Renoprotective Effect of Maydis Stigma on Puromycin Aminonucleoside-induced Nephrotic Syndrome (Puromycin Aminonucleoside에 의해 유도된 신증후군에 대한 옥미수(玉米鬚)의 보호효과)

  • Objectives: Nephrotic syndrome is a kidney disorder, which is characterized by proteinuria, edema (swelling), and hyperlipidemia. Maydis Stigma (Corn silk) has been widely used in Asia as a traditional medicine and is known to have a diuretic effect and is used for the treatment of edema and indigestion. Methods: The aim of this study is to investigate the improvement effect of Maydis Stigma in treating nephrotic syndrome induced by puromycin aminonucleoside. Sprague-Dawley rats were intravenously injected with 75 mg/kg/day puromycin aminonucleoside, then treated with either Losartan or 200 mg/kg/day Maydis Stigma for seven days. Results: Maydis Stigma significantly decreased ascites and proteinuria level. Plasma levels of blood urea nitrogen (BUN) and plasma creatinine reduced significantly by Maydis Stigma. In addition, treatment with Maydis Stigma attenuated histological damage. Treatment with Maydis Stigma also restored podocin expression and reduced inflammation markers such as intracellular adhesion molecules (ICAM-1), monocyte chemotactic protein-1 (MCP-1), tumor necrosis factor alpha (TNF-α) and high-mobility group box-1 (HMGB1). Conclusions: Maydis Stigma ameliorates kidney injury in nephrotic syndrome rat models. Maydis Stigma exerts a renoprotective effect owing to its anti-inflammatory effects and reductions of ascites and proteinuria. Thus, these results indicate that Maydis Stigma is likely to be a promising agent in the treatment of nephrotic syndrome.

  • Herbal medicines are widely used for centuries to treat illness and improve health in Asia and the use of these remedies has become a worldwide form of alternative therapy. However, there is very limited evidence from randomized controlled trials to support the efficacy of the vast majority of herbal products. Of the commonly used herbal medicines, systemic clinical experiences and promising experiments in animals have found potential evidence of efficacy for six herbal medicines (Unkei-to, Hachimijio-gan, Keishi-bukuryo-gan, Toki-shakuyaku-san, Shakuyaku-kanzo-to, Sairei-to) in the treatment for female reproductive dysfunction. Because herbs may contain potent bioactive substances, the physician should have an opportunity to outline more stringent regulation, similar to over-the counter drug.

Antibacterial Effects of Sasangjasaebang Aqueous Extracts Against Gardnerella Vaginalis ATCC14018, and Combination Effects with Metronidazole (사상자세방(蛇床子洗方)의 Gardnerella vaginalis에 대한 시험관 내 항균력 및 Metronidazole과 병용효과)

  • Purpose: The objective of this study was to observe the antibacterial effects of Sasangjasaebang (SSJSB) aqueous extracts, traditionally used for treating various gynecological diseases including vaginitis in Korea and their individual components - Cnidi Fructus(CF), Zanthoxyli Fructus(ZF) and Alumen(AL) against Gardnerella vaginalis ATCC14018, and combination effects of SSJSB extracts and Metronidazole were also monitored in this study. Methods: Antibacterial activities against Gardnerella vaginalis ATCC14018 of SSJSB (CF : ZF : AL = 10 : 3 : 3) aqueous extracts and their individual components were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were also monitored at MIC and $MIC{\times}2$ levels. The combination effects of SSJSB aqueous extracts and Metronidazole were observed by Checkboard Microtiter Assay and effects on bacterial growth curve treated with SSJSB aqueous extracts MIC + Metronidazole MIC, 1/2MIC and 1/4MIC, respectively. In the present study, Gardnerella vaginalis ATCC14018 were incubated under $37^{\circ}C$, 10% CO2 and bacterial growth curves were calculated at 24, 48, 72, 96 and 120 hrs after incubations. Results: MIC of CF aqueous extracts, ZF aqueous extracts, AL powders and SSJSB aqueous extracts against Gardnerella vaginalis ATCC14018 were detected as $27.5\;{\pm}\;13.693$(12.5~50), $6.875\;{\pm}\;3.423$(3.125~12.5), $4.375\;{\pm}\;1.713$(3.125~6.25) and $2.188\;{\pm}\;0.856(1.563{\sim}3.125)mg/m{\ell}$, respectively. MIC of Metronidazole was detected as $7.6\;{\pm}\;5.367(2{\sim}16){\mu}g/m{\ell}$ at same conditions. In addition, Metronidazole, CF aqueous extracts, ZF aqueous extracts, AL powders, and SSJSB aqueous extracts showed marked dosage-dependent inhibition of bacterial growth, and more dramatical inhibitions were detected in Metronidazole 1/2 MIC + SSJSB aqueous extracts MIC treatment as compared with each of single Metronidazole MIC and SSJSB aqueous extracts MIC treatments, respectively. Especially, quite similar inhibitory effects on bacterial growth were detected in Metronidazole 1/4 MIC + SSJSB aqueous extracts MIC treatment as compared with single Metronidazole MIC treatment in the present study. FIC index were detected as $0.475\;{\pm}\;0.137$(0.375~0.625) at Checkboard Microtiter Assay. Conclusion: The results obtained in this study suggest that CF aqueous extracts, ZF aqueous extracts, AL powders, and SSJSB aqueous extracts showed antibacterial effects against Gardnerella vaginalis ATCC14018, and they also showed dosage-dependent inhibitory effects on the bacterial growth. More potent antibacterial effects were detected in SSJSB aqueous extract as compared with individual components, respectively. In addition, combination treatment of SSJSB aqueous extract and Metronidazole showed more potent inhibitory effects on the growth of Gardnerella vaginalis with FIC index $0.475\;{\pm}\;0.137$(0.375~0.625). According to these results, the combination of SSJSB aqueous extract and Metronidazole is synergistic, and it is expected that effective dosages of Metronidazole could be reduced to 1/4 levels in combination with SSJSB extracts. And it might be needed to make further studies to seek the herbs which have antibacterial effects on the Gardnerella vaginalis ATCC14018.

  • Purpose : This study investigated reliability of Oriental DB & GY(obstetrics & gynecology) Questionnaires's items which was used by Dong-Eui OB & GY through analysis of oriental OB & GY books. Method : This study investigated differentiation of syndrome through analysis of oriental OB & GY book's. Result: This study investigated differentiation of syndrome through analysis of oriental OB & GY disease and pathogenesis. This study's pathogenesis was such that deficiency of Ki, deficiency of blood, stagnation of Ki, stagnated blood, deficiency of Yang, deficiency of Um, cold syndrome, heat syndrome, dampness, dryness, phlegm, kidney, liver, heart, spleen, wind, lung. We except lung from Questionnaires's pathogenesis because it is stuck for importance. We except wind from Questionnaires's pathogenesis because it is stuck for preguence. Oriental OB & GY Questionnaires's pathogenesis consist of 15 items such that deficiency of Ki, deficiency of blood, stagnation of Ki, stagnated blood, deficiency of Yang, deficiency of Um, cold syndrome, heat syndrome, dampness, dryness, phlegm, kidney, liver, heart, spleen. Oriental OB & GY Questionnaires construct pathogenesis's question and guide post through we examined it's reasonableness.

Effects of Angelicae Gigantis Radix on Gene Expression of Ovarian Tissue in Polycystic Ovary Syndrome Rats (당귀(當歸)가 다낭성난소증후군이 유발된 흰쥐 난소조직의 유전자 발현에 미치는 영향)

  • Objectives: This study was performed to investigate the effects of Angelicae gigantis Radix (AGR) which is one of the most useful herbal-drug to treat patients with Polycystic Ovary Syndrome (PCOS) in Oriental medicine on gene expression of ovary tissue. Methods: The effects of AGR on gene expression of ovary tissue resected from PCOS induced rats using single injection of $\ss$-Estradiol 17-valerate (EV) was measured using microarray technique, and the functional analysis on these genes was conducted. Results: Total 2,812 genes were up-regulated or down-regulated, 1,421 genes were up-regulated, 1,391 genes were down-regulated by induction of PCOS. Up-regulated genes were mainly involved in biological function such as cell signalling pathways and inflammatory response. Expression levels of 1,442 genes were restored to those of naive animals by administration of AGR. 558 genes were restored to those of naive animals, which were lowered by induction of PCOS. 884 genes were lowered to naive levels, which were elevated by induction of PCOS. The functions of restored genes were partially involved in the restoration of expression levels, which were changed by induction of PCOS. Especially, up-regulated gene by induction of PCOS were mainly involved in these changes. These results mean restorative effects of AGR on damaged functions by induction of PCOS. The network of total protein interactions was measured using cytoscape program, and some key molecules, such as IRS2, MCM10, ORC2L related in up-regulated genes, CTBP2, CD44, RHOA, related in down-regulated genes that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conclusion: Restored genes by AGR were thought to have common pathways related in regulation of gene expressions. Especially, genes in restored expression levels by AGR, which were up-regulated by induction of PCOS, were regulated by 9 of common transcription factors, genes in restored expression levels by AGR, which were down-regulated by induction of PCOS, were involved in 25 of common transcription factors.

The Effect of Dance Movement Therapy on Anxiety, Depression, and the Quality of Life in Climacteric Women (무용동작(舞踊動作) 치료(治療)가 폐경기 여성(女性)의 부안(不安), 우울(憂鬱) 및 삶의 질(質)에 미치는 효과(效果))

  • Objectives: The aim of this study is to find out the effects of dance movement therapy(DMT) on anxiety, depression, and life quality by applying DMT program to climacteric women. Methods: The subjects of the research are climacteric women between the ages of 45 and 55 who located in the city of oo in Chollabukdo. The program was conducted with the subjects of a total of 27 women less than one year after menopause, 14 in the experiment group and 13 in the comparison group, 90 minutes a session, once a week, for 15 weeks. Results: The results show that the points of anxiety and depression decreased significantly in the experiment group which participated in the DMT program (p<0.05). The points of life increased significantly in physical & mental wellbeing, competence, and stability (p<0.05). Vitality points increased, but not significantly. Conclusions: This means that DMT decreases anxiety and depression of climacteric women, has positive effects on life quality related to health, and can help increase physical, mental, and emotional wellbeing of climacteric women.

Antioxidant Effect of Paeonia Japonica Extracts on Mouse Embryonic Fibroblast Cells (백작약 에탄올 추출물이 mouse embryonic fibroblast cells에 미치는 항산화 효과)

  • Objectives: Paeonia japonica has been widely used for gynecopathy and analgesic effects in Korean Traditional Medicine. The aim of the present study is to determine the antioxidant effect of Paeonia japonica extracts(PJE) by using mouse embryonic fibroblast cells(MEF cells). Methods: We evaluated Radical Scavenging Activity of PJE by the DPPH assay. Protective effect of the PJE on the hydrogen peroxide($H_2O_2$) induced oxidative damage of MEF cells was analyzed by the MTT assay. The Morphological changes of MEF cells induced by P. japonica, $H_2O_2$ and P. japonica+$H_2O_2$ was evaluated by DAPI staining. And effect of PJE on the rate of apoptosis in MEF cells was measured using flow cytometry with Annexin V-FITC and PI double staining. Results: We observed that PJE contain significant DPPH radical scavenging activity. Cell viability of oxidative damaged cells treated with various concentrations of $H_2O_2$ was increased by treatment with PJE. Flow cytometric analysis of the cells treated with $H_2O_2$ in the absence or presence of PJE showed that the crumbled G1 peak was accumulated by the treatment with $H_2O_2$ alone, but restored by addition of PJE. Portion of cells that undergo apoptosis mediated by oxidative stress was decreased by treatment of PJE. The nuclear fragmentation occurred in the oxidative damaged MEF cells was also decreased by PJE treatment. Conclusions: Taken together, our results suggest that PJE exhibits significant antioxidant activity and functions to inhibit cell death mediated by oxidative damage induced apoptotic pathways.

Short-term Clinical Observation of the Early Postpartum Low Back Pain in the Korean Medical Hospital (한방치료를 받은 출산 초기 산모의 요통 경과 고찰)

  • Objectives: The purpose of this study was to observe the early postpartum low back pain and pelvic girdle pain in the Korean medical hospital for 2-weeks. Methods: Fifty-one postpartum women who admitted for postpartum care in Korean hospital from December 3rd, 2011 to January 31st, 2012 had written out pain measurement questionnaires(SF-MPQ, VAS, ODI) three times for 2-weeks. SPSS 18.0 for Windows was used to analyse the data and the independent samples t-test, paired T-test were used to verify the results. Results: 1. Evaluation of low back pain within the first week after birth was as follows: SF-MPQ sensory area was $5.06{\pm}4.41$ points; SF-MPQ emotional areas was $0.86{\pm}1.34$ points; VAS mean was $3.79{\pm}2.02$ points; and ODI mean was $10.27{\pm}6.69$. 2. Early postpartum low back pain and pelvic girdle pain were significantly decreased in SF-MPQ, VAS, and ODI after 2-weeks(p < 0.01). 3. Age, birth type, parity, weight, and BMI increased during pregnancy does not affect the postpartum low back pain and pelvic girdle pain. Conclusions: The top 10% of postpartum low back pain scale was 10.8 points or more in the sensory area of SF-MPQ, and 3 points or more in the affective area of SF-MPQ. In the case of VAS, it was more than 7 points, and ODI was more than 21.8 points. Postpartum back pain with oriental medical treatment was significantly improved.

The Study on Effectiveness of Oriental Medicine Treatment for Lumbar Disc Herniation Inpatients on 208 Cases (요추 추간판 탈출증 입원환자 208례를 통한 한방치료의 효과 연구)

  • This study was introduced the short-term effectiveness of Oriental medicine treatment for lumbar disc herniation inpatients. We selected 208 inpatients with a magnetic resonance imaging(MRI)-confirmed lumbar disc herniation since June 2012 until August 2012. Their demographics characterisitics, numeric rating scale(NRS) and Oswestry back-related disability index(ODI) for low or leg pain, lumbar flexion and extension angle were assessed at baseline and when discharged from the hospital. When it measured the straight leg raisingscale(SLR), the low side of the angle was examined. Treatment every patient has received as follow; herbal medicine, bee venom, acupuncture and physical examination, spinal manipulation. NRS of low back pain and leg pain and ODI is $5.6{\pm}2.3$, $4.9{\pm}2.8$ and $45.5{\pm}20.0$, respectively at baseline and $2.8{\pm}1.8$, $2.7{\pm}1.9$ and $27.2{\pm}14.1$, respectively when discharged from the hospital. range of motion(ROM) of lumbar flexion and extension angle and SLR is $70.0{\pm}27.1$, $15.4{\pm}7.2$ and $61.3{\pm}23.0$ respectively at baseline and $80.5{\pm}16.9$, $18.25{\pm}4.1$ and $73.2{\pm}14.0$, respectively when discharged from the hospital. It shows that statistically significant improved(P<0.001). Furthermore, regarding patient satisfaction with the treatment, excellent(33.7%), good(55.4%), normal(10.4%), poor(0.5%), it suggests that most patients(89%) satisfied with the treatment.

  • Byun Jeung Ok Ham, written by Jin Sa Tak is composed of four books. This book is estimated of being written after A.D 1688 and composed of internal medicine, obstetrics and gynecology, ophthalmology, dental surgery and so on, total 36 parts of clinical medicine. This is the last book of that four studying about definition on true and false of 15 diseases. 15 subjects follow, tumo, heat, faint, hematemesis and nosebleeding, insanity, great vomiting, great diarrhea, great thirst, scrotal hernea, uterus attacked by heat, dysentery, congestion, great swelling, malaria, attack by cold. This treatise is the last book of four which deals with jin ga translating into korean and studying about medical theories. In every parts, author's unique clinical theory appears affluently and in that periods his study developed a lot in those days' medical methods. The first book deals with eum yang. second book is weakness and strongness, third upper and low, fourth truth and untruth. First book, Jin Sa Tak says studying on eum yang is oriental medical basic theory and on incurable diseases or chronic symptoms, doctor must go back to that eumyang demonstration after can cure patients. Second parts are on weakness and strongness demonstration, that weakness is weakness of patients' energy and strongness is prosperous condition of diseases' attack. Third parts are on upper and low, that upper is upper parts of human body of painful parts and low is human low parts of pain, that is parts of under waist. And this treaties fourth parts are truth and untruth, true symptoms and untrue symptoms, that is the real reaction of human condition and the other way. Every prescriptions are author's creations. This book provides new viewpoints which surpasses original ancient medical theories. Author suggests new opinions about chronic and incurable diseases.
