A Study of the Growth Condition and Solubilization of Phosphate from Hydroxyapatite by Pantoea agglomeraus

  • The growth conditions of Pantoea aggicmerans, a phosphate solubilizing organism, were studied In our laboratory to determine the optimal conditions. Pantoea aggionerans showed the highest growth rate at 30$\^{C}$, pH 7.0 and 2 vvm, after 50 h cultivation. A certain relationship between pH and phosphate concentration was evident when the glucose concentration in the me dium was changed. Increasing glucose concentration increased the pH buffer action of the broth. At glucose concentrations higher than the optimum concentration of 0.2 M, the cell growth was retarded. P. agglomerans consumed glucose as a substrate to produce organic acids which caused the pH decrease in the culture medium. The phosphate concentration in the medium was increased by the presence of the organic acids, which solubilized insoluble phosphates such as hydroxyapa-tite.

  • pKi values and sensitivity of acid-base indicators in the mixed solvent systems of water-alcohols and water-acetone at differential pH and in concentration media were determined by spectrophotometric method. When the concentration of the organic solvents were increased, the pKi values of sulfophthalein indicators, thymol blue and bromcresol purple, became larger whereas those of an axo indicator, methyl yellow, became smaller nad the pH values of the equivalent points of acids became smaller and the pH values of the equivalent points of acids became higher than those in the aqueous solution. potassium bicarbonate, a standard compound used in the organic solvent systems without a prerequisite procedure of expelling carbon dioxide by boiling.

유기산성약물이 푸로세미드의 흡수, 배설및 이뇨효과에 미치는 영향 (The Effects of Organic Acidic Drugs on the Absorption, Excretion and Diuretic Action of Furosemide)

  • The effects of organic acidic drugs on the absorption, excretion and diuretic action of furosemide were studied. Cefalexin, p-aminohippuric acid (PAH), ibuprofen and p-amino salicylic acid (PAS) were selected as organic acidic drugs. The in situ absorption rate and absorption rate constant of furosemide (30$\mu{M}$) were significantly (p<0.05) decreased by 30$\mu{M}$ of cefalexin, PAH, ibuprofen and PAS in rat small intestine. The plasma concentration of furosemide was significantly (p<0.01) increased by cefalexin, PAH and ibuprofen in rabbits. But the urinary excretion rate, renal clearance and diuretic action of furosemide were significantly (p<0.05) decreased by cefalexin, PAH, ibuprofen and PAS in rabbits.

갈수기 정수장운영관리 사례 - 갈수기 pH저감제(황산)투입에 의한 정수처리효율 향상 (Improvement of Water Treatment Efficiency by pH Decreasing Agent (H2SO4) for Droughty Seasons)

  • Drinking water treatment is enhanced by coagulant dosages and chlorine injection because of pH increase in raw water in droughty seasons such as spring and fall. But water quality deterioration is occurred by increase in residual aluminium and disinfection by-products. Coagulation process can be used to control natural organic matter (NOM) during water treatment. The effect of coagulation process appeared to depend on the pH of water rather than coagulant dosages. In this study, for water treatment in high pH season $H_2SO_4$ was applied for pH adjustment at full scale. Before and after pH adjustment by $H_2SO_4$ injection, water quality of drinking water was evaluate. In the result of investigation of total organic carbon (TOC) removal in high pH season, TOC was removed approximately 30~40%, which showed decrease in water treatment efficiency. Also, it is increased both particle numbers and residual Al concentration in the water. After $H_2SO_4$ injection for adjustment to pH<7.5 in settled water, treated water turbidity decreased in 0.047 NTU from 0.059 NTU, and particle numbers of filtered water decreased in 20/mL from 90/mL. On the other side, TOC removal efficiency increased in approximately 10% after adjustment of pH. In the result of decrease in pH in raw water through more coagulants and prechlorine without $H_2SO_4$ injection, trihalomethanes (THMs) concentration increased in $16{\mu}g/L$ from $8{\mu}g/L$.

  • In order to clarify the impact of Ca-binding sites (Ca1 and 2) on the conformational stability of neutral proteases (NPs), we have analyzed the thermal, pH and organic solvent stability of a NP variant, V189P/A195E/G203D/A268E (Q-mutant), from Salinovibrio proteolyticus. This mutant has shown to bind calcium more tightly than the wild-type (WT) at Ca1 and to possess Ca2. Q-mutant was resisted against autolysis, thermoinactivation and pH denaturation in a Ca-dependent manner and exhibited better activity in organic solvents compared to the WT enzyme. These results imply that Ca1 and Ca2 are important for the conformational stability of NPs.

시화호 퇴적물에서 인의 지화학적 특성 (The geochemical properties of phosphorus from sediments of Lake Shihwa)

  • 시화호 퇴적물에서 인의 지화학적 특성을 연구하였다 퇴적인의 가장 큰 형태는 detrital apatite P와 Al-bound P이며 무산소환경이기 때문에 Fe-bound f는 그 비중이 낮았다. 퇴적총인과 유기물의 상관성으로 볼 때, 퇴적물로 제거되는 인의 거동은 유기물에 의해 조절되는 것으로 나타났다. 또한, 유기물 유입의 영향을 직접적으로 받는 정점 Cl에서 정점 C2에 비해 퇴적인의 함량, 유기물과의 상관성, 황산염환원에 의한 공극수내 인산염의 함량 및 플럭스가 높게 나타났다. 결국 시화호 퇴적물에서 인의 함량, 형태 및 공간에 따른 차이는 유기물에 의해 조절되는 것으로 밝혀졌다.

유기질비료의 시용과 수확시기가 Kenaf (Hibiscus cannabinus L.)의 생산성과 사료가치에 미치는 영향 (Response of Organic Fertilizer Application Rates and Different Harvesting Periods on Forage Yield and Quality of Kenaf (Hibiscus cannabinus L.))

  • 하계사료작물로 케나프를 재배할 시에 유기 및 화학비료의 질소공급원과 수확시기를 달리하였을 때 케나프의 생산성과 양질의 조사료를 얻기 위해 적정 질소시용수준과 수확시기를 구명하고자 실시되었다. 질소공급원에 따른 케나프의 건물수량은 화학비료 처리 구가 다소 높았지만, 유기질비료를 ha 당 200~250 kg의 수준에서는 화학비료 처리 구와 유의한 차이가 인정되지 않았고 사료가치 특히 조단백질과 TDN 함량은 오히려 유기질비료 처리 구에서 유의하게 높게 나타나(p<0.05), 반추동물에게 양질의 조사료 공급원으로 이용하고자 케나프가 재배된다면 유기질비료가 화학비료를 대체할 수 있음을 시사하였다. 한편 수확시기에 따른 건물수량은 100일째가 다른 수확시기보다 유의하게 높았지만 100~110일 수확시기가 수량구성요인 중 잎의 비율이 줄기보다도 높았고 사료가치 중 조단백질과 TDN 함량이 다른 수확시기보다 높게 나타났으며 ADF와 NDF 함량은 크게 감소하였다(p<0.05). 본 연구의 결과 특히 건물수량과 사료가치를 비교해 볼 때 적절한 질소공급원으로 유기질비료를 시용 시에는 ha당 200~250 kg 정도가 적절하고 수확시기는 100~110일이 적합하다고 사료된다.

Ethanol과 유기산의 첨가가 물김치의 품질에 미치는 영향 (Effect of Addition of Ethanol and Organic Acids on the Quality of Mul-kimchi)

  • 물김치에 ethanol. 또는 유기산 중 adipic acid, citric acid, acetic acid을 첨가하였을 때 김치의 발효 지연효과를 pH, 산도, 미생물수의 변화를 측정하여 조사하였으며 동시에 ethanol과 유기산을 첨가한 이 김치의 기호성을 살펴보아 다음과 같음 결과를 얻었다. 물김치에 ethanol을 0~5% 첨가하여 pH 변화를 조사한 결과 ethanol 함량이 높을수록 pH의 저하가 지연되었다. ethanol은 특히 발효 초기에 pH 저하 지연효과를 보여 2$0^{\circ}C$의 경우 발효 초기에 ethanol를 첨가하지 않은 김치는 pH 5.7에서 4.13까지 감소하는 반면 ethanol 5%를 첨가한 김치에서는 pH 5.8에서 5.14로 약간 감소하는데 그쳤다. 산도의 경우도 ethanol 함량이 높을수록 산도가 낮게 나타났으며 ethanol를 첨가하지 않은 김치에서는 산도 0.7~0.8%까지 증가하였으나 ethanol 5%를 첨가한 김치에서는 산도가 0.5~0.6%로 산도 증가가 억제되었다. 물김치에서 유기산 종류와 농도를 달리하여 첨가하였을 경우 사용된 유기산 중 adipic acid 첨가시 가장 발효 지연 효과가 컸으며 adipic acid 농도가 높아질수록 pH와 산도 변화가 적었다. Ethanol 2%와 adipic acid 0.1%를 병용 첨가한 결과에서는 유기산만을 첨가한 효과와 유사한 경향을 보였다. Ethanol를 2% 첨가한 물김치의 미생물 변화는 발효 중반이후 미생물 수가 낮게 나타났다. 물김치에 위에 사용된 유기산들을 0.025~0.2% 첨가하였을 때 대체적으로 유기산의 함량이 높을수록 균수가 감소하였으며 유기산 중 adipic acid에서 총균수와 젖산균수의 억제 효과가 가장 컸다. 물김치에 ethanol과 adipic acid을 첨가한 경우가 미생물 균수가 억제에 가장 효과가 있었다. Ethanol과 유기산 농도를 달리하여 제조한 김치의 기호도를 조사한 결과 맛과 향에서 control과 ethanol 2%를 첨가한 김치는 알콜 내, 알콜 맛에서만 유의적인 차이를 나타낸 반면 ethanol 2%와 adipic acid 0.1%를 병용 첨가한 김치는 전 항목에서 유의적인 차이를 나타내었다. 또한, 전반적인 기호도에서는 control이 3.4점으로 ethanol을 첨가한 김치와 같은 기호성을 나타내었고 ethanol 2%와 adipic acid 0.1%를 첨가한 김치도 3.2점으로 유사한 기호성을 보였다.

저압 나노여과 공정에서의 유해성 유기물질의 거동 (Behavior of Hazardous Organic Compounds in Low-Pressure Nanofiltration Process)

  • Behavior of hazardous organic compounds including bisphenol A, phtalic acid, and phosphoric acid in low pressure nanofiltration process were investigated. In the case of NTR729HF, rejection of all target organic compounds except 2-H-Benzothiazol and 2-isopropyl phenol was more than 90%. The lowest rejection for 2-H-Benzothiazol was observed in another membranes. The UTC60 and UTC20 showed similar rejection characteristics of hazardous organic compounds. Although the rejection of Bisphenol A, n-buthyl benzenesulfoneamide, N-ethyl-p-toluensulfonamide, 2-H-benzothiazol, p-t-butylphenol and 2-isopropyl phenol was less than 30%, the rejection of tributyl phosphate, triethyl phosphate, camphor, 2,2,4 trimethyl 1,3 pentandiol and diphenyl amine was more than 90% in the case of UTC60 and UTC20. The rejection characteristics of various hazardous organic compounds were converted into one parameter Ks, which was proposed in the diffusion-convection model. The Ks of hazardous organic compounds were discussed by comparing with their solute size represented by Stokes radius. The diffusion convection model considering Ks was successful to interpret rejection characteristics of hazardous organic compounds by low-pressure nanofiltration membranes.

Effect of slag on stabilization of sewage sludge and organic soil

  • Soil stabilization is one of the useful method of ground improvement for soil with low bearing capacity and high settlement and unrequired swelling potential. Generally, the stabilization is carried out by adding some solid materials. The main objective of this research was to investigate the feasibility of stabilization of organic soils and sewage sludge to obtain low cost alternative embankment material by the addition of two different slags. Slags were used as a replacement for weak soil at ratios of 0%, 25%, 50%, 75% and 100%, where sewage sludge and organic soil were blended with slags separately. The maximum dry unit weights and the optimum water contents for all soil mixtures were determined. In order to investigate the influence of the slags on the strength of sewage sludge and organic soil, and to obtain the optimal mix design; compaction tests, the California bearing ratio (CBR) test, unconfined compressive strength (UCS) test, hydraulic conductivity test (HCT) and pH tests were carried out on slag-soil specimens. Unconfined compressive tests were performed on non-cured samples and those cured at 7 days. The test results obtained from untreated specimens were compared to tests results obtained from soil samples treated with slag. Laboratory tests results indicated that blending slags with organic soil or sewage sludge improved the engineering properties of organic or sewage sludge. Therefore, it is concluded that slag can be potentially used as a stabilizer to improve the properties of organic soils and sewage sludge.
