Physical Characteristics of Stem and Fruit of Lycium Chinense Mill (구기자의 가지 및 열매의 특성에 관한 연구)

  • Physical characteristics of stem (ile., length and diameter) and branch, and detachment force of mature and immature fruits of 12 species of the Lycium chinese Mill at a cultivar were determined. The number of stems and branches were counted after the first and second pruning of each plant. Length and diameter of the stem were average of 113.5 cm and 9.5 mm for 12 species of the Lycium chinese Mill, respectively. Average number of stem and number of branch after the first and second pruning in each plant were 5, 30, and 61, respectively. Diameter of major and minor axis of the fruit was average of 13.8mm and 8.3mm, respectively, and the sphericity of fruits was average of 0.7 for 12 species of Lycium chinese Mill. Detachment force of mature and immature fruits was average of 1 N and 2.7 N, respectively, for 12 species of the Lycium chinese Mill. The maximum and minimum detachment force of the mature fruits was 2.06 N and 0.39 N, respectively, and that of the immature fruits was 3.72 N and 1.27 N, respectively. The force-weight ratio showed a decreasing trend as the weight of fruit increasing for all samples.

The Effects of Lycium chinese Mill., Morus alba L. and Their Combination on the Asthmatic Murine Model (천식동물모델을 이용한 지골피(地骨皮), 상백피(桑白皮) 및 두 배합약물의 실험적 연구)

  • Objectives : To clarify the possible effect of Lycium chinese Mill (LC)., Morus alba L (MA)., and Lycium chinese Mill. +Morus alba L. (LC+MA), we have examined their influence on the development of pulmonary eosinophilic inflammation in the asthmatic murine model. Methods : Female Balb/c mice (5weeks) were immunized on two different days (21 days and 7 days before inhalational exposure) by intraperitonial injections of 0.2ml alum-precipitated Ag containing $100{\mu}g$ of OVA bound to 4 mg of aluminum hydroxide in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 minutes/day on 3 days/week for 8 weeks (at a flow rate of 250 L/min, 2.5% ovalbumin in normal saline) and, LC, MA, and LC+MA (500 mg/kg) were orally administered 3 times per a week for 8 weeks. Results : The suppressive effect of LC, MA, and LC+MA were demonstrated by the accumulation of eosinophills into airways, with the reduction of eosinophil, total lung leukocytes numbers. These were correlated with the marked reduction of IL-5, IL-13 and IL-4 levels in the BALF and serum. OVA-specific IgE levels were also decreased in serum and BAL from these mice. LC, MA, and LC+MA decreased eosinophil CCR3 expression and CD11b expression in lung cells. Conclusions : These results indicate that LC, MA, and LC+MA have high inhibitory effects on airway inflammation and hyper-responsiveness in the asthmatic murine model. The suppression of IL-5, IgE, eosinophil CCR3 expression and CD11b expression, and the increase of IFN-${\gamma}$ production in BALF seem to contribute to this effect. Hence, the results indicated that LC, MA, and LC+MA could act as a immuno-modulator which possesses anti-inflammatory and anti-asthmatic property by modulating the imbalance between Th1 and Th2 cytokines.

Quality Characteristics of Gugija.Heukimja Jook Containing Different Levels of Black Sesame Powder (흑임자 첨가량을 달리한 구기자흑임자죽의 품질특성)

  • This study was to investigated optimum preparation conditions for Gugija Heukimja jook by response surface methodology (RSM). The important materials used for the Gugija Heukimja product were Lycium chinese, Seasamum indicum L., Ziziphus jujuba Mill. and rice. Moisture content, crude protein, crude fat, crude ash and crude fiber content were 15.62, 16.60, 13.62, 4.93 and 11.17% for Lycium chinese, 3.62, 20.94, 43.56, 5.67 and 10.72% for Seasamum indicum L., 24.12, 5.38, 0.73, 2.30 and 1.25% for Ziziphus jujuba Mill., and 12.36, 6.98, 0.46, 0.43 and 0.41% for rice, respectively. Optimal extraction conditions of brix, color, pH and total acidity in the materials for Gugija Heukimja jook were temperature $80^{\circ}C$, water volume 850 mL and extraction time of 63 min. The spreadability of jook with Lycium chinese and Ziziphus jujuba Mill. extracts was the highest when black sesame was 12 g pH was the lowest when black sesame was 9 g. PH showed a significant difference when the added black sesame ratio changed. The 'value L' of chromaticity was lower with an increase of the black sesame ratio, 'value a' was higher with an increase of the black sesame ratio, 'value b' was lower with an increase of the black sesame ratio. Brix was lower when black sesame in jook was added. Total acidity was highest (7.27%) when black sesame was 12 g. The degree of gelatinization tended to be lower with additional black sesame. Higher scores of sensory evaluation were found for the product containing 9 g black sesame with Lycii furctus and Ziziphus jujuba Mill. extracts ompared to the other samples with 3 and 6g. No significant difference in taste occurred for Gugija Heukimja jook prepared with over 9 g black sesame, while a great difference showed in color, pH and total acid. Therefore, the Gugija Heukimja jook prepared under the optimal conditions of 9 g black sesame with Lycium chinese and Ziziphus jujuba Mill. extracts exhibited higher qualities. Gugija Heukimja jook can use the easier for Yacksun jook. It is expected to become functional in the product's development.

Plant Regeneration from Leaf and Internode Segment Cultures of Boxthorn (Lycium chinense Mill.) (구기자나무의 잎과 마디절편체 배양에 의한 식물체 재생)

  • Callus and shoot formation from medicinal crop, Lycium chinense Mill. cv. 'Cheongyang', as influenced by various media, explant sources and plant growth regulators were investigated. The rate of shoots formation, number of shoots, and fresh weight of shoots were the best on MS medium followed by B$_{5}$, WPH, and SH. Callus induction was more effective in leaf than internode segments, and was the best on MS medium containing 0.5 mg/L NAA with 0.2 mg/L BA. Effects of plant growth regulators in shoot formation were more effective in BA than TDA combined with NAA. Shoot formation from callus induced in leaf and internode segments was the best on MS medium containing 0.01 mg/L NAA with 0.2 mg/L BA.

Two Species of Colletotrichum Associated with Anthracnose of Lycium chinense (구기자 탄저병에 관여하는 2종의 Colletotrichum에 관하여)

  • The symptom of anthracnose of chinese matrimony vine (Lycium chinense Mill) was darkbrown to black, water­soaked lesions with acervuli on fruits. Field survey revealed that the incidence rate of anthracnose ranged from $0.8\%\;to\;23\%$. to Colletotrichum gloeosporioides Penz and C. dematium(Fr.) Grove were identified as the causal organisms of the disease. C. gloeosporioides and C. dematium produced severe symptoms on fruits resulting in complete necrosis, but produced no or mild symptoms on leaves and stems of chinese matrimony vine. Of three isolates of C. dematium, isolate I and isolate III caused severe infection on immature fruits and isolate I was the most pathogenic on mature fruits of the plant. Isolates II of C. dematium caused relatively severe lesions on pepper fruit, but C. gloeosporioides was pathogenic on apple fruit.

Cultivar Discrimination of Korean and Chinese Boxthorn (Lycium chinense Mill. and Lycium barbarum L.) using SSR Markers (SSR 마커를 이용한 한국산과 중국산 구기자의 품종 판별)

  • This study was undertaken to develop a technique of discrimination using SSR makers in boxthorn cultivars. Forty one boxthorn cultivars, which were collected from Korea and China, were evaluated by 10 SSR markers. Total of 61 alleles were detected, ranging from 3 to 13 with an average of 6.1 alleles per locus. The averages of gene diversity and PIC values were 0.482 and 0.428, with a range from 0.25 (GB-LCM-022 and GB-LCM-087) to 0.83 (GB-LCM-167) and from 0.24 (GB-LCM-022 and GB-LCM-087) to 0.81 (GB-LCM-167), respectively. Five markers out of 10 markers, GB-LCM-022, GB-LCM-075, GB-LCM-104, GB-LCM-167 and GB-LCM-217, were selected as key markers for discrimination in boxthorn cultivars. All of boxthorn cultivars were individually distinguished by the combination of five SSR markers.

  • This report concerns with three species of eriophyid mites on Licium chinensis in Korea. Of them, the external morphology of Aceria tjyingi (Manson 1973) was observed with Scanning Electron Microscope. The author reviewed three species of Korean eryophid mites and suggests Korean names of Aceria kuko kishida, A. macrodonis Keifer, and A. tjyngi (Manson), respectively.

First Report of Anthracnose Caused by Colletotrichum fioriniae on Chinese Matrimony Vine in Korea

  • A fungus, Colletotrichum fioriniae, was isolated for the first time from fruits of Chinese matrimony vine (Lycium chinense Mill.) in Korea. It was classified as C. fioriniae based on the morphological characteristics and nucleotide sequence of glyceraldehyde-3-phosphate-dehydrogenase and ${\beta}-tubulin$. To the best of our knowledge, this is the first report of C. fioriniae causing anthracnose of Chinese matrimony vine in Korea.

  • Sterol compositions of three solanaceous plant seed oils i.e., tobacco(Nicotiana tabacum L.), datura (Datura stramonium L.) and Chinese lycium (Lycium chinense Mill.) were analyzed by thin layer chromatography, gas liquid chromatography and gas liquid chromatography-mass spectrometry. Cholesterol, cholest-7-enol, campesterol, 24-methylenecholesterol, stigmasterol, sitosterol, and 28-isofucosterot were identified in 4-des-methylsterol fraction. 31-Norlanost-8-enol, $4{\alpha}$-methylcholest-8-enol, lophenol, 31-norlanosterol, obtusifoliol, 31-norcycloartenol, cyloeucalenol, gramisterol (24-methylenelophenol), and citrostadienol in 4-monomethylsterol fraction, and lanost-8-enol, cycloartanol, lanosterol, ${\beta}$-amyrin, 24-methylenelanost-8-enol, cycloartenol, lupeol and 24-methylenecycloartanol in 4,4-dimethylsterol fraction were identified respectively.
