Production of Cloned Jeju Black Cattle (Korean Cattle) from SCNT Embryo using Vitrification, One-Step Dilution and Direct Transfer Technique (초자화 동결과 1-단계 융해된 체세포 핵이식란의 직접 이식 기술로 제주흑우 복제소 생산)

  • One-step dilution and direct transfer would be a practical technique for the field application of frozen embryo. This study was to examine whether Jeju Black Cattle (JBC, Korean Cattle) can be successfully cloned from vitrified and one-tep diluted somatic cell nuclear transfer (SCNT) blastocyst after direct transfer. For vitrification, JBC-SCNT blastocysts were serially exposed in glycerol (G) and ethylene glycol (EG) mixtures [10%, (v/v) G for 5 min., 10% G plus 20% EG (v/v) for 5 min., and 25% G plus 25% EG (v/v) for 30 sec.] which is diluted in 10% FBS added D-PBS. And then SCNT blastocysts were loaded in 0.25 ml mini straw, placed in cold nitrogen vapor for 3 min. and then plunged into $LN_2$. One-step dilution in straw was done in $25^{\circ}C$ water for 1 min, by placing vertically in the state of plugged-end up and down for 0.5 min, respectively. When in vitro developmental capacity of vitrified SCNT blastocyst was examined at 48 h after one-step dilution, hatched rate (56.4%) was slightly lower than that of control group (62.5%). In field trial, when the vitrified-thawed SCNT blastocysts were transferred into uterus of synchronized 5 recipients, a cloned female JBC was delivered by natural birth on day 299 and healthy at present. In addition, when the short tandem repeat marker analysis of the cloned JBC was evaluated, microsatellite loci of 11 numbers was perfectly matched genotype with donor cell (BK94-14). This study suggested that our developed vitrification and one-step dilution technique can be applied effectively on field trial for cloned animal production, which is even no longer in existence.

EFFECT OF A DESENSITIZER ON MICROTENSILE BOND STRENGTH OF DIFFERENT ADHESIVES (지각과민억제제 적용이 수종 접착제의 미세인장결합강도에 미치는 영향)

  • This study evaluated the influence of a desensitizer(MS coat) on microtensile bond strength of different adhesives:a three-step adhesive(All-Bond 2), a two-step adhesive(Single Bond), a one-step adhesive(One-up Bond F). Non-caries extracted human molars were used. Dentin surface was obtained by horizontal section on mid-portion of crown using a water-cooled low speed diamond saw. Teeth were randomly divided into 6 group. AMO(MS coat + All Bond), SMO(MS coat + Single Bond)- and OMO(MS coat + One-up Bond F)-dentin surface were treated with 17% EDTA before bonded adhesive. AMX-, SMX- and OMX-dentin surface were bonded with All-Bond 2, Single Bond and One-up Bond F, respectively. with no previous treatment with MS coat and 17% EDTA. About 1cm high resin composite($Z-250^{TM}$) were incrementally build-up on the treated surface. The specimens for the microtensile test were serially sectioned perpendicular to the adhesive layer to obtain $0.7{\times}0.7mm$ sticks. 30 sticks were prepared from each group. After that. tensile bond strength for each stick was measured with Microtensile Tester at a 1mm/min crosshead speed. Fractured dentin surfaces were observed under the SEM. The results were statistically analysed by using a One-way ANOVA and Tukey's test(p<0.05). Value in MPa were: $AMO-44.35{\pm}13.21;{\;}SMO-39.35{\pm}13.32;{\;}OMO-31.07{\pm}10.25;{\;}AMX-49.22{\pm}16.38;{\;}SMX-56.02{\pm}13.35;{\;}OMX-72.93{\pm}16.19$. Application of MS coat reduced microtensile bond strengths of both Single Bond and One-up Bond F, whereas microtensile bond strengths of All-Bond 2 were not affected significantly.

One-Step β-Li2SnO3 Coating on High-nickel Layered Oxides via Thermal Phase Segregation for Li-ion Batteries

  • The global energy storage markets have gravitated to high-energy-density and low cost of lithium-ion batteries (LIBs) as the predominant system for energy storage such as electric vehicles (EVs). High-Ni layered oxides are considered promising next-generation cathode materials for LIBs owing to their significant advantages in terms of high energy density. However, the practical application of high-Ni cathodes remains challenging, because of their structural and surface instability. Although extensive studies have been conducted to mitigate these inherent instabilities, a two-step process involving the synthesis of the cathode and a dry/wet coating is essential. This study evaluates a one-step β-Li2SnO3 layer coating on the surface of LiNi0.8Co0.2O2 (NC82) via the thermal segregation of Sn owing to the solubility limit with respect to the synthesis temperature. The doping, segregation, and phase transition of Sn were systematically revealed by structural analyses. Moreover, surface-engineered 5 mol% Sn-coated LiNi0.8Co0.2O2 (NC82_Sn5%) exhibited superior capacity retention compared to bare NC82 owing to the stable surface coating layer. Thus, the developed one-step coating method is suitable for improving the properties of high-Ni layered oxide cathode materials for application in LIBs.

MoS2 Layers Decorated RGO Composite Prepared by a One-Step High-Temperature Solvothermal Method as Anode for Lithium-Ion Batteries

  • A one-step high-temperature solvothermal approach to the synthesis of monolayer or bilayer $MoS_2$ anchored onto reduced graphene oxide (RGO) sheet (denoted as $MoS_2/RGO$) is described. It was found that single-layered or double-layered $MoS_2$ were synthesized directly without an extra exfoliation step and well dispersed on the surface of crumpled RGO sheets with random orientation. The prepared $MoS_2/RGO$ composites delivered a high reversible capacity of $900mAhg^{-1}$ after 200 cycles at a current density of $200mAg^{-1}$ as well as good rate capability as anode active material for lithium ion batteries. This one-step high-temperature hydrothermal strategy provides a simple, cost-effective and eco-friendly way to the fabrication of exfoliated $MoS_2$ layers deposited onto RGO sheets.

Correlation between agility and balance (민첩성과 균형감각 간의 상호관계)

  • Background: While agility is a critical factor in the performance of sport in the court field, due to the diversity of agility occurrences, it is generally difficult to develop by strength and conditioning training. Previous study reported the correlation with the static balance and agility. However, the correlation between dynamic balance and agility is insufficient. It is necessary to study how static and dynamic balance affect agility respectively, for agility development. Design: Cross sectional correlational study design Methods: Twenty young women participated in the study. Three tests were used : one leg stance(static balance),Y-balance(dynamic balance), side-step(agility). One leg stance measured time, Y-balance measured distance, and side-step measured number of times. Correlation between balance and agility was used by Pearson Correlation. Results: One leg stance and side steps were shown to be not correlated. The Y balance and the side step showed with a moderate positive correlation. Conclusion: Agility is a decisive factor in fast-moving sports performance, which is generally difficult to improve with muscle strength and conditioning training. Since the correlation between dynamic balance and agility was found in this study, the importance of dynamic balance was found to improve agility to promote coordination.

  • This study was to examine whether Hanwoo IVM/IVF/IVC blastocyst can be successfully survived in vitro/in vivo after vitrification and one-step dilution. For vitrification, blastocysts were serially exposed in glycerol (G) and ethylene glycol(EG) mixtures[10% (v Iv) G for 5 min., 10% G plus 20% EG (v/v) for 5 min., and 25% G plus 25% EG (v/v) for 30 sec.] which is diluted in 10% FBS added D-PBS. And then they were loaded in the straw, placed in cold nitrogen vapor for 3 min. and plunged into L$N_2$(-196$^{\circ}C$). One-step dilution within the straw was done in $25^{\circ}C$ and 36$^{\circ}C$ water for about 5 min. and 3 min., respectively. Recovered embryos after one-step dilution were cultured in cumulus cell mono-layered drop for 48 h or were transferred into recipient cows. When the embryo survival in vitro was assessed to re-expanded and hatched rates at 24 hand 48 h after one-step dilution, the results of vitrified group (85.4, 43.8%) was high, although these results were significantly lower than normal development (100.0, 63.3%) of control group, respectively (P<0.001, P<0.05). When in vitro survival of vitrified groups according to developmental stage was compared, the results of fast developed embryos (expanded blastocyst and early hatching blastocyst stage) were significantly higher than those of delayed developed one (early blastocyst stage) after one-step dilution (early hatching: 88.0, 48.0%: expanded: 81.1, 45.3%; early: 66.7, 14.3%) (P<0.05). Also, in case of in vitro survival of vitrified groups according to embryo age (day 7, 8 and 9), when embryo age was younger, in vitro survival was significantly higher (day 7: 67.3, 34.5%; day 8: 76.9, 40.7%; day 9: 60.9, 23.9%)(P<0.05). Finally, when in vivo development potential of vitrified and one-step diluted Hanwoo blastocysts was examined, 4 of 8 recipient (50%) cows became confirmed pregnant. These results demonstrated that our vitrification and one-step dilution technique can be applied easily and effectively on field trial without the equipment and embryological skills required for conservative dilution and transfer.

An Interactive Process Capability-Based Approach to Multi-Response Surface Optimization (대화식 절차를 활용한 공정능력지수 기반 다중반응표면 최적화)

  • Purpose: To develop an interactive version of the conventional process capability-based approach, called 'Interactive Process Capability-Based Approach (IPCA)' in multi-response surface optimization to obtain a satisfactory compromise which incorporates a decision maker(DM)'s preference information precisely. Methods: The proposed IPCA consists of 4 steps. Step 1 is to obtain the estimated process capability indices and initialize the parameters. Step 2 is to maximize the overall process capability index. Step 3 is to evaluate the optimization results. If all the responses are satisfactory, the procedure stops with the most preferred compromise solution. Otherwise, it moves to Step 4. Step 4 is to adjust the preference parameters. The adjustment can be made in two modes: relaxation and tightening. The relaxation is to make the importance of one of the satisfactory responses lower, which is implemented by decreasing its weight. The tightening is to make the importance of one of the unsatisfactory responses higher, which is implemented by increasing its weight. Then, the procedure goes back to Step 2. If there is no response to be adjusted, it stops with the unsatisfactory compromise solution. Results: The proposed IPCA was illustrated through a multi-response surface problem, colloidal gas aphrons problem. The illustration shows that it can generate a satisfactory compromise through an interactive procedure which enables the DM to provide his or her preference information conveniently. Conclusion: The proposed IPCA has two major advantages. One is to obtain a satisfactory compromise which is faithful to the DM preference structure. The other is to make the DM's participation in the interactive procedure easier by using the process capability index in judging satisfaction/unsatisfaction. The process capability index is very familiar with quality practitioners as well as indicates the process performance levels numerically.

A COMPARATIVE STUDY OF SHEAR BOND STRENGTH OF FLOWABLE RESIN ASSOCIATED WITH DENTIN ADHESIVE SYSTEMS WITH THERMOCYCLING EFFECT (상아질접착제와 열순환에 따른 유동성 레진의 전단결합강도 비교 연구)

  • Statement of problem : Limited research on flowable resin has been undertaken on its application directly on dentin associated with the adhesive systems. Purpose : This study was to evaluate the shear bond strengh and fracture aspect of flowable resin on human dentin with various types of dentin bonding adhesives with thermo cycling effect. Materials and methods: Filtek-Flow(3M ESPE, USA) was used as flowable resin and Eighty human molars were randomly divided into 4 groups : three dentin bonding adhesives (Scotchbond-Multipurpose : 3-step contentional system, One-Step : One-bottle system. Prompt L-Pop : All-in-one, self-etching primer) and 32% etching treatment without bonding adhesive as a control group. For evaluating their durability of bonding, each group was subdivided : storaging in the water at 37$^{\circ}C$(24 hours) and thermocycling (0$^{\circ}C$-55$^{\circ}C$, 30 seconds intervals, 1000 cycle). Shear bond strength tests were performed and resin-dentin interface and fracture mode were observed. Results were analysed by one-way ANOVA and Scheffe's multiple range test. Results and Conclusion : 1. At 0 cycle, the mean shear bond strength of One-Step exhibited the highest value of all groups(p<0.05), and there were no significant differences between Prompt L-Pop and Scotchbond-Multipurpose, Scotchbond-Multipurpose and control(p>0.05). After 1000 thermocycling, One-Step exhibited higher value than other groups(p<0.05), and there were no significant differences among other groups (p>0.05). 2. The shear bond strength of each group was significantly decreased after thermocycling except Scotchbond-Multipurpose (p>0.05). 3. The most common failure mode was adhesive type and mixed type, next in order.

  • The performance of one-column isothermal PSA process is assessed by dynamic simulator, gPROMS. The four-step and five-step processes are compared. A five-step process is employed in order to show the effect of the addtional cocurrent depressurization step on the four-step PSA process. Two processes parameters, purity and recovery of SO$_{2}$ are used for the performance comparison. The results of dynamic simulation show that four-step process is superior to five-step process in recovery, but not in purity.

Step Length Estimation Algorithm for Firefighter using Linear Calibration (선형 보정을 이용한 구난요원의 보폭 추정 알고리즘)

  • This paper presents a step length estimation algorithm for Pedestrian Dead Reckoning using linear calibrated ZUPT (zero velocity update) with a foot mounted IMU. The IMU consists of 3 axis accelerometer, gyro and magnetometer. Attitude of IMU is estimated using an inertial navigation algorithm. To increase accuracy of step length estimation algorithm, we propose a stance detection algorithm and an enhanced ZUPT. The enhanced ZUPT calculates firefighter's step length considering velocity error caused by sensor bias during one step. This algorithm also works efficiently at various motions, such as crawling, sideways and stair stepping. Through experiments, the step length estimation performance of the proposed algorithm is verified.
