Determining the doses of probiotics for application in Scylla tranquebarica (Fabricius 1798) larvae to produce crablet

  • Mass mortalities of mud crab Scylla spp. larvae due to pathogenic Vibrio spp. outbreaks have frequently occurred in hatcheries. To overcome this problem, probiotics containing Bacillus subtilis bacteria are applied to inhibit pathogenic ones. We tested different doses of probiotic-containing B. subtilis (108 CFU/g) on the Scylla tranquebarica larvae and investigated the microbiota population, including Vibrio. Water quality, larvae development, and crablet production were also monitored. The recently hatched larvae were grown in twelve conical fiber tanks filled with 200 L sterile seawater, with a salinity of 30 ppt at a stocking density of 80 ind/L. Four different doses of probiotics were applied in the larvae rearing, namely, A = 2.5 mg/L, B = 5 mg/L, C = 7.5 mg/L, and D = 0 mg/L, with three replicates. Next-generation sequencing analysis was used to obtain the abundance of microbes in the whole body of megalopa and the water media for larvae rearing after applying probiotics. Sixteen Raw Deoxyribonucleic Acid samples (eight from a whole body of megalopa extraction from four treatments of probiotics defined as A, B, C, D, and eight from water media extraction from four treatments of probiotic defined as E, F, G, H) were prepared. Then, they were sent to the Genetics Science Laboratory for NGS analysis. Ammonia, nitrite, total organic matter (TOM), larvae, and crablet production were monitored. Based on the Next-generation sequencing analysis data, the Vibrio spp. decreased significantly (p < 0.05) than control test (D) in megalopa-applied probiotics at the doses of 2.5 mg/L (A) and 7.5 mg/L (C) and in the water media for megalopa rearing treated with probiotics at the dosage of 5.0 mg/L (F). Ammonia in the zoea stage in B treatment and TOM in the zoea and megalopa stage in B and C treatments were decreased significantly (p < 0.05). It impacts the higher number of zoea survival in treatments B and C. Finally, it results in a significantly high crablet production in treatments B and C. Therefore, the dosage of 5 mg/L to 7.5 mg/L improves crablet S. tranquebarica production significantly.

  • Streptomycin, which is one of aminoglycoside antibiotics, has been widely used in the rearing of food-producing animals to prevent and treat diseases in cattle, pigs and poultry. Although not licensed in South Korea, streptomycin has also been used for the treatment of bacterial honeybee disease, such as European foulbrood in Third World countries. A reliable and effective method using liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed for the determination of streptomycin in honey. A established method was optimized the clean-up and extraction procedure for the trace determination, good precision and accuracy. And the chromatographic and tandem mass spectrometric parameters were also optimized. The precision (RSD) and accuracy (bias) in the concentration range of 5.0~50.0 ug/kg were 5.5~14% and -10.0~8.0%, respectively. Limit of detection was 0.75 ug/kg and recovery of streptomycin spiked at level of 10 ug/kg in honey was 74%. The established and validated method was applied to determine streptomycin in honey which was on the market.

  • The Korean rhinoceros beetle (Allomyrina dichotoma) is popular as a pet and as a food ingredient, and it is commercially distributed in Korea. It is also traditionally regarded as a medicine for liver-related diseases. Recently, the Oryctes rhinoceros nudivirus was introduced from Southeast Asia. This virus is reported as a disease factor for A. dichotoma in mass-rearing facilities, and economic losses due to this viral infection have been increasing in Korea since the 2010s. In this study, we observed serious structural changes in the fat body and the intestine of virus-infected beetles. We report five genes that are up-regulated by the viral infection in the intestine: BTF3H4-like (transcription factor BTF3 homolog 4-like), SPS-like (serine proteinase stubble-like), COPB1 (coatomer protein complex, subunit beta 1), T-CP (T-complex 1 subunit gamma), and HSP70 HSP70 (heat shock protein 70). The results may provide a clue for the early diagnosis and disease-treatment that occurs in mass-rearing facilities. The improvement of stable productivity will increase the farmers’ income, and quality control of beetle-breeding will help industries to utilize this beetle as a promising food ingredient.

Ecological Characteristics of Dorcus hopei(E Saunders) for the Development of Mass-rearing Technique in Korea (왕사슴벌레(Dorcus hopei)의 대량사육 기술개발을 위한 생태특성 조사)

  • This study was carried out to review the distributional data of Dorcus hopei in Korea and to investigate ecological characteristics to develop a mass rearing technique of the species. The coupling period of the male and female was needed at least 2 weeks in condition of 6 months after emergence, and the optimal sex ratio for oviposition was 2♀:1♂. Eggs were laid singly, up to 27.3 per female. The pre-ovipostion period was average 147.3 days, egg-period was 15.6 days on the average, and oviposition period was at least 120 days. The optimum size of oviposition room was 55${\times}$40${\times}$35 cm with 3-4 oviposition-trees cut in size of 15${\times}$15 cm. Quercus acutissima was highly preferred for the oviposition. Developing period of each instar in the insectary (25$^{\circ}C$, 75% R.H.. 16L:8D) was 24.1 days for the 1st instar,29.8 days for the 2nd, and 131.2 days for the 3rd instar, and 28.9 days for the pupa. The longevity of adults was longer than 35 months. The period of induced dormancy was needed at least 3-4 months.

  • Korean rhinoceros beetles (Allomyrina dichotoma), which can be found in broad-leaved forests in mountainous habitats and lives for around one year in wild. This beetle is currently popular as a pet and traditionally regarded as a medicine for liver-related diseases in Korea. It is reported that the economic losses in the mass-rearing facilities by virus infection have been increased since the 2010s in Korea. The causing virus for the A. dichotoma was firstly reported as an Oryctes rhinoceros nudivirus (OrNV) in 2015. We, here, observes that serious morphological changes in the intestinal tube from the OrNV-infected beetles, and report five genes, which are regulated by OrNV infection in the intestine; Krueppel-like factor 15 (Klf15), Endoplasmic reticulum aminopeptidase 2 (ERAP2), U5 small nuclear ribonucleoprotein 200 kDa helicase (Snrnp200), Muscleblind-like protein 2a (mbnl2a), and MIMI_L93. The results may provide a clue to the early diagnosis and disease treatment during the mass-rearing facilities of the A. dichotoma.

Development and Oviposition Characteristics of Protaetia brevitarsis (Coleoptera: Cetoniidae) Fed Fermented Mulberry Sawdust (뽕나무발효톱밥 급여에 따른 흰점박이꽃무지의 발육 및 산란 특성)

  • This study investigated the developmental and ovipositional characteristics of Protaetia brevitarsis individuals that were fed with fermented mulberry sawdust. The developmental periods of larvae were 164.0, 73.3, and 64.8 days at 25, 28, and $30^{\circ}C$, respectively. The maximum larval weight was 2.94 g at $25^{\circ}C$, and the weight of larvae decreased as temperature increased. The average weight of female adults were 0.94, 0.51, and 0.54 g at 25, 28, and $30^{\circ}C$, respectively. The weight of male adults was higher than that of females. The addition of 10% or 30% wheat bran to the fermented mulberry sawdust increased larval weight. It was possible to sell larvae from 75 days after mass rearing, when the cumulative rate of larvae that were heavier than 2.5 g was approximately 75%. The average number of eggs per female was 83.2 at $25^{\circ}C$, and this was the highest in the 5th week. Most of the eggs (73%) were oviposited between 3 and 8 weeks after adults emergence.

Differential Effects of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine on Motor Behavior and Dopamine Levels at Brain Regions in Three Different Mouse Strains

  • Developing an animal model for a specific disease is very important in the understanding of the underlying mechanism of the disease and allows testing of newly developed new drugs before human application. However, which of the plethora of experimental animal species to use in model development can be perplexing. Administration of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is a very well known method to induce the symptoms of Parkinson's disease in mice. But, there is very limited information about the different sensitivities to MPTP among mouse strains. Here, we tested three different mouse strains (C57BL/6, Balb-C, and ICR) as a Parkinsonian model by repeated MPTP injections. In addition to behavioral analysis, endogenous levels of dopamine and tetrahydrobiopterin in mice brain regions, such as striatum, substantia nigra, and hippocampus were directly quantified by liquid chromatography-tandem mass spectrometry. Repeated administrations of MPTP significantly affected the moving distances and rearing frequencies in all three mouse strains. The endogenous dopamine concentrations and expression levels of tyrosine hydroxylase were significantly decreased after the repeated injections, but tetrahydrobiopterin did not change in analyzed brain regions. However, susceptibilities of the mice to MPTP were differed based on the degree of behavioral change, dopamine concentration in brain regions, and expression levels of tyrosine hydroxylase, with C57BL/6 and Balb-C mice being more sensitive to the dopaminergic neuronal toxicity of MPTP than ICR mice.

Aerosol Emission from Road by Livestock Transport Vehicle Movement (축산관련차량 이동에 따른 도로의 에어로졸 발생량 분석)

  • Most of livestock houses are concentrated in certain area with mass rearing system resulting in rapid spread of infectious diseases such as HPAI (highly pathogenic avian influenza). The livestock-related vehicles which frequently travel between farms could be a major factor for disease spread by means of transmission of airborne aerosol including pathogens. This study was focused on the quantitative measurement of aerosol concentration by field experiment while vehicles were passing through the road. The TSP (total suspended particle) and PM10 (particle matter) were measured using air sampler with teflon filter installed downward the road with consideration of weather forecast and the direction of road. And aerosol spectrometer and video recorders were also used to measure the real-time distribution of aerosol concentration by its size. The results showed that PM2.5 was not considerable for transmission of airborne aerosol from the livestock-related vehicle. The mass generated from the road during the vehicle movement was measured and calculated to 241.4 ${\mu}g/m^3$ by means of the difference between TSP and PM2.5. The dispersion distance was predicted by 79.6 m from the trend curve.

  • Biotechnological researches in Central Asian Research Institute of Sericulture were developed since 1981 when laboratory of artificial diets for mulberry silk­worm was created. The researches were directed not only to creation of accessible artificial diets (AD) for off-season mass rearing of economical beneficial insect and for industrialization of sericulture but also to obtaining of easy reproducible source of animal proteins, carbohydrates, lipids, enzymes, enzymes inhibitors and other metabolites - bioresource for manufacture of oriental medicine preparations. Space experiments with silkworms became possible with use of AD. These experiments used mulberry silkworm as high effective board test organism with outstanding developmental, reproductive and transgenic properties having impor­tant significance in future of cosmonautics. Introduction to practice of bioprotective and ecologically pure method of drying and sterilizing of green cocoons and by-products of silk manufacture allows to increase efficiency of cocoons reeling technology as well as to preserve nativity of biological active substances in mulberry silkworm pupae that is unachievable by traditional methods of raw material processing. New spheres of application of two important silk proteins - fibroin and sericin in medicine, cosmetics, for bio- and nanotechnology are opening with use of biotechnological approaches. At present the achievements and experience of R & D in these fields are used in creation of modern pests biocontrol agents.   • 1. 침파리의 동계사육 결과 평균온도 $23.7^{\circ}C$에서 유충기간 7일, 용기간 9.2일, 전산란기간 14.2일, 성충수명 23.3일이었다. 2. 5월부터 7월까지 사육온도 $25\pm1^{\circ}C$로 고정시켰을 때의 생육기간은 유충기간 8.0일, 용기간 5.3일, 전산란기간 11.7일, 성충수명 30.1일 이었다. 3. 용 개체중량은 5월-7월의 $25\pm1^{\circ}C$에서 13.3mg으로서 현저한 증가를 볼 수 있었다. 4. 전산란 기간중의 성충사망율은 습도 조절(R. H.)에 따라서 큰 차이를 보여주었으며 누대사육을위해서는 $55-65\%$의 R.H.를 유지할 필요가 있을 것으로 본다. 5. 성충의 산란율은 산란 시작후 제8-10일에 제일 높다.
