Protective Effect of Sasa borealis Leaf Extract on AAPH-Induced Oxidative Stress in LLC-PK1 Cells

  • This study was designed to investigate the protective effect of Sasa borealis leaf extract on 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress in LLC-PK1 cells (porcine kidney epithelial cells). The butanol fraction from Sasa borealis leaf extract (SBBF) was used in this study because it possessed strong antioxidant activity and high yield among fractions. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in a significant decrease in cell viability, but SBBF treatment protected LLC-PK1 cells from AAPH-induced cell damage in a dose dependant manner. To determine the protective action of SBBF against AAPH-induced damage of LLC-PK1 cells, we measured the effects of SBBF on lipid peroxidation and antioxidant enzymes activities of AAPH treated cells as well as scavenging activities on superoxide anion radical and hydroxyl radical. SBBF had a protective effect against the AAPH-induced LLC-PK1 cellular damage and decreased lipid peroxidation and increased activities of antioxidant enzymes such as superoxide dismutase and glutathione peroxidase. Furthermore, SBBF showed strong scavenging activity against superoxide anion radical. The $IC_{50}$ value of SBBF was $28.45{\pm}1.28\;{\mu}g/mL$ for superoxide anion radical scavenging activity. The SBBF also had high hydroxyl radical scavenging activity ($IC_{50}=31.09{\pm}3.08\;{\mu}g/mL$). These results indicate that SBBF protects AAPH-induced LLC-PK1 cells damage by inhibiting lipid peroxidation, increasing antioxidant enzyme activities and scavenging free radicals.

Fucoidan Protects LLC-PK1 Cells against AAPH-induced Damage

  • This study was designed to investigate the protective effect of fucoidan against AAPH-induced oxidative stress in LLC-PK1 cells (porcine kidney epithelial cells). Oxidative stress was induced by exposing of LLC-PK1 cells to the 1 mM 2,2'-azobis(2-amidino propane) dihydrochloride (AAPH) for 24 hr. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in a significant (p<0.05) decrease in cell viability, but fucoidan treatment protected LLC-PK1 cells from AAPH-induced cell damage in a dose dependant manner. To investigate the protective action of fucoidan against AAPH-induced damage of LLC-PK1 cells, we measured the effects of fucoidan on lipid peroxidation and antioxidant enzymes activities of AAPH treated cells as well as scavenging activities on superoxide anion radical and hydroxyl radical. The $IC_{50}$ value of fucoidan was $48.37{\pm}1.54\;{\mu}g/mL$ for superoxide anion radical scavenging activity. The fucoidan also had high hydroxyl radical scavenging activity ($IC_{50}=32.03\;{\mu}g/mL$). These results indicate that fucoidan protects against AAPH-induced LLC-PK1 cell damage by inhibiting lipid peroxidation, increasing antioxidant enzyme activities and scavenging offree radicals.

HPLC-DAD를 이용한 차조기 잎의 Isoegomaketone 및 Perillaketone의 동시분석법 확립 (Simultaneous Determination of Isoegomaketone and Perillaketone in Perilla frutescens (L.) Britton Leaves by HPLC-DAD)

  • This study developed an HPLC analysis method for the determination of isoegomaketone (IK) and perillaketone (PK) in Perilla frutescens (L.) Britton leaves. P. frutescens ethanol extract was optimized through an HPLC analysis using a C18 column ($250{\times}4.6mml$, D, $S-5{\mu}m$, 12 nm) with gradient elution of water and acetonitrile as the mobile phase at a flow rate of 1 mL/min and a UV detection wavelength of 254 nm. The results of this method showed linearity in the calibration curve at a coefficient of correlation ($R^2$) of IK 0.9995, PK 0.9998. The limits of detection (LOD) for IK and PK were $0.234{\mu}g/mL$ and $0.952{\mu}g/mL$. The limits of quantification (LOQ) for IK and PK were $0.017{\mu}g/mL$ and $0.043{\mu}g/mL$. The inter-day precision RSDs of IK and PK in the P. frutescens were 1.25 to 2.69% and 0.36 to 1.10%, respectively, and the intra-day precision RSDs of IK and PK were 0.96 to 2.51% and 0.90 to 1.93%, respectively. The accuracies of IK and PK were 96.31 to 97.92% and 101.26 to 105.14%. In conclusion, this method was applied successfully to the detection of IK and PK in P. frutescens.

무막줄기세포추출물의 LLC-PK1 세포에서의 산화적 스트레스 개선 효과 (Protective Effect of Membrane-Free Stem Cell Extract against Oxidative Stress in LLC-PK1 Cells)

  • 신장에서 발생한 산화적 스트레스는 조직을 손상시키고 이는 만성신장질환으로 이어질 수 있다. 본 연구에서는 LLC-$PK_1$ 신장세포를 이용하여 산화적 스트레스 개선 효과를 살펴보았다. LLC-$PK_1$ 세포에 무막줄기세포추출물을 처리했을 때 체내 항산화 단백질인 heme-oxygenase-1, thioredoxin reductase 1, 및 NADPH quinine oxidoreductase-1의 발현이 증가함을 확인하였다. LLC-$PK_1$에 산화적 스트레스를 유도하기 위하여 3-morpholinosydnonimine (SIN-1)을 처리한 결과 세포생존율이 감소하여 산화적 스트레스로 인해 세포가 손상됨을 확인하였다. 그러나 무막줄기세포추출물을 처리하였을 때 세포생존율이 증가하였으며, $2.5{\mu}g/mL$에서 세포생존율이 58.84%에서 64.43%까지 증가하였다. 또한 무막줄기세포추출물은 LLC-$PK_1$ 세포에서 SIN-1으로 유도된 염증 및 세포사멸을 조절하였다. 염증 관련 단백질인 inducible nitric oxide synthase와 cyclooxygenase-2는 무막줄기세포 추출물을 처리했을 때 단백질 발현이 감소하였고, 세포사멸과 관련된 B-cell lymphoma-2-associated X protein/B-cell lymphoma-2 비율과 cleaved caspase-3, cleaved-poly (ADP-ribose) polymeras의 단백질 발현이 감소함을 확인하였다. 결과적으로 무막줄기세포출물은 SIN-1을 처리한 LLC-$PK_1$ 세포에서 산화적 스트레스에 대한 보호 효과가 있음을 알 수 있었으며, 이들 결과를 바탕으로 무막줄기세포추출물의 항산화 기능성 소재로서의 활용 가능성을 확인하였다.

FTY720에 의한 $LLC-PK_1$ 세포내 sphingoid bases의 축적 (Endogenous Sphingoid Bases Accumulation by FTY720 in $LLC-PK_1$ Cells)

  • FTY720, a novel immunosuppressant, elevated the level of endogenous sphingoid bases in a dose-dependent manner within 3 hr in $LLC-PK_1$ cells. The relative molar ratio of sphingoid bases expressed as sphingosine/sphinganine (SPN/SPA), a biomarker of altered sphingolipid biosynthesis, in $10{\;}{\mu}M$ of FTY720 showed tow-fold increase as compared with the one in control culture. FTY720 under the serum-free medium condition increased only cytosolic free sphingosine concentration, not sphinganine concentration in a time-dependent manner over the 8 hr incubation under the same condition as in serum free cultures, the SPN/SPA ratio began to fluctuate and the number of floating cells as an indicator of cytotoxicity was increased 8 hr after the addition of FTY720 to cultured cells. These results suggest that the process of FTY720-induced cell death in $LLC-PK_1$ cells.

공정능력지수 $C_{pk}$에 대한 보다 나은 비모수적 붓스트랩 신뢰구간에 관한 연구 (Better Nonparametric Bootstrap Confidence Intervals for Capability Index $C_{pk}$)

  • 공정능력지구 $C_{pk}$는 제조공정이 제품을 제대로 생산하고 있는지를 평가하기 위하여 널리 사용되고 있는 측도이다. 최근까지 공정능력지수 $C_{pk}$에 관한 추정문제들이 만히 연구되었는 바, 대부분의 이러한 연구들은 공정분포가 정규분포임을 가정하였다. 하지만 실제 품질관리 현장의 공정으로부터 얻어지는 특성치들이 정규분포를 따르지 않는 경우가 많이 발생하며, 이를 감지하기가 어려울 수 있다. 따라서 본 논문에서는 공정능력지수 $C_{pk}$에 대한 바람직한 구간추정 방법을 제안하기 위하여 6가지 형태의 비모수적인 붓스트랩 신뢰구간을 설정하고 세 가지 공정분포에 대하여 다양하고 포괄적인 모의실험을 통하여 그 효율성에 관하여 비교연구를 하였다.

  • In this paper we study two vector-valued process capability indices $C_{p}$=($C_{px}$, $C_{py}$ ) and $C_{pk}$=( $C_{pkx}$, $C_{pky}$) considering process capability indices $C_{p}$ and $C_{pk}$. Isolation and Antifungal and Antioomycete Activity of Streptomyces scabiei Strain PK-A41, the Causal Agent of Common Scab Disease

  • The actinomycete strain PK-A41 was isolated from a soil sample from pepper fields in Ko-yang, Korea. The strain PK-A41 inhibited the mycelial growth of some plant pathogenic fungi and oomycete, Alternaria mali, Colletotrichum orbiculare, Fusarium oxysporum f.sp. lycopersici, Magnaporthe grisea, Rhizoctonia solani, and Phytophthora capsici. The presence of LL-diaminopi-melic acid in the cell wall extract and the nucleotide sequence of the 16S rDNA region of the strain PK-A41 was assigned to Streptomyces scabiei. Further morpho-logical, biochemical, and pathological analyses also confirmed the strain PK-A41 to be S. scabiei, which is pathogenic to potato tubers. The maximum antibiotic production of the strain PK-A41 was achieved when grown on the glycerol peptone broth (GPB) medium for 9 days.

Production of Exo-polysaccharide from Submerged Culture of Grifola frondosa and Its Antioxidant Activity

  • Exo-polysaccharide isolated from the culture of Grifola frondosa was modified by sodium periodate ($NaIO_4$) and sodium chlorite ($NaClO_2$) to delete polysaccharide part and phenolic compound, respectively, and was investigated what effect has each part of exo-polysaccharide against 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress in porcine kidney epithelial cells (LLC-PK1). Oxidative stress on LLC-PK1 cell was measured by cell viability, lipid peroxidation, superoxide dismutase (SOD), and glutathione peroxidase (GSH-px) activity. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in significant decrease in cell viability, SOD, and GSH-px action, and significant increase in lipid peroxidation. The treatment of exo-polysaccharide and $NaIO_4$ modified sample protected LLC-PK1 cells from AAPH-induced cell damage such as cell viability, lipid peroxidation, SOD, and GSH-px activity in a dose dependant manner (10, 100, and $500{\mu}g/mL$). However, the treatment of $NaClO_2$ modified sample did not affect for cell viability, lipid peroxidation, SOD, and GSH-px activity. The antioxidant activity of exo-polysaccharide was significantly decreased on AAPH-induced LLC-PK1 cell system when phenolic compound was deleted. The antioxidant activity was significantly correlated with the content of phenolic compound of exo-polysaccharide.

Some articulatory reflexes observed in intervocalic consonantal sequences: Evidence from Korean place assimilation

  • This paper examines kinematic characteristics of /pk/ clusters, as compared to /kk/ and /pp/ with varying vowel contexts and speech rate. The results of EMMA data from eight Seoul-Korean speakers indicate as follows. Firstly, comparing /pk/ to /pp/ sequences, lips closing movement was faster and spatially greater in the /a/-to-/a/ context while temporally longer in the /i/-to-/i/ context. It was smaller in spatial displacement and shorter in temporal duration in /pk/ sequences. Peak velocity did not vary. Secondly, comparing /pk/ with /pp/ and /kk/ controls, lip aperture was less constricted in the /a/-to-/a/ context than /i/-to-/i/, but the maximum contact between the upper and lower lips was invariant across different vocalic contexts within /pk/ sequences (/apka/=/ipki/). Categorical reduction of C1 in /pk/ sequences fell in with the low-vowel and fast-rate conditions with across-/within-speaker variability. Gradient reduction of C1 was observed in all C1C2 types, being more frequent in fast rate. Lastly, the jaw articulator was a stable indicator of rate effects. The implication of the current study is that gestural reduction occurs with categorical reduction and general spatiotemporal weakening in the assimilating contexts, while quantitative properties of gestures may be a reason for gradient reduction, not necessarily confined to place assimilation.
