$Eudragit^{\circledR}$ 마이크로캅셀화에 의한 ${\beta}$-락탐계 항생물질의 방출제어제제 개발 (Preparation and Evaluation of Sustained-Release $Eudragit^{\circledR}$ Microcapsules Containing ${\beta}-Lactam$ Antibiotics)

  • Microencapsulations of amoxicillin and cephalexin, using Eudragit RS, RL, E, S and L were investigated. The microcapsules were prepared by the solvent evaporation process in liquid paraffin phase, which is based on dispersion of acetone/isopropanol containing the drug in liquid paraffin. Aluminium tristearate was used as an additive for the preparation of microcapsules. The size distribution, dissolution test and observation by SEM were examined. Good reproducibility in microcapsule preparation was observed. The microcapsules obtained were spherical and free-flowing particles. The dissolution rates of amoxicillin and cephalexin from the microcapsules were considerably decreased as compared with those from amoxicillin and cephalexin powder, respectively. As the dispersing agents (aluminium tristearate) increased, the particle size of microcapsules decreased and the dissolution rate increased. In order to control the release rate of drugs, microcapsules were prepared by mixing Eudragit RS/RL or Eudragit S/L. As Eudragit RL ratio in microcapsule of Eudragit RS/RL increased, the dissolution rate increased. As Eudragit L ratio in microcapsule of Eudragit S/L increased, the dissolution rate increased. Furthermore, the release rates of drugs from Eudragit RS/L or RS/polyelthylene glycol 1540 (PEG 1540) were examined. The dissolution rate of drugs increased with increasing of Eudragit L or PEG 1540 ratio. In conclusion, the release rates of drugs from Eudragit RS/RL or RS/PEG 1540 microcapsule could be controlled, and these microcapsules will be convenient for reducing frequency of administration.

Fabrication of Endothelial Cell-Specific Polyurethane Surfaces co-Immobilized with GRGDS and YIGSR Peptides

  • Polyurethane (PU) is widely used as a cardiovascular biomaterial due to its good mechanical properties and hemocompatibility, but it is not adhesive to endothelial cells (ECs). Cell adhesive peptides, GRGDS and YIGSR, were found to promote adhesion and spreading of ECs and showed a synergistic effect when both of them were used. In this study, a surface modification was designed to fabricate an EC-active PU surface capable of promoting endothelialization using the peptides and poly(ethylene glycol) (PEG) spacer, The modified PU surfaces were characterized in vitro. The density of the grafted PEG on the PU surface was measured by acid-base back titration to the terminal-free isocyanate groups. The successful immobilization of pep tides was confirmed by amino acid analysis, following hydrolysis, and contact angle measurement. The uniform distribution of peptides on the surface was observed by scanning electron microscopy (SEM) and atomic force microscopy (AFM). To evaluate the EC adhesive property, cell viability test using human umbilical vein EC (HUVEC) was investigated in vitro and enhanced endothelialization was characterized by the introduction of cell adhesive peptides, GRGDS and YIGSR, and PEG spacer. Therefore, GRGDS and YIGSR co-immobilized PU surfaces can be applied to an EC-specific vascular graft with long-term patency by endothelialization.

쥐 하이브리도마 세포배양을 위한 무혈청 배지개발( I ) -최적 배지성분의 결정- (Development of Serum-free Media for the Culture of Mouse Hybridoma (I) ; Determination of Optimal Media Composition)

  • 최근 단일클론항체의 상업적 이응가치가 증대하면서 그 수요량 또한 급격히 증가, 이를 뒷받침할 수 있는 대량생산 기술이 요구되고 있다. 본 연구에서는 쥐 하이브리도마 Alps 25-3의 대량배양을 위해 우선적으로 무혈청 배지 (serum-free media)의 개발을 시도하였다. 동물세포 배양액 IMDM과 Ham's F-12를 1:1로 혼합한 배지를 기본으로하여 ITES 용액 (insulin 10$\mu\textrm{g}$/$m\ell$, transferrin 10$\mu\textrm{g}$/$m\ell$, ethanolamine 10$\mu$M and selenium 30nM)을 첨 가하여 일차적으로 결정한 EBM(enriched basal medium)에 steroid hormones, vltamins, lipid, mineral 및 여러성분들을 각각 첨가하여 혈청과 대치될 수 있는 성분들의 최적농도를 조사하였다. 각 성분들의 개별효과와 함께 그들을 혼합하였을 때의 상호효과를 통해 혈청배지와 거의 동일한 성장을 보이는 무혈청배지 KM3(EBM with BSA 100$\mu\textrm{g}$/$m\ell$, mineral solution and 0.05% PEG)를 결정할 수 있었으며 이 배지에서의 최고 항체 생성량은 혈청배지의 약 80% 로 나타났다. 또한 무혈청 배지에서 여러 cell line의 성장과 그들의 항체 생성을 조사함으로써 다른 쥐 하이브리도마 세포배양에도 적응시킬 수 있음을 확인할 수 있었다.

Poly(benzyl-L-histidine)-b-Poly(ethylene glycol) Micelle Engineered for Tumor Acidic pH-Targeting, in vitro Evaluation

  • A polymeric micelle, based on the poly(benzyl-L-histidine)-b-poly(ethylene glycol) (polyBz-His-b-PEG) diblock copolymer, was designed as a tumor-specific targeting carrier. The micelles (particle size: 67-80 nm, critical micelle concentration (CMC); 2-3 $\mu$g/mL) were formed from the diafilteration method at pH 7.4, as a result of self-assembly of the polyBz-His block at the core and PEG block on the shell. Removing benzyl (Bz) group from polyBz-His block provided pH-sensitivity of the micellar core; the micelles were physically destabilized in the pH range of pH 7.4-5.5, depending on the content of the His group free from Bz group. The ionization of His group at a slightly acidic pH promoted the deformation of the interior core. These pHdependent physical changes of the micelles provide the mechanism for pH-triggering anticancer drug (e.g., doxorubicin: DOX) release from the micelle in response to the tumor’s extracellular pH range (pH 7.2-6.5).

플라스미드 유전자를 함유한 리포좀의 제조 및 특성 (Preparation and Characterization of Plasmid DNA Encapsulated in Liposomes)

  • The objective of this study was to construct the pegylated liposomes containing plasmid DNA with optimal encapsulation efficiency. Plasmid DNA $(pGL2\;clone\;753,\;{\sim}6\;kb)$ was encapsulated by the freeze/thawing method into liposomes composed of 1-palmitoyl-2-oleyl-sn-glycerol-3-phosphocholine (POPC), didodecyl dimethyl ammonium bromide (DDAB), distearoylphosphatidyl ethanolamine polyethylene glycol 2000 (DSPE-PEG 2000) and DSPE-PEG 2000-male-imide. The liposomes containing plasmid DNA were then extruded through two stacked polycarbonate filters with series of different pore sizes to control the liposome size. The plasmid DNA entrapped in the liposomes was separated from free plasmid DNA by Sephadex CL-4B column chromatography. The decreased pore size of polycarbonate filters resulted in the decreased size of liposomes. The encapsulation efficiency was markedly affected by the cationic lipid (DDAB) concentration, but to a low degree by the size of liposomes and by the amount of plasmid DNA.

  • The sensitivity of plasmid DNA (pDNA) to S1 nuclease, an enzyme to cleave a single-strand DNA, was dramatically modulated through a supramolecular assembly (polyion complex micelle) with a synthetic block copolymer, poly(ethylene glycol)-b-poly(L-lysine) (PEG-PLL). The pDNA condensed in stoichiometric charge ratio was cleaved into 7 fragments each being 10/12, 9/12, 8/12, 6/12, 4/12, 3/12, and 2/12 of the original DNA length, on the other hand, the pDNA condensed in higher charge ratios (>4), were digested into non-specific manner. Condensation of the pDNA was investigated from two viewpoints that how does the rigid DNA molecules fold and condense and how does the condensation influence their biological activity.

  • 반도체 소자의 집적도가 높아짐에 따라 3D SiP에 대한 관심이 높아지고 전기도금법을 이용한 구리 via filling이 활발히 연구되어왔다. Via filling시 via 입구와 바닥에 전류밀도 차이로 인해 via 내부에 결함이 발생하기 쉽다. 여러 가지 유기물 첨가제와 전류인가 방식의 변화를 통한 via filling을 하였다. 첨가된 유기물은 PEG, SPS, JGB, PEI를 사용하였다. 유기물이 첨가된 용액을 이용하여 펄스와 역펄스 방법을 이용하여 via filling을 하였다. 유기물의 첨가에 따른 도금된 구리 입자의 크기 및 형상에 관하여 고찰하였으며 도금 후 via 시편의 단면을 FESEM으로 관찰하였다. JGB에 비하여 PEI를 사용한 경우 치밀한 도금층을 얻을 수 있었다. 2 step via filling을 사용한 경우 via filling 시간을 단축시킬 수 있었다.

  • Buffalograss는 내한, 내서, 그리고 내한발에 우수한 주요 잔디 종 중 하나이다. 다양한 환경에서 buffalograss의 생리학적 조정(integration)을 이해하는 것은 균일한 잔디의 질을 도모하고 경종적 재배방법의 개발에 도움이 된다. 본 연구의 목적은 물 부족의 스트레스 처리에서의 buffalograss의 생리학적 조정과정에서 lipid peroxidation과 산화방지제의 연관성을 평가하였다. 한 실험에서 buffalograss는 네 개의 구분된 칸막이 성장 유닛의 중심에서 재배되었고, 일주일에 한번(+), 일주일에 두 번(-) 관수처리와 모래(S) 또는 피트(P)와를 혼합한 다섯 가지 토양 조합으로 처리하였다(P+S-P-S+, P+P+P+P+, S-S-S-S-, P-P-P-P-, and S+S+S+S+). 그 결과, 균일하게 혼합된 상토에서 생장한 줄기의 수가 네개의 단일 상토에 정착한 것보다 더 많았다. 두 번째 실험에서는 Hoagland 용액($S_o$), 또는 20% PEG-6000이 함유된 Hoagland용액($S_s$) 안에 하나의 라미트(무성생식체) 혹은 연결된 라마트를 다음과 같은 여러 가지 처리와 비교 실험하였다. 연결된 라미트들의 처리는 Hoagland 용액안의 어린 라미트($Y_{os}$)와 20%PEG-6000가 함유된 Hoagland 용액안의 성숙한 라미트($O_{os}$), Hoagland 용액 단독에 성숙한 라미트($O_{ys}$), 20%PEG-6000 함유된 Hoagland 용액안의 어린 라미트($Y_{ys}$)였다. Lipid peroxidation, antioxidants, proline은 각기 다른 수분 stress 정도에서 라미트들 간의 생리학적 활성을 보여 주었다. Superoxide dismutase (SOD), Guaiacol peroxidase (G-POD), malondi aldehyde (MDA), free proline의 활성도 처리 후 시간에 따라 상대적인 생리학적 활성을 보였다.

  • Electrophoretic deposition (EPD) of alcohol YBCO suspensions on the Ag wire electrode is studied. Poly(ethylene glycol) was coordinated to a structure formed by the EPD process with YBCO particles. The suspension is characterized in terms of zeta potential and conductivity. The d.c electric fields of 200-300 V/cm are applied for 1-10 min. The optimal condition for the EPD allows modifying the properties and microstructure of the deposited films. Superconducting coatings with nanometer-sized pores and a preferred orientation along the c-axis were prepared from the result with chemically modified precursor solution. In contrast, YBCO coatings of submicrometer-sized pores and randomly orientated grains were prepared from the solution without PEG.

  • A series of superabsorbents based on acrylic acid (AA), sodium acrylate, 2-acrylamido-2-methylpropane sulphonic acid, N,N'-methylene bis-acrylamide (MBA) were prepared by inverse suspension polymerization. These hydrogels were further crosslinked on the surface with polyethylene glycol-600 (PEG-600). The water absorbency or swelling behaviors for these xerogels in water and 0.9% saline solutions, both under free condition and under load were investigated. Absorption characteristics of these hydrogels were found to depend on nature and concentration of crosslinker in the system. It was also found that the saline absorption was significantly improved as the incorporation of AMPS in the polymer was increased. The surface crosslinking introduced in the polymers was found to improve the absorption under load characteristics without lowering the free water absorption capacities of the polymer to a considerable extent.
