A New Short Growth-Duration Rice Cultivar, "Keumo 3" (소득작물 전후작용 단기성 벼 품종 "금오3호")

  • A new rice cultivar "Keumo 3" was developed for adopting under double cropping system with after or before cash crop cultivation. It was selected from the cross-combination between YR17202 $F_2$/Shinkeumobyeo//YR15727-B-B-B-102. The parent, YR17202 $F_2$ individual plant, was used for tolerance to lodging, it derived from a cross between Nonganbyeo/Shinkeumobyeo. Nonganbyeo is well known to lodging tolerance cultivar, as well as biotic stress, because it was developed by crossing with Tongil type. And the YR15727-B-B-B-102 line used as another parent with short growth duration, likewise highly resistance to rice blast disease. The pedigree derived from the cross-combination YR17202 $F_2$/Shinkeumobyeo//YR15727-B-B-B-102 were generated to $F_7$, and a best line among them named as Milyang 201. After a series of yield trials, including local adaptability test conducted throughout the peninsular of Korea, Milyang 201 was registered with the name of "Keumo 3" in 2005. The cultivar belongs to a early maturing group and heads 4 days earlier than Keumobyeo, a standard cultivar. It has short culm, and less spikelet number per panicle than Keumobyeo. However, its milled rice yield grown under extremely late transplanting time, 10. July, over the 3 local sites for 2003-2005 years, averaged 4,48 MT/ha, which is 6% higher than the standard, Keumobyeo. "Keumo 3" has showed a durable resistance to leaf blast disease during fourteen blast nurseries screening covered from south to north in Korea for 2003-2007 years. And it was confirmed harbours pi-zt, a durable blast resistance gene. Moreover it was incompatible with 19 blast isolates under artificial inoculation, except one isolate, K1101. Additionally, "Keumo 3" exhibits resistance to $K_1$, $K_2$ and $K_3$ of bacterial blight pathogen, as well as strip virus disease resistance, and moderate resistance to dwarf virus disease. Consequently, the new rice cultivar "Keumo 3" would be well adopted where a bio stress makes a big problem annually.

Inter-ramet Physiological Integration Detected in Buffalograss(Buchloe dactyloides (Nutt.) Engelm.) under Water Stress (수분스트레스 하에 있는 버팔로그래스에서 검출된 무성생식체의 생리학적 조정)

  • Buffalograss is an important turfgrass species with excellent cold, heat, and drought tolerance. Understanding the physiological integration of buffalograss under heterogeneous conditions helps to develop cultural practices that better use limited resources for uniform turf quality. The objective of this study was to evaluate physiological integration of buffalograss under water deficit stress and the involvement of lipid peroxidation and antioxidants in the process. In one experiment, buffalograss was planted in the center of a four-compartment growth unit. Watering frequencies, once a week(+) and once in two weeks(-), were combined with the sand(S) or peat(P) in each unit to generate five total treatments(P+S-P-S+, P+P+P+P+, S-S-S-S-, P-P-P-P-, and S+S+S+S+). The average number of shoot established from the heterogeneous root-zone medium was higher than the average of four possible homogeneous media. In second experiment, single ramet in Hoagland solution($S_0$) or single ramet in Hoagland solution with 20% PEG-6000($S_s$) were compared with two connectedramets under different treatments. Treatments for connected ramets were young ramet in Hoagland solution($Y_{os}$) and old ramet in Hoagland solution with 20% PEG-6000($O_{os}$), and old ramet in Hoagland solution($O_{ys}$) and young ramet in Hoagland solution with 20% PEG-6000($Y_{ys}$). Lipid peroxidation, antioxidants, and proline showedphysiological integration between ramets subjected to different levels of water stress. Superoxide dismutase(SOD), Guaiacol peroxidase(G-POD), malondialdehyde(MDA), and free proline also showed different time courses and relative activities during the physiological integration.

The Effects of Increased Temperature on Yield Properties, Antioxidant Contents, and Pollen Viability of Adzuki Bean (Vigna angularis L.) Responses in Temperature Gradient Greenhouse and Growth Periods (온도구배온실에서 온도상승이 생육시기별 팥의 수량, 항산화 성분, 화분 임성에 미치는 영향)

  • The quality and yield of crops produced using field cultivation are expected to decrease due to the recent global climate change caused by extreme weather. The plant reproductive stage associated with crop yields is a highly vulnerable period to global warming caused by high temperatures. This study analyzed the adzuki bean's yield properties, antioxidant contents, and pollen viability of adzuki bean (Vigna angularis L.) under high-temperature stress and growth periods in a temperature gradient greenhouse that forms 0 to 4℃ above the outside temperatures. As the main variety of red beans cultivated in Korean farms, the "Arari" red bean was grown in the rain shield greenhouse and the temperature gradient greenhouse from 2021 to 2022 in Milyang, Korea. Compared to 2022, it showed a 0 - 1.0℃ lower temperature during the whole growth period in 2021. However, its average temperatures were 0 - 3.7℃ higher in the vegetative stage and 0.4 - 2.4℃ higher in the anthesis stage in 2021. The lowest yield (6.8 ± 0.7 g) was at the highest temperature (T4: low, 23.6℃; average, 28.5℃; high, 35.8℃) during the anthesis stage in 2021. The temperatures of the mature stage were 1.7 - 3.9℃, which were higher in 2022 than in 2021, although the low temperatures of 2022 were lower than in 2021. The yields of the mature stage in 2022 increased more than in 2021 because of the high temperature of the mature stage. The growth and yield were good at 40.5℃ in the vegetative stage. However, growth was poor when the average temperature was 27.0℃ or higher, and yields decreased during the flowering period. Total polyphenol and flavonoid contents were increased, and the pollen viability was 40.75% in the whole growth period at high temperature (T4: low, 22.9℃; average, 28.8℃; high, 36.9℃). These results showed that the antioxidant levels increased when the antioxidant component was affected at higher temperatures than at normal. In contrast, the pollen viability-related yield decreased as the temperature increased. Our results are the basic data for field growers and the breeding of thermos-tolerance in adzuki beans to prepare for the changeable future climate.

Evaluations on agronomic traits of rice transgenic lines (벼 형질전환계통의 주요 작물학적 특성에 대한 고찰)

  • This study was conducted to evaluate the performances of rice transgenic lines along with their wild types in terms of agronomic traits. A total of 32 rice transgenic lines, through previously conducted collaborative researches between molecular biologists and conventional rice breeders, were selected as promising lines. As the introduced functional genes, 17 genes, which were putatively related with high yield, disease and herbicide resistance, abiotic stress tolerance, and diversifying endosperm starch components, were transformed into three Japonica cultivars, Nipponbare, Nagdongbyeo, and Dongjinbyeo. The transgenic lines exhibited significantly deviated performances from their wild types on agronomic traits such as days to heading, culm length and yield potential. Multivariate analyses on transgenic lines to the evaluated agronomic traits also indicated random manner of phenotypic deviations from their wild type in terms of deviation directions and degrees. Our results suggested that, therefore, breeding strategies to control unexpected deleterious phenotypic performances among transgenic lines would be critical as much as the functions and proper expressions of the transformed genes.

Analysis of Growth Response and Gene Expression by Waterlogging Stress on B73 Maize (침수 처리에 따른 B73 옥수수의 생육 반응 및 유전자 발현 분석)

  • Maize is thought to be an alternative crop to rice in paddy fields for efficient field management and maintenance of rice production at appropriate levels in Korea. Thus efforts to breed waterlogging-tolerant maize cultivars have been ongoing. However, molecular studies related to waterlogging tolerance are limited for developing molecular markers to select waterlogging tolerant maize varieties. In this study, we examined molecular biological changes of B73 in the V3 stage after immersion treatment for 7 days. Overall growth of maize was lower in treated samples compared to non-immersed control samples. The length of leaf and root decreased by 21.3% and 50.6%, respectively and the weight of root reduced by 21.6%. Soil plant analysis development (SPAD) value and chlorophyll content of leaf also decreased by 55.7% and 35.3%, respectively. Reactive oxygen species (ROS) content of root increased by 46.5% at 2 hours in immersion treatment. In addition, immersed roots were 2.5-fold thickened with additional aerenchyma formation in the cortex. In order to identify the causes of these changes, we performed a microarray and found increased expression of genes, such as WIP1, PMIP2, EXPA1, TPS1, and MAS1, in immersed samples. These differentially expressed genes and expression of previously reported genes, including ALDH2, Wusl1032, UP-1, UP-2, and CAT2 were further confirmed with qRT-PCR. Here, we report 7 differentially expressed genes after immersing treatment, which may be utilized as useful resources for breeding waterlogging- tolerant maize.

Endophytes from Natural Festuca spp. in Southwest China and Their Compatibility with Tall Fescue Cultivars (남서중국의 자생페스큐의 엔도파이트와 톨체스큐 품종과의 공생)

  • Investigating endophyte distribution, naturally occurring in native grasses, is important for understanding endophyte-grass associations and using the beneficial effects of endophytes in cultivated plants. The goal of this study was to investigate endophytes from natural Festuca spp. in Yunnan, Guizhou, and Sichuan Provinces of China, and to study the compatibility between the endophytes and turf type tall fescue (Festuca arundinacea) which is widely used for lawn and athletic fields in that region. Of 628 accessions in F. ovina, 421 had endophytes identified in leaf sheath from on-site microscopic examination. From Festuca spp, three isolates were obtained from the seeds and ninety isolates were obtained from seedlings established from the collected seeds. The isolates from F. ovina and F. stapfii were tentatively identified as Neotyphodium typhinum and Neotyphodiumstarii, respectively. We tested compatibility of the two fungal species with seven tall fescue cultivars, Little Hero, Sub Boy, Eldorado, Arid III, Millennium, Crossfire, and Fawn. N. typhinum or N. starii did not infect 'Fawn' with either seed injection or seed soaking method. The highest infection rate byboth N. typhinum and N. starii was in 'Sun Boy' and 'Eldorado'. There were significant interaction effects between tall fescue cultivar and type of endophyte on infection.

Antioxidant capacity in seedling of colored-grain wheat under water deficit condition

  • Nutritious and functional foods from crop have received great attention in recent years. Colored-grain wheat contains high phenolic compound and a large number of flavonoid. The anthocyanin and polyphenolic synthesis and accumulation is generally stimulated in response to biotic or abiotic stresses. Here, we analyzed genome wide transcripts in seedling of colored-grain wheat response to ABA and PEG treatment. About 900 and 1500 transcripts (p-value < 0.05) from ABA and PEG treatment were aligned to IWGSC1+popseq DB which is composed of over 110,000 transcripts including 100,934 coding genes. NR protein sequences of Poaceae from NCBI and protein sequence of transcription factors originated from 83 species in plant transcription factor database v3.0 were used for annotation of putative transcripts. Gene ontology analysis were conducted and KEGG mapping was performed to show expression pattern of biosynthesis genes related in flavonoid, isoflavonoid, flavons and anthocyanin biopathway. DroughtDB (http://pgsb.helmholtz-muenchen.de/droughtdb/) was used for detection of DEGs to explain that physiological and molecular drought avoidance by drought tolerance mechanisms. Drought response pathway, such as ABA signaling, water and ion channels, detoxification signaling, enzymes of osmolyte biosynthesis, phospholipid metabolism, signal transduction, and transcription factors related DEGs were selected to explain response mechanism under water deficit condition. Anthocyanin, phenol compound, and DPPH radical scavenging activity were measured and antioxidant activity enzyme assays were conducted to show biochemical adaptation under water deficit condition. Several MYB and bHLH transcription factors were up-regulated in both ABA and PEG treated condition, which means highly expressed MYB and bHLH transcription factors enhanced the expression of genes related in the biosynthesis pathways of flavonoids, such as anthocyanin and dihydroflavonols in colored wheat seedlings. Subsequently, the accumulation of total anthocyanin and phenol contents were observed in colored wheat seedlings, and antioxidant capacity was promoted by upregulation of genes involved in maintaining redox state and activation of antioxidant scavengers, such as CAT, APX, POD, and SOD in colored wheat seedlings under water deficit condition. This work may provide valuable and basic information for further investigation of the molecular responses of colored-grain wheat to water deficit stress and for further gene-based studies.

Interpreting Soil Tests for Turfgrass (잔디 토양 분석의 해석)

  • Soil testing laboratories unfamiliar with turfgrasses will often overestimate the plant's need for phosphorus and underestimate the need for potassium. This is partly due to differences in rooting between grasses and many garden plants and crops. The grasses are generally more efficient in extracting phosphorus from the soil, reducing their need for phosphorus fertilizer. The fact that crop yield is often the primary objective in field crop production, and is usually of little interest in turfgrass management, may affect soil test interpretation for potassium. Potassium levels above those required for maximum tissue yield of grasses may improve stress tolerance and turfgrasses will usually benefit from higher applications of this element. There are also diffrrences in soil testing philosophies. Some laboratories use the sufficiency level of available nutrients(SLAN) approach, whereas others prefer the basic cation saturation ratio(BCSR) approach. Some will use a combination of the two methods. The use of the BCSR theory easily lends itself to abuse and questionable fertilizer applications and products are sometimes recommended citing imbalances in cation ratios. The usefulness of the BCSR ratio theory of soil testing varies with soil texture and interpretations on tests performed on sand-based media are particularly a problem. Other soil testing problems occur when sand-based media used on sports fields and golf greens contain free calcium carbonate. The ammonium acetate extractant at pH 7.0 dissolves excessive amounts of calcium that can bias cation exchange capacity measurements and measurements of cation ratios. Adjusting the pH of the extractant to 8.1 can improve the accuracy of the testing procedure for calcareous media.

Evaluation of Field Application of Soil Conditioner and Planting Chrysanthemum zawadskii on the Roadside Soils Damaged by Deicing Agents

  • Background and objects: Soil contamination caused by CaCl2 that is used to deice slippery roads in winter is now recognized as one of the major causes of damage of roadside plants. The aim of this study is to identify the salt mitigation effects of planting Chrysanthemum zawadskii and using a soil conditioner. Methods: The study was conducted at the site where Pinus densiflora f. multicaulis was planted on the roadside between Konkuk University Sageori and Danwol Samgeori located in Chungju-si. We classified the soils collected from the field experimental site according to the degree of the damage caused by deicing agents and divided the site into six blocks of three 80 × 80 cm plots replicated by treatment type. Three selected plots were treated with loess-balls on the soil surface (high salinity with loess-balls, medium salinity with loess-balls, low salinity with loess-balls) and three were left as an untreated control (H = high salinity, M = medium salinity, L = low salinity). The soil properties were measured including pH, EC and exchangeable cations as well as the growth of Chrysanthemum zawadskiia. Results: In the results of soil analysis, pH before planting Chrysanthemum zawadskiia was 6.39-6.74 and in September, five months after planting, the acidity was reduced to 5.43-5.89. Electrical conductivity (EC) was measured to be H > M > L with the higher degree of damage by deicing agents. The analysis of deicing exchangeable cations showed that the content of Ca2+ of soils were significantly correlated to deicing exchangeable cations (Ca2+, Na+, Mg2+) in the shoot part of Chrysanthemum zawadskii. The loess-ball treatment showed a lower content of deicing exchangeable cations than the treatment where Chrysanthemum zawadskiia was planted. Conclusion: In this study, the use of a new system made of loess-balls is proposed as a soil conditioner to protect soils from the adverse effects of road deicing salts. These data suggest that treatment of soil conditioners and planting Chrysanthemum zawadskiia are effective in mitigation of salt stress on the soils damaged by deicing agents.

Resistance and Survival of Cronobacter sakazakii under Environmental Stress of Low Temperature (저온 환경에서 Cronobacter sakazakii의 저항과 생존)

  • Cronobacter sakazakii has been isolated from a wide range of environmental sources and from several foods of animal and plant origin. The objective of this study was to determine the resistance of C. sakazakii (ATCC 12868, ATCC 29004, and ATCC 29544) in cold, cold-freeze thaw, cold-acid, and cold starvation-freeze thaw stress. The number of C. sakazakii decreased to 1 log CFU/mL at $5^{\circ}C$ (cold storage) for 10 days. When C. sakazakii was cultivated at a low temperature ($13^{\circ}C$), the population of C sakazakii ATCC 12868 and 29004 increased to $10^9$ CFU/mL, and the population of C. sakazakii ATCC 29544 increased to $10^8$ CFU/mL. For C. sakazakii ATCC 12868 and 29004, the cold-adapted cells ($5^{\circ}C$ 24 hr) decreased by 4 log CFU/mL, and the low-temperature-cultivated cells ($13^{\circ}C$) decreased by 0.5 log CFU/mL. In this study, low-temperature cultivation enhanced the freeze-thaw cross-resistance due to the metabolic changes in the cells. Cold stress ($5^{\circ}C$ 48 hr, $13^{\circ}C$ cultivation) enhanced the cold-acid cross-resistance. The cold-starved cells in the sterilized 0.1% peptone water enhanced the freeze-thaw cross-resistance with significant differences (p<0.05). Therefore, the increased tolerance of the cold-adapted or low-temperature-cultivated C. sakazakii cells to freeze-thaw, acid, or starvation suggests that such environments should be considered when processing minimally processed foods or foods with extended shelf life.
