Nuclear Localization Signals in Prototype Foamy Viral Integrase for Successive Infection and Replication in Dividing Cells

  • We identified four basic amino acid residues as nuclear localization signals (NLS) in the C-terminal domain of the prototype foamy viral (PFV) integrase (IN) protein that were essential for viral replication. We constructed seven point mutants in the C-terminal domain by changing the lysine and arginine at residues 305, 308, 313, 315, 318, 324, and 329 to threonine or proline, respectively, to identify residues conferring NLS activity. Our results showed that mutation of these residues had no effect on expression assembly, release of viral particles, or in vitro recombinant IN enzymatic activity. However, mutations at residues 305 (R ${\rightarrow}$ T), 313(R ${\rightarrow}$ T), 315(R ${\rightarrow}$ P), and 329(R ${\rightarrow}$ T) lead to the production of defective viral particles with loss of infectivity, whereas non-defective mutations at residues 308(R ${\rightarrow}$ T), 318(K ${\rightarrow}$ T), and 324(K ${\rightarrow}$ T) did not show any adverse effects on subsequent production or release of viral particles. Sub-cellular fractionation and immunostaining for viral protein PFV-IN and PFV-Gag localization revealed predominant cytoplasmic localization of PFV-IN in defective mutants, whereas cytoplasmic and nuclear localization of PFV-IN was observed in wild type and non-defective mutants. However sub-cellular localization of PFV-Gag resulted in predominant nuclear localization and less presence in the cytoplasm of the wild type and non-defective mutants. But defective mutants showed only nuclear localization of Gag. Therefore, we postulate that four basic arginine residues at 305, 313, 315 and 329 confer the karyoplilic properties of PFV-IN and are essential for successful viral integration and replication.

고나트륨과 저나트륨 식이시 성인 여성의 혈중 아미노산 농도변화에 관한 연구 (Effects of High and Low Sodium Diet on Plasma Amino Acid Levels in Korean Adult Women)

  • This study was performed to determine the effect of dietary Na levels on plasma amino acid levels. Plasma amino acid levels were compared in 20 adult healthy women subjects who were given high Na diet (290.48 mEq/day : NaCl 17g) or low Na diet (51.26 mEq/day : NaCl 3g) for subsequent 6 days. Plasma essential amino acids levels were significantly decreased (36%) while plasma non-essential amino acids levels were significantly increased (22%) when subjects were given low Na diet (p<0.001). Among essential amino acids, threonine was decreased (74%) significantly when subjects were given low Na diet (p<0.01). Among nonessential amino acids, serine (49%), proline (20%) and aspartic acid (14%) were increased (p<0.01), while arginine (48%) and glutamic acid (27%) were decreased (p<0.001). In conclusion, dietary Na contents seemed to be an important factor to affect plasma amino acid levels. It would be appropriate to decrease the dietary Na intakes level considering the various clinical effects of dietary Na on the body fluid. For the patients who need low Na diet, it would be suggested that the level of dietary proteins should be carefully considered along with dietary Na manipulation. 혼합영양 배양에서 Chlorella protothecoides의 GABA를 포함한 아미노산 함량 분석 (The Content Analysis of Amino Acids Including GABA of Chlorella protothecoides under Mixtrophic Culture)

  • Chlorella is quantitatively and qualitatively high in protein with balanced essential amino acid profiles, vitamins and minerals. ${\gamma}-Aminobutyric$ acid (GABA) is broadly distributed in nature and fulfills multi-physiological functions including effect such as a health-promoting functional compound. To improve the GABA production, Chlorella protothecoides were grown through the modified mixtrophic culture medium containing 2L of sterilized bristol medium with 0.01% urea and 4.0% glucose in a 5L fermenter. The results showed that nineteen kinds of amino acid including GABA at C. protothecoides sample were analyzed using high performance liquid chromatography (HPLC). Glutamic acid in total concentration (%) of amino acid is the most abundant amino acid (33.10%), followed by alanine (20.48%) and GABA (17.48%). Three amino acids including GABA were responsible for more than 70% total concentration in C. protothecoides sample including eight essential and nine non-essential amino acids: aspartic acid, asparagine, serine, glutamine, histidine, glycine, threonine, arginine, tyrosine, valine, methionine, tryptophan, phenylalanine, isoleucine, leucine, lysine. As a result of this experiment, it is expected that Chlorella will be developed to a critical product having high value as, GABA, functional food materials.

  • This study was conducted to evaluate the effects of SaSang constitutional diet for essential hypertension on blood pressure, fat, and serum lipid on the subjects with aerobic exercise and low salt diet at the same time. Method: A non-equivalent control group time-series design was used. For the experimental group, aerobic exercise and SaSang constitutional diet were taught by researcher at health center. Test for hypothesis was done by repeated measured ANOVA. Result: There was significant decrease in systolic, diastolic, and MAP between the experimental group and control group over three different times. There was significant decrease in body weight, BMI, and body composition over three different times. But, there was no significant difference between groups and interaction by groups or over time. There was significant decrease in T/G and cholesterol over three different times. But, there was no significant difference in T/G, HDL, LDL, and total cholesterol between groups and interaction by groups or over time. Conclusion: Findings indicate that this study will contribute to develop nursing strategies for the regulation of the blood pressure as a means of alternative- complementary nursing intervention.

사자발쑥 정유를 첨가한 기능성 필름에 의한 토마토의 품질 특성 (Quality Characteristics of Tomato Packaged with Functional Film Applied to Essential Oil of Artemisia Princeps Pampanini)

  • This research was conducted to investigate the effect of essential oil (EO) from Artemisia Princeps Pampanini on quality of tomato during storage. EO was extracted by steam distillation and diluted by 1 and 2%. Total phenolic contents and antioxidative activities of EO were analyzed. Tomato was packaged corrugated box covered with polypropylene (PP) film containing EO (1 and 2%) and then stored at room temperature for 10 days. During storage, appearance, weight loss rate, soluble solid contents, pH and a (redness) value were measured for tomato. Total phenolic contents of EO increased in a dose-dependence. The DPPH and ABTS radical scavenging activities also showed a concentration-dependent increase. The weight loss rate was lowest using the film packaged in 1% EO. During the storage period, soluble solid contents and pH increased in all treatments. The a value increased regardless of EO concentration in films. Especially, tomato packaged with EO film showed a lower a value than non EO-packaged ones. This result suggests that application of EO to film is effective for tomato quality preservation.

  • 현대 사회는 사물인터넷, 빅데이타 등으로 급속하게 발전하고 있으며 코로나 시대에도 발전하는 산업이 있다. 그러나, 교회는 코로나 시대에 발전하지 못하고 충분히 대응하지 못하고 있으며 교회의 존립 자체를 흔들고 있다. 교회의 본질은 예배, 전도, 교육, 봉사, 교제로 구성된다. 비대면의 안전을 중요시하는 코로나 시대에는 본질은 변화하지 않지만 사회적 책임감이 있게 교회의 본질을 구현하는 노력이 필요하다. 본 논문은 이러한 측면에서 교회의 5대 본질 요소를 중심으로 교회의 사회적 책임을 논한다. 이를 위해 교회의 본질적 요소를 살펴보고, 코로나 시대에 적합한 구현 방법을 연구한다.

Effect of Turmeric Extracts on Nutritional and Antioxidant Properties of Germinated Korean Brown Rice

  • Brown rice is more nutritious than the commonly consumed white rice, however, the former is not widely accepted because of its coarse texture and difficulty in cooking. Germination is an easy technique to minimize the drawback of normal brown rice. The objective of this study was to investigate the effect of three different concentrations (1, 3, and 5% w/v) of turmeric extracts on the nutrient and antioxidant activities of germinated brown rice (GBR). The b (yellowness) values of the turmeric-treated GBR were significantly higher than that of the untreated control. Lower concentration (1%) slightly increased (3395.5 mg/kg) but higher concentrations (3 and 5%) reduced (1735.8 - 2393.7 mg/kg) the total mineral content in GBR, as compared to the control (3377.4 mg/kg). The amount of essential, non-essential, and total amino acids, including GABA, were increased with the concentration of turmeric extracts. The amount of essential amino acids was increased by 58.3, 71.5, and 88.3% with the application of 1, 3, and 5% extracts, respectively. The antioxidant potential of GBR was also enhanced with turmeric treatment. Overall results indicated that 1 or 3% turmeric treatment could be appropriate to enhance the nutritional and functional value of GBR.

Accumulation and distribution of nutrients, radionuclides and metals by roots, stems and leaves of plants

  • In the process of growth and development, plants not only absorb essential nutritional elements, but also absorb radioactive and non-essential elements from the environment, and their distribution varies in different parts of the plant. In this study, neutron activation analysis and gamma spectrometry were performed on stems, roots, and leaves of vegetables. The results indicate that the accumulation of radionuclides and multi-elements depends on the plant type and plant parts. Activity concentrations of 226Ra and 232Th in plants were accumulated in the following order: Roots > Stems > Leaves. The highest concentrations of 40K and 210Pb were observed in the stems and leaves of plants, respectively. Essential nutrient requirements of plants are in the following order: K > Ca > Mg > Fe > Zn > Mn. Among the nonessential metals, the concentration of Na in the vegetable sample was much greater than those of the other elements. The K/Na ratio in the plant depends on the type of plant and the translocation within the plant.

The test object can be material, component, plant, earth, environment etc. Total qualify management, total productivity management, concurrent engineering and many other essential ingredients of success in plant engineering and manufacturing industry are dependent on NDE far success and good returns on investments.

효모의 급여가 육계의 성장 및 장내 대장균의 변화에 미치는 영향 (Effect of Feeding Yeast (Saceharomyces eerevisiae) on Growth Performance and Changes of Intestinal E. coli in Broiler Chicks)

  • In order to investigate the effect of feeding live yeast (Sacckaromyces cerevisiae) on the growth performance and changes of intestinal microorganism (E. coli), a growth assay was conducted with 144 broiler chicks. Treatments were consisted of corn-soybean meal control, 0.05% live yeast, and 0.05% dead yeast. Most of the chick protein of the live yeast was in the pure protein form, and had a high amino acid composition with 47% of essential amino acids and 53% of non-essential amino acids. No differences in growth performance were shown among dietany treatments. Total number of E. coli in the small intestine of chicks fed either live or dead yeast was significantly reduced compared to chicks fed the control diet. Although the changes of E. coli in the cecum were not identical to differences in the small intestine, the changes of E. coli in the cecurn had a similar trend.
