Comparison of Total Phenol, Flavonoid Contents, and Antioxidant Activities of Orostachys japonicus A. Berger Extracts (와송(Orostachys japonicus A. Berger) 추출물의 총 페놀, 플라보노이드 함량 및 항산화 활성 비교)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of Orostachys japonicus A. Berger. Orostachys japonicus A. Berger have been known to contain functional materials such as kaempferol, hydroquinone, methyl gallate, quercetin, gallic acid etc. To identify the main functional materials of Orostachys japonicus A. Berger, the contents of flavonoid and phenol were measured. We extracted Orostachys japonicus A. Berger powder from four solvents such as chloroform:methanol (CM, 2:1, v/v), distilled water (DW), 70% methanol, 70% ethanol. After that, this study determined tannin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power and reducing power of Orostachys japonicus A. Berger extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of Orostachys japonicus A. Berger extracts was higher in the order of 70% ethanol, 70% methanol, DW and CM (p<0.05). The results showed that antioxidant activity of Orostachys japonicus A. Berger extracts supposed to affect by the total phenol and flavonoid contents.

A Study on the Antioxidative Effect of Orostachys Japonicus A. Berger Ethyl Acetate Fraction (와송 에틸아세테이트 분획물의 항산화 효능에 관한 연구)

  • In this study Orostachys japonica A. Berger used is a medicinal herb that has long been used as a folk remedy for cancer treatment. In this study, the antioxidant efficacy of the ethyl acetate fraction of Orostachys japonica A. Berger was confirmed. The results of the Orostachys japonica A. Berge ethyl acetate fraction of antioxidant activity assays showed Antioxidant effect of Orostachys japonica A. Berger EtOAc fraction extract at 0.10 mg/mL was showed a DPPH radical scavenging rate of 78.54% and ABTS+ radical scavenging rate of 73.48%. Also, the toxicity result of Orostachys japonica A. Berger EtOAc fraction extracts using alternative experimental animal model zebrafish, confirmed a 100% the survival of the zebrafish embryo was shown that there was no coagulation and no hatching delay at all concentrations. also ROS generation induced by UV-B irradiation was confirmed that the fluorescence intensity decreased as a whole in all larvae treated with Orostachys japonica A. Berger EtOAc fraction extracts. In particular, it was confirmed that ROS generation was effectively suppressed by showing a 35.7% reduction rate compared to the positive control at a concentration of 3 ㎍/mL. These results were confirmed that Orostachys japonica A. Berger EtOAc fraction extracts has the possibility of application in the cosmetics field as a natural antioxidant.

Total Phenol Contents and Antioxidant Activity in Orostachys japonicus A. Berger Grown under various Cultivation Conditions (재배 조건에 따른 바위솔의 총페놀함량 및 항산화효과)

  • Changes in the contents of total phenolic compounds in as Orostachys japonicus well as their antioxidant capacity according to the control of night-break, day-length control, variation in luminosity and amount of fertilizer used were examined. The results were compared with those obtained from wild Orostachys japonicus. The contents of the total phenolic compounds in Orostachys japonicus were roughly proportional to the increasing level of irradiation and nitrogen fertilization. The total content of phenolic compounds in the wild Orostachys japonicus of was higher than cultivated under various conditions. The antioxidant capacities of the Orostachys japonicus were increased roughly with increasing level of irradiation and nitrogen fertilization. Overall, the total content of phenolic compounds of Orostachys japonicus and their antioxidant capacity were not significantly affected by the amount of phosphorus and potassium fertilizer used.

Antioxidant activities and Quality characteristics of Yanggaeng Added with Orostachys japonicus Powder (와송 분말 첨가 양갱의 항산화 활성 및 품질특성)

  • This study was conducted to investigate the antioxidant effect and sensory characteristics of yanggaeng made with Orostachys japonicus powder results are as follows. Antioxidant effect study, the total polyphenol, DPPH radical scavenging activity, and total flavonoid content showed higher antioxidant effects as the amount of Orostachys japonicus powder added increased. Results of the quality characteristic study, the moisture content increased as the amount of Orostachys japonicus powder added increased, and the pH decreased. In the Hunter's color value results, the L, a, and b values all decreased as the amount of Orostachys japonicus powder added increased. As a result of texture measurement, hardness and chewiness increased with the addition of Orostachys japonicus powder, Cohesiveness decreased, and Springiness showed no significant difference between the control and added groups (p<0.05). The sensory evaluation showed that the color, taste, and overall acceptability were highest in the 4% addition group, the flavor was highest in the 2% addition group, and the softness was highest in the control group. Therefore, it was concluded that it is reasonable to decide on an addition amount of around 4% when manufacturing yanggaeng by adding Orostachys japonicus powder.

The Effects of Artermisiae Capillaris Herba, Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba on Expression of Angiogenic Factors in HepG2 Cells (인진(茵蔯), 울금(鬱金), 상기생(桑寄生), 와송(瓦松)이 HepG2 cell의 혈관생성인자 발현에 미치는 영향)

  • Objectives : This study was designed to investigate the effects of Artermisiae Capillaris Herba, Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba on expression of angiogenic factors in HepG2 cells. Materials and Methods : The mRNA expression level and protein secretion level of angiogenic factors were measured using quantitative RT-PCR, western blot and ELISA assay respectively in Artermisiae Capillaris Herba, Curcumae Rhizoma, Loranthi Ramulus, Orostachys Herba -treated and untreated HepG2 cells. Results : Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba reduced mRNA expression level and protein secretion level of angiogenic factors, but Artermisiae Capillaris Herba increased it, especially VEGF and bFGF in HepG2 cells. Conclusions : The results indicate that Artermisiae Capillaris Herbapromotes expression of angiogenic factors but Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba inhibit expression of angiogenic factors in HepG2 cells. The result is expected that Curcumae Rhizoma, Loranthi Ramulus, Orostachys Herba have an inhibitive effect of angiogenesis in HCC.

Genetic variation in five species of Korean Orostachys (Crassulaceae) (한국산 바위솔속(돌나물과) 5종에 대한 유전적 변이)

  • Starch gel electrophoretic studies using 24 populations of five Korean Orostachys species were conducted to investigate allozyme variation and to test hypotheses of systematic relationships among species. The resulting phenogram showed that the populations of five Korean Orostachys species were divided into two major groups. And they were concordant with molecular and morphological data in suggesting that Orostachys was divided into two groups corresponding to the subsect. Appendiculatae and subsect. Orostachys. The low genetic identities among Korean Orostachys species indicated that the species of Orostachys have diverged gradually through the model of geographical species. Comparing the previous genetic data from the species with similar life history and mode of reproduction, most of Korean Orostachys species revealed a significant low genetic variation, while the widespread O. japonicus showed a relatively high genetic variation among the Korean species. This kind of genetic variation pattern might be the results of the isolated habitats, limited numbers of individuals within the populations, destruction of habitats, inbreeding and asexual reproduction in Korean Orostachys populations. The Jungdongjin population (POP 21) of O. malocophyllus was genetically unrelated with remaining populations of the same species, and this interpretation was consistent to the results from the previous palynological and morphological studies. Our allozyme data supported the taxonomic treatment of recently proposed taxa, O. iwarenge (Makino) Hara for. magnus and O. margaritifolius.

Prediction and discrimination of taxonomic relationship within Orostachys species using FT-IR spectroscopy combined by multivariate analysis (FT-IR 스펙트럼 데이터의 다변량 통계분석 기법을 이용한 바위솔속 식물의 분류학적 유연관계 예측 및 판별)

  • To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts can be used to discriminate cultivars metabolically, leaves of nine commercial Orostachys plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from leaves were analyzed by principal component analysis (PCA) and Partial least square discriminant analysis (PLS-DA). The dendrogram based on hierarchical clustering analysis of these PLS-DA data separated the nine Orostachys species into five major groups. The first group consisted of O. iwarenge 'Yimge', 'Jeju', 'Jeongsun' and O. margaritifolius 'Jinju' whereas in the second group, 'Sacheon' was clustered with 'Busan,' both of which belong to O. malacophylla species. However, 'Samchuk', belong to O. malacophylla was not clustered with the other O. malacophylla species. In addition, O. minuta and O. japonica were separated to the other Orostachys plants. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from leaves represented the most probable chemotaxonomical relationship between commercial Orostachys plants. Furthermore these metabolic discrimination systems could be applied for reestablishment of precise taxonomic classification of commercial Orostachys plants.

A Study on the Evaluation of Antimicrobial Effect of Orostachys Japonicus A. Berger Ethyl Acetate Fraction (와송 에틸아세테이트 분획물의 항균효능 평가에 관한 연구)

  • In this study, a study was conducted to utilize Orostachys japonica A. Berger EtOAc fraction extract as an antibacterial activity and cosmetic ingredient. As a result of measuring the antimicrobial activity of Orostachys japonica A. Berger EtOAc, the growth of S. aureus, S. epidermidis, and P. aeruginosa was inhibited. Among them, S. aureus was an extract of 18.35 ± 1.5 mm Orostachys japonica A. Berger EtOAc fraction at a concentration of 0.5 g / mL, showing superior antibacterial activity than methyl paraben (16.83 ± 1.0 mm), and was shown as a positive control. As a result of evaluating the MIC of the Orostachys japonica A. Berger EtOAc fraction extract through MIC measurement, the remaining strains excluding Candida. A showed a MIC of 17.5 mg/mL. As a result of evaluating the cosmetic preservation effect through the challenge test applied to the cosmetic emulsion formulation, the growth inhibitory effect of S. aureus in the emulsion containing 0.3% Orostachys japonica A. Berger EtOAc fraction extract 7 days after microbial inoculation was 100%.

Morphological Characteristics and Material Comparison of Native Orostachys species (Orostachys japonica (Maxim.) A. Berger, Orostachys minuta (Kom.) A. Berger, Orostachys chongsunensis Y. N. Lee) (자생 바위솔(바위솔, 정선바위솔, 좀바위솔)의 형태적 특성과 성분 비교)

  • Orostachys species grow from anywhere in Korea. This variety, which lives in the harsh areas of the beach and mountains, is known to have as much change as it was isolated. It has been used as a medicine for a long time, and now the flower is beautiful and has been developed for horticulture. As a result of morphological characteristics and component analysis of three Orostachys species, the following results were obtained. 1. Orostachys species showed two types of leaves according to the growing environment. O. japonica (Maxim.) A. Berger and O. minuta (Kom.) A. Berger were lanceolate, and O. chongsunensis Y. N. Lee was obovate. 2. The leaves were all green. However, some of the O. japonica (Maxim.) A. Berger had white powder. Some of the O. minuta (Kom.) A. Berger was green with red spots on the back. O. chongsunensis Y. N. Lee showed strong gray before flowering but weak green when flowering. 3. The thorns of the leaves were O. japonica (Maxim.) A. Berger and O. minuta (Kom.), but O. chongsunensis Y. N. Lee was not. 4. Whether it was a leaf dot or not, there were dots in O. japonica (Maxim.) A. Berger and there were dots on the O. minuta (Kom.) and the O. chongsunensis Y. N. Lee. 5. The flowers were all white, and the stamens were red in O. japonica (Maxim.) A. Berger and O. minuta (Kom.) A. Berger and white in O. chongsunensis Y. N. Lee. 6. It was confirmed that the ingredients in Orostachys species contained kaempferol, quercetin, flavonoid etc., which have high antioxidant activity.

Determination of Steroids and Tetracyclic Triterpenes in Orostachys japonicus A. Beger Grown under Various Cultivation Conditions Using Gas Chromatography (여러가지 조건하에서 재배한 바위솔에서 스테로이드와 테트라싸이클릭 트리테르펜의 가스 크로마토 그래피를 이용한 분석)

  • The content of two steroids-campesterol (1) and ${\beta}$-sitosterol (2), and four triterpenes-taraxetrone (5), ${\beta}$-amyrin (6), (-)-friedelin (7), glutinol (8) in the Orostachys japonicus A. Berger cultivated under various conditions was estimated and compared with those in wild one. The present investigation disclosed that there are no significant difference in their contents between cultivated Orostachys japonicus A. Berger and wild one. from viewpoint of the content of the steroids 1 and 2, and the triterpenes 5-8, the quality of cultivated Orostachys japonicus A. Berger is not inferior to the wild one.
