Pseudomonas sp. LG2의 Ferulic acid esterase 및 Xylanase 유도와 부분적 특성 (Partial Characterization and Induction of Ferulic Acid Esterase and Xylanase from Pseudomonas sp. LG2)

  • 리그닌 분해 세균인 Pseudomonas sp. LG2는 lignocellulose 기질을 분해하여 APPL 화합물을 생성하는 균주이다. 이 균주를 BSG(brewer's spent grain)가 함유된 배지에서 배양한 배양액에서 APPL 화합물을 확인하였다. 세포외 조효소들의 유도에 관한 여러 가지 탄소원의 영향을 조사한 결과 glucose 배지에서는 xylanase의 효소활성만 확인 되었고 xylose, arabinose에서 배양한 조효소에서는 FAE 및 xylanase의 효소활성이 없었다. Oat spelt xylan, HBSG I(hydrolyzed brewer's spent grain I), HBSG II(hydrolyzed brewer's spent grain II) 및 AFBSG(autoclaved fraction from brewer's spent grain)를 탄소원으로 배양한 조효소에서는 FAE 및 xylanase의 효소활성이 확인됐다. Pseudomonas sp. LG2를 oat spelt xylan, HBSG I, HBSG II 및 AFBSG를 탄소원으로 사용하여 14일 동안 배양하면서 배양기간에 따른 세포외 효소들의 FAE와 xylanase 활성을 조사하였다. Xylanase의 최고 활성은 xylan을 탄소원으로 6일간 배양 했을때 5.3 U/mg으로 가장 높았으며, FAE의 최고 활성은 AFBSG를 탄소원으로 배양했을 때 배양 8일째 15.4 mU/mg으로 가장 높았다. Oat spelt xylan, HBSG I, HBSG II 및 AFBSG를 탄소원으로 사용하여 배양한 배지에 분리된 ferulic acid가 확인되었다. 세포외 효소의 FAE 활성은 methyl ferulic acid, methyl caffeic acid, methyl p-coumaric acid에 대해 esterase의 활성을 보였으나, methyl sinapinic acid, methyl vanillic acid 및 methyl gallic acid에 대해서는 esterase의 활성이 없었다.

자외선 LED를 이용한 자유 시안의 광촉매 산화 (Photocatalytic Oxidation of Free Cyanide Using UV LED)

  • 본 연구는 기존의 광촉매 산화 기술에서 주로 사용된 자외선램프의 단점을 보완할 수 있는 대체 광원인 자외선 LED를 사용하여 자유 시안을 폐수로부터 제거하는 공정을 평가하고자 수행되었다. 특히 광촉매 산화 공정의 다양한 영향 인자들에 대해 살펴보았다. 연구 결과, 자외선 LED는 기존의 광원인 자외선램프를 대체할 수 있는 적용성을 확인할 수 있었다. 뿐만 아니라 LED 개수를 증가할수록 광촉매 산화 반응의 효율은 증가하였으나, 공정의 경제성과 효율성을 동시에 만족시키기 위해서는 최적의 LED의 개수를 선정할 필요가 있다는 것을 확인하였다. 광촉매로 이용된 아나타제(anatase), 루틸(rutile), Degussa P25 등 세 종류의 $TiO_2$ 중 Degussa P25가 가장 높은 성능을 보였으며, 아나타제(anatase)와 루틸(rutile)을 특별한 전처리 과정없이 단순하게 혼합하였을 때는 Degussa P25 만큼의 공정의 효율은 얻지 못했다. 또한 $TiO_2$의 입자 크기가 작을수록 광촉매 산화 반응이 더욱 활발하게 이루어졌다. 그리고 광촉매 산화 반응에 있어 주로 전자 수용체 역할을 수행하는 산소를 주입함으로써 공정의 효율이 증진되는 효과를 얻을 수 있었다.

지하수 중 탄소원으로 fumarate 주입과 유전자분석을 통한 질산성질소 자연저감도 평가 (Evaluation of Natural Attenuation by Addition of Fumarate as Carbon Source and Gene Analysis in Groundwater Sample)

  • In the results of monitoring nitrate concentration in more than 8,000 groundwater wells around agro-livestock, the average and maximum nitrate concentration was 9.4 mg/L and 101.2 mg/L, respectively. Since about 31% of the monitoring wells was exceed the quality standard for drinking water, nitrate control such as remediation or source regulation is required to conserve safe-groundwater in South Korea. Typical nitrate-treatment technologies include ion exchange, reverse osmosis, and biological denitrification. Among the treatment methods, biological denitrification by indigenous microorganism has environmental and economic advantages for the complete elimination of nitrate because of lower operating costs compared to other methods. Major mechanism of the process is microbial reduction of nitrate to nitrite and nitrogen gas. Three functional genes (nosZ, nirK, nirS) that encode for the enzyme involved in the pathway. In this work, we tried to develop simple process to determine possibility of natural denitrification reaction by monitoring the functional gene. For the work, the functional genes in nitrate-contaminated groundwater were monitored by using PCR with specific target primers. In the result, functional genes (nosZ and nirK) encoding denitrification enzymes were detected in the groundwater samples. This method can help to determine the possibility of natural-nitrate degradation in target groundwater wells without multiplex experimental process. In addition, for field-remediation application we selected nitrate-contaminated site where 200~600 mg/L of nitrate is continuously detected. To determine the possibility of nitrate-degradation by stimulated-natural attenuation, groundwater was sampled in two different wells of the site and nitrate concentration of the samples was 300 mg/L and 616 mg/L, respectively. Fumarate for different C/N ratio was added into microcosm bottles containing the groundwater to examine denitrification rate depending on carbon concentration. In the result, once 1.5 times more than amount of fumarate stoichiometry required was added, the 616 mg/L of nitrate and 300 mg/L of nitrate were completely degraded in 8 days and 30 days. The nitrite, byproduct of denitrification process, was also completely degraded during the experimental period.

액화천연가스(LNG)를 사용한 수소 생산 및 액화 공정 개발 (Design and Analysis of Hydrogen Production and Liquefaction Process by Using Liquefied Natural Gas)

  • 액체 상태의 수소는 기체 상태의 수소에 비해 수송이 용이하고 에너지 밀도가 높으며 폭발 위험성이 낮다. 하지만 수소 액화 공정은 냉각 사이클에 많은 양의 에너지가 소모된다. 반면에 액화천연가스(LNG; Liquefied Natural Gas)는 재기화 과정에서 다량의 냉열이 버려진다. 따라서 LNG 냉열을 회수하여 수소 냉각에 활용한다면 공정 효율을 높일 수 있다. 또한, 천연가스 개질을 통한 수소 생산은 가장 경제성 있는 방법으로 평가받고 있으며, 이러한 측면에서 LNG를 수소 생산의 원료로 사용할 수 있다. 본 연구에서는 LNG를 원료 및 냉열원으로 사용하여 수소를 생산 및 액화시키는 공정을 개발하고 열역학적 관점에서 공정을 평가하였다. 공정 개발을 위해 기존의 탄화 수소 혼합 냉매와 헬륨-네온 냉매를 이용한 수소 액화 공정을 비교 공정으로 선정하였다. 이후 LNG를 원료 및 수소 예냉의 냉열원으로 사용하는 새로운 공정을 설계하여 에너지 소모량 및 엑서지 효율 측면에서 기존 공정과 비교, 분석하였다. 제안된 공정은 기존 공정 대비 약 17.9%의 에너지 절감 및 11.2%의 엑서지 효율이 향상된 결과를 나타내었다.

Insecticidal Activity of Bacillus thuringiensis 656-3 Strain to Mushroom Flies in Oyster Mushroom House

  • Bacillus thuringiensis 656-3 which was isolated from a soil sample of mushroom house and showed high toxicity to mushroom flies, Lycoliella mali and Coboldia fuscipes, was surveyed for insecticidal effect in the oyster mushroom house.3. thutingiensis 656-3 was mass-cultured in the fermenter containing soybean cake(2%) and wheat bran (2%) as media source. Semi-for-mutation of B. thuringiensis 656-3 was performed with metamorphic starch only. When the formulation suspension containing $5{\times}{10^7}$ cfu was sprayed on the mushroom in mushroom house, the insecticidal effect of B. thuringiensis 656-3 to mushroom flies,1. mali and C. fuscipes, was maintained over 90% by the fifth day after starting spraying. The yield of oyster mushroom house with B. thuringiensis 656-3 was significantly increased compared to control. B. thuringiensis 656-3 represents a powerful biological insecticide for the control of mushroom flies.

Anti-inflammatory and Antimicrobial activities of Petroleum ether and Ethanol extracts of Scutia myrtina (Rhamnaceae)

  • The aim of the present study is to investigate the anti-inflammatory and antimicrobial activity of petroleum ether and ethanol extracts of Scutia myrtina (Family: Rhamnaceae). In anti-inflammatory activity carrageenan and histamine induced paw oedema and cotton pellet induced granuloma for acute and chronic inflammatory models were studied in Wister albino rats. Based on the results of the present study it can be concluded that petroleum ether and ethanol extract of Scutia myrtina at 400 mg/kg has potential anti-inflammatory effect and they act in a dose dependent manner. Both extracts of Scutia myrtina showed antimicrobial activity against all bacterial and fungal strains tested at the concentration of $100\;{\mu}g$/ml. From the result, it can be concluded that the Scutia myrtina contain antibacterial and antifungal principle. Further more, besides the confirmation of the popular use; the obtained results demonstrate this herbal drug to represent a new source of antimicrobial and anti-inflammatory agent.

Effect of natural pre-converted nitrite sources on color development in raw and cooked pork sausage

  • Objective: The effect of pre-converted nitrites from natural sources (spinach, lettuce, celery, and red beet) on color development in raw and cooked pork sausage was investigated in this study. Methods: The pork sausage was manufactured with six treatments: NC (negative control, nitrite free), PC (positive control, 150 ppm sodium nitrite), FS (3.0% fermented spinach extracts), FL (3.0% fermented lettuce extracts), FC (3.0% fermented celery extracts), and FR (3.0% fermented red beet extracts). Results: The pH value of the pre-converted nitrites groups was lower than those treated with 150 ppm sodium nitrite (p<0.05). The color values of raw and cooked pork sausage added with pre-converted nitrite showed slightly lower and/or similar lightness, lower redness, and higher yellowness values than PC. Color development (redness values) of cooked samples added with FS was higher than those of the NC and other treatments (FL, FC, and FR). Additionally, treatments with FS and FL were most effective for reducing thiobarbituric acid reactive substances and volatile basic nitrogen than the NC. Conclusion: Effects of natural nitrites from fermented vegetables on shelf stability of raw and cooked pork sausages were investigated. Fermented spinach extract was much more useful for maintaining the color development, but also inhibiting lipid and protein oxidation of cooked pork sausage. Therefore, pre-converted nitrite from spinach as a natural nitrite could be used as another natural nitrite source for making processed meat products.

GIS를 이용한 2차원 수치모형의 자연하천 적용 (Application of 2D Numerical Model for Natural Rivers using GIS)

  • 현재까지 개발되고 있는 대부분의 유한체적모형은 가상하도 및 실험하도와 같은 단순하도에 적용하여 흐름율과 생성항의 균형문제를 해결하기 위한 여러 가지 노력들이 있어 왔다. 하지만, 실제 자연하천에서의 적용에서는 단순하도에서의 적용에서 나타나지 않았던 여러 가지 문제점들이 발생하며, 이러한 문제점들을 수치적으로 해결하여야 비로소 자연하천에서의 적용이 가능하다. 본 연구에서는 자연하천에 적용가능한 2차원 유한체적모형을 개발하여 2차원 부분적 댐 붕괴에 적용하여 개발모형의 정확성을 검증하고, 한강의 측량단면을 GIS를 이용하여 2차원 격자에 정확하게 반영할 수 있는 간단하고 효율적인 2차원 격자생성기법을 제안하였다. 그리고 제안된 기법으로 생성된 2차원 격자로 한강에 대한 흐름모의를 수행하고 계산수위와 실측수위를 비교하여 자연하천에 대한 개발모형의 정확성 및 적용성을 입증하였다.

배지내 천연유기물 첨가가 딸기 "설향" 배양묘 기내 생육에 미치는 영향 (Effect of Natural Additives on In Vitro Growth Medium of Strawberry 'Seolhyang')

  • Strawberry which is the genus Fragaria under family Rosaceae is one of the most important fruit plants for both fresh consumption and food processing in the temperate and subtropical countries. Propagation of strawberry is achieved either through runners or by in vitro micropropagation. Meristem tips, generally obtained from runners of virus-free plants, are commonly used to establish in vitro cultures, which are employed for mass propagation or as a source of plant material for regeneration and transformation experiments. This study was conducted to determine the optimal natural additives strength to improve sprouting shoot rate of apical meristem of strawberry 'Seolhyang'. Strawberry apical meristem at size (0.2 mm to 0.3 mm) with leaf primordials were cultured on the 1/3MS(Murashige & Skoog) medium supplemented with five natural additives such as coconut milk, maple sap, banana powder and peptone. The sprouting ratio and growth characteristics were evaluated after eight weeks after in vitro culture. Shoot ratio of 'Seolhyang' apical meristem was 72.9% in 1/3MS medium supplemented with maple sap. On the other hand, the low shoot ratio was observed 47.7% in 1/3MS medium supplemented with banana powder. Shoot length was different as natural additives but numbers of leaf was not significantiy different among the natural additives. As a result, the sprouting ratio and plant growth were enhanced effectively in 1/3MS medium with maple sap compared to the others.

Steroid Components of Marine-Derived Fungal Strain Penicillium levitum N33.2 and Their Biological Activities

  • Genus Penicillium comprising the most important and extensively studied fungi has been well-known as a rich source of secondary metabolites. Our study aimed to analyze and investigate biological activities, including in vitro anti-cancer, anti-inflammatory and anti-diabetic properties, of metabolites from a marine-derived fungus belonging to P. levitum. The chemical compounds in the culture broth of P. levitum strain N33.2 were extracted with ethyl acetate. Followingly, chemical analysis of the extract leaded to the isolation of three ergostane-type steroid components, namely cerevisterol (1), ergosterol peroxide (2), and (3β,5α,22E)-ergosta-6,8(14),22-triene-3,5-diol (3). Among these, (3) was the most potent cytotoxic against human cancer cell lines Hep-G2, A549 and MCF-7 with IC50 values of 2.89, 18.51, and 16.47 ㎍/mL, respectively, while the compound (1) showed no significant effect against tested cancer cells. Anti-inflammatory properties of purified compounds were evaluated based on NO-production in LPS-induced murine RAW264.7 macrophages. As a result, tested compounds performed diverse inhibitory effects on NO production by the macrophages, with the most significant inhibition rate of 81.37±1.35% at 25 ㎍/mL by the compound (2). Interestingly, compounds (2) and (3) exhibited inhibitory activities against pancreatic lipase and α-glucosidase enzymes in vitro assays. Our study brought out new data concerning the chemical properties and biological activities of isolated steroids from a P. levitum fungus.
