Effect of substrate bias voltage on a-C:H film (기판 bias 전압이 a-C:H 박막의 특성에 미치는 영향)

  • Hydrogenated amorphous carbon(a-C:H) films were deposited on p-type Si(100) by DC saddle-field plasma enhanced CVD to investigate the effect of substrate bias on optical properties and structural changes. They were deposited using pure methane gas at a wide range of substrate bias at room temperature and 90 mtorr. The substrate bias voltage ($V_s$) was employed from $V_s=0 V$ to $V_s=400 V$. The information of optical properties was investigated by photoluminescence and transmitance. Chemical bondings of a-C:H have been explored from FT-IR and Raman spectroscopy. The thickness and relative hydrogen content of the films were measured by Rutherford backscattering spectroscopy (RBS) and elastic recoil detection (ERD) technigue. The growth rate of a-C:H film was decreased with the increase of $V_s$, but the hydrogen content of the film was increased with the increase of $V_s$. The a-C:H films deposited at the lowest $V_s$ contain the smallest amount of hydrogen with most of C-H bonds in the of $CH_2$ configuration, whereas the films produced at higher $V_s$ reveal dominant the $CH_3$ bonding structure. The emission of white photoluminescence from the films were observed even with naked eyes at room temperature and the PL intensity of the film has the maximum value at $V_s$=200 V. With $V_s$ lower than 200 V, the PL intensity of the film increased with V, but for V, higher than 200 V, the PL intensity decreased with the increase of $V_s$. The peak energy of the PL spectra slightly shifted to the higher energy with the increase of $V_s$. The optical bandgap of the film, determined by optical transmittance, was increased from 1.5 eV at $V_s$=0V to 2.3 eV at $V_s$=400 V. But there were no obvious relations between the PL peak and the optical gap which were measured by Tauc process.

Anaerobic Digestion Biochemical Sludge Produced from Municipal Sewage Treatment Process (하수처리시설에서 발생된 약품 잉여슬러지의 혐기성 소화 특성)

  • This study was carried out to get the characteristics of anaerobic digestion for chemical/biological sludge produced from municipal sewage treatment plant for phosphorus. Anaerobic mesophilic batch tests showed that the ultimate biodegradability of waste activated sludge showed 31%, PACl sludge 24%, Alum sludge 26%, respectively. At the S/I 1.0, 75% of total biodegradable volatile solids (TBVS) of waste activated sludge was degraded with an initial rapid decay coefficient, k1 of $0.1129day^{-1}$ and 74% of TBVS of PACl sludge with k1 of $0.0998day^{-1}$, and 76% of TBVS of Alum sludge with k1 of $0.1091day^{-1}$ for 20 days. During the operation of SCFMRs, the 3 reactor (Control, PACl, Alum) pH maintained 6.7~7.0 and the reactor alkalinity maintained 1,800~ 2,200 mg/L as $CaCO_3$. The average biogas production rates of SCFMRs fed with PACl sludge and Alum sludge were 0.089 v/v-d and 0.091 v/v-d, respectively, which was 27~28% lower than that of the control (0.124 v/v-d) at an HRT (hydraulic retention times) of 20 days. And the methane content during the operation ranged 70~76% in 3 reactor. The average TVS removal efficiency of SCFMRs fed with PACl sludge and Alum sludge were 19.6% and 19.9%, respectively, at an HRT of 20 days, which showed 4% lower than that of the control (23.8%). The average BVS removal efficiency of SCFMRs fed with PACl sludge and Alum sludge were 25.8% and 26.9%, respectively, at an HRT of 20 days, which was 8~9% lower than that of the control (34.5%).

Study for Clean Energy Farming System by Mass and Energy Balance Analysis in the Controlled Cultivation of Vegetable Crop (Cucumber) (물질 및 에너지 수지 분석을 통한 시설채소(오이)의 청정에너지 농업 시스템 구축을 위한 기초 연구)

  • Clean energy farming is the agricultural activity to improve an efficiency of agricultural energy use and to replace fossil fuels. This study was carried out to establish the clean energy farming system in the controlled cultivation of vegetable crop (cucumber) adopting the biogas production facility. In order to design the clean energy farming system, mass and energy balance was analyzed between the controlled cultivation system and the biogas production facility. Net yearly heating energy demands ($E_{YHED}$) of forcing and semi-forcing cultivation types were 48,697 and $13.536Mcal\;10^{-1}$ in the controlled cultivation of vegetable cucumber. To cover these $E_{YHED}$, the pig slurry of 511 and $142m^3\;10a^{-1}$ (biogas volume of 9,482 and $2,636Nm^3\;10a^{-1}$, respectively, as 60% methane content) were needed in forcing and semi-forcing cultivation types. The pig slurry of $511m^3\;10a^{-1}$ caused N 1,788, $P_2O_5$ $511kg\;10a^{-1}$ in the forcing cultivation type, and the pig slurry of $142m^3\;10a^{-1}$ caused N 497, $P_2O_5$ $142kg\;10a^{-1}$ in the semi-forcing cultivation type. The daily heating energy demand ($E_{i,DHED}$) by the time scale analysis showed the minimum $E_{i,DHED}$ of $7.7Mcal\;10a^{-1}\;day^{-1}$, the maximum $E_{i,DHED}$ of $515.1Mcal\;10a^{-1}\;day^{-1}$, and the mean $E_{i,DHED}$ of 310.2 in the forcing cultivation type. And the minimum $E_{i,DHED}$, the maximum $E_{i,DHED}$, and the mean $E_{i,DHED}$ were 5.3, 258.0, and $165.1Mcal\;10a^{-1}\;day^{-1}$ in the semi-forcing cultivation type, respectively. Input scale of biogas production facility designed from the mean $E_{i,DHED}$ were 3.3 and $1.7m^3\;day^{-1}$ in the forcing and the semi-forcing cultivation type. The maximum $E_{i,DHED}$ gave the input scale of 5.4 and $2.7m^3\;day^{-1}$ in the forcing and the semi-forcing cultivation type.

A Study on Increasing the Efficiency of Biogas Production using Mixed Sludge in an Improved Single-Phase Anaerobic Digestion Process (개량형 단상 혐기성 소화공정에서의 혼합슬러지를 이용한 바이오가스 생산효율 증대방안 연구)

  • In this study, we attempted to improve the biogas production efficiency by varying the mixing ratio of the mixed sludge of organic wastes in the improved single-phase anaerobic digestion process. The types of organic waste used in this study were raw sewage sludge, food wastewater leachate and livestock excretions. The biomethane potential was determined through the BMP test. The results showed that the biomethane potential of the livestock excretions was the highest at $1.55m^3CN4/kgVS$, and that the highest value of the composite sample, containing primary sludge, food waste leachate and livestock excretions at proportions of 50%, 30% and 20% respectively) was $0.43m^3CN4/kgVS$. On the other hand, the optimal mixture ratio of composite sludge in the demonstration plant was 68.5 (raw sludge) : 18.0 (food waste leachate) : 13.5 (livestock excretions), which was a somewhat different result from that obtained in the BMP test. This difference was attributed to the changes in the composite sludge properties and digester operating conditions, such as the retention time. The amount of biogas produced in the single-phase anaerobic digestion process was $2,514m^3/d$ with a methane content of 62.8%. Considering the value of $2,319m^3/d$ of biogas produced as its design capacity, it was considered that this process demonstrated the maximum capacity. Also, through this study, it was shown that, in the case of the anaerobic digestion process, the two-phase digestion process is better in terms of its stable tank operation and high efficiency, whereas the existing single-phase digestion process allows for the improvement of the digestion efficiency and performance.

In order to obtain useful mutants such as early maturity, resistance to lodging, high protein and oil content, and capability of high yield, dormant seeds of two soybean varieties, Jang Dan Baik Mok and Clark, were treated with ${\gamma}$-ray, Ethyl Methane Sulfonate(EMS), Ethylene Imine(EI)and combinations of ${\gamma}$-ray and EMS or EI. 2. The germination rate and survival rate in a variety Jang Dan Baik Mok were significantly decreased with ${\gamma}$-ray treatment while it was not the same in the Clark variety. A significant decrease for seedling height measured at 14 and 21 days after sowing was found with the increase of ${\gamma}$-ray dose in both varieties. 3. Germination rates in both varieties were significantly decreased as EI concentration increases, particularly severe damage in germination was observed at 0.008 Mo. concentration. Germination rate damages were found with EMS concentration increases in the variety Jang Dan Baik Mok while no regular responses in seedling height were observed in the variety Clark. 4. Germination rate was significantly lowered with the combined treatment of EMS and ${\gamma}$-ray 24KR than that of EMS alone. In the treatments of ${\gamma}$-ray with three levels of EI concentration, the combined treatments except 24KR+EI 0.002 Mol. resulted in better germination than of EI alone. In both varieties, significant reduction in seedling height was observed in the combined treatments of ${\gamma}$-ray with various concentrations of EMS, whereas stimulation effect on seedling height was found with treatment of ${\gamma}$-ray EI +0.08Mol. 5. Germination rate, survival rate, and plant height as criteria of radio sensitivity, variety Jang Dan Baik Mok is moze sensitive to ${\gamma}$-ray, EMS, and EI than the variety Clark, and the varietal difference in responsibility to mutagen may be due to the genetic-constitution of the varieties.

Development and Characterization of EMS-induced Mutants with Enhanced Salt Tolerance in Silage Maize (EMS 유도 내염성 증진 사료용 옥수수 돌연변이체 선발 및 특성 분석)

  • Maize (Zea mays L.) is one of the most valuable agricultural crops and is grown under a wide spectrum of environmental conditions. However, maize is moderately sensitive to salt stress, and soil salinity is a serious threat to its production worldwide. In this study, we used ethyl methane sulfonate (EMS) to generate salt-tolerant silage maize mutants. We screened salt-tolerant lines from 203 M3 mutant populations by evaluating the morphological phenotype after salt stress treatment and selected the 140ES91 line. The 140ES91 mutant showed improved plant growth as well as higher proline content and leaf photosynthetic capacity compared with those of wild-type plants under salt stress conditions. Using whole-genome re-sequencing analysis, 1,103 single nucleotide polymorphisms and 71 insertions or deletions were identified as common variants between KS140 and 140ES91 in comparison with the reference genome B73. Furthermore, the expression patterns of three genes, which are involved in salt stress responses, were increased in the 140ES91 mutant under salt stress. Taken together, the mutant line identified in our study could be used as an improved breeding material for transferring salt tolerance traits in maize varieties.

Plasma-assisted Catalysis for the Abatement of Isopropyl Alcohol over Metal Oxides (금속산화물 촉매상에서 플라즈마를 이용한 IPA 저감)

  • This work investigated the plasma-catalytic decomposition of isopropyl alcohol (IPA) and the behavior of the byproduct compounds over monolith-supported metal oxide catalysts. Iron oxide ($Fe_2O_3$) or copper oxide (CuO) was loaded on a monolithic porous ${\alpha}-Al_2O_3$ support, which was placed inside the coaxial electrodes of plasma reactor. The IPA decomposition efficiency itself hardly depended on the presence and type of metal oxides because the rate of plasma-induced decomposition was so fast, but the behavior of byproduct formation was largely affected by them. The concentrations of the unwanted byproducts, including acetone, formaldehyde, acetaldehyde, methane, carbon monoxide, etc., were in order of $Fe_2O_3/{\alpha}-Al_2O_3$ < $CuO/{\alpha}-Al_2O_3$ < ${\alpha}-Al_2O_3$ from low to high. Under the condition (flow rate: $1L\;min^{-1}$; IPA concentration: 5,000 ppm; $O_2$ content: 10%; discharge power: 47 W), the selectivity towards $CO_2$ was about 40, 80 and 95% for ${\alpha}-Al_2O_3$, $CuO/{\alpha}-Al_2O_3$ and $Fe_2O_3/{\alpha}-Al_2O_3$, respectively, indicating that $Fe_2O_3/{\alpha}-Al_2O_3$ is the most effective for plasma-catalytic oxidation of IPA. Unlike plasma-alone processes in which tar-like products formed from volatile organic compounds are deposited, the present plasma-catalyst hybrid system did not exhibit such a phenomenon, thus retaining the original catalytic activity.

  • Biogasification is a technology that produces environmentally friendly fuel using methane gas generated in the process of stably decomposing and processing organic waste. Biogasification is the most used method for energy conversion of organic waste with high moisture content, and is a useful method for organic waste treatment following the prohibition of direct landfill (2005) and marine dumping (2013). Due to African Swine Fever (ASF), which recently occurred in Korea, recycling of wet feed is prohibited, and consumers such as dry feed and compost are negatively recognized, making it difficult to treat food waste. Accordingly, biogasification is attracting more attention for the treatment and recycling of food waste. Korea's energy consumption amounted to 268.41 106toe, ranking 9th in the world. However, it is an energy-poor country that depends on foreign imports for about 95.8% of its energy supply. Therefore, in Korea, the Renewable Energy Portfolio Standard (RPS) is being introduced. The domestic RPS system sets the weight of the new and renewable energy certificate (REC, Renewable energy certificate) of waste energy lower than that of other renewable energy. Therefore, an additional incentive system is required for the activation of waste-to-energy. In this study, the operation of an anaerobic digester that treats food waste, food waste Leachate and various organic wastes was confirmed. It was intended to be used as basic data for preparing the waste-to-energy incentive system through precise monitoring for a certain period of time. Four sites that produce biogas from organic waste and use them for power generation and heavy gas were selected as target facilities, and field surveys and sampling were conducted. Basic properties analysis was performed on the influent sample of organic waste and the effluent sample according to the treatment process. As a result of the analysis of the properties, the total solids of the digester influent was an average of 12.11%, and the volatile solids of the total solids were confirmed to be 85.86%. BOD and CODcr removal rates were 60.8% and 64.8%. The volatile fatty acids in the influent averaged 55,716 mg/L. It can be confirmed that most of the volatile fatty acids were decomposed and removed with an average reduction rate of 92.3% after anaerobic digestion.

Effect of Byproducts Supplementation by Partically Replacing Soybean Meal to a Total Mixed Ration on Rumen Fermentation Characteristics In Vitro (대두박 대체 부산물 위주의 TMR 사료가 반추위 내 미생물의 In Vitro 발효특성에 미치는 영향)

  • This study was performed to evaluate the effects of replacing basic total mixed ration (TMR) with fermented soybean curd, Artemisia princeps Pampanini cv. Sajabal, and spent coffee grounds by-product on rumen microbial fermentation in vitro. Soybean in the basic TMR diet (control) was replaced by the following 9 treatments (3 replicates): maximum amounts of soybean curd (SC); fermented SC (FSC); 3, 5, and 10% FSC + fermented A. princeps Pampanini cv. Sajabal (1:1, DM basis, FSCS); and 3, 5, 10% FSC + fermented coffee meal (1:1, DM basis, FSCC) of soybean. FSC, FSCS, and FSCC were fermented using Lactobacillus acidophilus ATCC 496, Lactobacillus fermentum ATCC 1493, Lactobacillus plantarum KCTC 1048, and Lactobacillus casei IFO 3533. Replacing dairy cow TMR with FSC treatment led to a pH value of 6 after 8 h of incubation-the lowest value measured (p<0.05), and FSCS and FSCC treatments were higher than SC and FSC treatment after 6 h (p<0.05). Gas production was higher in response to 3% FSC and FSCC treatments than the control after 4-10 h. Dry matter digestibility was increased 0-12 h after FSC treatment (p<0.05) and was the highest after 24 h of 10% FSCS treatment. $NH_3-N$ concentration was the lowest after 24 h of FSC treatment (p<0.05). Microbial protein content increased in response to treatments that had been fermented by the Lactobacillus spp. compared to control and SC treatments (p<0.05). The total concentration of volatile fatty acids (VFAs) was increased after 6-12 h of FSC treatment (p<0.05), while the highest acetate proportion was observed 24 h after 5% and 10% FSCS treatments. The FSC of propionate proportion was increased for 0-10 h compared with among treatments (p<0.05). The highest acetate in the propionate ration was observed after 12 h of SC treatment and the lowest with FSCS 3% treatment after 24 h. Methane ($CH_4$) emulsion was lower with A. princeps Pampanini cv. Sajabal and spent coffee grounds treatments than with the control, SC, and FSC treatments. These experiments were designed to replace the by-products of dairy cow TMR with SC, FSC, FSCS, and FSCC to improve TMR quality. Condensed tannins contained in FSCS and FSCC treatments, which reduced $CH_4$ emulsion in vitro, decreased rumen microbial fermentation during the early incubation time. Therefore, future experiments are required to develop a rumen continuous culture system and an in vivo test to optimize the percentages of FSC, FSCS, and FSCC in the TMR diet of the dairy cows.

Effect of Protein Fractionation and Buffer Solubility of Forage Sources on In Vitro Fermentation Characteristics, Degradability and Gas Production (조사료 자원의 단백질 분획 및 Buffer 추출이 In Vitro 발효 성상, 분해율 및 Gas 생성량에 미치는 효과)

  • Buffer solubility and protein fractionation were evaluated from the hays (timothy, alfalfa and klein) and straws (tall fescue and rice), and $In$ $vitro$ trial was conducted to examine the effect of buffer extraction on fermentation characteristics, degradability and gas ($CO_2$ and $CH_4$) production. Buffer soluble protein (SP) content and A fraction in total protein were highest in alfalfa hay as 61% and 41.77%, respectively while lowest in rice straw (42.8% and 19.78%, respectively). No difference was observed in B1 fraction among forages but B2 fraction was slightly increased in klein hay (12.34%) and tall fescue straw (10.05%) compared with other forages (6.34~8.85%). B3 fraction of tall fescue was highest as 38.49% without difference among other forages while C fraction was highest in rice straw. pH in incubation solution was higher in all forages after extraction than before extraction at 3h (P<0.01) and 6h (P<0.05), and pH from hays of timothy and alfalfa was higher than the other forages at 6h (P<0.05) and 12h (P<0.001). Regardless of extraction, ammonia-N concentration from alfalfa hay was increased at all incubation times and extraction effect was appeared only at 3h incubation time (P<0.01). Total VFA concentration from alfalfa hay was highest up to 24h incubation while those from tall fescue straw and rice straw were lowest. Buffer extraction decreased (P<0.01~P<0.001) the total VFA concentration. Acetic acid proportion was increased (P<0.001) before extraction of forages but no difference was found between forages. Propionic acid($C_3$) proportion was also increased(P<0.001) before extraction in all forages than in straws at 3h, 24h and 48h incubations, and $C_3$ from hays were mostly higher (P<0.05) than from straws. Butyric acid proportion, however, was not affected by extraction at most incubation times. Parameter 'a' regarding to the dry matter (DM) degradation was increase (P<0.001) in all forages before extraction, and was decreased (P<0.05) in tall fescue straw and rice straw compared with hays. Parameter 'b' was also increased (P<0.001) before extraction but no difference was found between forages. Effective degradability of DM (EDDM) was higher (P<0.001) before extraction in most forages except for rice straw. Buffer extraction decreased (P<0.05) all parameters (a, b, and c) regrading to the crude protein (CP) degradation but no difference was found between forages. Effective degradation of CP (EDCP) was lower (P<0.05) in straws than in hays. Parameters 'a' and 'b' regarding to the NDF degradation (P<0.01) and effective degradability of NDF (EDNDF, P<0.001) were also higher in forages before extraction than after extraction but no difference was found between forages. Buffer extraction reduced (P<0.05~P<0.001) $CO_2$ production from all the forages uo to 24h incubation and its production was greater (P<0.05~P<0.01) from hays than straws. Methane ($CH_4$) production was also greater (P<0.01~P<0.001) in all forages at all incubation times, and its production was greater (P<0.05) from hays than from straws at most incubation times. Based on the results of the current study, it can be concluded that buffer solubility and CP fractionation might be closely related with $In$ $vitro$ VFA concentration, degradability and gas ($CO_2$ and $CH_4$) production. Thus, measurement of buffer solubility and protein fractionation of forages might be useful to improve TMR availability in the ruminants.
