Preparation of Three Different Style Packed Capillary Frits

  • Three different style capillary columns, a packed capillary with temporary quartz wool frit, a packed capillary with immobilized frit, and an immobilized packed-capillary, were easily prepared with a commercially available (S)-N-(3,5-dinitro-benzoyl)leucine-N-phenyl-N-alkylamide derived chiral stationary phase. Liquid chromatographic chiral separations of some racemic amino acid derivatives on these columns were performed and the results were compared to each other. The packed capillary with immobilized frit showed some merits in chiral chromatography.

Simultaneous Gas Chromatographic Analysis of Ethanol and Acetic Acid in Vinegar (식초의 에탄올 및 초산 함량의 동시분석)

  • A method is developed for simultaneously determining ethanol and acetic acid in vinegars by quantitative packed-column gas chromatography. Vinegars were filtrated and directly chromatographed on a $2\;m{\times}2\;mm$ stainless steel column packed with Tenax-GC, 80/100. Ethanol, isopropy alcohol as an internal standard, and acetic acid were completely separated within 20 min of running time without any interfering peaks. The accuracy of packed column gas solid chromatography (PCGSC) was discussed compared to the analytical data by titration, high performance liquid chromatography and capillary column gas liquid chromatography (CCGLC).

Studies on Analysis of Sterols in Mayonnaise by GLC with Packed and Capillary Column (충전컬럼 및 모세관컬럼 가스크로마토그라프에 의한 마요네즈 중의 스테롤 분석연구)

  • Sterols in mayonnaise were determined by the gas liquid chromatographic(GLC) analysis using two different columns, packed column(silicone SE30) and fused silica capillary column(CBPl). Four kinds of sterol(cholesterol, campesterol, stigmasterol and ${\beta}-sitosterol$) were detected in mayonnaise. The method using capillary column proved to be superior in rapidity, reproducerbility and separability for the determination of sterols. The ranges of concentration of egg york in mayonnaise predicted from cholesterol content using packed and capillary columns were $2.69{\sim}7.11%\;and\;2.42{\sim}6.08%$, respectively. Analyzing the composition of campesterol, stigmasterol and (${\beta}-sitosterol$ in mayonnaise, it could be known that soybean oil and cottonseed oil are commonly used for the manufacture of mayonnaise.

A Capillary Electrochromatographic Microchip Packed with Self-Assembly Colloidal Carboxylic Silica Beads

  • An electrochromatographic microchip with carboxyl-group-derivatized mono-disperse silica packing was prepared from the corresponding colloidal silica solution by utilizing capillary action and self-assembly behavior. The silica beads in water were primed by the capillary action toward the ends of cross-patterned microchannel on a cyclic olefinic copolymer (COC) substrate. Slow evaporation of water at the front of packing promoted the self-assembled packing of the beads. After thermally binding a cover plate on the chip substrate, reservoirs for sample solutions were fabricated at the ends of the microchannel. The packing at the entrances of the microchannel was silver coated to fix utilizing an electroless silver-plating technique to prevent the erosion of the packed structure caused by the sudden switching of a high voltage DC power source. The electrochromatographic behavior of the microchip was explored and compared to that of the microchip with bare silica packing in basic borate buffer. Electrophoretic migration of Rhodamine B was dominant in the microchip with the carboxyl-derivatized silica packing that resulted in a migration approximated twice as fast, while the reversible adsorption was dominant in the bare silica-packed microchip. Not only the faster migration rates of the negatively charged FITC-derivatives of amino acids but also the different migration due to the charge interaction at the packing surface were observed. The electrochromatographic characteristics were studied in detail and compared with those of the bare silica packed microchip in terms of the packing material, the separation potential, pH of the running buffer, and also the separation channel length.

  • 본 연구에서는 이온 크로마토그래피로 환경오염물질들을 분석하고자 할때의 단전들을 해소하기 위하여 모세관 컬럼 이온 크로마토그래피를 개발하였다. 모세관 컬럼은 두가지 형태로 개발하였는데 하나는 packed capillary 컬럼이며 다른하나는 open tubular capillary 컬럼이다. 또한 위와 같이 모세관 컬럼을 개발하여 사용하게됨을써, 부수적으로 펌프, 시료주입기, 억압장치 그리고 전도도 셀 등을 모두 적은 부피를 다룰 수 있는 구조로 바꾸어 주었다. 따라서 본 연구의 결과로 미량환경오염 물질의 분석에 모세관 컬럼 이온크로마토그래피를 이용할 수 있는 가능성을 확인할 수 있었고, 나아가 이온 크로마토그래피의 소형화로 현장에서 직접 분리, 분석을 할 수 있으리라 기대된다.

Capillary Hemangioma of the Thoracic Spinal Cord

  • Capillary hemangiomas are common soft tissue tumors on the skin or mucosa of the head and neck in the early childhood, but very rare in the neuraxis. A 47-year-old man presented with one month history of back pain on the lower thoracic area, radiating pain to both legs, and hypesthesia below 17 dermatome. Thoracic spine MRI showed $1{\times}1.3{\times}1.5\;cm$, well-defined intradural mass at T6-7 disc space level, which showed isointensity to spinal cord on T1, heterogeneous isointensity on T2-weighted images, and homogeneous strong enhancement. The patient underwent T6-7 total laminotomy, complete tumor removal and laminoplasty. Histologically, the mass showed a capsulated nodular lesion composed of capillary-sized vascular channels, which were tightly packed into nodules separated by fibrous septa. These features were consistent with capillary hemangioma.

  • In the refrigerator, the two-phase refrigerant-induced noise of the capillary tube in an evaporator-inlet pipe has been a great concern. The capillary tube is usually covered with insulation-foam packed in the space between inner and outer cabinets without any vibration isolation. Therefore, the refrigerant-induced vibration of the capillary tube can be easily transferred to the outer cabinet, which may increase the radiated noise. In this paper, the characteristics of transferred vibration through the insulation-foam are investigated experimentally by using the refrigerantsupplying equipment. The frequency characteristics, such as dynamic Young's modulus and loss factor, of the insulation-foam are also discussed.

On-Channel Micro-Solid Phase Extraction Bed Based on 1-Dodecanethiol Self-Assembly on Gold-Deposited Colloidal Silica Packing on a Capillary Electrochromatographic Microchip

  • A fully packed capillary electrochromatographic (CEC) microchip with an on-column micro-solid phase extraction (SPE) bed for the preconcentration and separation of organic analytes was prepared. A linear microchannel with monodisperse colloidal silica packing was formed on a cyclic olefinic copolymer microchip with two reservoirs on both ends. Silver-cemented silica packing frit structure was formed at the entrance of the microchannel by electroless plating treatment as a base layer. A gold coating was formed on it by reducing $Au^{3+}$ to gold with hydroxylamine. Finally micro-SPE bed was formed by self-assembly adsorption of 1-dodecanethiol on it. Micro-SPE beds were about 100-150 ${\mu}m$ long. Approximately $10^3$ fold sensitivity enhancements for Sulforhodamine B, and Fluorescein in nM concentration levels were possible with 80 s preconcentration. Basic extraction characteristics were studied.

  • The effects of column length and particle size on the efficiency of separation and characterization of phospholipids (PLs) are investigated using nanoflow liquid chromatography-electrospray ionization-tandem mass spectrometry (nLC-ESI-MS-MS). Since PLs are associated with cell proliferation, apoptosis, and signal transduction, it is of increasing interests in lipidomics to establish reliable analytical methods for the qualitative and quantitative profiling of PLs related to biomarker development in adult diseases. Due to the complexity of PLs, the preliminary separation of PLs is necessary prior to MS analysis. In this study, length of capillary column and the particle size of reversed phase ($C_{18}$) packing materials are varied to find a reliable condition for the high speed and high resolution separation using 8 PL standard mixtures. From experiments, it was found that a capillary column of nLC-ESI-MS-MS analysis for PL mixtures can be minimized to a 5 cm long pulled tip column packed with 3 ${\mu}m$ $C_{18}$ particles without losing resolution.

Development of Ultra-High Pressure Capillary Reverse-Phase Liquid Chromatography/Tandem Mass Spectrometry for High-Sensitive and High-Throughput Proteomics

  • Recently mass spectrometry and separation methods such as liquid chromatography have become major tools in the field of proteomics. In this report, we describe in detail our efforts to develop ultra-high pressure capillary reverse-phase liquid chromatography (cRPLC) and its online coupling to a mass spectrometer by a nanoelectrospray (nanoESI) interface. The RPLC system is constructed in house to deliver LC solvents at the pressure up to 20,000 psig, which is four times higher than conventional RPLC systems. The high operation pressure allows the efficient use of packed micro-capillary columns (50, 75 and 150 ${\mu}$m i.d., up to 1.5 m long). We will discuss the effect of column diameter on the sensitivity of cRPLC/MS/MS experiments and the utility of the developed technique for proteome analysis by its application in the analysis of proteome samples having different levels of complexity.
