OVA로 유도된 천식 모델 생쥐에서 목천료자(木天蓼子)가 조절 T 세포, NK T 세포 및 gammadelta T 세포수 변화에 미치는 영향 (Effects of APF and CsA on the number of regulatory T cells, NK T cells and gammadelta T cells in OVA-induced murine model of asthma)

  • Objectives : To clarify the effects of Actinidia polygama and CsA on OVA-induced asthma model, we examined the influence of Actinidia polygama fructus extract (APF) and CsA on the number of regulatory T cells, NKT cells and ${\gamma}{\delta}$ T cells in murine model of asthma. Methods : All mice were immunized on two different days (21 days and 7 days before inhalational exposure) by i.p. injections of OVA in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 min/day on 3 days/week for 12 weeks and APF (400, 40 mg/kg) were orally administered 3 times a week for 8 weeks. Results : The suppressive effects of APF on asthma model were demonstrated by the increase the number of regulatory T cells, ${\gamma}{\delta}$ T cells and by reducing the number of NK T cells. Conclusion : These results indicate that APF has a deep inhibitory effect on airway inflammation and hyperresponsiveness in murine model of asthma by increase the number of regulatory T cells, and ${\gamma}{\delta}$ T cells and by reducing the number of NK T cells.

Enhancement of NK Cytotoxicity, Antimetastasis and Elongation Effect of Survival Time in B16-F10 Melanoma Cells by Oregonin

  • We investigated the antitumor activity of oregonin, a diarylheptanoid derivative purified from Alnus hirsuta Turcz, Betulaceae. Oregonin is a potential novel immunomodulator, which augments the activation of natural killer (NK) cells, and thereby leads to a powerful antitumor activity. To evaluate the cytotoxicity of oregonin against tumor cells, we examined the effectiveness of NK cells and determined that oregonin could increase NK cell cytotoxicity. This was confirmed by MTT assay. In addition, the survival time of C57BL/6 mice were measured by inoculating 816-F10 melanoma cells to mice via intra muscular (i.m.) injection. Oregonin treatment after 10 hours of inoculation at 10 mg/kg dosage showed a significant extension of survival time by up to 51.32%, when compared to the control group. Moreover, oregonin significantly reduced the incidence of pulmonary metastasis, which may be developed from 816-F10 melanoma cells. These findings suggest that oregon in may be classified as a new and novel immunomodulator due to its potential antitumor activity.

COVID-19 Vaccination Alters NK Cell Dynamics and Transiently Reduces HBsAg Titers Among Patients With Chronic Hepatitis B

  • Coronavirus disease 2019 (COVID-19) vaccination may non-specifically alter the host immune system. This study aimed to evaluate the effect of COVID-19 vaccination on hepatitis B surface Ag (HBsAg) titer and host immunity in chronic hepatitis B (CHB) patients. Consecutive 2,797 CHB patients who had serial HBsAg measurements during antiviral treatment were included in this study. Changes in the HBsAg levels after COVID-19 vaccination were analyzed. The dynamics of NK cells following COVID-19 vaccination were also examined using serial blood samples collected prospectively from 25 healthy volunteers. Vaccinated CHB patients (n=2,329) had significantly lower HBsAg levels 1-30 days post-vaccination compared to baseline (median, -21.4 IU/ml from baseline), but the levels reverted to baseline by 91-180 days (median, -3.8 IU/ml). The velocity of the HBsAg decline was transiently accelerated within 30 days after vaccination (median velocity: -0.06, -0.39, and -0.04 log10 IU/ml/year in pre-vaccination period, days 1-30, and days 31-90, respectively). In contrast, unvaccinated patients (n=468) had no change in HBsAg levels. Flow cytometric analysis showed that the frequency of NK cells expressing NKG2A, an NK inhibitory receptor, significantly decreased within 7 days after the first dose of COVID-19 vaccine (median, -13.1% from baseline; p<0.001). The decrease in the frequency of NKG2A+ NK cells was observed in the CD56dimCD16+ NK cell population regardless of type of COVID-19 vaccine. COVID-19 vaccination leads to a rapid, transient decline in HBsAg titer and a decrease in the frequency of NKG2A+ NK cells.

Protective Antitumor Activity through Dendritic Cell Immunization is Mediated by NK Cell as Well as CTL Activation

  • Dendritic cells (DCs) are potent professional antigen-presenting cells (APC) capable of inducing the primary T cell response to antigen. Although tumor cells express target antigens, they are incapable of stimulating a tumor-specific immune response due to a defect in the costimulatory signal that is required for optimal activation of T cells. In this work, we describe a new approach using tumor-DC coculture to improve the antigen presenting capacity of tumor cells which does not require a source of tumor-associated antigen. Immunization of a weakly immunogenic and progressive tumor cocultured with none marrow-derived DCs generated an effective tumor vaccine. Immunization with the cocutured DCs was able to induce complete protectiv immunity against tumor challenges and was effective for the induction of tumor-specific CTL (cytotoxic T lymphocyte) activity. Furthermore, high NK cell activity was observed in mice in which tumors were rejected. In addition, immunization with tumor-pulsed DC s induced delayed tumor growth, but not tumor eradication in tumor-bearing mice. Our results demonstrate that coculture of DCs with tumors generated antitumor immunity due to the NK cell activation as well as tumor-specific T cell. This approach would be used for designing tumor vaccines using DCs when the information about tumor antigens is limited.

자유생활아메바의 병원성에 따른 자연살세포의 활성도 (Natural killer cell activity in mice infected with free-living amoeba with reference to their pathogenicity)

  • 원발성 아메바성 수막뇌염을 일으키는 병원성 자유생활아메바인 Naegleria fowleri 및 Acanthamoeba culbertsoni를 CSH/HeJ 마우스에 감염시켜 자연살세포를 활성화시킬 수 있는 감염 최소량 및 활성도를 최대치가지 이르게 하는 감염 최대량을 결정하여 수막뇌염의 발생여부 및 사망율을 조사하고, 이와 함께 비병원성 자유생활아메바인 Naegleria fowleri를 감염시켜 자연살세포가 활성화되는지 조사하였다 자연살세포 활성도를 병원성 자유생활아메바 감염군과 비교하였으며, 병원성, 비병원성 자유생활아메바 감염군에서의 자연살세포의 세포독성의 변화를 단세포 독성검사법을 이용하여 표적세포 결합능, 활성 자연살세포, 더 나아가 최대 재순환능을 측정하여 조사하였다. Naegleria fowleri 감염 군에서 자연살세포를 활성화 시킬 수 있는 감염 최소량인 아메바 영양형 $1{\times}10^4$개 감염 군에서 사망률이 5.9%이었으며 최대량인 $1{\times}10^5$개 영양형 감염군에서는 72.2%이었다. Acnnthamoebn culbertsoni 감염 군에서의 감염 최소량인 아메바 영양형 $1{\times}10^3$개 감염군에서의 사망률은 6.9%이었으며 최대량인 $1{\times}10^5$개 감염군에서는 65.5%이었다. 자연살세포의 세포독성은 병원성 자유생활아메바 감염군 모두에서 감염후 1일째에 대조군과 비교하여 통계학적으로 유의하게 증가하였으며, 감염후 2일과 5일째에는 감소하였고, 아메바 감염량간에는 유의한 차이를 발견할 수 없었다. 아메바 감염 군에서의 자연살세포의 활성도의 증가는 표적세포 결합능 및 활성 자연살세포의 증가에 기인한 것이었으며 재순환능에는 차이가 없음을 알 수 있었다. 비병원성 자유생활아메바인 Naegleria fowleri 감염 군에서는 자연살세포의 활성도는 대조군과 비교하여 유의한 차이를 발견할 수 없었으며 병원성 자유생활아메바 감염 군과는 자연살세포의 세포독성 통계학적으로 유의한 차이가 있음을 알 수 있었다.

Characterization of the Immunologically Active Components of Glycyrrhiza uralensis Prepared as Herbal Kimchi

  • A crude polysaccharide fraction (GU-3) from the roots of Glycyrrhiza uralensis (licorice root), a screened herbal plant used in the preparation of herbal kimchi, enhanced Peyer's patch mediated bone marrow cell proliferation and NK cell-mediated tumor cytotoxicity against Yac-1 cells. GU-3 was further purified by DEAE-Sepharose CL-6B yielding fractions designated as GU-3I, and 3IIa∼3IIe. GU-3IIa is mainly composed of arabinose, galactose and galacturonic acid, and showed the highest bone marrow cell proliferation activity. In addition, GU-3IIb had arabinose, galactose, rhamnose and galacturonic acid as the component sugars with a small quantity of protein; GU-3IIb also enhanced activity of NK cell-mediated tumor cytotoxicity. After these fractions were further fractionated via gel filtration on Sepharose CL-6B or Sephacryl S-300, two immunological active polysaccharides, GU-3IIa-2 and 3IIb-1 were purified from the respective fractions. GU-3IIa-2 mostly contained neutral sugars (75%) such as arabinose and galactose (molar ratio; 1.0 : 0.7) in addition to a considerable amount of galacturonic acid (20%), whereas GU-3IIb-1 was composed of arabinose, galactose, rhamnose and galacturonic acid (molar ratio; 0.3 : 0.5 : 0.1 : 1.0). Methylation analysis indicated that GU-3IIa-2 was composed mainly of terminal, 4- or 5-linked and 3,4- or 3,5-branched arabinose, 3-linked, 4-linked and 3,6-branched galactose, and terminal and 4-linked galacturonic acid whereas GU-3IIb-1 contained various glycosidic linkages such as terminal and 4- or 5-linked arabinose, 2,4-branched rhamnose, terminal and 4-linked galactose, and terminal and 4-galacturonic arid. Single radial gel diffusion indicated that only GU-3IIa-2 strongly reacted with β-D-glucosyl-Yariv antigen. These results suggest that bone marrow cell proliferating activity and enhancement of NK cell-mediated tumor cytotoxicity of GU-3 are caused by polysaccharides containing a pectic arabinogalactan (GU-3IIa-2) and pectic polysaccharide (GU-3IIb-1).

흡연자에 있어 비타민 C 보충이 면역능력에 미치는 영향 (Effects of Vitamin C Supplementation on Immune Status in Smoking and Nonsmoking Male College Students)

  • This study investigated the effects of vitamin C supplementation on immune status in smoking and nonsmoking male college students. Blood samples were obtained from 15 healthy smoking and from 15 healthy nonsmoking volunteers before and 4 weeks after vitamin C supplementation (1000mg/day). Daily nutrient intakes was also calculated. Plasma vitamin C, E, and A concentrations, white blood cells and subpopulations, lymphocytes and subpopulations, NK cell percentage,plasma immunoglobulin A, G, M and complement C3, plasma interleukin-2 , and prostagladin E2 were measured . Daily vitamin C consumption was lower in smokers than in nonsmokers. Vitamin C supplementation significantly increased plasma vitamin C in smokers, and increased NK cell percentage in smokers and nonsmokers. Plasma IL-2 concentrations were significantly increased by vitamin C supplementation in nonsmokers, and decreased helpe $r^pressor T cell ratio were decreased by vitamin C supplementatiion , but the differences were not significant. White blood cells and subpopulation percentage and plasma prostaglandin E2 concentration were not affected by smoking and vitamin C supplementation. These findings as nonsmokers.rs.

Effect of Propolis Volatiles from a Stingless Honeybee(Apidae : Meliponinae) on the Immune System of Elderly Residents in a Nursing Home

  • We report an evaluation of the immunostimulatory effect of propolis volatiles from a stingless honeybee. We studied 34 elderly residents at a special nursing home. Twenty-one subjects were treated with propolis, 8 with $Binch\hat{o}$ charcoal and 5 subjects acted as controls. Subjects treated with either propolis or Bincho charcoal were housed in rooms separated from the other non-study residents in the nursing home. The effects of each treatment on natural killer (NK) cell activity and lymphocyte levels were examined after 2 months and then for a longer period. The results indicated that NK cell activity was significantly improved to that within the normal range only after propolis treatment.

가미청간탕이 N-nitrosomorpholine으로 유도된 전암성 간병변에 미치는 영향 (Effect of Kamicheonggan-tang on Pre-hepatocarcinogenesis Induced by N-nitrosomorpholine)

  • The purpose of this study was to evaluate the protective effect of Kamicheonggan-tang(KCGT) on pre-hepatocarcinogenesis induced by N-nitrosomorpholine. The studied using blood chemistry, lipidperoxidation, antioxidant, immunohistochemistry and morphological change. The results were obtained as follows. In the pre-hepatocarcinogenesis induced by NMP, serum AST, ALT, ALP and total bilirubin were not changed in NMP and NK treated group after 1 st week, but desreased in NK treated group after 4th week as compared with NMP treated 4th week group. The content of GSH was similary to in NK treated groups as compares with data of normal group. The content of MDA was increased in NK treated group after 1st and 4th week, and more increased in the NMP treated group than those of their NK treated group. The immunohistochemically, stain of GST-p, positive lesions of KCGT were significantly decreased than those of NMP treated group. The histopathologically, fat changes, nucleotic changes, oval cell and inflammatory cells in periportal were observed in NMP treated fater 4th week, but those were significantly decreased from 4th week in the NK treated group. And the enlarged nucleus was not changed in KCGT treated group, but increased in NMP treated group after 1st and 4th week. The ultrastructurally, nucleotic changes, glycogen degeneration, lipid droplet and rER fragmentation were observed in NMP treated group after 4th week, but those changes were significantly decreased from 4th week in the NK treated group. These results suggested that KCGT extracts has protective effect on prehepatocarcinogenesis by NMP, might be usefully applied for clinical treatment of hapatic disease and also it was necessary to do more studies about its mechanisms.

Enhancement of Anticancer Activities of Kimchi by Manipulating Ingredients

  • To enhance the antitumor activity of Chinese cabbage kimchi, four kinds of kimchi, which ere differently prepared in kinds and levels of sub-ingredients, were fermented at 15$^{\circ}C$ for 1 day and then at 5$^{\circ}C$ up to pH 4.3. The solid tumor formation, hepatic glutathione S-transferase activity and glutathione contents in the liver, and natural killer (NK) cell activity of spleen were determined from the sarcoma-180 cell injected Balb/c mice that were treated with methanol extracts of the kimchi samples. Kimchi IV, prepared with organically cultivated Chinese cabbage, red pepper powder, garlic, Chinese pepper powder mustard leaf and heat processed salt (Gueun salt), reduced the tumor formation by 39.3% compared to the sarcoma-180 cell treated group, resulting in the smallest tumor weight. Methanol extracts of the kimchi III and kimchi IV recovered the activities of hepatic glutathione S-transferase(GST) that was decreased by the transplantation of the sarcoma-180 cells to th mice. The injections of methanol extracts of kimchi II and kimchi IV increased glutathione contents in sarcoma-180 cells treated mice. The methanol extract of kimchi IV increased the natural killer (NK) cell activity of spleen lymphocytes a more effectively (p<0.05) than those the other kimchi samples. These results suggest that the anticancer activities of kimchi can be increased by changing the kinds and levels of sub-ingredients.
