Changes in Taste Components of Kanjang Made with Barley Bran during Fermentation (보리등겨로 제조한 간장의 발효기간별 맛성분 변화)

  • The changes in taste components of kanjang made with barley bran during fermentation time were examined. The pH was gradually decreased and total nitrogen content reached to 0.7% at 90 days fermentation. Five kinds of free sugars, three kinds of volatile organic acid, and eight kinds of non-volatile organic acid were detected. Lactic acid known as abundant component in kanjang was not detected in kanjang made with barley bran. The content of free amino acid was $422.9{\sim}803.6\;mg%$. Glutamic acid was most abundant component among the amino acids, followed by proline and phenylalanine. Essential amino acid content was revealed $34.3{\sim}37.3%$. Based on result of sensory evaluation, it was most comfortable to eat sample of fermentation $45{\sim}60$ days.

The Importance of Work Capability and the Educational Needsfor Optometric Duty (안경사 직무에 관한 작업수행의 중요도와 교육의 필요도)

  • Purpose: The purpose of this study was to document the importance of work capability and the educational needs for optometric duty. Methods: Questionnaires were distributed to 813 optometrists and 89 full-time faculties on nation wide in July, 2008. Results: Statistical analysis showed that high mean of 3.53, maximum of 4, for the importance of work capability, and 3.46 for the educational needs as for the total optometric duty. Especially, optometric dispensing was the highest on the importance of work capability and the educational needs, where as medical ethics and optometric jurisprudence and business management was the lowest. Conclusions: Both optometrists and full-time faculties agreed to the importance of work capability and the educational needs, and therefore it should be reflected in optometric education program.

Comparative serology of human brucellosis in Korea

  • The study was carried out to evaluate the diagnostic efficacy of the tube agglutination test (TAT), enzyme-linked immunosorbent assay (ELISA) and the 2-Mercaptoethanol agglutination test (2-MAT) to detect human brucellosis patients in Korea. We examined 87 serum samples of people in the rural farm areas where bovine brucellosis had been reported. People in this study were divided into seven groups- farmers and their families, veterinarians, veterinary quarantine workers, livestock health control officers, artificial inseminators, livestock traders and healthy control individuals. Among 87 people, 65 were males and 22 were females ranging in age from 13 to 72 years. Of 87 serum samples, ELISA detected 21.84%, TAT detected 11.50% and 2-MAT detected 8.05% Brucella positive sera. Brucella specific IgG ELISA antibody titer was recorder higher in the individuals between the ages of 50 and 65 years. The highest prevalence rate of brucellosis(29.4%) was recorded in the cattle farmers and their family members followed by quarantine veterinary office workers (25%) and practicing veterinarians 01.1%). The majority of the Brucella sero-positive individuals in this study had a history of direct contact with animals.

Purification and Characterization of an Acid Deoxyribonuclease from the Cultured Mycelia of Cordyceps sinensis

  • A new acid deoxyribonuclease (DNase) was purified from the cultured mycelia of Cordyceps sinensis, and designated CSDNase. CSDNase was purified by $(NH_4)_2SO_4$ precipitation, Sephacryl S-100 HR gel filtration, weak anion-exchange HPLC, and gel filtration HPLC. The protein was single-chained, with an apparent molecular mass of ca. 34 kDa, as revealed by SDS-PAGE, and an isoelectric point of 7.05, as estimated by isoelectric focusing. CSDNase acted on both double-stranded (ds) and single- stranded (ss) DNA, but preferentially on dsDNA. The optimum pH of CSDNase was pH 5.5 and its optimum temperature 55. The activity of CSDNase was not dependent on divalent cations, but its enzymic activity was inhibited by high concentration of the cation: $MgCl_2$ above 150 mM, $MnCl_2$ above 200 mM, $ZnCl_2$ above 150 mM, $CaCl_2$ above 200 mM, NaCl above 300 mM, and KCl above 300 mM. CSDNase was found to hydrolyze DNA, and to generate 3-phosphate and 5-OH termini. These results indicate that the nucleolytic properties of CSDNase are essentially the same as those of other well-characterized acid DNases, and that CSDNase is a member of the acid DNase family. To our knowledge, this is the first report of an acid DNase in a fungus.

  • The purpose of our study is to explore dynamic mechanisms within families and self-authored decisions among Korean female college students. In particular, we investigated how parent-child interactions influence the development of self-authorship and self-authored decisions within Korean culture through interviews with 14 female college students. Results showed unique cultural influences on how female college students interacted with parents and interpreted and reflected parents' opinions in their self-authored decisions. These results suggest further research on gender differences and complex family influences on self-authored decisions within cultural contexts.

  • Phytochemical study of the aerial parts of Lotus lalambensis Schweinf resulted in the isolation and identification of two new flavonol glycosides; kaempferol $3-O-(5"-acetyl)-apioside-7-O-{\alpha}-L-rhamnopyranoside$ (1) and kaempferol $3-O-{\alpha}-[{\beta}-D-xylosyl-(1""{\rightarrow} 2")-L-rhamnopyranoside]-7-O-{\alpha}-L-rhamnopyranoside$ (2). Structures were determined utilizing different physical, chemical, spectroscopic data including 2D-NMR experiments and HRFABMS.

Seasonal and Sexual Variation of Ginkgolides Contents in Ginkgo Leaves (계절, 성에 따른 은행잎 중의 Ginkgolides 함량의 변화)

  • The contents of ginkgolides were determined in the leaves of male and female Ginkgo biloba from late spring until mid-autumn. Ginkgolides were detected during the whole growing season in the leaves of each tree. Ginkgolides content was low in late spring, gradually increased to reach a maximum in august and decreased thereafter. The male trees have two or three times higher ginkgolides content than the female trees. Comparing our results with that of previously reported values, the sexual variation of ginkgolides content seemed not to be genetic.

Bilobalide Attenuates Glutamate-Induced Neurotoxicity in Primary Cultures of Rat Cortical Cells (빌로바라이드가 글루타메이트에 의한 신경독성에 미치는 영향)

  • The neurotoxicity induced by L-glutamate in primary cultures of rat cortical cells could be attenuated by sesquiterpene constituent of Ginkgo biloba leaves, bilobalide. At the c oncentration of 100 nM, Bilobalide elevated the combined levels of reduced/oxidized glutathione in rat cortical cells exposed to 100 ${\mu}$M glutamate. Furthermore, bilobalide promoted a reduction in superoxide dismutase activity in glutamate-treated cells. Finally, bilobalide markedly inhibited the production of malondialdehyde. a measure of lipid peroxidation, in glutamate-treated rat cortical cells.

Rhapontigenin Production by Bioconversion and Inhibition of Melanin Synthesis (생물전환에 의한 Rhapontigenin의 생산 및 멜라닌 합성저해)

  • Rhapontin is the glycosylated stilbene compound, and comprising major component of rhubarb root extract. Rhapontin has been used as a raw material of skin-whitening cosmetics in Korea. Rhapontigenin, the aglycone of rhapontin, has been suggested to be more active than its glycosylated form. Therefore, the rhubarb root extract was treated with commercial enzyme, Pectinex to remove glycosylated moiety of rhapontin and rhapontigenin was prepared. The resulting material was analysed and identified as rhapontigenin by proton NMR and MALDI-Mass. Rhapontigenin exhibited tyrosinase inhibitory activity with an $IC_{50}$ of $126.72{\mu}g/mL$. The tyrosinase inhibitory activity of rhapontigenin was six times higher than that of rhapontin. In melanin biosynthesis inhibition assay using Streptomyces bikiniensis, rhapontigenin showed wider inhibition zone than that of rhapontin. From these results, we expect that rhapontigenin has stronger skin whitening effect than rhapontin and has advantages in cosmetic industry.

Reliability and responsiveness of Equivital Lifemonitor and photoplethysmography based wristwatch for the assessment of physiological parameters during a simulated fatigue task

  • Objective: To investigate test-retest reliability and responsiveness of Equivital Lifemonitor and photoplethysmography based wristwatch tools in assessing physiological parameters during a simulated fatigue task. Methods: Ten university students (Mean age, 30.6 ± 1.7 years) participated in this pilot study. Participants were asked to perform a 30-minute of a simulated fatigue task in an experimental setup in a lab. The physiological parameters (e.g., heart rate, heart rate variability, respiratory rate, electrodermal activity, and skin temperature) were measured at baseline and immediately after the fatigue task. An intraclass correlation coefficient (ICC2,1) was used to evaluate the test-retest reliability of each tool in assessing physiological measures. In addition, the responsiveness of each tool to measure changes from baseline to posttest was calculated using a standardized response mean. Results: The Equivital Lifemonitor has shown good to excellent test-retest reliability for the assessment of heart rate (ICC, 0.97), heart rate variability (ICC, 0.86), respiratory rate (ICC, 0.77), and local skin temperature (ICC, 0.76). However, photoplethysmography based wristwatch showed moderate to good test-retest reliability for the assessment of heart rate (ICC, 0.71), heart rate variability (ICC, 0.73), electrodermal activity (ICC, 0.80), and skin temperature (ICC, 0.72). A large standardized response mean (>0.8) indicates that both tools can capture the changes in heart rate, heart rate variability, respiratory rate, skin temperature, and electrodermal activity after a 30-minute of fatigue task. Conclusions: The Equivital Lifemonitor and photoplethysmography based wristwatch devices are reliable in measuring physiological parameters after the fatigue task. Additionally, both devices can capture the fatigue response after a simulated construction task. Future field studies with a larger sample should investigate the sensitivity and validity of these tools in measuring physiological parameters for fatigue assessment at construction sites.
