Porosome: the Universal Molecular Machinery for Cell Secretion

  • Porosomes are supramolecular, lipoprotein structures at the cell plasma membrane, where membrane-bound secretory vesicles transiently dock and fuse to release inravesicular contents to the outside during cell secretion. The mouth of the porosome opening to the outside, range in size from 150 nm in diameter in acinar cells of the exocrine pancreas, to 12 nm in neurons, which dilates during cell secretion, returning to its resting size following completion of the process. In the past decade, the composition of the porosome, its structure and dynamics at nm resolution and in real time, and its functional reconstitution into artificial lipid membrane, have all been elucidated. In this mini review, the discovery of the porosome, its structure, function, isolation, chemistry, and reconstitution into lipid membrane, the molecular mechanism of secretory vesicle swelling and fusion at the base of porosomes, and how this new information provides a paradigm shift in our understanding of cell secretion, is discussed.

Computational Modelling of Droplet Dynamics Behaviour in Polymer Electrolyte Membrane Fuel Cells: A Review

  • Polymer Electrolyte Membrane Fuel Cells (PEMFC) is one of the leading advanced energy conversion technology for the use in transport. It generates water droplets through the catalytic processes and dispenses the water through the gas-flowed microchannels. The droplets in the dispensing microchannel experience g-forces from different directions during the operation in transport. Therefore, this paper reviews the computational modelling topics of droplet dynamics behaviour specifically for three categories, i.e. (i) the droplet sliding down a surface, (ii) the droplet moving in a gas-flowed microchannel, and (iii) the droplet jumping upon coalescence on superhydrophobic surface; in particular for the parameters like hydrophobicity surfaces, droplet sizes, numerical methods, channel sizes, wall conditions, popular references and boundary conditions.

  • Flow-field design has much influence over the performance of proton exchange membrane fuel cell (PEMFC) because it affects the pressure magnitude and distribution of the reactant gases. To obtain the pressure magnitude and distribution of reactant gases in four kinds of flow-field designs without additional measurement equipment, computational fluid dynamics (CFD) analysis was performed. After the CFD analysis, the performance values of PEMFC according to the flow-field configurations were measured via a single cell test. As expected, the pressure differences due to different flow-field configurations were related to the PEMFC performance because the actual performance results showed the same tendency as the results of the CFD analysis. A large pressure drop resulted in high PEMFC performance. So, the single serpentine configuration gave the highest performance. On the other hand, the parallel flow-field configuration gave the lowest performance because the pressure difference between inlet and outlet was the lowest.

Refinement of Protein NMR Structure under Membrane-like Environments with an Implicit Solvent Model

  • Refinement of NMR structures by molecular dynamics (MD) simulations with a solvent model has improved the structural quality. In this study, we applied MD refinement with the generalized Born (GB) implicit solvent model to protein structure determined under membrane-like environments. Despite popularity of the GB model, its applications to the refinement of NMR structures of hydrophobic proteins, in which detergents or organic solvents enclose proteins, are limited, and there is little information on the use of another GB parameter for these cases. We carried out MD refinement of crambin NMR structure in dodecylphosphocholine (DPC) micelles (Ahn et al., J. Am. Chem. Soc. 2006, 128, 4398-4404) with GB/Surface area model and two different surface tension coefficients, one for aquatic and the other for hydrophobic conditions. Our data show that, of two structures by MD refinement with GB model, the one refined with the parameter to consider hydrophobic condition had the better qualities in terms of precision and solvent accessibility.

  • 수소경제 시대의 도래와 함께 연료전지에 관한 연구가 크게 주목받고 있다. 그중 실험적으로 분석이 어려운 부분에 관하여 비용과 시간이 요구되는 실험적인 방법을 배제할 수 있는 모델링 기법인 전산유체역학(computational flow dynamics, CFD)이 큰 관심을 받고 있다. 연료전지의 연구에 주로 사용되는 전산유체역학에 관한 연구는 열분포, 유체의 흐름, 각종 반응물의 농도, 그리고 전기화학반응 등의 실험적인 분석이 현실적으로 불가능한 부분의 분석으로 통하여 실험을 줄이고도 많은 결과를 얻을 수 있는 연구가 활발하게 진행되고 있다. 본 기고문에서는 전산유체역학을 이용한 연료전지 내부에서 벌어지고 있는 각종 유체, 열, 전기화학반응 등에 관한 연구동향을 소개하고자 한다.

  • Water management is one of many operating parameters, which influences the performance and stability of a proton exchange membrane fuel cell (PEMFC). Local humidity condition including liquid water saturation has profound impacts on the distributions of overpotentials, current density, and membrane water content. Computational fluid dynamics simulations were conducted to investigate the effect of the inlet humidity variation on the performance of a PEMFC of $9\;cm^2$ active cell area with serpentine flow fields. The results showed that the performance of the simulated PEMFC remained at an almost same level when the cathode inlet humidity was changed from 100% to 60%, while reaching its maximum at air humidity of 80%. However, further decrease in the cathode inlet humidity below 40% started to significantly deteriorate the performance of the PEMFC. The variations of overpotentials, membrane water content, etc. due to the change in the cathode inlet humidity were also discussed.

  • Design of a reformer consisting of combustion chamber and reforming chamber was investigated for a 1 kW and a 5 kW polymer electrolyte membrane fuel cell (PEMFC), respectively, using the computational fluid dynamics (CFD). First, the 1kW reformer was considered to obtain the reliability of the numerical study. It was modeled, calculated and compared with experimental data. Second, the 5kW reformer was considered for a geometric study. Three tip sizes (35, 40, and 45 mm) and five aspect ratios was selected. It was found that the optimum was at tip sizes of 40 and 45 mm, at aspect ratios of -10% and -20% of the standard length.

EphB/ephrinB Signaling in Cell Adhesion and Migration

  • Eph receptors and their ligands, ephrins, represent the largest group of the receptor tyrosine kinase (RTK) family, and they mediate numerous developmental processes in a variety of organisms. Ephrins are membrane-bound proteins that are mainly divided into two classes: A class ephrins, which are linked to the membrane by a glycosylphosphatidylinositol (GPI) linkage, and B class ephrins, which are transmembrane ligands. Based on their domain structures and affinities for ligand binding, the Eph receptors are also divided into two groups. Trans-dimerization of Eph receptors with their membrane-tethered ligands regulates cell-cell interactions and initiates bidirectional signaling pathways. These pathways are intimately involved in regulating cytoskeleton dynamics, cell migration, and alterations in cellular dynamics and shapes. The EphBs and ephrinBs are specifically localized and modified to promote higher-order clustering and initiate of bidirectional signaling. In this review, we present an in-depth overview of the structure, mechanisms, cell signaling, and functions of EphB/ephrinB in cell adhesion and migration.

전산유체역학을 이용한 공압시스템용 제습장치의 형상 개선에 관한 연구 (A Study on Shape Improvement of Dehumidifier for Pneumatic System using Computational Fluid Dynamics)

  • In this study, flow analysis and dehumidification experiment were conducted on hollow fiber membrane module to determine the dehumidification characteristics of its various configurations. A quantitative analysis of the CFD for four different models with a temperature of $30^{\circ}C$ and 30%RH inlet humidity was conducted. Each model has different shape parameters i.e. the number of hollow fiber membranes and the presence or absence of baffles. After comparison between the flow analysis results and dehumidification experiment results, the percentage error was found to be approximately 2%. The moisture removal rate for each model was calculated using flow analysis data. It was found that the moisture removal rate of refined model with three baffles and eight hollow fiber membranes was highest among the four modeled modules of hollow fiber membrane one, i.e. about 60%.

Volatile Memristor-Based Artificial Spiking Neurons for Bioinspired Computing

  • The report reviews recent research efforts in demonstrating a computing system whose operation principle mimics the dynamics of biological neurons. The temporal variation of the membrane potential of neurons is one of the key features that contribute to the information processing in the brain. We first summarize the neuron models that explain the experimentally observed change in the membrane potential. The function of ion channels is briefly introduced to understand such change from the molecular viewpoint. Dedicated circuits that can simulate the neuronal dynamics have been developed to reproduce the charging and discharging dynamics of neurons depending on the input ionic current from presynaptic neurons. Key elements include volatile memristors that can undergo volatile resistance switching depending on the voltage bias. This behavior called the threshold switching has been utilized to reproduce the spikes observed in the biological neurons. Various types of threshold switch have been applied in a different configuration in the hardware demonstration of neurons. Recent studies revealed that the memristor-based circuits could provide energy and space efficient options for the demonstration of neurons using the innate physical properties of materials compared to the options demonstrated with the conventional complementary metal-oxide-semiconductors (CMOS).
