New Evaluation of Initial Growth Mechanisms of Hydroxyapatite on Self-assembled Collagen Nanofibrils by Using ToF-SIMS and AFM Techniques

  • Bone is considered as hierarchically organized biocomposites of organic (collagen) and inorganic (hydroxyapatite) materials. The precise structural dependence between hydroxyapatite (HAp, $Ca_{10}(PO_4)_6(OH)_2)$ crystals and collagen fibril is critical to unique characteristics of bone. To meet those conditions and obtain optimal properties, it is essential to understand and control the initial growth mechanisms of hydroxyapatite at the molecular level, such as other nano-structured materials. In this study, collagen fibrils were prepared by adsorbing native type I collagen molecules onto hydrophobic surface. Hydrophobicity was introduced on the Si wafer surface by using PECVD (plasma enhanced chemical vapor deposition) method and cyclohexane as a precursor. Biomimetic nucleation and growth of HAp on the self-assembled collagen nanofibrils were occurred through incubation of the sample in SBF (simulated body fluid). Chemical and morphological evolution of HAp nanocrystals was investigated by surface-sensitive analytical techniques such as ToF-SIMS (Time-of-Flight Secondary Ion Mass Spectrometry) and AFM (Atomic Force Microscopy) in the early growth stages (< 24 hrs). The very initial stages (< 12 hrs) of mineralization could be clearly demonstrated by ToF-SIMS chemical mapping of surface. In addition to ToF-SIMS and AFM measurement, scanning electron microscopy, energy dispersive spectroscopy and X-ray diffraction analysis were conducted to characterize the HAp layer in the late stages. This study is of great importance in the growth of real bone-like materials with a structure analogous to that of natural bones and the development of biomimetic nanomaterials.

Effect of Additive Ammonium Hydroxide on ZnO Particle Properties Synthesized by Facile Glycol Process

  • ZnO particles are successfully synthesized at 150 ℃ for 30 min using zinc acetate as the Zn source and 1,4-butanediol as solvent using a relatively facile and convenient glycol process. The effect of ammonium hydroxide amounts on the growth behavior and the morphological evolution of ZnO particles are investigated. The prepared ZnO nanoparticle with hexagonal structure exhibits a quasi-spherical shape with an average crystallite size of approximately 30 nm. It is also demonstrated that the morphology of ZnO particles can be controlled by 1,4-butanediol with an additive of ammonium hydroxide. The morphologies of ZnO particles are changed sequentially from a quasi-spherical shape to a rod-like shape and a hexagonal rod shape with a truncated pyramidal tip, exhibiting preferential growth along the [001] direction with increasing ammonium hydroxide amounts. It is demonstrated that much higher OH- amounts can produce a nano-tip shape grown along the [001] direction at the corners and center of the (001) top polar plane, and a flat hexagonal symmetry shape of the bottom polar plane on ZnO hexagonal prisms. The results indicate that the presence of NH4+ and OH- ions in the solution greatly affects the growth behaviors of ZnO particles. A sharp near-band-edge (NBE) emission peak centered at 383 nm in the UV region and a weak broad peak in the visible region between 450 nm and 700 nm are shown in the PL spectra of the ZnO synthesized using the glycol process, regardless of adding ammonium hydroxide. Although the broad peak of the deep-level-emission (DLE) increases with the addition of ammonium hydroxide, it is suggested that the prominent NBE emission peaks indicate that ZnO nanoparticles with good crystallization are obtained under these conditions.

BAT AGN Spectroscopic Survey - The parsec scale jet properties of the ultra hard X-ray selected local AGNs

  • We have conducted a 22 GHz very long baseline interferometry (VLBI) survey of 281 local (z < 0.05) active galactic nuclei (AGNs) selected from the Swift Burst Alert Telescope (BAT) 70-month ultra hard X-ray (14-195 keV) catalog. The main goal is to investigate the relation between the strengths of black hole accretion and the parsec-scale nuclear jet, which is expected to tightly correlate but has not been observationally confirmed yet. The BAT AGN Spectroscopic Survey (BASS) provides the least biased AGN sample against obscuration including both Seyfert types, hence it makes an ideal parent sample for studying the nuclear jet properties of an overall AGN population. Using the Korean VLBI Network (KVN), the KVN and VERA Array (KaVA), and the Very Long Baseline Array (VLBA), we observed 281 objects with a 22 GHz flux > 30 mJy, detecting 11 targets (~4% of VLBI detection rate). This implies that the fraction of X-ray AGNs which are currently ejecting a strong nuclear jet is very small. Although our 11 sources span a wide range of pc-scale morphological types, from compact to complex, they lie on a tight linear relation between accretion luminosity and nuclear jet luminosity. Our finding may indicate that the power of nuclear jet is directly responsible for the amount of black hole accretion. We also have probed the fundamental plane of black hole activity in VLBI scale (e.g., few milli-arcsecond). The results from our high-frequency VLBI radio study support that the change of jet luminosity and size follows what is predicted by the AGN evolution scenario based on the Eddington ratio (ƛ$_{Edd}$) - column density ($N_H$) plane, proposed by a previous study.

New Records of Two Arcuospathidium Subspecies (Ciliophora: Haptoria: Arcuospathidiidae) from Korea

  • Arcuospathidium is a haptorian ciliate genus composed of 18 species, and only one species has been reported in Korea. Here, we identify two unrecorded Arcuospathidium subspecies by morphological observation of both living and protargol-impregnated specimens with the small subunit ribosomal RNA (18S rRNA) gene sequence. These subspecies, Arcuospathidium cultriforme cultriforme (Penard, 1922) Foissner, 1984 and A. cultriforme scalpriforme (Kahl, 1930) Foissner, 2003, were isolated from various terrestrial habitats in July and August 2013, respectivley. Arcuospathidium cultriforme cultriforme is similar to A. cultriforme scalpriforme by a knife-shaped body, a twisted-shaped macronucleus, number of dorsal brushes, position of dorsal brushes, and shape of macronucleus but former mainly differs from the body length to oral bulge length ratio (27-38% vs. 41-53%), extrusome (one types vs. three types), cyst shape (roughly faceted wall vs. smooth surface and thin wall) and number of somatic kinety rows(18-30 vs. 30-44). Additionally, we analyzed the 18S rRNA gene sequences of two A. cultriforme subspecies and compared them with the sequences from GenBank to confirm their identification at the molecular level. As the results of genetic analysis, the 18S rRNA gene sequence of the Korean A. cultriforme cultriforme population is most similar to that of Austrian population. Also, the sequence of the Korean A. cultriforme scalpriforme population is most similar to that of another population with some nucleotide differences.

Geographic Variation and Interspecific Hybridization between Two Species of the Genus Agkistrodon (Crotalidae) in Korea (한국산 살모사속(뱀과) 2종의 지리적 변이 및 종간 잡종)

  • Genetic and morphometric comparison between two species of the genus Agkistrodon were performed to estimate the degree of genic variation and to clarify the taxonomic status of Cheju population, a suspected hybrid form between A. blomhoffii brevicaudus and A u ussuriensis. A sum of 147 specimens representing six populations of A.b. brevicaudus and two populations of A ussuriensis was used in this study. Out of the 21 loci examined, 5 loci (Adh, Ldh-2, Mdh-2, Mpi, Pgi) were monomorphic with identical mobility in both species. O Other Sioci (Aco, Gp, Fum, [po and Xdh in A b. brevicaudus; Est, Got-l, Gp, Mdh-l and 6Pgd in A ussuriensis) were highly poplymorphic. The degree of genic variation of Ab. b brevicaudus and A ussuriensis was A=1.SS, P=42.1%, HD=0.096, HG=O.l1S, and A= 1.4S, P = 31.6%, HD=0.117, HG =0.121, repectively. These values are twofold higher t than those of other reptilian species reported (Selander, 1976) including Korean species of Rhabdophis tigrina and Elaphe dione (Paik and Yang, 1986, 1987). The average values of t the genetic similarities among six populations of A b. brevicaudus and two populations of A ussuriensis were S=0.919 and S=0.962, respectively, whereas the value between species was S = 0.662. Presumed divergent time estimate (Nei, 1975) of these two species was about 1 1.8 million years ago. The analysis of the geographic variations of various morphological c characters was based on Gloyd’s criteria (Gloyd, 1972). Coloration of tongue and tail tip, t the number of ventral scales, and the number of subcaudal scales are good diagnostic c characters to identify these two species. The pattern of cross band is, however, highly variable within and between populations of both species. No hybrid was detected and the s suspected hybrid form of Cheju population falls within the range of A. ussuriensis in the g genetic and morphometric analyses.

  • The partial sequences of mtDNA cytochrome b gene in two subspecies of striped field mouse(Apodemus agrarius coreae and A. agrarius manchuricus) from Korea and northeast China were analyzed to determine the degree of genetic diversity in ech subspecies and to confirm their subspecific difference. In 18 specimens of A. agrarius coreae, ten haplotypes were resulted, and in two specimens of A. agrarius manchuricus. one haplotype was revealed. Tamura-Nei nucleotide distance among ten haplotypes in subspecies coreae ranged 0.36 to 1.86%. and nucleotide distance between two subspecies (coreae and manchuricus) was 0.37 to 1.47%: maximum infrasubspecific divergence in coreae was greater than maximum intersubspecific difference between two subspecies. Moreover, no major subgroup was resulted when 11 haplotypes in two subspecies were compared. Our sequence result was not cancordant with the morphological data studied so far, and it is concluded that cytochrome b gene sequence is not a good genetic marker to distinguish two subspecies of A. agrarius. In futurem, mtDNA control region analyses seemded to be necessary to reveal genetic relationship between these two subspecies of A. agrarius.

The Morphological Change by Slope Erosional Processes in the Dokdo Seamount (독도해산의 사면침식으로 인한 지형변화)

  • The purpose of this study is to analyze landform characteristics and geomorphic processes in Dokdo seamount. For geomorphic analysis, bathymetry data were collected by multi-beam echosounder and the seismic survey was also conducted. Through the detailed analysis of depth, slope, aspect and erosional landform, Dokdo Seamount is characterized by a flat or gently sloping top of $2^{\circ}$ or less and seamount slope with $14{\sim}40^{\circ}$ gradient. There are protrusion of landform around the Dokdo on top of the Dokdo seamount. It is inferred that the features are formed by collapsed debris deposits or remained bedrocks by differential erosion in the past. The massmovement topography including slump and slide is shown on seamount slope with $14{\sim}40^{\circ}$ gradient. In addition, gullies with various length are developed on the Dokdo seamount slope. Slope erosional processes occur more actively along the submarine gullies on the Dokdo seamount. It is inferred that the massmovement processes on the slope of Dokdo seamount are related to earthquake activities and evolution of submarine volcano. Consequently, slope of the Dokdo seamount has retreated by erosional processes of mass-movement and submarine gullies.

Genetic Relationship of the Ampelopsis brevipedunculata var. heterophylla and Vitis thunbergii var. sinuata with the Other Vitis Plants (개머루와 까마귀머루의 유전적 유연관계 분석)

  • DNA sequences of the intergenic spacer 1 and intergenic spacer 2 of the nineteen plants belonging Vitis genus were collected from the Genbank. DNA sequences of the same regions of Vitis thunbergii var. sinuata and Ampelopsis brevipedunculata var. heterophylla, both common plants in Korea, were not available in Genbank. Those two plants were collected, their genomic DNA encoding 18S rRNA, intergenic spacer 1, 5.8S rRNA, intergenic spacer 2 and part of 28S rRNA amplified and DNA sequence determined. DNA sequences of twenty-one plants including two Korean plants were aligned by the Multiple sequence comparison by log-expectation(MUSCLE) algorithm and the alignment was used to calculate neighbor-joining tree and pairwise distance. The results indicate DNA sequences of the two Korean plants are highly homologous with each other, but they are quite distantly related to the other Vitis plants. Distant relationship of the two Korean plants with the other Vitis plants might be due to independent evolution of those two plants in geographically isolated environment. Those two Korean plants are classified in different genera based on the morphology, one in Vitis genus and the other in Ampelopsis genus, providing another example of discrepancy between morphological and genetic classification.

Systematic Relationships Among Ten Species of Subgenus Drosophila (Diptera: Drosophilidae) in Korea (한국 초파리아속(쌍시목: 초파리과) 10종에 대한 계통적 유연관계)

  • This study sought to observe systematic relationships through taximetrical analyses by morphological characters among the 10 species, on three species in the virilis section (D. virilis, D. tsigana and D. lacertosa) and seven species in the quinaria section (D. angularis, D. brachvnephros, D. curvispina, D. kuntzei, D. nigromaculata, D. takadai and D. unispina) of the subgenus Drosophila. In the cluster and the cladistic analysis among the members of subgenus Drosophila, 10 species was divided into the 1st group of D. virilis, D. tsigana, and D. lacertosa and the 2nd group of D. angularis, D. brachynephros, D. curvispina, D. kuntzei, D. nigromaculata, D. takadai, and D. unispina. In cluster analysis, the 2nd group had three sister groups; one sister group that clustered D. angularis and D. brachynephros then D. unispina was clustered to them, another sister group clustered D, curvispina and D. takadai then D. kuntzei was clustered to them and the other sister group of D. nigromaculata. In the 10 species, D. virilis and D. lacertosa were the first to be divided and then D. tsigana. Although 1st group which D. virilis was belonged can be determined as more primitive than the 2nd group, it seemed that this group was not the direct ancestor of the 2nd group, rather there should be another ancestor. Among the quinaria species group, D. nigromaculata was the first to be divided and D. kuntzei was the most recent species to be divided.

Elevation of Properties of Al-Nb-Ar alloys Fabricated by Mechanical Alloying Metho (기계적합금화법을 이용한 고온 고강도 Al-Nb-Zr 합금 제조 및 특성 평가)

  • Recently there have been many investigations on the synthesis and properties of transition metal trialuminides based on Ti, Zr, V, Nb and Ta for use aircraft structure materials in an elevated environment. The effect of Zr additions on the formation behaviour of Al-Nb alloy was investigated. Al-1.3at.%(Nb+Zr) alloys with different Nb to Zr atomic 1:3, 1:1 and 3:1 were prepared by mechanical alloying(MA). The morphological changes and microstructural evolution of Al-Nb-Zr powders during MA were investigated by SEM, XRD and TEM. The intermetallic compound phase of $Nb_2Al\; and\; Al_3Zr_4$ was identified by X-ray diffraction. The intemetallic compound of $Al_3Zr,\; Al_3Nb$ and $Al_3Zr_4$ were formed by heat treatment for 1 hour at $500^{\circ}C$. The size of intermetallic compounds observed by TEM were approximately below 100nm, when they were heat treated after mechanically alloying for 30 hours.
