This study assessed the effects of oxygen and ozone nanobubbles on gill morphology, weight gain, and mortality of Pacific white shrimp (Penaeus vannamei), as well as the level of Vibrio parahaemolyticus and water quality of shrimp culture tanks under lab conditions. Two experiments were carried out with oxygen macrobubble, ozone macrobubble, oxygen nanobubble, ozone nanobubble, and control treatments (air-stone macrobubble). Experiments were done in triplicate in 100 L tanks with 15‰ saline water, and 20 shrimp per tank. Tanks in Experiment 1 were not inoculated with bacteria; tanks in Experiment 2 were inoculated with V. parahaemolyticus at a concentration of 106 CFU/mL. The results revealed that short treatments with ozone nanobubbles had minimal impact on shrimp gills, mortality, and growth rates, reduced V. parahaemolyticus concentration in water compared to the other groups, and improve water quality. These laboratory results indicate that ozone nanobubble treatment may be useful for controlling V. parahaemolyticus. More work is needed to find the best protocol to apply the technology on a commercial scale.
This study investigated species identification and labeling compliance of 48 shrimp products sold in the Korean online markets. Species identification was conducted using the standard DNA barcoding method, using the cytochrome c oxidase subunit I gene. The obtained sequences were compared with those deposited in the NCBI GenBank and BOLD Systems databases. Additionally, phylogenetic analysis was performed to further verify the identified shrimp species. Consequently, 16 shrimp species were identified, including Penaeus vannamei, Pandalus borealis, Palaemon gravieri, Leptochela gracilis, Penaeus monodon, Pleoticus muelleri, Metapenaeopsis dalei, Euphausia pacifica, Lebbeus groenlandicus, Trachypenaeus curvirostris, Argis lar, Metanephrops thomsoni, Metapenaeopsis barbata, Alpheus japonicus, Penaeus chinensis, and Mierspenaeopsis hardwickii. The most prevalent species was Penaeus vannamei, found in 45.8% of the analyzed products. A significant mislabeling rate of 72.9% was found; however, upon excluding generic names such as shrimp, the mislabeling rate reduced to 10.4%. The mislabeling rate was higher in highly-processed products (89.3%) compared with that in minimally-processed products (50%). No correlation was found between the country of origin and mislabeling rate. The results of this study provide crucial data for future monitoring of shrimp products and improving the labeling of shrimp species in Korea.
This study investigated the effects of dietary supplementation with Sargassum horneri celluclast hydrolysate (SHCH) on the growth performance, feed utilization, non-specific immunity and antioxidant capacity of the Pacific white shrimp Penaeus vannamei. Six diets were prepared by SHCH supplemention levels of 0, 5, 10, 20, 40 and 80 g/kg (designated as Con, S5, S10, S20, S40 and S80, respectively). A total of 540 shrimp were randomly distributed in triplicate across 18 tanks and fed one of the diets for five weeks. Weight gain did not differ significantly among the groups except for S80. The feed conversion ratio was significantly lower in S10 than in S80. Phenoloxidase and nitro-blue tetrazolium activities were significantly higher in S5 than in Con. The prophenoloxidase gene was significantly upregulated in S10 compared to that in Con. The gene expression of crustin, glutathione peroxidase, lipopolysaccharide and β-1,3-glucan-binding protein were significantly upregulated in S5 compared to Con. The superoxide dismutase gene was significantly upregulated in S20 and S80 compared to Con. Supplementing shrimp diets with up to 8% of SHCH did not impair non-specific immunity or antioxidant capacity. However, based on growth results, supplementation up to 4% was considered appropriate.

In this study, whiteleg shrimp (Penaeus vannamei) imported from Vietnam were collected from South Korean markets, and examined for 2 viruses: infectious hypodermal and hematopoietic necrosis virus (IHHNV, recently classified as decapod penstyldensovirus-1), and white spot syndrome virus (WSSV). Among 58 samples, we detected IHHNV in 23 samples and WSSV in 2 samples, using polymerase chain reaction and sequencing analyses. This is the first report of IHHNV and WSSV detection in imported shrimp, suggesting that greater awareness and stricter quarantine policies regarding viruses infecting shrimp imported to South Korea are required.
Vibrio parahaemolyticus associated with acute hepatopancreatic necrosis disease (VpAHPND) and Enterocytozoon hepatopenaei (EHP) are the two most important pathogens in shrimp aquaculture and they have caused enormous losses to the shrimp industry worldwide. In ponds, the major target organ for the two pathogens is the hepatopancreas, and infection with EHP is a known potential risk factor for VpAHPND infection. This study aimed to develop a PCR (polymerase chain reaction)-based diagnostic method for simultaneously detecting VpAHPND and EHP. The newly developed PCR diagnostic method could be used to test various samples, such as seawater, shrimp, and feces. The diagnostic method exhibited high sensitivity and specificity for both pathogens. This will help reduce the potential economic losses that may have been caused by the two major shrimp pathogens, VpAHPND and EHP, and will allow for the efforts and time spent combatting them to be dedicated elsewhere.
A study was conducted to evaluate dietary valine (Val) requirement for Pacific white shrimp (Penaeus vannamei). Five isonitrogenous (353 g/kg) and isocaloric (4.08 kcal/g) semi-purified diets containing graded levels of Val (2.7, 5.1, 8.7, 12.1 or 16.0 g/kg) were formulated. Quadruplicate groups of 12 shrimp (average body weight: 0.46 ± 0.00 g) were fed one of the experimental diets (2%-5% of total body weight) for 8 weeks. Maximum weight gain was observed in 8.7 g/kg Val group. However, the growth performance was reduced when Val concentration in diets were higher than 12.1 g/kg. Feed conversion ratio was significantly increased with 2.7 and 16.0 g/kg Val inclusion. Shrimp fed the diets containing 2.7 g/kg Val showed significantly lower protein efficiency ratio, whole-body crude protein and Val concentrations. Dietary inclusion of Val significantly improved the relative expression of insulin-like growth factor binding protein and immune-related genes (prophenoloxidase, lysozyme and crustin) in the hepatopancreas and 8.7 g/kg Val group showed highest expression among all the groups. The dietary requirement of Val for maximum growth of juvenile P. vannamei, estimated using polynomial regression analysis on growth, was 9.54 g/kg of Val (27.2 g/kg based on protein level) and maximum growth occurred at 9.27 g/kg of Val (26.2 g/kg based on protein level) based on broken-line regression analysis.
DNA-based molecular markers for differentiation of five penaeid shrimps (Penaeus monodon, P. semisulcatus, Feneropenaeus merguiensis, Litopenaeus vannamei and Marsupenaeus japonicus) were developed based on polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and single-stranded conformation polymorphism (SSCP) of 16S ribosomal (r) DNA. Differentiation of P. monodon, P. semisulcatus and L. vannamei can be unambiguously carried out by PCR-RFLP of 16S $rDNA_{560}$ whereas P. semisulcatus and M. japonicus shared a BABB mitotype. These shrimps were successfully discriminated by SSCP analysis of 16S $rDNA_{560}$. Nevertheless, the amplification success for L. vannamei and F. merguiensis was not consistent when tested against larger sample sizes. As a result, 16S $rDNA_{560}$ of an individual representing the most common mitotype of each species was cloned and sequenced. The new primer pair was designed and tested against the large sample sizes (312 bp product, N = 185). The amplification success was consistent across all species. PCR-RFLP of 16S $rDNA_{312}$ was as effective as that of 16S $rDNA_{560}$. Differentiation of all shrimp species were successfully carried out by SSCP analysis.

Plastic waste has emerged as a major environmental concern in recent years. As plastic waste discharged into the marine environment, it undergoes a breakdown process, eventually accumulating in aquatic organisms in the form of microplastics (MPs). To date, reduced food intake, nutritional absorption, and impaired immune system are known adverse effects of MPs-exposed aquatic organisms. This study aims to investigate whether MP exposure accelerated white spot syndrome virus (WSSV) infection in Pacific white shrimp (Penaeus vannamei) via laboratory tests. Briefly, experimental shrimp were divided into four groups; WSSV (group 1); MP (group 2); WSSV + MP (group 3); and Control (group 4). No mortality was observed in group 2, group 4, and even in group 1. However, group 3 showed a cumulative mortality of 50% during the experimental period. The PCR assay results showed no WSSV in the other three groups (groups 1, 2, and 4), but the dead and alive shrimp collected from group 3 were confirmed to be infected with the virus. Histopathological examination revealed normal structures in the hepatopancreas, gill, and muscle tissues of group 4, whereas numerous abnormally shaped nuclei were detected in the gill tissue of group 2. Moreover, group 1 showed minor WSSV-related lesions with few basophilic inclusion bodies in the gills, interestingly, group 3 exhibited severe lesions with numerous basophilic inclusion bodies in the gills. In conclusion, this study confirmed the correlation between the viral disease of shrimp and MPs, which can cause significant economic losses to the shrimp aquaculture industry.
This study involves a blending of intensive and extensive shrimp culture techniques for a hypothetical shrimp farm which uses a combination of heated raceway nurseries and extensive grow-out ponds per year. The present value method of economic analysis is used to determine economic feasibility. The biological data in this reports were obtained from published or personal communications from leaders in the field of shrimp aquaculture. The proposed system showed economic feasibility using the present value method with discount rates of 10% and 12%. The most profitable scenario, the culture of three crops of Penaeus vannamei showed a 1.26 year payback period and 120% annual average rate of return. The breakeven price was $1.25/1b., which is $1.52 less than the market price of $2.77. Breakeven production was 724 1bs/acre, which is 8761bs. less than the assumed 1,600 1bs/acre. All other scenarios 1.2 and 3 crops for P. stylirostris and P. setiferus showed economic feasibility also.

The increasing economic losses associated with growth retardation caused by Enterocytozoon hepatopenaei (EHP), a microsporidian parasite infecting penaeid shrimp, require effective monitoring. The internal transcribed spacer (ITS)-1 region, the non-coding region of ribosomal clusters between 18S and 5.8S rRNA genes, is widely used in phylogenetic studies due to its high variability. In this study, the ITS-1 region sequence (~600-bp) of EHP was first identified, and primers for a polymerase chain reaction (PCR) assay targeting that sequence were designed. A newly developed nested-PCR method successfully detected the EHP in various shrimp (Penaeus vannamei and P. monodon) and related samples, including water and feces collected from Indonesia, Thailand, South Korea, India, and Malaysia. The primers did not cross-react with other hosts and pathogens, and this PCR assay is more sensitive than existing PCR detection methods targeting the small subunit ribosomal RNA (SSU rRNA) and spore wall protein (SWP) genes. Phylogenetic analysis based on the ITS-1 sequences indicated that the Indonesian strain was distinct (86.2% nucleotide sequence identity) from other strains collected from Thailand and South Korea, and also showed the internal diversity among Thailand (N = 7, divided into four branches) and South Korean (N = 5, divided into two branches) samples. The results revealed the ability of the ITS-1 region to determine the genetic diversity of EHP from different geographical origins.
