사람의 눈지방유래 줄기세포의 간세포 분화 (Hepatic Differentiation of Human Eyelid Adipose-Derived Stem Cells)

  • A variety of stem cells has been emerging as therapeutic cells that can replace organ transplantation in human liver diseases. The present study focused on whether human eyelid adipose-derived stem cells (HAD) might differentiate into functional hepatocyte-like cells in vitro. HAD were isolated from human eyelid adipose tissue. Effect of dimethyl sulfoxide (DMSO), fibroblast growth factor (FGF)-2 and FGF-4 on the hepatic differentiation of HAD have been examined in vitro. Immunocytochemical analysis and PAS staining showed that HAD cultured in both DMSO and FGF-4 exhibited the most intense staining than HAD of the other experimental groups. These HAD expressed numerous hepatocyte-related genes. Immunoblotting analyses showed that HAD cultured in the presence of DMSO and FGF-4 secreted higher amount of human albumin than HAD cultured in other conditions. Urea analysis also demonstrated that these HAD produced higher amount of urea than any other groups of HAD. In conclusion, combined treatment of DMSO and FGF-4 could effectively induce the functional differentiation of HAD into hepatocyte-like cells.

늑골에서 발생한 개연골육종의 증례 (A case of canine chondrosarcoma from the rib)

  • We report large tumors of the left thoracic cavity, which arose from the ribs, were diagnosed as chondrosarcoma in a 4-year-old male mixed dog. The dog was presented with swelling in the left side of the chest wall and lameness. The masses were found to be circumscribed, whitish-grey, large and firm. The cut surface revealed whitish-grey lobules of varying size with cartilaginous consistency. Those were subsequently metastasized to the lung and mandibular lymph node. Histologically, the tumors were made up of clusters of chondroid cells having pleomorphic hyperchromatic nuclei with occasional mitosis. A special and immunohistochemical staining was performed to confirm the diagnosis. Chondroid matrix in tumor showed a positive reaction for alcian blue-periodic acid-Schiff staining. Immunoreactivity to S-100 proteins was present in the cytoplasm and/or nucleus of chondrocytes in tumor. The final diagnosis was grade III chondrosarcoma in the rib, considering histological features in grading criteria.

혈당 조절이 불량한 제1형 당뇨병 환자에서 발생한 간의 당원축적증 (Hepatic glycogenosis in a patient with poorly controlled type 1 diabetes mellitus)

  • 당뇨병 환자에서 간비대와 간 기능 이상이 동반될 경우 간 내 당원축적증과 비알콜성 지방간염을 감별해야 한다. 저자들은 인슐린 치료를 제대로 받지 않아 혈당 조절이 불량한 환자에서 저혈당과 간비대로 발현한 간 내 당원축적증을 경험하여 보고한다. 만 6세경 1형 당뇨병으로 진단받은 환아로 10세경 우상복부 통증과 반복되는 고혈당과 저혈당으로 내원하였으며, 당시 진찰상 둥근 얼굴, 우측 늑골연 하방 6 cm 정도에서 만져지는 간비대가 있었다. 검사상 소변 케톤 양성, 당화혈색소는 9.6%였다. 간 효소치가 증가되어 있었고, 혈중 콜레스테롤/중성지방 250/267 mg/dL였다. 바이러스성 간염, 자가면역성 간염, 대사성 간 질환에 대한 검사는 모두 정상이었다. 복부 초음파 및 컴퓨터 단층 촬영상 지방간과 간비대 소견을 보였다. 간 조직 검사상 PAS 염색에서 양성, diastase-PAS 염색에서는 음성으로 전형적인 당원축적증의 소견을 보였다. 고혈당을 보일 때마다 초속효성 인슐린을 투여하고 심한 저혈당의 증상이 동반된 경우 글루카곤을 투여하였으며 생옥수수 전분 가루를 간식으로 먹이면서 저혈당 횟수가 감소하여 퇴원하였다. 현재 외래에서 초속효성 인슐린으로 혈당 조절하면서 관찰 중이다. 제1형 당뇨병 환자에서 당원축적증은 간비대와 간 효소치의 상승을 야기할 수 있는 원인이다. 본 증례에서는 심한 저혈당이 반복되어 하루에도 수 차례 글루카곤을 투여하였으며, 생옥수수 전분 가루을 투여하면서 저혈당이 호전되었다. 엄격한 혈당 관리로 간 비대 및 간 기능 이상이 호전될 수 있으므로 적절한 인슐린 치료로 혈당을 조절하고 저혈당을 예방해야 하겠다.

조피볼락, 용치놀래기, 송곳니베도라치 및 졸복 장관 점액질에 대한 조직화학적 연구 (Histochemical Study of the Intestinal Mucosubstances in Sebastes schlegeli, Halichoeres poecilopterus, Bryzoichthys lysimus and Takifugu pardalis)

  • 경골어류 4종(조피볼락, 용치놀래기, 송곳니베도라치 및 졸복)의 장관 상피 선조연 및 배상세포의 점액질의 조직화학적 성상을 밝히기 위해 PAS 반응, AB pH 1.0 및 pH 2.5, AB pH 2.5-PAS, AF pH 1.7-AB pH 2.5 및 HID-AB pH 2.5 염색법을 사용하였다. 장관 선조연의 점액질은 조피볼락의 근위장과 직장은 중성점액질만을, 중간장과 원위장은 중성점액질과 산성점액질의 혼합성이었으나 용치놀래기는 전 장관에서 중성점액질과 산성점액질을 함유하고 있었으며, 송곳니베도라치와 졸복의 전 장관은 중성점액질만을 함유하고 있었다. 중성점액질의 양은 조피볼락과 용치놀래기는 중등량 내지 상당량, 송곳니베도라치와 졸복은 미량 내지 소량이었다. 장 배상세포 점액질의 양과 성상은 어종 및 장 부위에 따라 차이가 있어 송곳니베도라치와 졸복은 중성점액질만을 함유하고 조피볼락과 용치놀래기는 중성 점액질, sulfomucin 및 sialomucin의 혼합성이었다. 중성점액질의 양은 졸복의 원위장 및 직장은 상당량 내지 다량이었고 조피볼락의 전장, 용치놀래기의 근위장, 원위장 및 직장, 졸복의 근위장 및 중간장은 중등량 내지 상당량이었으며 용치놀래기의 중간장은 미량 내지 소량이었다. 조피볼락의 장 배상세포는 미량의 강 sulfomucin, 약 sulfomucin 및 미량 내지 소량의 sialomucin을 함유하고 있었으며 직장을 제외한 용치놀래기의 장 배상세포는 미량 내지 소량의 강 sulfomucin과 sialomucin을 함유하고 있었다.

가토 저작근 근섬유 분류에 관한 광학현미경적 연구 (Light Microscopic Study on Muscle Fiber Classification of Rabbit Masticatory Muscles)

  • In order to study of muscle fiber proportion of masticatory muscle, 6 rabbits masticatory muscles (masseter, temporal, internal pterygoid, external pterygoid) were excised. Muscle specimens were fixed in 10% neutral buffered formalin fixer and sectioned $5{\mu}$ for PAS staining. With the light microscopic photograph the proportion of muscle fibers of each muscle were computed. The results were as follow; 1. Average classical red fiber proportion of rabbit masticatory muscles was 85.7% 2. Masseter muscle revealed 90.3% of classical red fiber in the rabbit masticatory muscles.

Isoproterenol이 백서악하선(白鼠顎下腺) 및 치주조직(齒周組織)에 미치는 영향(影響) (The Effect of Isoproterenol on Rat Submaxillary Gland and Periodontium)

  • The author has studied the histochemical changes on rat submaxillary gland histological changes on rat periodontium after administration of isoproterenol. The results are as follows: 1) After administration of isoproterenol, the rat submaxillary glands weight of the first and second group were increased two times compared with control group. 2) The acinar cells of rat submaxillary gland were shown the hypertrophy and enlargement. 3) The PAS staining reaction of the acinar cells of rat submaxillary gland were shown the strong positive reaction. 4) The blood vessels of rat periodontium were dilated and alveolar bone resorption was occurred.

백서악하선적출이 갑상선에 미치는 영향

  • It has been said that thyroid gland controls the function of salivary gland and vice versa. In an attempt to investigate the role of submaxillary gland on the function of thyroid gland, the authors have observed the changes of thyroid gland histologically and histochemically at 70 days after removal of submaxillary gland in rats. The results are as follows: 1. There are no significant changes of thyroid cells after submaxillary gland removal. 2. The colloidal concentration of thyroid follicles is markedly decreased after submaxillary gland removal. 3. The colloids of thyroid follicles show intensive reaction to PAS staining in intact rats, but moderate or minimal reaction in submaxillary removal rats.

Purification and characterization of glucosyltransferase and fructosyltransferase from Leuconostoc mesenteroides NRRL B-1149

  • A glucan and a fructan producing enzymes from Leuconostoc mesenteroides NRRL B-1149 were prepared and concentrated from the cu1ture of 1.5% sucrose using polysulfone ultrafiltration hollow fiber in the presence of 0.1% (w/v) Tween 80, 1 mM $CaCl_2$, and 0.02% $NaN_3$. The molecular masses of the enzymes were estimated to be about 213.6 kDa and 180 kDa, respectively, based on the PAS staining for the glucosyltransferase and Mukasa method for fructosyltransferase. Polymers produced by the enzymes showed different solubility; an insoluble glucan and a soluble fructan. The linkages of polymers were determined by methylation using Hakomori reagent and following acid hydrolysis. The glucan was composed of ${\alpha}$-1,6 and 1,3 linkages and the fructan showed similar linkage data of levan.

개에서의 myxoid liposarcoma 증례 보고 (Myxoid liposarcoma in a dog)

  • The pathological features of a mass in the back skin region of an 8-year-old castrated male dog are described herein. The cut section of the tumor was white to tan with a soft multilobulated mass containing hemorrhagic and necrotic foci and a mucinous-like composition. Microscopically, the tumor was composed of a mixture of lipocytes, lipoblasts, spindle cells and stellate cells and had a myxoid background. Oil red O staining revealed that the cytoplasm of neoplastic cells contained large numbers of lipid droplets. Immunohistochemically, tumor cells were positive for vimentin and S-100 protein. The skin mass was diagnosed as myxoid liposarcoma.

생쥐, 쥐 및 돼지의 난소내 난포의 폐쇄에 관한 조직화학적 연구 (Histochemical Study of the Atresia of Ovarian Follicles)

  • In order to study the mechanism of follicle growth and maturation, and also to supplement the criteria identifying the follicle state of normal of atretic, the histochemical investigation on the ovarian follicles according to the ovarian cycle of mouse, rat and pig has been done. The intercellular space of granulosa cells, especailly Call-Exner body, and follicular fluid in the antrum showed positive to PAS, and blue stain by trichrome dye. The resutls suggest that the mucous polysaccharide was synthesized by the granulosa cells, and secreted into the antrum through Call-Exner body so as to be the components of the follicular fluid as the follicles proceeded to growth and maturation. The further the follicles proceeded to atresia the more densely their theca externa were stained blue by follicles proceeded to atresia the more densely their theca externa were stained blue by trichrome dye, and the more densely the granulosa cells were stained red by oil red 0 dye. Therefore, these staining methods can be applied to the criteria identifying the follicle atresia.
