Peroxynitrite Scavenging Activity of Sabohwanin Lipopolysaccharide-Induced Oxidatively-Stressed Mice (Lipopolysaccharide로 산화 스트레스를 유도한 Mouse에서 사보환(四補丸)의 Peroxynitrite 억제 효과)

  • Objectives : Peroxynitrite (ONOO-), superoxide anion radical (?O2-) and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging process, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate Sabohwan's activity for scavenging ONOO- and its precursors. NO and ?02-. Methods : For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4.5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Results : Sabohwanblocked tert-butylhydroperoxide (t-BHP)-induced cell death in a dose-dependent fashion. It scavenged t-BHP-induced ONOO-, NO and ?O2- in YPEN cells. Sabohwan inhibited the generation of ONOO-, NO and ?O2- in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondria both in vitro and in vivo. The lipid peroxide level increased and glutathione level decreased in the LPS-treated mice, whereas the ones in the Sabohwanadministered group among the LPS-treated mice reversed toward their natural levels. Conclusions : These results suggest that Sabohwanis an effective ONOO-, ?O2- and NO scavenger, and thereby it might have a potential role as a therapy against the aging process and age-related diseases.

Antioxidant and inflammation inhibitory effects from fruiting body extracts of Ganoderma applanatum

  • In this study, the antioxidant and anti-inflammatory effects of methanol extract (ME) and hot water extracts (HE) from the fruiting bodies of Ganoderma applanatum were investigated. The 1,1-diphenyl-2-picryl-hydrazy (DPPH) radical scavenging activity of 2.0 mg/mL ME (94.83%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME and HE were similar to that of BHT at 2.0 mg/mL, whereas lipid peroxidation activity of the ME and HE were significantly lower than that of BHT. High-performance liquid chromatography analysis showed that the G. applanatum fruiting bodies contained nine phenolic compounds, which might contribute to antioxidant and anti-inflammatory activities. The survival rate of RAW 264.7 macrophages treated with 2.0 mg/mL ME and HE were 65.23 to 68.12% at 2.0 mg/mL, thereby indicating that the extracts were slightly cytotoxic at the concentration tested. The extracts also inhibited the nitric oxide (NO)-mediated expression of inducible nitric oxide synthase (iNOS) protein in lipopolysaccharide-induced RAW 264.7 macrophages and carrageenan-induced paw edema in rats. The study results demonstrated that the fruiting bodies of G. applanatum possessed good antioxidant and anti-inflammatory activities, which might be used to develop novel anti-inflammatory agents.

Anti-Oxidant Activity and Anti-Inflammatory Effects of Spiraea fritschiana Schneid Extract (참조팝나무 추출물의 항산화 활성 및 항염증 효과)

  • Background : We studied the anti-oxidant activity and anti-inflammatory effects of Spiraea fritschiana Schneid extract (SFSE). Methods and Results : The SFSE was prepared using methanol and was evaluated for its total phenol and flavonoid content, DPPH (1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity, reducing power, and effect on nitric oxide (NO) production, and cell viability by using real-time polymerase chain reaction (PCR). The total phenol content was $212.78{\mu}g{\cdot}galli$c acid equivalent (GAE)/mg and the total flavonoid content was $66.84{\mu}g{\cdot}quercetin$ equivalent (QE)/mg. The extract showed antioxidant activity (DPPH free-radical scavenging activity) with $RC_{50}$ value of $76.61{\mu}g/m{\ell}$. The reducing power of the extract was Abs 0.58 at $250{\mu}g/m{\ell}$. Cell viability was determined using the MTT 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. To evaluate anti-inflammatory activity, we examined the inhibitory effects on lipopolysaccharide-(LPS)-induced NO production in RAW 264.7 cells. The NO inhibition rate was 90% at $200{\mu}g/m{\ell}$ SFSE. At the same concentration, the expression of pro-inflammatory genes such as inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 also decreased. Conclusions : Our results suggest that SFSE is a novel resource for the development of foods and drugs that possess anti-oxidant and anti-inflammatory activity.

Comparison of Activity of Angelica Gigas and Angelica Acutiloba from Kangwon (강원도 재배 참당귀 및 일당귀의 활성에 대한 비교연구)

  • In genus Angelica, three species have been used and cultivated for medical material in orient, A. gigas in korea, acutiloba in Japan and sinensis in China. The plant material of Angelica spp. is used for the treatment of women's disease as a hematic. The extracts from A. gigas and acutiloba were fractionated aqueous partitions. And study was performed to examine DPPH scavenging activities, BSA degradadion, anti-apoptosis and NO scavenging. DPPH radical scavenger activity was measured by DPPH method, it was shown dose-dependently effect. and BSA degradadion was shown same result. Treatment of cells with hydrogen peroxide, a reactive oxygen species was to indiced cell death and pretreatment with Angelica gigas and angelica acutiloba extract attenuated the occurrence of $H_2O_2$-induced cell death. In vitro nitric oxide (NO) scavenging effect on Angelica gigas and angelica acutiloba extracts. All extracts effectively reduced the generation of NO radicals in a dose-dependant manner.

Characterization of anti-oxidative effects of Mori Cortex Radicis

  • We tested to determine if Mori Cortex Radicis extract has antioxidant activities and its potential mechanism of action was explored. Anti-oxidative effects were tested by measuring free radical and nitric Oxide (NO) scavenging activity, and reducing power. Since iNOS and COX-2 are important enzymes responsible for the production of free radicals in the cell, Mori Cortex Radicis extract was tested as to whether it could inhibit iNOS and COX-2 expression in LPS stimulated Raw cells. 70% methanolic extract of Mori Cortex Radicis exerted significant DPPH free radical and NO scavenging activities. In addition, the Mori Cortex Radicis extract exerted dramatic reducing power with maximal activity observed at 1 mg/ml (11-fold over control). Production of iNOS induced by LPS was significantly inhibited by the Mori Cortex Radicis extract, suggesting it could inhibit NO production by suppressing iNOS expression. COX-2 induced by LPS was also significantly inhibited by the Mori Cortex Radicis extract. The extract contains well known antioxidant components including phenolics, flavonoids and anthocyanin at the concentration of 0.23 mg/g, 42.97 mg/g and 12.08 mg/g, respectively. These results suggest that 70% methanolic extract of Mori Cortex Radicis exerts significant anti-oxidant activity via inhibiting iNOS and COX-2 induction.

Phenolic Compounds from Barks of Actinidia arguta Planchon Growing in Korea and its Anti-Oxidative and Nitric Oxide Production Inhibitory Activities (국내산 다래나무 수피의 페놀성 화합물의 항산화 및 Nitric Oxide 생성 억제 활성)

  • Phytochemical examination of the barks of Actinidia arguta led to the isolation of five flavonoids. Structures of compounds were elucidated as catechin (1), (-)-epicatechin (2), quercetin (3), $quercetin-3-O-{\beta}-D-glucopyranoside$ (4), $quercetin-3-O-{\beta}-D-galactopyranoside$ (5) by comparison with previously reported spectral evidences. To investigate the anti-oxidative effect and nitric oxide (NO) production inhibitory activity of these compounds, DPPH radical scavenging activity and nitric oxide production inhibitory activity in $IFN-{\gamma}$, LPS stimulated RAW 264.7 cell were examined. The $IC_{50}s$ were determinied as follows : $1\;$IC_{50}=26.61\;{\mu}g/ml$, $2\;IC_{50}=25.30\;{\mu}g/ml$, $3\;IC_{50}=20.41\;{\mu}g/ml$, $4\;IC_{50}=18.23\;{\mu}g/ml$ , $5\;IC_{50}=30.46\;{\mu}g/ml$, $6\;IC_{50}=28.0;{\mu}g/ml$, $7\;IC_{50}=27.24\;{\mu}/ml$. These NO production inhibitory effects were significantly different compared with the positive control, L-NMMA $(IC_{50}=20.77\;{\mu}g/ml)$, respectively. Compound $1\;(IC_{50}=6.19\;{\mu}g/ml)$, $2\;(IC_{50}=8.98\;{\mu}g/ml)$, $3\;(IC_{50}=7.30\;{\mu}g/ml)$ and $4\;(IC_{50}=7.64\;{\mu}g/ml)$ also showed potent antioxidative activities similar level to ascorbic acid $(IC_{50}=9.22\;{\mu}g/ml)$. These results suggest that barks of A. arguta have a potent anti-oxidative and anti-inflammatory activity.

The Anti-inflammatory Effect and DPPH Free Radical Scavenging Capability of Rhizoma drynariae Aqueous Extract (골쇄보 열수추출물 약침액의 항염증반응 및 DPPH 라디칼 소거능 활성에 대한 연구)

  • Objectives : This study is to investigate the effects of Rhizoma drynariae aqueous extract(RDA) on cell cytotoxicity, Nitric Oxide (NO) and Prostaglandin $E_2(PGE_2)$ production and 1,1-diphenyl-2-picryl ghdrazyl(DPPH) free radical scavenging capability. Methods : Cell cytotoxicity was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay. The production of NO was measured by Griess assay. The production of $PGE_2$ was measured by immunoassay. And, the anti-oxidant activity was measured by the DPPH method. Results : Cell cytotoxicity in 50, 100, 200 and $400{\mu}g/ml$ RDA did not increase significantly compared to the RDA untreated group. RDA($200{\mu}g/ml$ and $400{\mu}g/ml$) inhibited NO and $PGE_2$ production in lipopolysaccharide-stimulated RAW 264.7 cells. RDA had high DPPH free radical scavenging capability. Conclusions : This study indicates that RDA inhibits NO and $PGE_2$ production in lipopolysaccharide-stimulated RAW 264.7 cells and improve DPPH free radical scavenging capability. RDA may have an anti-inflammation effect and an anti-oxidant activity.

Biological Activities of the Water Extract and its Fractions from Taraxacum coreanum Nakai (흰민들레 열수 추출물 및 분획물의 피부 관련 생리활성)

  • In this study, we investigated on antioxidative activity, tyrosinase inhibitory activity and nitric oxide(NO) production inhibitory activity in the hot water extract of Taraxacum coreanum and its fractions. The total polyphenol and flavonoid contents of the extract were found to be 65.42 mg/g and 9.83 mg/g, respectively. And the total polyphenol and flavonoid contents of the ethyl acetate fraction were found to be 168.23 mg/g and 31.92 mg/g, respectively. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate and butanol fraction were exhibited 64.65 ${\mu}g/ml$ and 277.42 ${\mu}g/ml$, respectively. Moreover, ethyl acetate fraction showed higher inhibitory activity than other samples in tyrosinase inhibitory activity. In NO production inhibitory activity, the extract and its fractions showed NO production inhibitory effect. Especially, the ethyl acetate, chloroform and butanol fraction was exhibited higher NO production inhibitory activity than other samples. As a result, the ethyl acetate and butanol fraction from the water extract of T. coreanum could be applicable to functional materials for skin-related fields.

Antioxidant activity and NO production of the Alisma orientale Juzep fermented by Paenibacillus kribbensis AM49 (Paenibacillus kribbensis AM49로 발효시킨 택사의 항산화 활성 및 NO 생성 효과)

  • Objectives : The purpose of this study was to investigate the antioxidant activities and nitric oxide (NO) production in RAW 264.7 macrophages in extract of Alisma orientale Juzep (EAOJ) and fermented extract (FAOJ) by Paenibacillus kribbensis AM49 (P. kribbensis AM49). Methods : The Alisma orientale Juzep was fermented with P. kribbensis AM49 at 37℃ for 72 hours. We measured total polyphenol and total flavonoid, DPPH radical scavenging activity, FRAP activity and reducing power by spectrometric assay in EAOJ and FAOJ at concentrations at 0.5, 1, 5, 10 mg/㎖. Positive control was used ascorbic acid. Furthermore, we examined effect of EAOJ and FAOJ on the cell viability and NO production in RAW 264.7 macrophages. Results : The total polyphenol and total flavonoids content of FAOJ were increased 9.16 mg/g, 2.59 mg/g to 12.58 mg/g, 3.45 mg/g. DPPH radical scavenging activity, FRAP activity and reducing power were dose dependently increased according to the treatment concentration (0.5, 1, 5, 10 mg/㎖) of EAOJ and FAOJ. In particular, DPPH radical scavenging activity, FRAP activity of FAOJ was significantly increased at 5, 10 mg/㎖. Reducing power of FAOJ at 10 mg/㎖ was similar to ascorbic acid at 0.1 mg/㎖. In addition, the cell viability and NO production in RAW 264.7 macrophages were significantly increased at the concentrations of 250, 500, 1000 ㎍/㎖. Conclusions : These results suggest that FAOJ by P. kribbensis AM49 has effects to antioxidant activity. In addition, the cell viability and NO production in RAW 264.7 macrophages were significantly increased.

Evaluation of Oxidation Inhibition and Nitrogen Oxide Scavenging Activity from Curcuma longa L. Extracts (울금(Curcuma longa L.) 추출물의 산화억제 및 질소산화물 소거활성)

  • The aim of the present investigation was to assess the oxidation inhibition by nitrogen oxide scavenging activity and physiological activities. Bioactive compound of proanthocyanidin $69.000{\pm}2.737mg$ catechin equivalents (CE)/g dry weight. Antioxidant effects (nitric oxide radical scavenging activity, nitrite scavenging activity, ${\beta}$-carotene bleaching assay and lipid peroxidation inhibition activity) of distilled water (DW), 70% ethanol and n-butanol extract of turmeric (Curcuma longa L.). Turmeric extracts yield were DW 17.11%, 70% ethanol 15.26% and n-butanol 4.12%, respectively. Oxidation inhibition activity of the samples exhibited a dose-dependent increase. However, in the current study, none of the samples evaluated showed activity as strong as the BHA and trolox. Total flavonoid content was the highest in the n-butanol extract, followed by 70% ethanol and DW extract. Further, nitrite scavenging activity was the highest for the n-butanol extract. As a result of this experiment, the turmeric can be utilized as a valuable and potential natural oxidation inhibition for the functional food industry.
