatly affected below 40% moisture. Root yield showed maximum at 60% moisture and significantly coorelated with stem diameter and root length. Rotten root rate and die back rate of top were least at 60% and gradually increased with either increase or decrease of soil moisture.

  • Removal of heavy metals by water hyacinth, Eichhornia crassipes Solms-Laub, was examined with two heavy metals Cd, Cu under laboratory conditions. Cd in culture solution was reducd to 0.116, 0.873, 2.015, 3.755 and 4,747 mg/L from 1, 2, 3, 4 and 5 mg/L for 24 hrs, after cultivating of water hyacinth respectively And, Cu was reduced to 0.086, 0.600, 2.174, 3.473, and 4.365 mg/L from 1, 2, 3, 4, and 5 mg/L for 24 hrs, after the cultivation, respectively. Cu was removed faster than Cd nd airating cultivation was effected higher than fixing cultivation. Removal effect of heavy metals by water hyacinth was higher in low a heavy metal cocentration than in high concentration
