녹장균(Metarhizium ansiopliae)을 생물살충제로서 활용할 목적으로 녹강균의 균사생육 및 포자형성에 미치는 환경요인과 균사외 분비 효소 활성에 대하여 조사였다. M. anisopliae DGUM35001의 균사 생장을 보여주었으며, 공중균사는 SDPY 사용시 가장 우수하엿다. 최소배지로 Czapek-Dox 한천배지를 사용하여 탄소원 이용성 실험의 결과, 단당류에서 glucoserk 이당류에서 sucrose가 가장 우수한 탄소원이었다. 고분자 물질로 chitin 사용시 우수한 균사 생육을 보여주었다. 유기질소원으로 bacto-peptone과 soytone아 무기질소원으로 urea와 ammonium phosphate가 우수한 균사생육을 나타내었으며, 아미노산으로 serine 첨가시 가장 우수한 균사 생육을 나타내었다. Starch배지를 사용하였을 때 가장 우수한 포자형성의 결과를 얻었다. 액체배양후 군사를 제거한 상등액을 조효소액으로 사용시 analyase와 prorease 효소 활성이 우수하였다. 그러나,;ipase와 chitinase 효소활동은 상대적으로 낮았다.
Livestock manure is generally beneficial to soil and crop production when appropriate amount is applied, but excessive application may be detrimental to soil and water environments. A proper protocol of livestock waste treatment is required to manage the quality of soil and water. A trickling filter system using rice straw media was employed to treat piggery wastewater from small-scaled livestock farms as an alternative to the currently available methods. Batches of piggery wastewater were treated with this system, and the byproducts of rice straw media and trickling filtrate were applied to the soil with cultivating rye (Secale cereale L.). Objective of this research was to characterize these byproducts for the sustainable soil amendments and rye production. Both the treated straw medium and filtrate were proven to be effective organic fertilizers for rye plant development, with the enhanced but balanced absorption of nutrients. The synergistic effects of filtrate in addition to straw application did not show, but the filtrate appeared to lead to a higher water content of the plant. No specific nutrient deficiency or toxicity symptom was shown due to the salts derived from the byproducts applied. Chemical parameters of the soil quality were significantly improved with the application of straw medium either with or without the filtrate. Judging from parameters relating to the salt accumulations, such as sodium adsorption ratio (SAR), electrical conductivity (EC), exchangeable sodium percentage (ESP), potassium adsorption ratio (KAR), and residual P concentrations, the byproducts from piggery wastewater exhibited no detrimental effects on soil quality within the ranges of treatments used. In addition to the effectiveness of the rice straw trickling filter system for the small-scaled swine farms, both rice straw medium and filtrate could be recycled for the sustainable soil amendment and plant nutrition.
여과공정에서 망간제거특성을 고찰하기 위하여 모래, 망간사 등의 4가지 여재를 사용한 여과실험이 실시되었다. 여과속도는 123 m/d였고, 칼럼당 유입량은 $3.9m^3/d$이었다. 실험기간은 약 1년이었으며, 실험기간 동안 여재별로 이산화망간 부착량, 탁도제거율, 망간제거율, 유기물제거율 등을 고찰하였다. 평균 망간농도 0.208 mg/L의 여과지 유입수(잔류염소 1.0 mg/L)를 사용하여 여과실험한 결과 모래+망간사 칼럼은 98.9%, 망간사칼럼은 99.2%의 망간제거율을 보였다. 기존 모래여과지를 망간제거를 위해 사용할 경우 여층의 전부를 망간사로 교체하지 않고 일부만 교체하여도 망간제거에 충분한 효과가 나타나는 것으로 조사되었다.
미디어 빅뱅이란 말이 나올 만큼 미디어가 폭발적으로 늘어나고 있다. 하지만 정작 저널리즘은 위기를 맞는 역설적 현상이 나타나고 있다. '사람들(수용자)이 자유로워지고 자신을 스스로 통제하는 데에 필요한 정보를 제공하는' 저널리즘의 목적에 비추어 볼 때, 저널리즘의 위기는 곧바로 '수용자 복지'의 훼손으로 귀결된다. 따라서 저널리즘 위기의 실체를 정확히 규명하는 일이 저널리즘의 공공성을 확보할 수 있는 선행과제다. 이 논문은 저널리즘의 존재 원칙에서 진실과 공정을 기본윤리로 개념화하고, 한국 저널리즘이 얼마나 그 기준에 부합하는지를 분석했다. 그 결과 우리 시대가 풀어가야 할 주요 의제에 대해 한국 저널리즘은 진실과 공정의 기본 윤리조차 지키지 않고 있음을 확인했다. 저널리즘 위기의 실체를 근본적으로 '존재 원칙의 위기'로 규명한 뒤 이를 극복하기 위한 방안을 제시했다. 미국의 '저널리즘을 걱정하는 언론인위원회'의 연구를 소개하며, 한국 언론학계가 언론 단체와 유기적 결합을 통해 저널리즘 살리기에 나설 것을 제안했다.
꽃송이 버섯의 균사생장은 $25^{\circ}C$의 온도와 pH4의 조건에서 가장 양호하였으며, Hamada 배지에서 가장 조밀한 생장을 나타낸 반면, Hoppkins 배지에서 가장 큰 직경생장을 나타내었다. Maltose, Arabinose 및 Mannitol 같은 탄소원은 꽃송이 버섯의 양호한 생장을 촉진하였다. 또한, 질소원의 하나인 Glycine도 양호한 균사생장을 나타내었다. 배지에 탄소원을 1%로 고정시킨후 C/N비를 20:1로 하였을 때, 꽃송이 버섯 균사의 생장이 가장 양호하였다. 유기산으로서 Fumaric acid와 Lactic acid의 처리를 하였을 때, 꽃송이 버섯의 생장이 좋았다.
The availability of large quantities of waste woods provides an impetus for investigating woody biomass potential uses. Polyurethane (PU) foams are prepared with reacting isocyanates and polyols, and are used. in various industry fields. Thus, lignocellulosic waste raw-materials are proposed as replacement for synthetic polyol to PU foam formulation. In this study PU foams were manufactured from liquefied woods, methanediisocyanate(MDI), catalyst, foaming stabilizer, and viscosity aids. The polyol content, isocyanate.hydroxyl group (NCO/OH) ratio, and water content were varied to evaluate their effects on the foaming and water absorption of the PU foams. Less than 400 Molecular weight. of polyethylene glycol(PEG) and 1 to 3 solvent to woody raw-material ratio were desirable for liquefying woody materials. Liquefying rate was increased with more than 3 % addition of inorganic and organic catalysts and raising reaction temperature more than $150^{\circ}C$. Addition of starch enhanced liquefying of woody materials. Fourty percents of starch resulted in about 90% liquefying rates. Foaming rates were increased with increasing moisture contents of liquefied wood. Moisture contents of 0.6% resulted in 5 time-foaming rates, and seven percents of moisture contents more than 30 time-foaming rates. But, an increase in water content may result in a decrease in cross-links between wood polyol and isocyanate, because the NCO/OH ratio is constant. Increasing moisture contents have significantly decreased density of PU foams. The optimum water content should be about 2.5% or less in this adopted condition.
Horizontal-Flow Roughing Filtration (HRF) is one of altemative pretreatment methods e.g. prior to Slow Sand Filtration (SSF). However, some of its limitations are that the effluent quality drops drastically at higher turbidity (>200 NTU) and at higher filtration rate (>1 m/h). To overcome these drawbacks, we suggested Direct Horizontal-Flow Roughing Filtration (DHRF), which is a modified system of Horizontal-Flow Roughing (HRF) by addition of low dose of coagulant prior to filtration. In this study to optimize the DHRF configuration, a conceptual and mathematical model for the coarse compartment has been developed in analogy with multi-plate settlers. Data from simple column settling test can be used in the model to predict the filter performance. Furthermore, the model developed herein has been validated by successive experiments carried out. The conventional column settling test has been found to be an handy and useful to predict the performance of DHRF for different raw water characteristics (e.g. coagulated or uncoagulated water, different presence of organic matter, etc.) and different inital process conditions (e.g. coagulant dose, mixing time and intensity, etc.). An optimum filter design for the coarse compartment (grain size 20mm) has been found to be of 3 m/h filtration rate with filter length of 4-4.5 m.
Previously, Pseudomonas plecoglossicida YJR13 and Pseudomonas putida YJR92 from a sequential screening procedure were proven to effectively control Phytophthora blight caused by Phytophthora capsici. In this study, we further investigated the anti-oomycete activities of these strains against mycelial growth, zoospore germination, and germ tube elongation of P. capsici. We also investigated root colonization ability of the bacterial strains in square dishes, including cell motility (swimming and swarming motilities) and biofilm formation. Both strains significantly inhibited mycelial growth in liquid and solid V8 juice media and M9 minimal media, zoospore germination, and germ tube elongation compared with Bacillus vallismortis EXTN-1 (positive biocontrol strain), Sphingomonas aquatilis KU408 (negative biocontrol strain), and MgSO4 solution (untreated control). In diluted (nutrient-deficient) V8 juice broth, the tested strain populations were maintained at >108 cells/ml, simultaneously providing mycelial inhibitory activity. Additionally, these strains colonized pepper roots at a 106 cells/ml concentration for 7 days. The root colonization of the strains was supported by strong swimming and swarming activities, biofilm formation, and chemotactic activity towards exudate components (amino acids, organic acids, and sugars) of pepper roots. Collectively, these results suggest that strains YJR13 and YJR92 can effectively suppress Phytophthora blight of pepper through direct anti-oomycete activities against mycelial growth, zoospore germination and germ tube elongation. Bacterial colonization of pepper roots may be mediated by cell motility and biofilm formation together with chemotaxis to root exudates.
국내에 문제가 되는 담배가루이(Bemisia tabaci)의 살충성 Lecanicillium lecanii Btab01 균주의 생육 특성과 살충력을 검정하였다. PDA, TSA, SDA+Y배지에서 L. lecanii Btab01은 $20{\sim}25^{\circ}C$에서 균사성장이 가장 빨랐으며 $28^{\circ}C$이상에서 성장이 억제되었다. L. lecanii Btab01는 고체배양과 액체배양 시 담배가루이 약충에 대해 살충률에 큰 차이를 보이지는 않았다(93.9%, 98.3%). 농도별로는 $10^5$, $10^6$, $10^7$, $10^8$ cfu/ml에서 각각 5.8%, 33.8%, 77.3%, 98.5%의 살충률을 보였고 각각 16.1일, 7.3일, 5.1일, 3.5일의 $LT_{50}$값을 나타내었다. L. lecanii Btab01을 약충에 처리 후 포화습도 조건을 0 시간 24 시간, 168 시간 유지하였을 때, 0%, 20.3%, 100%의 살충률을 보였다. 균주에 0.5% 식용유를 첨가하였을 때 포자의 발아가 2 배 가량 증가하였으며 균주에 0.1% 식용유를 첨가하여 약충에 처리했을 때(98.6%)가 첨가하지 않았을 때(79.9%)보다 살충력이 향상됨을 알 수 있었다.
양돈폐수의 효율적인 혐기성 처리를 위하여 실험실 규모 UASB, AF, 그리고 이단 ASBF 생물반응기를 구성하고 HRT 1일과 OLR $5.1kg-COD/m^3{\cdot}d$까지 200일간 연속운전하고 운전인자가 공정에 미치는 영향과 처리효율을 비교하였다. 양돈폐수는 유기물, 고형물, 그리고 암모니아를 고농도로 함유하고 있으며 축사의 형태와 계절의 변화에 큰 영향을 받는다. 하수종말처리장 혐기성 소화조 잉여 슬러지에 의한 혐기성 생물반응기의 식종은 효과적이었으며 적응기간은 약 40일 정도 소요되었으나 media로 충전된 필터형 생물반응기인 AF와 이단 ASBF의 적응 속도가 UASB보다 빨랐다. 유입수 T-N 370~800mg/L에서 생물반응기는 77~91% 의 높고 안정된 COD 제거율을 보였으나 유입수 T-N 760~1,310 mg/L에서 COD 제거율은 24~94%로 불안정하였으며 전반적으로 감소되었다. 양돈폐수에 함유된 암모니아는 메탄균에 대한 저해작용을 나타낼 수 있는 충분한 농도이므로 효율적인 혐기성 소화처리를 위하여 전처리로서 암모니아의 제거는 필수적이다. 생물반응기는 $1.1{\sim}2.2kg-COD/m^3{\cdot}d$의 비교적 낮은 OLR에서 COD 제거효율은 78.9~81.5%, 그리고 biogas 발생량은 $0.39{\sim}0.59m^3/kg-COD_r$로 효과적이었으나 유입수 COD의 증가 혹은 HRT 단축운전에 의한 OLR의 증가에 따라 COD 제거효율은 감소하였으며 UASB는 AF와 이단 ASBF보다 COD 제거효율 감소폭이 컸다. AF와 이단 ASBF는 충전된 media와 이단화에 의하여 biomass의 보유능력이 우수하며 높은 OLR에도 안정적이며 높은 COD 제거율올 나타내므로 고농도의 유기물을 함유하며 축사의 형태와 계절에 따라 특성의 변화가 큰 양돈폐수의 처리에 효율적이다.
